そこで、スタチンとは作用機序が異なり、かつ、強力な血中LDLコレステロール低下作用を有する医薬が求められている。
従って、PCSK9タンパク量の低下作用や機能阻害作用を有する化合物は、上記疾患に対する医薬の有効成分としての利用も期待できる。

また、別の観点から、本発明は、trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸の左旋性のエナンチオマー、若しくはその塩、又はそれらの溶媒和物(好適には、実質的に光学的に純粋なtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸の左旋性のエナンチオマー、若しくはその塩、又はそれらの溶媒和物)を提供するものである。
また、本発明は、有効成分としての(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物、及び製薬上許容される担体を含有する医薬組成物を提供するものである。
当該医薬及び医薬組成物は、血中LDLコレステロールを低下させることにより、高血中LDLコレステロール状態に起因する疾患(例えば、高LDL血症、脂質異常症(高脂血症)、動脈硬化症、アテローム性動脈硬化症、末梢血管疾患、高コレステロール血症、家族性高コレステロール血症、心臓血管障害、狭心症、虚血、心虚血、血栓症、心筋梗塞、再灌流障害、血管形成性再狭窄、高血圧等)の予防及び/又は治療のための医薬として用いることができる。
さらに、本発明は、高血中LDLコレステロール状態に起因する疾患の予防及び/又は治療のための医薬を製造するための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物の使用;高血中LDLコレステロール状態に起因する疾患の予防及び/又は治療に用いるための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物を提供するものである。
さらに、本発明は、PCSK9 mRNA発現抑制剤、PCSK9タンパク量の低下剤、PCSK9タンパク産生抑制剤又はLDL受容体量の増加剤を製造するための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物の使用;及びPCSK9 mRNA発現抑制剤、PCSK9タンパク量の低下剤、PCSK9タンパク産生抑制剤又はLDL受容体量の増加剤の有効成分として使用するための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物を提供するものである。
さらに、本発明は、PCsの関与する疾患の予防及び/又は治療のための医薬を製造するための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物の使用;及びPCsの関与する疾患の予防及び/又は治療に用いるための(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物を提供するものである。
また、本発明は、HMG-CoA還元酵素mRNA発現抑制剤、HMG-CoA還元酵素産生抑制剤、又はHMG-CoA還元酵素mRNA発現に起因する疾患の予防及び/又は治療のための医薬を製造するためのラセミ体化合物(I)、特許文献2 実施例44記載の化合物、若しくはそれらのエナンチオマー、若しくはそれらの塩、又はそれらの溶媒和物の使用;及びHMG-CoA還元酵素mRNA発現抑制剤、HMG-CoA還元酵素産生抑制剤、又はHMG-CoA還元酵素mRNA発現に起因する疾患の予防及び/又は治療のための医薬の有効成分として使用するためのラセミ体化合物(I)、特許文献2 実施例44記載の化合物、若しくはそれらのエナンチオマー、若しくはそれらの塩、又はそれらの溶媒和物を提供するものである。
さらに、本発明は、ヒトを含む哺乳類動物におけるPCsの関与する疾患の予防及び/又は治療方法であって、(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物の有効量をヒトを含む哺乳類動物に投与する工程を含む方法を提供するものである。
すなわち、一般論として、実質的に光学的に純粋な化合物は、そのラセミ体を合成した後にキラルカラムにより光学分割して製造することができる場合があることが知られている。
しかしながら、キラルカラムを用いる光学分割法は、化合物によっては分割条件の設定が非常に困難な場合があり、また、工業的な大量スケールでの製造には不向きである。加えて、キラルカラムを用いる光学分割法による、実質的に光学的に純粋な(S)体化合物(III)又は(R)体化合物(II)の製造は、その分割条件の設定が非常に困難であった。すなわち、キラルカラムの種類(例えば、CHIRALCEL OD-H、CHIRALCEL OJ-Hなど)、移動相として用いる溶媒の種類・組成(例えば、MeOH/TFA混液、EtOH/TFA混液など)、移動相の流速等の条件を種々変更して、特許文献2 実施例45記載の方法に従って製造したラセミ体化合物(I)からの各エナンチオマーの分取が試みられたが、殆どの条件では分離が上手くいかなかった。そのような中、後記実施例1-1に記載した条件であれば各エナンチオマーの分離が可能であることが判明したものの、当該条件では分解物(エチルエステル体)が生じることが判明した。
また、メタンスルホニル基の代わりにトルエンスルホニル基、クロロメタンスルホニル基又は2,4,6-トリイソプロピルベンゼンスルホニル基等の脱離基を有する光学活性ベンジル化剤を用いて検討を行なったが、1-[3,5-ビス(トリフルオロメチル)フェニル]-1-メタンスルホニルオキシエタンの場合と同様に、実質的に光学的に純粋な(S)体化合物(III)又は(R)体化合物(II)を得ることはできなかった。
しかしながら、一般的に臭化物イオンが脱離する求核置換反応では、反応により生じた臭化物イオンが反応系中でベンジルブロミドと反応し、ラセミ化が進行することが知られている。また、一般にベンジル位での求核置換反応は、ベンジルカチオンの安定化によりSN1型の置換反応も競合するため、部分的にラセミ化が起こることが知られている。
ここで、部分ラセミ化により光学純度が低下した結果得られる、中程度の光学純度を有する化合物(本明細書において、「中程度の光学純度を有する化合物」とは10%ee程度以上90%ee未満、好適には20~80%ee程度、特に好適には40~70%ee程度の光学純度を意味しており、中程度の光学純度を有する化合物を、以下、「セミキラル体」とも称する。さらに、当該セミキラル体において、下記の部分構造中、*印で示される不斉炭素原子がS配置の化合物が、R配置の化合物よりも過剰量存在する場合を特に「(S)体優位のセミキラル体」と称する。一方、当該セミキラル体において、*印で示される不斉炭素原子がR配置の化合物が、S配置の化合物よりも過剰量存在する場合を特に「(R)体優位のセミキラル体」と称する。)
しかしながら、本発明者らの検討によれば、ラセミ体化合物(I)及びそのエチルエステル誘導体では結晶化が進行せず、優先晶析法によって光学純度を高めることは出来なかった。
そして、下記のスキーム2に示すように、アリールアルキル又はヘテロアリールアルキルエステル化合物のセミキラル体(IV)を製造し、ラセミ体が成分として優位な低光学純度の結晶(以下、本明細書において「ラセミ体優位の結晶」と称する場合がある。)を晶析させた後除去することにより、高い光学純度のアリールアルキル又はヘテロアリールアルキルエステル体(V)又は(V´)を得、これを原料とすることによって、ラセミ体化合物(I)の所望のエナンチオマー((S)体化合物(III)及び(R)体化合物(II))を実質的に光学的に純粋な形態として製造することに成功した。

また、(S)体化合物(III)は、PCsの関与する疾患、より具体的には癌、肥満、糖尿病、アルツハイマー病、又はウイルス感染症に対する予防及び/又は治療のための医薬の有効成分としても有用である。
さらに、本発明の製造方法によれば、ラセミ体化合物(I)の所望のエナンチオマー((S)体化合物(III)及び(R)体化合物(II))を実質的に光学的に純粋な形態として簡便に製造することができ、例えば、医薬の有効成分等として有用な、実質的に光学的に純粋な(S)体化合物(III)を製造する方法として好適に利用できる。
従って、例えば「実質的に光学的に純粋な(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸」、「実質的に光学的に純粋なtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸の左旋性のエナンチオマー」及び「実質的に光学的に純粋な(S)体化合物(III)」とは、光学純度が90%ee以上、好ましくは95~100%ee、特に好ましくは97~100%eeである(S)体化合物(III)を意味する。
本発明において、(S)体化合物(III)の光学純度としては、良好なPCSK9タンパク量の低下作用及び/又はLDL受容体量の増加作用を得る観点から、下記実施例1-1に示すキラルHPLC分析条件において98%ee以上であるのが好ましく、99%ee以上であるのが特に好ましい。係る光学純度であれば、他方のエナンチオマー((R)体化合物(II))を実質的に含有しないこととなる。
また、一般式中、nが示す整数としては、1が好ましい。
また、一般式中、R1におけるC1-6アルキル基としては、C1-4アルキル基が好ましく、エチル基がより好ましい。
また、一般式中、Xが示すハロゲン原子としては、塩素原子又は臭素原子が好ましく、臭素原子がより好ましい。
本工程はアミン(XI)を塩基存在下、光学活性なベンジルハロリド(X)又は(X´)と反応させ、セミキラル体(IX)を製造する工程である。化合物(X)又は(X´)の量は、化合物(XI)に対して1.0~3.0モル当量、好ましくは1.5~2.5モル当量用いればよい。
なお、アミン(XI)は公知の化合物であり、その製造方法は、例えば特許文献2に記載されている。
本工程はセミキラル体(IX)を加水分解し、セミキラル体(VII)を製造する工程である。
本工程はセミキラル体(VII)とアルコール(VIII)とを縮合し、セミキラル体(VI)を製造する工程である。アルコール(VIII)の量は、化合物(VII)に対して、0.8~2.0モル当量、好ましくは1.0~1.2モル当量用いればよい。
本工程はセミキラル体(VI)の硫黄原子を酸化することにより、セミキラル体(IV)を製造する工程である。
本工程はセミキラル体(IV)からラセミ体優位な低光学純度の結晶を優先晶析することにより、高光学純度の光学活性体(V)又は(V´)を製造する工程である。
溶媒としては、メタノール、エタノール、n-プロパノール、イソプロパノール等のアルコール類が挙げられ、直鎖状又は分岐鎖状の炭素数1~6のアルコールが好ましく、エタノール、イソプロパノールが特に好ましい。溶媒の量は、化合物(IV)に対して、2~20倍量(V/W)、好ましくは4~10倍量(V/W)、より好ましくは5~8倍量(V/W)である。
また、本工程Eで得られるラセミ体優位の結晶をそのままラセミ体の種晶として使用してもよいが、前記一般式(IV)で表される化合物のラセミ体を別途製造し結晶化して得られる結晶を、ラセミ体の種晶として用いてもよい。係る一般式(IV)で表される化合物のラセミ体は、例えば、下記に示す方法で製造することができる。
すなわち、ラセミ体(IV)を溶媒に溶解し、10~40℃、好ましくは15~20℃で、30分~2日間、好ましくは15~24時間攪拌すればよく、結晶の収量を高めたければさらに温度を-10~10℃、好ましくは-5~5℃に冷却して、30分~24時間、好ましくは2~5時間攪拌してもよい。また、溶媒としては、メタノール、エタノール、n-プロパノール、イソプロパノール等のアルコール類が挙げられ、直鎖状又は分岐鎖状の炭素数1~6のアルコールが好ましく、エタノール、イソプロパノールが特に好ましい。溶媒の量は、ラセミ体(IV)に対して、2~20倍量(V/W)、好ましくは4~10倍量(V/W)、より好ましくは5~8倍量(V/W)である。
<工程F>
本工程は高光学純度の化合物(V)又は(V´)を脱保護することにより、実質的に光学的に純粋な(S)体化合物(III)又は(R)体化合物(II)を製造する工程である。
以下に、ハロゲン化剤として三臭化リンと臭化水素を用いる場合、及び1,2-ジブロモ-1,1,2,2-テトラクロロエタンとトリフェニルホスフィンを用いる場合について特に具体的に説明する。
ハロゲン化剤として三臭化リンと臭化水素 (30%酢酸溶液)を用いる場合は、三臭化リンはフェニルエタノール(XII)又は(XII´)に対して0.3~2.0モル当量使用されるが、好ましくは0.4~0.6モル当量を用いるのがよい。臭化水素はフェニルエタノール(XII)又は(XII´)に対して0.7~3.0モル当量使用されるが、好ましくは0.8~1.2モル当量を用いるのがよい。
ハロゲン化剤として1,2-ジブロモ-1,1,2,2-テトラクロロエタンとトリフェニルホスフィンを用いる場合は、1,2-ジブロモ-1,1,2,2-テトラクロロエタンはフェニルエタノール(XII)又は(XII´)に対して1.0~3.0モル当量使用されるが、好ましくは1.0~1.2モル当量を用いるのがよい。トリフェニルホスフィンはフェニルエタノール(XII)又は(XII´)に対して1.0~3.0モル当量使用されるが、好ましくは1.0~1.2モル当量を用いるのがよい。
本発明は下記推察に何ら拘束されるものではないが、(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物は、PCSK9タンパクの産生を抑制することにより、LDL受容体の分解を抑制し、LDL受容体の量の増加させる結果、血中LDLのLDL受容体への取り込みを促進する。そして、斯かる血中LDLのLDL受容体への取り込みの促進が、血中LDLコレステロール値の低下作用を発揮させる要因の一つとなっているものと推察される。
従って、(S)体化合物(III)、若しくはその塩、又はそれらの溶媒和物を有効成分として含有する医薬や医薬組成物は、高LDL血症のほか、脂質異常症(高脂血症)、動脈硬化症、アテローム性動脈硬化症、末梢血管疾患、高コレステロール血症、家族性高コレステロール血症、心臓血管障害、狭心症、虚血、心虚血、血栓症、心筋梗塞、再灌流障害、血管形成性再狭窄、又は高血圧等の疾患の予防及び/又は治療のための医薬として用いることができる。

実施例1-1:キラルカラムを用いた光学分割
下記条件であれば、特許文献2(国際公開第2008/129951号パンフレット) 実施例45に記載の方法に従って製造したラセミ体化合物(I)からの各エナンチオマーの分離が可能であり、キラルHPLC分析法による(S)体化合物(III)及び(R)体化合物(II)の光学純度の測定に用いることができることが明らかとなった。なお、本発明は、下記キラルHPLC分析条件(特に、下記カラムと下記移動相の組合せ)を用いる、ラセミ体化合物(I)、(S)体化合物(III)若しくは(R)体化合物(II)、若しくはそれらの塩、又はそれらの溶媒和物の光学純度測定方法を提供する。

従って、上記条件でのキラルカラムを用いた光学分割法は光学純度の測定(キラルHPLC分析)には用いることができるものの、実質的に光学的に純粋な各エナンチオマーの製造方法、特に大量スケールでの製造方法としては不適切なものであった。
本発明者らが実施した、優先晶析法による実質的に光学的に純粋な(S)体化合物(III)の製造方法の概略をスキーム5として以下に示す。
なお、各化合物の絶対配置は、工程1で確認された(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタンの絶対配置から判断した。
また、工程6で得られた(S)体化合物(III)((S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸)の光学純度は、上記1-1に記載の条件によるキラルHPLC分析により決定した。
さらに、工程1で得られた1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタン、工程4及び5で得られたtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステルの光学純度は、それぞれ下記条件によるキラルHPLC分析により決定した。なお、本発明は、下記キラルHPLC分析条件(特に、下記カラムと下記移動相の組合せ)を用いる、各化合物、若しくはそれらの塩、又はそれらの溶媒和物の光学純度測定方法を提供する。

(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタンを下記1-(a)の方法により製造し、その絶対配置を以下の通り確認した。すなわち、得られた(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタンを(S)-1-[3,5-ビス(トリフルオロメチル)フェニル]エチルアミンに導いたうえで、市販されている絶対配置既知の(S)-1-[3,5-ビス(トリフルオロメチル)フェニル]エチルアミンの標準品と比旋光度の実測値の符号を比較することで確認した。
また、(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタンを別途下記1-(b)の方法によっても製造した。
アルゴン雰囲気下、1,2-ジブロモ-1,1,2,2-テトラクロロエタン(7.57g、23.2mmol)をトルエン(12.5mL)に溶解し、0℃にてトリフェニルホスフィン(6.1g、23.2mmol)を加え30分間撹拌した。これに(S)-1-[3,5-ビス(トリフルオロメチル)フェニル]エタノール(1)(5.0g、19.4mmol、>99.5%ee)のトルエン溶液(12.5mL)を0℃で10分以上かけて滴下した後、室温まで昇温し、同温にて1時間撹拌した。反応液にn-ヘキサン(25mL)を加え、セライトろ過した。ろ液を水、飽和重曹水、飽和食塩水で順次洗浄し、硫酸ナトリウムで乾燥後、減圧留去した。得られた残渣を減圧蒸留(56℃、0.7mmHg)することで、5.52gの(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタン(2)を無色油状物として得た(収率:88.6%)。

上記1-(a)で得られた(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタン(2)(106mg、0.336mmol、99%ee)のN,N-ジメチルホルムアミド溶液(1mL)にアジ化ナトリウム(64.4mg、0.990mmol)を加え-18~-15℃にて4時間撹拌した。反応溶液を酢酸エチル/n-ヘキサン(1:1)及び水より抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥後、減圧濃縮することで111.5mgの1-[3,5-ビス(トリフルオロメチル)フェニル]エチルアジド(粗生成物:111.5mg)を得た。

アルゴン雰囲気下、(S)-1-[3,5-ビス(トリフルオロメチル)フェニル]エタノール(1)(300g、1.16mol、96%ee)に水浴下、20℃以下で三臭化リン(157.3g、0.58mol)を滴下し、19~22℃で30分撹拌した。反応液を冷却し、0℃以下で臭化水素(30%酢酸溶液)(228mL、1.16mol)を滴下し、13~15℃で16時間撹拌した。反応液を氷水に注加し、n-ヘキサン(3L×2)で抽出した。有機層を合わせ、飽和重曹水(3L)、飽和食塩水(3L)で順次洗浄し、無水硫酸マグネシウムで乾燥後、減圧下(90~100mmHg)濃縮し、389.2gの粗体を得た。得られた粗体をカラムクロマトグラフィー(シリカゲル900g、展開溶媒:n-ヘキサン)で精製することにより、349.8gの(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタン(2)を無色油状物として得た(収率93.8%)。
なお、下記の通りキラルHPLC分析において、第一ピークがメインピークとして現れたことから、1-(b)で製造した1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタンも1-(a)と同様に、(R)体であることが確認された。

アルゴン雰囲気下、特許文献2(国際公開第2008/129951号パンフレット)記載の方法により合成したtrans-[4-([(エチル){2-[({5-[2-(メチルチオ)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}アミノ]メチル)シクロヘキシル]酢酸エチル(3)(565.4g、0.99mol)の無水テトラヒドロフラン(THF、2.26L)溶液に氷冷下、NaH(60% in oil、119g、2.98mol)を加え、室温にて1時間撹拌した。反応液を-30℃に冷却し、工程1で得られた(R)-1-ブロモ-1-[3,5-ビス(トリフルオロメチル)フェニル]エタン(2)(682g、1.99mol、93.9%ee)の無水N,N-ジメチルホルムアミド(4.53L)溶液を反応系内が-15℃以下となるように滴下し、-15℃~-1℃で5時間撹拌した。反応液を氷水(35L)とトルエン(30L)の混合溶液に注加し、pHが6.9になるまでクエン酸を加え、有機層を分離した。
アルゴン雰囲気下、工程2で得られたtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルチオ)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸の、(S)体優位のセミキラル体(4)(744.1g、0.95mol)の無水ジクロロエタン(11.6L)溶液に、氷冷下、ベンジルアルコール(113.1g、1.05mol)、WSC・HCl(200.5g、1.05mol)及びDMAP(11.9g、98mmol)を加え、室温で終夜撹拌した。反応液に水(10L)を加え、クロロホルム(19L、14L)にて抽出し、有機層を飽和食塩水(12L)で洗浄し、無水硫酸マグネシウムで乾燥後、減圧濃縮し、得られた残渣をカラムクロマトグラフィー(シリカゲル 28g、展開溶媒:ヘプタン/酢酸エチル=6/1)で精製することにより、trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルチオ)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステルのセミキラル体(5)を得た(黄色油状物、745.8g、収率90%)。
アルゴン雰囲気下、工程3で得られたtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルチオ)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステルの、(S)体優位のセミキラル体(5)(745.8g、0.87mol)の2-プロパノール(15.2L)溶液に、五塩化タンタル(31.3g、87.3mmol)及び30%過酸化水素水(496mL、4.38mol)を加え、室温にて5時間撹拌した。反応液を飽和亜硫酸水素ナトリウム水溶液(3.1L)でクエンチし、水(15L)を加え、クロロホルム(14L、12L)で抽出し、有機層を飽和食塩水(20L)で洗浄し、無水硫酸マグネシウムで乾燥後、減圧濃縮し、得られた残渣をカラムクロマトグラフィー(シリカゲル 26kg、展開溶媒:ヘプタン/酢酸エチル=3/1→2/1)で精製することにより、trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステルのセミキラル体(6)を得た(黄色アモルファス、619.5g、収率79%)。

工程4で得られたtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステルの、(S)体優位のセミキラル体(6)(111.7g、123.7mmol、67.7%ee)をエタノール(825mL)に溶解し、別途調製した種晶(下記工程7で製造したラセミ体結晶)2.0mgを15℃~20℃にて加え、同温にて21時間、0℃にて3時間撹拌した。析出物をろ別し、冷エタノール(165mL)で洗浄した後、母液を減圧濃縮することにより、実質的に光学的に純粋なtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステル(7)を得た(黄色アモルファス、66.38g、収率59%)。
なお、得られたtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステル(7)は、(S)体優位のセミキラル体(6)からラセミ体優位の結晶をろ別して得られていることから、(S)体である。

窒素雰囲気下、工程5で得られた(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステル(7)(34.2g、37.88mmol、>99%ee)のエタノール(340mL)溶液に、10%Pd-C(wet)(3.4g)を加え、水素置換後、室温にて2時間撹拌した。反応懸濁液をセライトろ過し、エタノール(50mL)で洗浄し、洗液を減圧濃縮することにより、実質的に光学的に純粋なtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸(III)を得た(白色アモルファス、31.78g、収率100%)。
なお、得られた化合物は、下記比旋光度に示す通り、左旋性の化合物であった。また、得られた化合物は、(S)体であるベンジルエステル(7)よりエステルを脱保護して得られたことから、同様に(S)体である。

特許文献2(国際公開第2008/129951号パンフレット) 実施例45記載の方法により合成したtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸(ラセミ体化合物(I))(20g、24.61mmol)の無水ジクロロメタン(200mL)溶液に、氷冷下、ベンジルアルコール(2.93g、27.07mmol)、DMAP(300mg、2.46mmol)及びWSC・HCl(5.19g、27.07mmol)を加え、室温に昇温し、16時間撹拌した。反応液に水(100mL)を加え、クロロホルム(500mL)で抽出し、有機層を2M 塩酸水溶液(100mL)及び飽和食塩水(100mL)で洗浄し、無水硫酸マグネシウムで乾燥後、減圧濃縮し、得られた残渣をカラムクロマトグラフィー(シリカゲル 350g、展開溶媒:N-ヘキサン/酢酸エチル=3/1→1/1)で精製することにより、trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸ベンジルエステル(21.15g、収率95.2%)を白色アモルファスとして得た。
ヒト肝癌細胞株であるHepG2細胞に被験化合物を添加し、48時間培養後のPCSK9タンパク量及びLDL受容体(LDLR)量をウエスタンブロット法にて測定することにより、被験化合物がPCSK9タンパク量、LDL受容体量に及ぼす影響を確認した。
すなわち、HepG2細胞を6穴プレートに5×105cells/wellの濃度で播種し一晩培養した後、ジメチルスルホキシド(DMSO)に溶解した被験化合物、又はDMSOのみを培養液に対して1000分の1倍量加えた。細胞をCO2 インキュベーターにて37℃で48時間培養した後、RIPA緩衝液(50mM Tris-HCl、pH7.8、150mM NaCl、1% NP-40、0.5% sodium deoxycholate、0.1% SDS、proteinase inhibitor)を100μL加えて破砕して、タンパク質を抽出した。抽出したタンパク質を10000×gで遠心後、上清を回収し、SDSサンプル緩衝液(60mM Tris-HCl、pH6.8、2% SDS、10% Glycerol、3% メルカプトエタノール)を加え、8%アクリルアミドゲルを用いたSDS-PAGE(SDS-ポリアクリルアミドゲル電気泳動法)で分離した。分離終了後、iBlotゲルトランスファーシステム(インビトロジェン社)を用いてニトロセルロースメンブレンにタンパク質を固定し、ブロックエース(DSファーマバイオメディカル社、カタログ番号 UK-B 80)を用いてブロッキングを行った。
PCSK9タンパク、LDLR、β-Actinタンパクの検出及び量の測定は、それぞれ1次抗体にAnti-PCSK9(Cayman社、カタログ番号1000718)、Anti-LDLR(BioVision社、カタログ番号3839-100)又はAnti-β-Actin(Sigma社、カタログ番号A5316)を用い、2次抗体にAnti-Rabbit IgG-HRP(Sigma社、カタログ番号A0545)又はAnti-Mouse IgG-HRP(Sigma社、カタログ番A4416)を用いてメンブレン上のタンパク質を抗体で標識し、化学発光試薬(HRPの基質)をメンブレン上の2次抗体と反応させた後、ルミノ・イメージアナライザーLAS‐3000(富士写真フィルム株式会社製)を用いてシグナル強度を測定することにより行なった。得られたシグナル強度は、画像解析ソフトScience Lab 2002 Multi Gauge(富士写真フィルム株式会社製)を用いて数値化した。
結果を表1に示す。

(S)体化合物(III)(光学純度>99%ee)を終濃度10μMとして培養液に添加した。
(R)体化合物(II)(光学純度≧98%ee)を終濃度10μMとして培養液に添加した。
ラセミ体化合物(I)を終濃度10μMとして培養液に添加した。
以上の試験結果から、(S)体化合物(III)は、PCSK9タンパク量の低下作用、LDL受容体量の増加作用を有することが明らかとなった。
なお、本発明は以下の推察に何ら拘束されるものではないが、ラセミ体化合物(I)はおよそ50%の(S)体化合物(III)を含むものであるにも拘わらずPCSK9タンパク量の低下作用を殆ど示さず、また、LDL受容体量の増加作用を全く示さなかったことから、ラセミ体化合物(I)においては、その構成成分たる(R)体化合物(II)が(S)体化合物(III)の作用発現を阻害しているものと推察された。そして、(S)体化合物(III)の光学純度を高くして(R)体化合物(II)の含有量を減らすことが、特にLDL受容体量の増加作用を高めるうえで好ましいものと推察された。
上記実施例2で確認されたPCSK9タンパク量の低下作用のメカニズムを確認するため、HepG2細胞に被験化合物を添加し、24時間培養した後のPCSK9 mRNA発現量を定量リアルタイムPCR法により確認した。
すなわち、HepG2細胞を24穴プレートに2×105cells/wellの濃度で播種して一晩培養した後、ジメチルスルホキシド(DMSO)に溶解した被験化合物、又はDMSOのみを培養液に対して1000分の1倍量加えた。細胞をCO2インキュベーターにて37℃で24時間培養した後、ISOGEN(ニッポンジーン社、カタログ番号31‐02501)を500μL加えてtotal RNAを抽出した。抽出したtotal RNAからHigh Capacity cDNA Reverse Transcription kit(アプライドバイオシステムズ社、カタログ番号4368813)を用いてcDNAを合成した。ヒトPCSK9 mRNA発現量は、ヒトPCSK9に特異的なプライマー(Kourimate S. et.al., J Biol Chem. Vol.283, p9666)及びFast SYBR Green master mix(アプライドバイオシステムズ社、カタログ番号4385614)を用いた定量リアルタイムPCR法により測定した。測定機器としては7900HT Fast Realtime PCR systemを用いた。
結果を表2に示す。
1:(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸((S)体化合物(III))
(S)体化合物(III)(光学純度>99%ee)を終濃度10μMとして培養液に添加した。
従って、実施例2で確認された、(S)体化合物(III)の有するPCSK9タンパク量の低下作用の少なくとも一部はPCSK9遺伝子発現抑制作用に基づくものであり、(S)体化合物(III)はPCSK9タンパクの産生抑制作用を有するものと考えられた。
(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸((S)体化合物(III):光学純度>99%ee)を0.5%メチルセルロース溶液に懸濁させた後、正常ハムスター(雄性Syrian Hamster)に金属ゾンデを用いて1日1回、14日間反復経口投与した。最終投与の4時間後に血液を採取し、血漿を得た。血漿中のリポタンパク質の分析は、J. Lipid. Res., 43, p805- 814に記載の方法に準じ、ポストラベル法を用いたHPLCシステムにより自動測定することで行なった。すなわち、血漿サンプル15μLを1mM EDTAを含むPBSで10倍希釈後、HPLCシステム(送液ユニット:Shimadzu LC‐20A system、島津製作所)に接続したゲルろ過カラム(Superose6カラム(カラムサイズ:10×300mm)、GEヘルスケアバイオサイエンス製)に80μL注入した。ランニングバッファとして1mM EDTAを含むPBSを用い、流速0.5mL/min、カラム温度40℃にて分離を行なった。カラムからの溶出液に対しコレステロール測定試薬(コレステロールE テストワコー、和光純薬工業社製)を流速0.25mL/minで混合し、反応コイル(0.5mm×15m)中にて送液下40℃にて反応させた。反応コイルからの溶出液中のコレステロールを、波長600nmで検出した。得られたコレステロールのピーク総面積に占めるLDL画分の面積割合を求め、あらかじめコレステロールE テストワコーを用いて測定した総コレステロール量に、LDL画分の面積割合を乗じてLDLコレステロール量を算出した。
以上の試験結果から、(S)体化合物(III)は、血中LDL低下作用を有する医薬の有効成分等として有用であることが明らかとなった。
HepG2細胞に被験化合物を添加し、8時間培養した後のHMG-CoA還元酵素mRNA発現量を定量リアルタイムPCR法により測定した。
すなわち、HepG2細胞を24穴プレートに2×105cells/wellの濃度で播種し一晩培養した後、ジメチルスルホキシド(DMSO)に溶解した被験化合物、又はDMSOのみを培養液に対して1000分の1倍量加えた。細胞をCO2 インキュベーターにて37℃で8時間培養後、ISOGEN(ニッポンジーン社、カタログ番号31-02501)を500μL加えてtotal RNAを抽出した。抽出したtotal RNAからHigh Capacity cDNA Reverse Transcription kit(アプライドバイオシステムズ社、カタログ番号4368813)を用いてcDNAを合成した。ヒトHMG-CoA還元酵素mRNA発現量は、ヒトHMG-CoA還元酵素に特異的な以下のプライマーセット:5'‐GGTGTTCAAGGAGCATGCAAAG‐3'及び5'‐TGACAAGATGTCCTGCTGCCA‐3'、並びにFast SYBR Green master mix(アプライドバイオシステムズ社、カタログ番号4385614)を用いた定量リアルタイムPCR法により測定した。測定機器としては7900HT Fast Realtime PCR systemを用いた。
結果を表4に示す。
1:trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸(ラセミ体化合物(I))
ラセミ体化合物(I)を終濃度10μMとして培養液に添加した。
特許文献2 実施例44記載の化合物を終濃度10μMとして培養液に添加した。
Claims (19)
- 実質的に光学的に純粋な(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸、若しくはその塩、又はそれらの溶媒和物。
- 実質的に光学的に純粋なtrans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸の左旋性のエナンチオマー、若しくはその塩、又はそれらの溶媒和物。
- 光学純度が99%ee以上である、請求項1又は2記載の化合物、若しくはその塩、又はそれらの溶媒和物。
- 請求項1~3のいずれか1項に記載の化合物、若しくはその塩、又はそれらの溶媒和物を有効成分とする医薬。
- 高LDL血症、脂質異常症、動脈硬化症、アテローム性動脈硬化症、末梢血管疾患、高コレステロール血症、家族性高コレステロール血症、心臓血管障害、狭心症、虚血、心虚血、血栓症、心筋梗塞、再灌流障害、血管形成性再狭窄及び高血圧から選ばれる疾患の予防及び/又は治療に用いられるものである、請求項4に記載の医薬。
- 請求項1~3のいずれか1項に記載の化合物、若しくはその塩、又はそれらの溶媒和物を有効成分とするPCSK9 mRNA発現抑制剤。
- 請求項1~3のいずれか1項に記載の化合物、若しくはその塩、又はそれらの溶媒和物を有効成分とするPCSK9タンパク量の低下剤。
- 請求項1~3のいずれか1項に記載の化合物、若しくはその塩、又はそれらの溶媒和物を有効成分とするPCSK9タンパク産生抑制剤。
- 請求項1~3のいずれか1項に記載の化合物、若しくはその塩、又はそれらの溶媒和物を有効成分とするLDL受容体量の増加剤。
- 実質的に光学的に純粋な(S)-trans-{4-[({2-[({1-[3,5-ビス(トリフルオロメチル)フェニル]エチル}{5-[2-(メチルスルホニル)エトキシ]ピリミジン-2-イル}アミノ)メチル]-4-(トリフルオロメチル)フェニル}(エチル)アミノ)メチル]シクロヘキシル}酢酸、若しくはその塩、又はそれらの溶媒和物の製造方法であって、下記の一般式(IV):
- 一般式(V)で表される化合物から、-(CH2)n-Rで表される基を除去する工程をさらに含む、請求項10に記載の方法。
- (S)-1-[3,5-ビス(トリフルオロメチル)フェニル]エタノールを、ハロゲン化剤の存在下でハロゲン化して、一般式(X)で表される化合物を製造する工程をさらに含む、請求項15に記載の方法。
- 請求項10に記載の一般式(IV)で表される化合物、若しくはその塩、又はそれらの溶媒和物。
- 請求項10に記載の一般式(V)で表される化合物、若しくはその塩、又はそれらの溶媒和物。
- Rがフェニル基であり、nが1である請求項17又は18に記載の化合物、若しくはその塩、又はそれらの溶媒和物。
