a.ホスト基含有モノマー
本発明のホスト基含有モノマーは、ホスト基を1つ以上(好ましくは1つ)含有するビニル系モノマーである。
Qは、NHであることが好ましく、上記式(1)で表されるモノマーとしては、6-アクリルアミド-α-シクロデキストリン、6-アクリルアミド-β-シクロデキストリンが好ましい。
本発明のゲスト基含有モノマーは、ゲスト基を1つ以上(好ましくは1つ)含有するビニル系モノマーである。
本発明のアクリル系モノマーとは、アクリル酸又はメタクリル酸、及びそれらの誘導体(エステル、アミド等)である。
a.溶解
本発明の製造方法においては、上記ホスト基含有モノマー、ゲスト基含有モノマー及びアクリル系モノマーを、水系溶媒に混合し、溶解して、水系溶媒溶液を製造する。当該モノマーを水系溶媒中に溶解することにより、ホスト基とゲスト基が包接錯体を形成する。
本発明の製造方法においては、上記モノマーを、水系溶媒中に溶解後、次いでこれらのモノマーを共重合させることにより、上記モノマーからなるゲルを製造することができる。
本発明のゲルは、上記のとおり、ホスト基含有モノマー、ゲスト基含有モノマー及びアクリル系モノマーを、水系溶媒中に溶解し、次いでこれらのモノマーを共重合させることにより得られる。

サンプル瓶(3 mL)に、国際公開2012/036069号に記載の製造方法により得られた6-アクリルアミド-α-シクロデキストリン92mg(0.09mmol)と、アクリル酸n-ブチル(東京化成工業株式会社製)12mg(0.09mmol)と、全試薬量が0.5 mmolとなるようにアクリルアミド(和光純薬工業株式会社製)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、80℃に加温して1時間80℃に保ち、前記モノマーを水に溶し込んだ。このモノマーの溶解液を25℃まで放冷後、過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、6-アクリルアミド-α-シクロデキストリン由来単位:アクリル酸n-ブチル由来単位:アクリルアミド由来単位=12:11:77(モル組成比)からなるゲルを得た。

サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン104mg(0.088mmol)と、国際公開2012/036069号に記載の製造方法により得られたN-(1-アダマンチル)アクリルアミド18mg(0.087mmol)と、全試薬量が0.5 mmolとなるようにアクリルアミド(和光純薬工業株式会社製)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、80℃に加温して1時間80℃に保ち、前記モノマーを水に溶し込んだ。このモノマーの溶解液を25℃まで放冷後、過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、6-アクリルアミド-β-シクロデキストリン由来単位:N-(1-アダマンチル)アクリルアミド由来単位:アクリルアミド由来単位=7:6:87(モル組成比)からなるゲルを得た。
(1)N-ベンジルアクリルアミドの合成
得られた化合物の1H NMR(500MHz、DMSO-d6、25℃)を、図9に示した。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、上記(1)で得られたN-ベンジルアクリルアミド4.0 mg(0.025 mmol)と、全試薬量が0.5 mmolとなるようにアクリルアミド31.9 mg(0.45 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、70℃に加温して5時間70℃に保ち、前記モノマーを水に溶し込んだ。このモノマーの溶解液を25℃まで放冷後、過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、ゲルを得た。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、実施例5の(1)で得られたN-ベンジルアクリルアミド4.0 mg (0.025 mmol)と、全試薬量が0.5 mmolとなるようにアクリル酸(ナカライテスク(株)社製)31μl(0.45 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、70℃に加温して5時間70℃に保ち、前記モノマーを水に溶し込んだ。このモノマーの溶解液を25℃まで放冷後、過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、70℃で1時間加熱することで、ゲルを得た。
(1)N-1-ナフチルメチルアクリルアミドの合成
得られた化合物の1H NMR(500MHz、DMSO-d6、25℃)を、図10に示した。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、上記(1)で得られたN-1-ナフチルメチルアクリルアミド5.4 mg (0.026 mmol)と、全試薬量が0.5 mmolとなるようにアクリルアミド31.8 mg (0.45 mmol)とを加え、溶液濃度が1MとなるようにDMSO/ H2O = 1/ 2の混合溶媒(0.5 mL)を加えた。これを25℃で1日間撹拌後、70℃に加温して2時間70℃に保ち、前記モノマーを混合溶媒に溶し込んだ。このモノマーの溶解液を25℃まで放冷後、過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、6-アクリルアミド-β-シクロデキストリン由来単位:N-1-ナフチルメチルアクリルアミド由来単位:アクリルアミド由来単位=4:2:94(モル組成比)からなるゲルを得た。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、スチレン3μl(0.026 mmol)と、全試薬量が0.5 mmolとなるようにアクリルアミド32.5 mg(0.46 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、50℃に加温して24時間50℃に保ち、前記モノマーを水に溶し込んだ。過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、ゲルを得た。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、スチレン3μl(0.026 mmol)と、全試薬量が0.5 mmolとなるようにアクリル酸メチル41μl (0.46 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、50℃に加温して24時間50℃に保ち、前記モノマーを水に溶し込んだ。過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、25℃で24時間重合させ、ゲルを得た。

サンプル瓶(3 mL)に、6-アクリルアミド-α-シクロデキストリン25.6 mg (0.025 mmol)と、スチレン3μl(0.026 mmol)と、全試薬量が0.5 mmolとなるようにメタクリル酸2-ヒドロキシエチル54μl(0.45 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、50℃で1日間撹拌することで、前記モノマーを水に溶し込んだ。過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、50℃で24時間、70℃で2時間重合させ、ゲルを得た。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン29.7 mg (0.025 mmol)と、スチレン3μl(0.026 mmol)と、全試薬量が0.5 mmolとなるようにメタクリル酸2-ヒドロキシエチル54μl(0.45 mmol)とを加え、溶液濃度が1Mとなるように純水(0.5 mL)を加えた。これを25℃で1日間撹拌後、50℃で1日間撹拌することで、前記モノマーを水に溶し込んだ。過硫酸アンモニウム5.7mg(0.025mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン2.9mg(0.025mmol)を添加し、50℃で24時間、70℃で2時間重合させ、ゲルを得た。
(1)ゲスト基(アダマンチル基)を2つ含有する化合物の製造
MALDI TOF MS: m/z = 510.10 ([C36H44O2N2Br2 + H]+ = 510.71, 0.23 % error)
melting point: over 300 ℃
Elemental Anal. Calced for C34H42O2N2Br2(H2O)1 : C, 59.31; H, 6.44; N, 4.07
Found: C, 59.31; H, 6.27; N, 4.05. (0.17 % error).
得られた化合物の1H NMR(500MHz、DMSO-d6、30℃)を、図11に示した。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン59 mg (0.05 mmol)と、上記(1)で得られたアダマンチル基を2つ含有する化合物13 mg (0.025 mmol)と、6-アクリルアミドーβ―シクロデキストリンとアクリルアミドの全試薬量が0.5 mmolとなるようにアクリルアミド34mg(0.48 mmol)を加え、溶液濃度が1Mとなるように純水(500μL)を加えた。これを25℃で1日間撹拌後、45℃のオイルバス中で24時間加熱し、前記モノマーを水に溶し込んだ。このモノマーの溶解液に、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]ジヒドロクロライド8.1mg(0.025mmol)を添加し、30分間アルゴンバブリングを施した後、42℃のオイルバス中に静置し、24時間重合させ、ゲルを得た。
(1)ゲスト基(アダマンチル基)を2つ含有する化合物の製造
MALDI TOF MS: m/z = 536.10 ([C36H44O2N2Br2 + H]+ = 537.35, 0.23 % error)
melting point: over 300 ℃
Elemental Anal. Calced for C36H44O2N2Br2(H2O)0.5 : C, 61.28; H, 6.43; N, 3.97.
Found: C, 61.33; H, 6.22; N, 4.05. (0.2 % error).
得られた化合物の1H NMR(500MHz、DMSO-d6、30℃)を、図12に示した。
サンプル瓶(3 mL)に、6-アクリルアミド-β-シクロデキストリン59 mg (0.05 mmol)と、上記(1)で得られたアダマンチル基を2つ含有する化合物13 mg (0.025 mmol)と、6-アクリルアミドーβ―シクロデキストリンとアクリルアミドの全試薬量が0.5 mmolとなるようにアクリルアミド34mg(0.48 mmol)とを加え、溶液濃度が1Mとなるように純水(500μL)を加えた。これを25℃で1日間撹拌後、45℃のオイルバス中で24時間加熱し、前記モノマーを水に溶し込んだ。このモノマーの溶解液に、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]ジヒドロクロライド8.0mg(0.025mmol)を添加し、30分間アルゴンバブリングを施した後、42℃のオイルバス中に静置し、24時間重合させ、ゲルを得た。
サンプル瓶 (10 mL) に、6-アクリルアミド-α-シクロデキストリン92mg(0.09mmol)と、アクリル酸n-ブチル(東京化成工業株式会社製)12mg(0.09mmol)を、全試薬量が 1mmol となるようにアクリルアミド(和光純薬工業株式会社製)を加え、溶液濃度が1Mとなるように純水 (1 mL) を加え、25℃で1日間撹拌したが、前記モノマーは溶解せず、懸濁液であった。そこに、過硫酸アンモニウム11.4mg(0.05mmol)、[2-(ジメチルアミノ)エチル]ジメチルアミン5.8mg(0.05mmol)を添加し、25℃で24時間重合させたが、ゲルは得られなかった。
(1)実施例1において、6-アクリルアミド-α-シクロデキストリン及びアクリル酸n-ブチルの使用量を、それぞれ、[51mg (0.050mmol):6.3mg(0.050mmol)]、[77mg(0.075mmol):9.4mg(0.075mmol)]、[92mg(0.090mmol):12mg(0.091mmol)]、[103mg(0.1mmol):13mg(0.1mmol)]とし、それぞれ、6-アクリルアミド-α-シクロデキストリン由来単位:アクリル酸n-ブチル由来単位=(0.7:0.3)、(4:5)、(9:9)、(12:11)(モル組成比)のゲルを得た。
(1)実施例1で得られたゲルを5mm×5mm×10mmの立方体にカットし、くさび型治具(P-28;株式会社山電製)で中央から2つに破断した。破断面を合わせ、25℃で、24時間水中で静置したところ、ゲルは再接着した。ゲルの破断と再接着の様子を図4に示した。
(1)実施例1で得られたゲルを5mm×5mm×5mmの立方体にカットし、カミソリで中央から2つに破断した。一方の破断面に1-へキサノールを添加した後、破断面を合わせ、25℃で、24時間水中で静置したが、ゲルは再接着しなかった。
(1)試験例2の(1)における、切断前のゲルと再接着後のゲルについて、それぞれクリープメータ(RE2-33005B;株式会社山電製)で、破断時の応力(以下、破断応力と称することもある)を測定し、切断前のゲルの破断応力に対してどの程度、応力が戻っているかを計算したところ、応力回復率は72%であった。結果を図6に示した。
実施例4において、6-アクリルアミド-β-シクロデキストリン及びN-(1-アダマンチル)アクリルアミドの使用量を、それぞれ、[6mg (0.005mmol):1mg(0.005mmol)]、[29mg(0.026mmol):6mg(0.026mmol)]、[45mg(0.038mmol):8mg(0.038mmol)]、[59mg(0.050mmol):10mg(0.050mmol)]とし、それぞれ、6-アクリルアミド-β-シクロデキストリン由来単位:N-(1-アダマンチル)アクリルアミド由来単位=(0.1:0.1)、(0.3:0.4)、(1.0:1.4)、(1.6:1.9)(モル組成比)のゲルを得た。
試験例5で調製した4種の組成を有するゲルを、それぞれ、競争分子(α-シクロデキストリン、β-シクロデキストリン、γ-シクロデキストリン、1-アダマンタンカルボン酸ナトリウム)の10mM水溶液に、それぞれ1時間浸漬した。その後、試験例6と同様の方法で、それぞれのゲルの破断強度を測定したところ、β-シクロデキストリン、1-アダマンタンカルボン酸ナトリウムの溶液に浸漬した場合に、破断応力が低下した。
試験例5で調製した、6-アクリルアミド-β-シクロデキストリン由来単位:N-(1-アダマンチル)アクリルアミド由来単位=(0.3:0.4)のゲル(10mm×10mm×5mm)を、クリープメータ(RE2-33005B;株式会社山電製)を用いて、応力 0.1Nから1Nで引っ張った後、応力をかけない状態で保持した。引張り時から、応力をかけずに保持した時のひずみを計測することで、ゲルの形状回復性を評価した。0.1Nで、180%まで伸ばした後に、応力をかけない状態で保持すると、200秒後にひずみ0%を示した。
実施例4で得られたゲルを、5mm×5mm×10mmの立方体にカットし、それぞれくさび型の冶具(P-28;株式会社山電製)で中央から2つに破断した状態で、25℃で、それぞれ1時間、3時間、6時間、12時間、15時間、18時間、24時間、水中で静置した。各時間経過後、それぞれ破断面を合わせ、水中で24時間静置し、再接着したゲルについて、それぞれクリープメータ(RE2-33005B;株式会社山電製)で、破断時の応力を測定し、切断前のゲルの破断応力に対してどの程度、応力が戻っているかを計算したところ、破断後の経過時間に関係なく、ゲルの応力は回復した。結果を図13に示した。
試験例5で調製した、6-アクリルアミド-β-シクロデキストリン由来単位:N-(1-アダマンチル)アクリルアミド由来単位=(0.3:0.4)のゲル(10mm×10mm×3mm)を、2枚のガラス製スライドガラスの間に挟み、水中で1日間、150gで圧力をかけた後、1日間25℃、湿度42%で乾燥させた。水中での圧縮後と乾燥後の写真を、図14に示した。
Claims (10)
- ホスト基含有モノマー、ゲスト基含有モノマー及びアクリル系モノマーを、水系溶媒に混合する工程、及び
前記工程で得られた混合物を、40~80℃に加温して、前記モノマーの水系溶媒溶液を得る工程、
を備えた、ホスト基含有モノマー、ゲスト基含有モノマー及びアクリル系モノマーの水系溶媒溶液の製造方法。 - ホスト基含有モノマーを30~0.5モル%、ゲスト基含有モノマーを30~0.5モル%、及びアクリル系モノマーを40~99モル%の割合で用いる、請求項1~4のいずれか1項に記載の方法。
- 水系溶媒が、水、又は、水及び水と相溶性のある有機溶媒との混合溶媒である、請求項1~5のいずれか1項に記載の方法。
- 請求項1で得られた水系溶媒溶液中のモノマーを共重合させることにより、ホスト基含有モノマー、ゲスト基含有モノマー及びアクリル系モノマーからなるゲルを製造する方法。
- 0~100℃で共重合を行う、請求項7に記載の方法。
- 請求項7又は8に記載の方法により得られるゲル。
- ホスト基含有モノマー由来の単位を20~0.1モル%、ゲスト基含有モノマー由来の単位を20~0.1モル%、及びアクリル系モノマー由来の単位を60~99.8モル%含有する、請求項9に記載のゲル。
