散乱光強度測定は高感度であるものの、ダイナミックレンジが極めて狭い。そのためアナライトを高濃度含有する試料を測定する場合には、測定時のアナライトの濃度が測定系の測定範囲内の濃度となるまで試料の希釈と測定を繰り返す必要があり、測定結果の報告までに時間を要するという欠点があった。
一方、吸光度測定は散乱光強度測定と比較して、ダイナミックレンジはやや拡大できるものの、アナライト低濃度域での測定精度に劣ることは当業者によく知られている。
これら粒子増強免疫凝集測定法の課題を解決する手段として特許文献1では、複数の波長を用いて、高感度化およびダイナミックレンジを拡大しようとする測定方法が開示されている。
また、特許文献2、特許文献3においては、平均粒子径が大小2種類の担体粒子とアナライトに対する反応性の異なる複数種類の抗体を組み合わせることによってダイナミックレンジを拡大しようとする粒子増強免疫凝集測定法用の測定試薬が開示されている。
また、特許文献2、特許文献3に開示されている測定試薬に含まれる小粒子は、凝集に伴う光学的変化が小さくまた大量に処方できることから、また反応性の弱い抗体は、凝集力が弱いことから、いずれの発明においてもダイナミックレンジの拡大には効果があると考えられる。しかしながら小粒子の使用や反応性の弱い抗体の使用は、光学的変化や凝集力を弱めることになるため、感度に関する性能については期待した性能を得ることはできない。
以上のように、高感度化とダイナミックレンジの拡大を両立させることは困難に等しく、これらを両立させた粒子増強免疫凝集測定法はこれまで実用化することができなかった。
これまでにも、例えば特開2001-141654号公報記載のように、散乱光強度と吸光度をほぼ同時に測定できる分析装置は開示されているが、当該分析装置において散乱光強度は免疫凝集測定に用いられるものの、吸光度は酵素反応や化学反応に基づくスペクトル変化を測定する分光光度測定に用いられ、単に2種類の分析方法を同一の装置で実施することのみを目的としており、両測定法を一つの測定で併用する粒子増強免疫凝集測定法は記載も示唆もされておらず、そもそもこれまで想起されることもなかった。
すなわち本発明の目的は、高感度な散乱光強度測定と、ダイナミックレンジ確保に特化した分析条件による吸光度測定を一つの測定で併用し、従来よりも簡便に、高感度かつダイナミックレンジの広い粒子増強免疫凝集測定法を提供することにある。
〔1〕粒子増強免疫凝集測定法であって、アナライトを含む試料溶液と、アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液とを混合して混合液を調製する工程と、第1、第2の時点間の散乱光強度差から前記混合液の(イ)散乱光強度の変化量を測定する工程と、第3、第4の時点間の吸光度差から前記混合液の(ロ)吸光度の変化量を測定する工程と、測定された前記(イ)散乱光強度の変化量および前記(ロ)吸光度の変化量を、散乱光強度変化量に基づく検量線および吸光度変化量に基づく検量線を用いて試料中のアナライトの存在量と関連付ける工程と、を具備する粒子増強免疫凝集測定法。

〔3〕前記(ロ)を算出する2時点の間隔は、前記(イ)を算出する2時点よりも間隔が短い前記〔1〕または〔2〕に記載の測定法。
〔4〕前記(イ)及び前記(ロ)の測定を共通の波長で行う前記〔1〕~〔3〕のいずれかに記載の測定法。

実施形態にかかる粒子増強免疫凝集測定法は、アナライトを含む試料溶液と、アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液とを混合して混合液を調製する工程と;
第1、第2の時点間の散乱光強度差から混合液の(イ)散乱光強度の変化量を測定する工程と;
第3、第4の時点間の吸光度差から混合液の(ロ)吸光度の変化量を測定する工程と;
測定された(イ)散乱光強度の変化量および(ロ)吸光度の変化量を、散乱光強度変化量に基づく検量線および吸光度変化量に基づく検量線を用いて試料中のアナライトの存在量と関連付ける工程と;を有する。
本実施形態によれば、このような工程を有することから、実質的に低濃度から高濃度までを包含する検量線を得ることができ、高感度かつダイナミックレンジの広い粒子増強免疫凝集測定を行うことができる。
ここで、第1、第2、第3、第4の時点は混合液の調製開始から1000秒後までの間からそれぞれ選ばれることが好ましい。混合液の調製開始から1000秒以内とすることにより、測定試薬設計の自由度を確保しながら所望の感度と所望のダイナミックレンジの両方を満たすことが可能となるからである。
(イ)散乱光強度の変化量および(ロ)吸光度の変化量の測定は、共通の波長を用いて行うことが好ましい。また(イ)散乱光強度の変化量および(ロ)吸光度の変化量の測定は、550から900nmの波長の範囲内でおこなうことが好ましい。
なお本明細書において「一つの測定」とは、一つの反応槽において行われる一連の反応と測定を意味する。自動分析装置における測定を例にとれば、第一試液と試料との混合と、それに引き続く第二試液(アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液)の添加混合、散乱光強度の変化量の測定および吸光度の変化量の測定を一つの反応槽で行うことを意味する。
また、本発明における「アナライトを含む試料溶液」には、上記のように第一試液(緩衝液)と混合され希釈された試料溶液も含まれるものとする。
また、本明細書において「散乱光強度」の語は、「散乱光度」と記載することもあるが、同義である。
(不溶性担体粒子)
本発明の粒子増強免疫凝集測定法において、不溶性担体として用いられる素材は、測定試薬の成分として利用可能な物質であれば特に制限はないが、具体的にはラテックス、金属コロイド、シリカ、カーボンなどが挙げられる。不溶性担体粒子の平均粒子径は0.05~1μmまで適宜選択できるが、本発明の免疫凝集測定法用の測定試薬においては散乱光強度測定の照射光波長よりも250~450nm小さい粒子径が好適であり、具体的には300~450nm小さい粒子径が好適である。例えば、照射波長が700nmである場合の平均粒子径は250nm~400nmとなる。不溶性担体粒子の平均粒子径は粒度分布計や透過型電子顕微鏡像などで一般に使用される方法により確認することができる。
本発明の粒子増強免疫凝集測定法は種々の生体試料を測定対象とし、特に限定されないが、例えば血液、血清、血漿、尿などの体液である。
本発明の粒子増強免疫凝集測定法のアナライトはタンパク質、ペプチド、アミノ酸、脂質、糖、核酸、ハプテンなど、理論的に粒子増強免疫凝集測定法により測定可能な分子であれば特に制限はない。例としてCRP(C反応性タンパク質)、Lp(a)(リポプロテイン(a))、MMP3(マトリクスメタロプロテイナーゼ3)、抗CCP(環状シトルリン化ペプチド)抗体、抗リン脂質抗体、抗梅毒抗原抗体、RPR、IV型コラーゲン、PSA、AFP、CEA、BNP(脳性ナトリウム利尿ペプチド)、NT-proBNP、インスリン、マイクロアルブミン、シスタチンC、RF(リウマチ因子)、CA―RF、KL-6、PIVKA―II、FDP、Dダイマー、SF(可溶性フィブリン)、TAT(トロンビン-アンチトロンビンIII複合体)、PIC、PAI、XIII因子、ペプシノーゲンI・IIや、フェニトイン、フェノバルビタール、カルバマゼピン、バルプロ酸、テオフィリンなどが挙げられる。
本発明の粒子増強免疫凝集測定法に供される結合パートナーとしては、アナライトに結合する物質としてタンパク質、ペプチド、アミノ酸、脂質、糖、核酸、ハプテンなどが挙げられるが、特異性および親和性から抗体や抗原の利用が一般的である。また分子量の大小、天然や合成といった由来に特に制限はない。
本発明の粒子増強免疫凝集測定法に供される測定試薬の構成に関して特に制限はないが、臨床検査の分野で汎用される自動分析装置での使用を考慮した場合、緩衝液からなる第一試液(R1)とアナライトに対する結合パートナーを担持した担体粒子を含む第二試液(R2)の2液で構成された測定試薬が一般的である。
本発明の不溶性担体粒子を用いる測定試薬の成分は、反応の主成分である結合パートナーを担持した不溶性担体の他に、試料のイオン強度や浸透圧などを緩衝する成分として、例えば、酢酸、クエン酸、リン酸、トリス、グリシン、ホウ酸、炭酸、及びグッドの緩衝液や、それらのナトリウム塩、カリウム塩、カルシウム塩などを含んでも良い。また凝集形成を増強する成分としてポリエチレングリコール、ポリビニルピロリドン、リン脂質ポリマーなどの高分子を含んでも良い。また、凝集の形成をコントロールする成分として高分子物質、タンパク質、アミノ酸、糖類、金属塩類、界面活性剤類、還元性物質やカオトロピック物質など汎用される成分を1種類、または複数の成分を組合わせて含んでも良い。また消泡成分を含んでも良い。
本発明の粒子増強免疫凝集測定法には、測定に要する総反応時間が10分以内と迅速かつ簡便な自動分析装置の利用が適しており、特に特開2013-64705号公報に開示されるような、散乱光強度と吸光度をほぼ同時に測定できる自動分析装置が好適である。
本発明に用いられる散乱光強度測定の散乱角度に特に制限はないが、15度~35度にするのが望ましく、より好ましくは20度~30度である。散乱角度を前記の範囲にすることによって、散乱光を検知するための受光部において透過光の影響を強く受けず、また散乱光を受光する能力に関しても有利となる。
本発明に用いられる散乱光強度測定の光源や照射光波長には特に制限はないが、粒子増強免疫凝集測定法には可視光領域が適しており、特に650~750nmが好適である。散乱光強度の変化量を測定する2時点の測光間隔に特に制限はなく、一般的には間隔が長い方がより高感度となる。
前述の自動分析装置においては、アナライトを含む試料溶液と、アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液との混合直後から、最大1000秒までの任意の2時点における散乱光強度測定と吸光度測定の変化量を、それぞれ測定できるが、混合直後から300秒以内の2時点の散乱光強度の変化量と吸光度の変化量の両方をそれぞれ測定することにより、第一試液、第二試液を通じた一つの測定(一試料)あたりの総測定時間を10分以内とすることができ、市販されている各種自動分析装置の最大検体処理速度の利益を享受することができる。
本発明に用いられる吸光度測定の波長に特に制限はないが、散乱光強度の変化量を測定する波長に対して±25%の範囲内にある同一または別の波長が適しており、550nmから900nmが好適である。より好適な範囲としては570nmから800nmである。また、本発明に用いられる吸光度測定の波長としては、前記の範囲内において1波長測定あるいは、散乱光強度測定に用いる波長と比較して短波長側の主波長と、長波長側の副波長とを組合せた2波長測定を利用することができる。例えば、散乱光強度測定の波長を700nmに設定した場合、吸光度測定の主波長を550から699nm、副波長を701から900nmの範囲で設定することができる。
吸光度の変化量を測定する2時点の測光間隔に特に制限はないが、散乱光強度測定よりも短い方が適しており、散乱光強度測定の1/2以下の測光間隔が望ましい。また、散乱光強度測定の1/3以下の測光間隔が望ましい場合もある。例えば、散乱光強度測定の測光間隔を300秒に設定した場合、吸光度測定の測光間隔は150秒以下である事が望ましく、100秒以下であることがより望ましい場合もある。また、測光開始時点はアナライトを含む試料溶液と、アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液との混合直後が望ましい。
本発明に用いられる光量(散乱光強度および吸光度)の変化量は、2時点間の差や比、単位時間あたりの換算値など、粒子増強免疫凝集測定法に適用可能な算出法であれば特に制限はない。
本発明の粒子増強免疫凝集測定法においては、既知濃度のアナライト含有試料を用いて、散乱光強度測定、吸光度測定それぞれの検量線を個別に作成し、アナライト低濃度域は高感度な散乱光強度測定の検量線、アナライト高濃度域はダイナミックレンジの広い吸光度測定の検量線に基づき濃度を算出する。ダイナミックレンジが広い吸光度測定では、より広い濃度範囲で検量線を作成することができる。
感度とは測定可能な最小のアナライト量を意味し、一般的に当該アナライト量における光量変化が大きいほど高感度である。本発明の粒子増強免疫凝集測定法においては、低濃度のアナライト測定値の正確性や再現性が良好であることが高感度の指標となる。
またダイナミックレンジとは測定可能な最大のアナライト量までの範囲を意味する。本発明の粒子増強免疫凝集測定法のダイナミックレンジは、アナライト濃度に比例した光量変化が検出できる範囲となる。
粒子増強免疫凝集測定法の感度ならびにダイナミックレンジは、測定試薬に含まれる結合パートナーを担持した不溶性担体粒子に依存し、前述のように両性能は二律背反の関係にある。
(調製例:PSA測定試薬の調製)
1.第2試液(R2):抗体結合ラテックス溶液の調製
(1)抗PSAモノクローナル抗体#79および#91と、定法に従って合成した平均粒子径320nmのラテックス粒子とをそれぞれ20mM グリシン緩衝液(pH9)で希釈し、0.7mg/mLの各抗体溶液と1%(w/w)ラテックス溶液を調製した。それぞれの抗体溶液に等量のラテックス溶液を混合し、約1時間攪拌した。
(2)前記(1)の各混合液に、それぞれブロッキング液(10%BSA)を同量添加し、約1時間攪拌した。
(3)前記(2)の各混合溶液を遠心分離して上清を除去後、5mM MOPS緩衝液(pH7.0)溶液で再懸濁し、波長600nmの吸光度が1.5Abs/mLとなるように含量調整した後、両溶液を等量混合して第二試液:抗体結合ラテックス溶液(R2)を得た。
2.第一試液(R1)の調製
1Mの塩化カリウム、0.1%のBSAを含む30mM Tris-HCl緩衝液(pH8.5)を調製し、第一試液とした。
特開2013-64705号公報記載の自動分析装置を用いて一つの測定について散乱光強度と吸光度両方の測定をおこなった。散乱光強度測定の各条件は、照射波長700nm、散乱角度30°とし、吸光度測定の各条件は、主波長570nm、副波長800nmの2波長として測定した。PSAを含む試料8μLにR1 100μLを添加撹拌して37℃で約300秒間インキュベーション後、R2 100μLを添加撹拌し、37℃で約300秒間インキュベーションしている間の任意の2時点間の光量の差から散乱光強度ならびに吸光度の変化量を算出した。
PSAキャリブレーター(積水メディカル社製)を用いてスプライン演算した散乱光強度、吸光度それぞれに基づく検量線を用いて試料中のPSA濃度をそれぞれ算出した。なお検量線の濃度範囲は各測定条件下におけるダイナミックレンジに応じて都度選択した。
散乱光強度ならびに吸光度の光量変化を、本発明の測定方法を用いて本実施例のPSAを測定する際に最大感度が得られると考えられる測光間隔である、R2添加後約30秒後から270秒間で測定した。異なる2濃度(0.4ng/mLおよび1ng/mL)のPSAを含有する試料を10回連続測定し、散乱光測定の場合と吸光度測定の場合の同時再現性を確認した(表1)。PSAの臨床的なカットオフ値は4ng/mLとされているが、感度に優れる散乱光強度測定は1ng/mL以下の濃度でも良好な正確性と再現性を示した。
測光間隔を変動させた次の測定条件、散乱光強度(測光間隔 R2添加約30秒後(第1の時点)から270秒間(第2の時点))、吸光度1(本発明の条件1:測光間隔 R2添加約30秒後(第3の時点a)から約90秒間(第4の時点a))、吸光度2(本発明の条件2:測光間隔 R2添加約15秒後(第3の時点b)から約90秒間(第4の時点b))、吸光度3(従来条件(比較例):測光間隔 R2添加約30秒後(比較例第3の時点)から270秒間(比較例第4の時点))、でダイナミックレンジを比較した(図1)。
散乱光強度測定(-●-)におけるダイナミックレンジは狭くPSA濃度25ng/mLをピークとして変化量は顕著に低下した。従来条件(比較例)である吸光度3(-×-)も同様であったが、高濃度域の光量変化の低下の度合いは緩やかであった。本発明の吸光度条件1(―□―)、2(―△―)では濃度依存的に高濃度域まで光量変化の上昇が認められ、広いダイナミックレンジを得られることが確認された。
本発明の測定条件、吸光度1ならびに2においてPSA濃度100ng/mLから3000ng/mLを超える試料(総称してPSA超高濃度試料という)を測定しプロゾーン現象を確認した(図2A、図2B)。プロゾーン現象とは粒子増強免疫凝集測定法において抗原量過剰によって発生する見かけの測定値が低下する現象で、陽性が陰性と判定され誤診を招く可能性があるため臨床検査の現場では大きな問題となっている。
PSA超高濃度試料を測定した結果、条件2の方がプロゾーン現象による測定値の低下が緩やかでダイナミックレンジが広いことが確認された。本結果を踏まえると、吸光度1、2の実用的な測定上限はそれぞれ50ng/mL程度(図2A)、100ng/mL程度(図2B)と考えられ、吸光度2の測定条件は測定範囲をより高濃度側に設定できることが確認された。
本発明の粒子増強免疫凝集測定法でPSA濃度既知のPSA陽性検体を測定し低濃度域(~10ng/mL以下)は散乱光強度による測定値(●)を、高濃度域(10.1ng/mL以上)は吸光度2による測定値(△)を参照して相関性を確認した(図3)。
相関性は良好であり、本発明によって高感度かつダイナミックレンジの広い粒子増強免疫凝集測定法が達成された。

Claims (7)
- 粒子増強免疫凝集測定法であって、
アナライトを含む試料溶液と、アナライトとの結合パートナーを担持した不溶性担体粒子を含む溶液とを混合して混合液を調製する工程と、
第1、第2の時点間の散乱光強度差から前記混合液の(イ)散乱光強度の変化量を測定する工程と、
第3、第4の時点間の吸光度差から前記混合液の(ロ)吸光度の変化量を測定する工程と、
測定された前記(イ)散乱光強度の変化量および前記(ロ)吸光度の変化量を、散乱光強度変化量に基づく検量線および吸光度変化量に基づく検量線を用いて試料中のアナライトの存在量と関連付ける工程と、を具備することを特徴とする粒子増強免疫凝集測定法。 - 前記第1、第2、第3、第4の時点は、前記混合液の調製開始から1000秒後までの間から選ばれるものであることを特徴とする請求項1に記載の測定法。
- 前記(ロ)を算出する2時点の間隔は、前記(イ)を算出する2時点よりも間隔が短いことを特徴とする請求項1または2に記載の測定法。
- 前記(イ)及び前記(ロ)の測定を共通の波長で行うことを特徴とする請求項1~3のいずれかに記載の測定法。
