<15>前記一般式(1)、(2)に表される化合物、前記<2>、<3>、<4>、<6>、<7>に定義される化合物、その異性体及び/又はそれらの薬理学的に許容される塩を、色素沈着に対する予防又は改善が必要な対象に投与することを含む、色素沈着予防又は改善方法。
本発明の皮膚外用剤は、前記一般式(1)に表される化合物、その異性体及び/又はそれらの薬理学的に許容される塩よりなる色素沈着予防又は改善剤を含有することを特徴とする。本発明の色素沈着予防又は改善剤には、既に形成された色素沈着を薄くする又は消去する色素沈着改善作用に加え、色素沈着を予防する作用も包含される。本発明の色素沈着予防又は改善剤としては、前記一般式(1)に表される化合物、その異性体及び/又はそれらの薬理学的に許容される塩であって、色素沈着を予防又は改善する作用を有する成分であれば特段の限定なく適応することが出来るが、より好ましくは、後記の「有色モルモットを用いた紫外線照射による色素沈着抑制作用評価」において色素沈着抑制作用を有する成分が好適に例示出来る。前記の色素沈着抑制作用評価において色素沈着抑制作用を有する成分とは、コントロール群(溶媒対照群)に比較し、評価物質投与群に色素沈着抑制作用が認められる成分を意味し、より好ましくは、コントロ-ル群に比較し、評価物質投与群の色素沈着抑制作用に統計的な有意差が認められる成分が好ましい。

本発明の皮膚外用剤は、前記一般式(1)、(2)に表される化合物、前記<2>、<3>、<4>、<6>、<7>に定義される化合物、その異性体及び/又はそれらの薬理学的に許容される塩よりなる色素沈着予防又は改善剤を含有することを特徴とする。
[工程1] p-メチルベンゾイルクロリドの合成
十分に乾燥させたナスフラスコにp-トルイル酸(100g、0.734mol)(東京化成工業株式会社)とトルエン(500mL)(和光純薬工業株式会社)を入れ、p-トルイル酸を溶解させた。この溶液に塩化チオニル(132.4mL、1.84mol)(和光純薬工業株式会社)を1時間掛け滴下した。滴下後、2時間加熱還流を行った。反応後、室温まで冷却した後、ロータリーエバポレータで残存する塩化チオニル及びトルエンを留去した。濃縮液にトルエン(200mL)を添加し、濃縮を2回繰り返した。最終的に得られた残渣をテトラヒドロフラン(200mL)(和光純薬工業株式会社)に溶解し、次工程に付した。
ナスフラスコにL-セリン(100g、0.952mol)(和光純薬工業株式会社)、炭酸カリウム(131.5g、0.952mol)(和光純薬工業株式会社)、水1Lを入れ、激しく撹拌した。この溶液に、工程1で調製したp-メチルベンゾイルクロリドをテトラヒドロフラン(和光純薬工業株式会社)に溶解し、30分掛けて滴下した。途中、炭酸カリウムを追添加しながら、pH8付近を維持した。滴下終了後、1時間撹拌した。反応液を別容器に準備した水1Lに添加後、塩酸にてpH3以下にし、4℃に冷却した。析出した結晶をろ過した後、エタノール(和光純薬工業株式会社)/水=6/4の混合溶媒にて再結晶し、目的物を106.0g(収率 64.7%)で得た。
安息香酸及びL-セリンを用い、前記化合物1と同様の方法に従い、化合物2を合成した。
p-トルイル酸及びDL-セリンを用い、前記化合物1と同様の方法に従い、化合物3を合成した。
L-セリン(2.01g、19.1mmol)(株式会社ペプチド研究所)及び炭酸カリウム(2.89g、20.9mmol)(和光純薬工業株式会社)を秤量後、水(10mL)を加えた。氷冷下、撹拌しながら、p-フルオロベンゾイルクロリド(3.61g、22.8mmol)(和光純薬工業株式会社)/テトラヒドロフラン(10mL)(和光純薬工業株式会社)溶液を9分掛けて滴下した。水浴をはずし、室温に戻して43.5時間撹拌後、減圧下にてテトラヒドロフランを留去した。氷冷下、撹拌しながら、塩酸(4mL)(和光純薬工業株式会社)を加え、pH2以下にした。水(40mL)を加え、固体を濾取し、これを水にてよく洗浄した。4時間減圧下にて乾燥後、酢酸エチル(200mL)(和光純薬工業株式会社)に溶解し、飽和食塩水(50mL)及び5N塩酸(5mL)(和光純薬工業株式会社)の混合溶液及び飽和食塩水(100mL×2)にて順次洗浄した。有機層を無水硫酸ナトリウム(和光純薬株工業式会社)にて乾燥後、濾過し、濾液を減圧下にて濃縮した。濃縮残渣にtert-ブチルメチルエーテル(東京化成工業株式会社)を加え、不溶物を濾取し、化合物4を1.65g(収率 38.0%)で得た。
p-メトキシベンゾイルクロリド及びL-セリンを用い、前記化合物4と同様の方法に従い、化合物5を合成した。
L-セリンメチルエステル塩酸塩(1.55g、9.96mmol)(東京化成工業株式会社)をジクロロメタン(30mL)(和光純薬工業株式会社)に分散し、トリエチルアミン(2.25g、22.2mmol)(和光純薬工業株式会社)を加え、氷冷下、撹拌しながら、p-メチルベンゾイルクロリド(1.78g、11.5mmol)(アルドリッチ)/ジクロロメタン(5mL)溶液を3分掛けて滴下した。水浴をはずし、室温にて戻して6時間撹拌後、反応液を酢酸エチル(100mL)(和光純薬工業株式会社)にて希釈し、飽和炭酸水素ナトリウム溶液(30mL)、1N塩酸(50mL)及び飽和食塩水(30mL、60mL×2L)にて順次洗浄した。有機層を無水硫酸ナトリウム(和光純薬工業株式会社)にて乾燥後、濾過し、濾液を減圧下にて濃縮した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=1:2)に付し、目的物を含むフラクションを集め、減圧下にて濃縮し、化合物6を1.88g(収率 79.5%)で得た。
L-セリン(1.18g、11.2mmol)(株式会社ペプチド研究所)及び炭酸カリウム(1.71g、12.4mmol)(和光純薬工業株式会社)を秤量後、水(5mL)及びテトラヒドロフラン(5mL)(和光純薬工業株式会社)を加えた。氷冷下、撹拌しながら、p-エチルベンゾイルクロリド(2.24g、13.3mmol)(和光純薬工業株式会社)を5分掛けて滴下した。水浴をはずし、室温に戻して16時間撹拌後、減圧下にてテトラヒドロフランを留去した。氷冷下、撹拌しながら、塩酸(3mL)(和光純薬工業株式会社)を加え、pH2以下にした。水(20mL)を加え、固体を濾取し、これを水にてよく洗浄した。tert-ブチルメチルエーテル(20mL)(東京化成工業株式会社)にて加熱溶解した後、飽和食塩水(10mL)にて洗浄した。有機層を無水硫酸ナトリウム(和光純薬工業株式会社)にて乾燥後、濾過し、濾液を減圧下にて濃縮した。濃縮残渣にtert-ブチルメチルエーテルを加え、不溶物を濾取し、化合物7を1.08g(収率 40.5%)で得た。
p-(トリフルオロメチル)ベンゾイルクロリド及びL-セリンを用い、前記化合物7と同様の方法に従い、化合物8を合成した。
1H-NMR(CD3OD):δ 4.02(2H、m)、4.74(1H、m)、7.81(2H、d)、8.07(2H、d).
p-メチルベンゾイルクロリド及びDL-O-メチルセリンを用い、前記化合物7と同様の方法に従い、化合物9を合成した。
L-セリン(2.00g、19.0mmol)(株式会社ペプチド研究所)をテトラヒドロフラン(19mL)(和光純薬工業株式会社)に分散し、氷冷下、撹拌しながら、2N 水酸化ナトリウム水溶液(19mL)を加えた。ついで、2-ナフトイルクロリド(3.64g、19.1mmol)(東京化成工業株式会社)を加えた。水浴をはずし、室温に戻して16時間撹拌後、減圧下にてテトラヒドロフランを留去した。氷冷下、撹拌しながら、塩酸(4mL)(和光純薬工業株式会社)を加え、pH2以下にした。固体を濾取し、これを水にてよく洗浄した。tert-ブチルメチルエーテル(30mL)(東京化成工業株式会社)を加え、不溶物を濾取した。これをtert-ブチルメチルエーテルにて徹底的に洗浄した。更に、tert-ブチルメチルエーテル:酢酸エチル(=4:1)及びn-ヘキサンにて順次洗浄し、化合物10を2.92g(収率 59.2%)で得た。
p-メチルベンゾイルクロリド及びO-アセチル-L-セリン塩酸塩用い、前記化合物6と同様の方法に従い、化合物11を合成した。
2-ナフトイルクロリド及びL-セリンメチルエステル塩酸塩を用い、前記化合物6と同様の方法に従い、化合物12を合成した。
2-ナフトイルクロリド及びDL-O-メチルセリンを用い、前記化合物4と同様の方法に従い、化合物13を合成した。
4-フェニルベンゾイルクロリド及びL-セリンを用い、前記化合物4と同様の方法に従い、化合物14を合成した。
ナスフラスコにD-セリン(1.67g、15.9mmol)(東京化成工業株式会社)、水(9mL)及び炭酸カリウム(16.6g、16.6mmol)(和光純薬工業株式会社)、テトラヒドロフラン(9mL)(和光純薬工業株式会社)を加え、氷冷下、撹拌しながら、p-メチルベンゾイルクロリド(2.59g、16.7mmol)(アルドリッチ)を2分掛けて滴下した。水浴をはずし、室温にて戻して26時間撹拌後、反応液に塩酸(4mL)(和光純薬工業株式会社)を加え、pH2以下にし、20分間氷冷下にて撹拌した。析出した結晶を濾取し、これを水にて洗浄した。固体を減圧下にて乾燥した後に、酢酸エチル12mL(和光純薬工業株式会社)を加え、不溶物を濾取した。これを酢酸エチルにて洗浄した。24時間、減圧下にて乾燥した後に、エタノール(和光純薬工業株式会社)/水=5/5の混合溶媒にて再結晶し、化合物15を2.20g(収率 62.0%)で得た。
ベンゾイルクロリド及びDL-O-メチルセリンを用い、前記化合物7と同様の方法に従い、化合物16を合成した。
p-メチルベンゾイルクロリド及びL-セリンエチルエステル塩酸塩を用い、前記化合物6と同様の方法に従い、化合物17を合成した。
有色モルモット8匹の背部皮膚を電気バリカンとシェーバーで除毛及び剃毛し、この部位を2×2cmの照射窓を上下左右2ヶ所ずつの計4個有する黒布で覆った後に、FL20S・E30ランプを光源として300mJ/cm2の紫外線を照射した。この操作を試験1、3、5、8日に繰り返し行い、4ヶ所の試験部位に色素沈着を誘導した。評価化合物(化合物1)を3%(w/v)となるようにエタノ-ルに溶解し、調製した。また、溶媒対照群としてエタノ-ルを塗布した。試験1日目の紫外線照射終了時より、各評価化合物の塗布を開始した。各評価化合物は、所定の試験部位に1日1回、30μLずつ塗布し、これを5週間(試験35日目まで)継続して実施した。塗布開始日(試験1日目)の紫外線照射前及び5週間後(試験36日目)に、色彩色差計(CR-200、コニカミノルタ株式会社)を用いて各試験部位の皮膚明度(L*値)を測定し、試験36日目のL*値から紫外線照射前のL*値を差し引いたΔL*値を求めた。すなわち、L*値は、色素沈着の程度が強いほど低い値となるため、ΔL*値が大きい程、色素沈着が抑制されたとすることができる。結果を表2に示す。
表3に記載の処方に従って、本発明の皮膚外用剤である化粧料(ローション)を作製した。即ち、処方成分を80℃に加熱し、攪拌し、溶解させ、攪拌冷却し、化粧料(ローション1)を得た。同様にして、「本発明の色素沈着抑制剤(化合物1)」を水に置換した比較例1のローション、「本発明の色素沈着抑制剤(化合物1)」をアルブチンに置換した比較例2も作製した。
表4に示す処方に従って、油中水クリームを作成した。即ち、イ、ロの成分をそれぞれ80℃に加熱し、イにロを徐々に加え、乳化し、ホモジナイザーで粒子を均一化した後、撹拌冷却して化粧料(クリーム1)を得た。
表5に示す処方に従って、本発明の皮膚外用組成物である化粧料(乳液1)を作製した。即ち、イ、ロ及びハの成分を80℃に加熱し、ロにハを攪拌しながら徐々に加え中和した後、イを徐々に攪拌しながら加え、ホモミキサーにより乳化粒子を均一化し乳液を得た
ローション1、比較例1及び比較例2の化粧料を用いて、色素沈着抑制効果を調べた。自由意思で参加したパネラーの上腕内側部に1.5cm×1.5cmの部位を上下2段に分け、2ヶ所ずつ合計4ヶ所設け、最少紅斑量(1MED)の紫外線照射を1日1回、3日連続して3回照射した。照射終了後1日より、1日1回28日連続してサンプル50μLを塗布した。1部位は無処置部位とした。塗布終了24時間後に色彩色差計(CR-300、コニカミノルタ株式会社)にて各試験部位の皮膚明度(L*値)を測定し、無処置部位のL値に対するΔL*値を算出した。L*値は、色素沈着の程度が強いほど低い値となる。従って、ΔL*値が大きい程、色素沈着が抑制されたと判断することができる。結果を表6に示す。これにより、本発明の皮膚外用剤である化粧料(ローション1)は優れた色素沈着抑制効果を有することが分かる。これは、ローション1に含有される前記化合物1の色素沈着抑制作用によると考えられる。
Claims (13)
- 前記一般式(1)において、
R1が、無置換の又は炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有する芳香族基、
R2が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、tert-ブチル基、アセチル基、プロピオニル基又はブチリル基、
R3が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基又はtert-ブチル基、
であることを特徴とする、請求項1に記載の色素沈着予防又は改善剤。 - 前記一般式(1)において、
R1が、無置換の又は炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有するフェニル基、ナフチル基又はビフェニル基、
R2が、水素原子、メチル基又はアセチル基、
R3が、水素原子、メチル基又はエチル基、
であることを特徴とする、請求項2に記載の色素沈着予防又は改善剤。 - 前記一般式(1)に表される化合物が、N-ベンゾイル-セリン、N-(p-メチルベンゾイル)セリン、N-(p-エチルベンゾイル)セリン、N-(p-メトキシベンゾイル)セリン、N-(p-フルオロベンゾイル)セリン、N-(p-トリフルオロメチルベンゾイル)セリン、N-(2-ナフトイル)セリン、N-(4-フェニルベンゾイル)セリン、N-(p-メチルベンゾイル)セリン メチルエステル、N-(p-メチルベンゾイル)セリン エチルエステル、N-(2-ナフトイル)セリン メチルエステル、N-ベンゾイル-O-メチルセリン、N-(p-メチルベンゾイル)-O-メチルセリン、N-(p-メチルベンゾイル)-O-アセチルセリン、N-(2-ナフトイル)-O-メチルセリン、その異性体及び/又はそれらの薬理学的に許容される塩であることを特徴とする、請求項1~3の何れか一項に記載の色素沈着予防又は改善剤。
- 前記一般式(2)において、
R4が、無置換の又は炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有する芳香族基(ただし、無置換のフェニル基を除く)、
R5が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、tert-ブチル基、アセチル基、プロピオニル基又はブチリル基、
R6が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基又はtert-ブチル基であって、
R4が置換基を有するフェニル基又は無置換のナフチル基の場合、R5又はR6の少なくとも一つは水素原子以外である、
ことを特徴とする、請求項5に記載の色素沈着予防又は改善剤。 - 前記一般式(2)において、
R4が、炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有するフェニル基、無置換の又は前記の置換基を有するナフチル基又はビフェニル基、
R5が、水素原子、メチル基又はアセチル基、
R6が、水素原子、メチル基又はエチル基であって、
R4が置換基を有するフェニル基又は無置換のナフチル基の場合、R5又はR6の少なくとも一つは水素原子以外である、
ことを特徴とする、請求項6に記載の色素沈着予防又は改善剤。 - 請求項1~7のいずれか一項に記載の色素沈着予防又は改善剤を含有することを特徴とする、皮膚外用剤。
- 皮膚外用剤全量に対し、前記色素沈着予防又は改善剤を、0.0001質量%~20質量%含有することを特徴とする、請求項8に記載の皮膚外用剤。
- 化粧料(但し、医薬部外品を含む)であることを特徴とする、請求項8又は9に記載の皮膚外用剤。
- 前記一般式(2)において、
R4が、無置換の又は炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有する芳香族基(ただし、無置換のフェニル基を除く)、
R5が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、tert-ブチル基、アセチル基、プロピオニル基又はブチリル基、
R6が、水素原子、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基又はtert-ブチル基、
R4が置換基を有するフェニル基又は無置換のナフチル基の場合、R5又はR6の少なくとも一つは水素原子以外である、
ことを特徴とする、請求項11に記載の化合物。 - 前記一般式(2)において、
R4が、炭素数1~6の直鎖又は分岐のアルキル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルコキシ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアルキルアミノ基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するアシル基、炭素数1~6の直鎖又は分岐のアルキル鎖を有するエステル基、ハロゲン原子又はハロゲン化アルキル基、ヒドロキシ基若しくはアミノ基である置換基を有するフェニル基、無置換の又は前記の置換基を有するナフチル基又はビフェニル基を表し、
R5が、水素原子、メチル基又はアセチル基を表し、
R6が、水素原子、メチル基又はエチル基を表し、
R4が置換基を有するフェニル基又は無置換のナフチル基の場合、R5又はR6の少なくとも一つは水素原子以外である、
ことを特徴とする、請求項12に記載の化合物。
