Description
また、標識物質により標識化した抗体又は抗原を免疫反応により試料中の被検出物質に結合させ、この結合状態にある標識物質等を測定する方法も知られており、標識物質として放射性同位元素を用いる放射免疫測定法、酵素を用いる酵素免疫測定法、螢光物質を用いる螢光免疫測定法なども採用されている。
これらの免疫測定法では、被検出物質と標識物質により標識化した抗体等との反応工程、被検出物質と結合状態にある標識物質と結合状態にない標識物質との分離工程が必要となるが、これらの工程を、クロマトグラフィーの原理を応用して、固定相とそれに接して連続的に流れる移動相からなる系で行う方法として、イムノクロマトグラフ法が知られている。
(1)被検出物質である抗原に特異的に結合する抗体を固定化試薬とし、この固定化試薬をクロマトグラフ媒体の所定の部位に所定の形で塗布すること等により、クロマトグラフ媒体の任意の位置に反応部位を形成する。
(2)一方、被検出物質と特異的に結合する抗体を検出試薬とし、この検出試薬を酵素等の標識物質により標識する、又は、検出試薬を不溶性担体等の標識物質に感作することにより、標識化した検出試薬を調製する。
(3)移動相を構成する展開液を、被検出物質を含む試料及び標識化した検出試薬と共に、固定相であるクロマトグラフ媒体上を展開させる。
以上の操作により、クロマトグラフ媒体に形成された反応部位において、被検出物質である抗原が、反応部位に固定した固定化試薬である抗体と結合することにより捕捉されると共に、この抗原と、標識化した検出試薬である抗体とによって抗原-抗体反応が生ずる結果、当該反応部位においては固定化試薬(固定化した抗体)-被検出物質(抗原)-検出試薬(標識化した抗体)の三者のサンドイッチ型結合体が生成し、試料中に被検出物質が存在するときに反応部位に間接的に標識物質が結合することによって所定のシグナルが現れ、これによって被検出物質の検出を行うことができる。
また、不溶性担体を標識物質に用いる場合、酵素を標識物質に用いた場合に比べて、反応部位に検出されるシグナルが弱いという問題点もあった。酵素を標識物質に用いた場合には、反応部位において酵素反応により生じた不溶性物質が蓄積するため、陽性シグナルの増幅が見られ、比較的高感度な測定が可能となる。一方、不溶性担体を標識物質に用いた場合には、反応部位に捕捉された不溶性担体の数に応じた陽性シグナルしか得ることができず、特に、体積の小さい不溶性担体を用いた場合においてこの問題点が顕著であった。
また、本発明は、上記展開液が移動相を構成することを特徴とする、イムノクロマトグラフ法に関する。
さらに、本発明は、イムノクロマトグラフ法におけるクロマトグラフ媒体、及び前記した本発明の展開液を含んでなるイムノクロマトグラフ法による検出キットに関する。
また、本発明は、酸素原子含有極性基を有するビニル系水溶性ポリマー及び非イオン性界面活性剤を含む、イムノクロマトグラフ法における試料希釈液に関する。
(1)酸素原子及び窒素原子含有極性基を有するビニル系水溶性ポリマー及びHLB値が13~18である非イオン性界面活性剤を含む、不溶性担体で標識化された検出試薬を用いるイムノクロマトグラフ法における展開液。
(2)前記不溶性担体が、コロイド状金属粒子である、前記(1)に記載の展開液。
(3)前記コロイド状金属粒子が、コロイド状金粒子である、前記(2)に記載の展開液。
(4)移動相を構成する展開液として前記(1)~(3)のいずれかに記載の展開液を使用することを特徴とする、イムノクロマトグラフ法。
(5)イムノクロマトグラフ法におけるクロマトグラフ媒体、及び前記(1)~(3)のいずれかに記載の展開液を含んでなるイムノクロマトグラフ法による検出キット。
(6)酸素原子含有極性基を有するビニル系水溶性ポリマー及び非イオン性界面活性剤を含む、イムノクロマトグラフ法における試料希釈液。
(7)前記酸素原子含有極性基を有するビニル系水溶性ポリマーが、酸素原子及び窒素原子含有極性基を有するビニル系水溶性ポリマーである、前記(6)に記載の試料希釈液。
(8)非イオン性界面活性剤が、HLB値13~18である非イオン性界面活性剤である、前記(6)又は(7)に記載の試料希釈液。
また、本発明の展開液は展開液としてだけでなく、試料希釈液としても使用することができ、試料の希釈に用いた希釈液をそのままイムノクロマトグラフ法における展開液として使用することができる。特に、被検出物質がインフルエンザウイルス抗原である場合には、鼻腔又は咽頭拭い液等の試料希釈液としてだけでなく、ウイルス抗原の溶解性に優れた被検出物質を含む試料の溶解液として使用することができる。
クロマトグラフ媒体
本発明のイムノクロマトグラフ法において用いるクロマトグラフ媒体は、毛管現象を示す微細多孔性物質からなる不活性のものであって、使用される検出試薬、固定化試薬、被検出物質などと反応しないものであり、短時間での判定で十分な感度が得られる展開速度を有していれば、特にその素材が限定されるものではない。
本発明において、クロマトグラフ媒体としては、シリカ、チタニア、ジルコニア、セリア、アルミナ等のセラミック微粒子又は有機高分子の微粒子、ポリウレタン、ポリエステル、ポリエチレン、ポリ塩化ビニル、ポリフッ化ビニリデン、ナイロン、ニトロセルロース又は酢酸セルロース等のセルロース誘導体等で構成される繊維状又は不織繊維状マトリクス、膜、濾紙、ガラス繊維濾紙、布、綿等が挙げられる。微粒子はそれ自体が多孔性でなくとも、充填された状態では微粒子間に空隙が生じクロマトグラフ媒体として機能する。好ましくはセルロース誘導体やナイロンの膜、濾紙、ガラス繊維濾紙等であり、より好ましくはニトロセルロース膜、混合ニトロセルロースエステル(ニトロセルロースと酢酸セルロースの混合物)膜、ナイロン膜、濾紙である。
本発明において用いるクロマトグラフ媒体上には、被検出物質と特異的に結合する物質、例えば抗体が固定化試薬として任意の位置に固定化された反応部位が形成される。固定化試薬をクロマトグラフ媒体に固定化する方法としては、固定化試薬をクロマトグラフ媒体に物理的又は化学的手段により直接固定化する方法と、固定化試薬をラテックス粒子などの微粒子に物理的又は化学的に結合し、この微粒子をクロマトグラフ媒体に捕捉して固定化する間接固定化方法がある。
直接的に固定化する方法としては、物理吸着を利用しても良いし、共有結合によってもよい。一般にクロマトグラフ媒体がニトロセルロース膜又は混合ニトロセルロースエステル膜の場合、物理吸着を行うことができる。共有結合ではクロマトグラフ媒体の活性化には一般的に臭化シアン、グルタルアルデヒド、カルボジイミド等が用いられるが、いずれの方法も用いることができる。間接的に固定化する方法としては、不溶性微粒子に固定化試薬を結合した後に、クロマトグラフ媒体に固定化する方法がある。不溶性微粒子の粒径はクロマトグラフ媒体に捕捉されるが移動することのできないサイズのものを選択することができ、好ましくは平均粒径10μm程度以下の微粒子である。これらの粒子としては抗原抗体反応に使用されるものが種々知られており、本発明でもこれら公知の粒子を使用することができる。例えば、ポリスチレン、スチレン-ブタジエン共重合体、スチレン-メタクリル酸共重合体、ポリグリシジルメタクリレート、アクロレイン-エチレングリコールジメタクリレート共重合体などの乳化重合法によって得られる有機高分子ラテックス粒子などの有機高分子物質の微粒子、ゼラチン、ベントナイト、アガロース、架橋デキストランなどの微粒子、シリカ、シリカ-アルミナ、アルミナなどの無機酸化物や無機酸化物にシランカップリング処理などで官能基を導入した無機粒子等が挙げられる。本発明においては、感度調整の容易さ等から直接固定化の方が好ましい。また、クロマトグラフ媒体への固定化試薬の固定化には、いろいろな方法が使用できる。例えば、マイクロシリンジ、調節ポンプ付きペン、インキ噴射印刷等、種々の技術が使用可能である。反応部位の形態としては特に限定されないが、円形のスポット、クロマトグラフ媒体の展開方向に垂直にのびるライン、数字、文字や+、-などの記号等として固定化することもできる。
その他、クロマトグラフ媒体には、必要に応じて、被検出物質を含む試料を添加するための試料添加部位(サンプルパッド等)、試料中の血球等の固形成分を除去する部位(血球分離部位等)、展開液を添加するための展開液添加部位、反応部位に捕獲されなかった被検出物質や展開液を吸い取る吸収部位(吸収パッド等)、測定が正常に行われたことを示す対照部位等を組み入れてもよい。これらの部位の部材は、毛管現象により試料液や展開液が移動できれば特に限定されず、一般的には、ニトロセルロース膜、濾紙、ガラス繊維濾紙等の複数の多孔性物質からその目的に応じたものを選択して用い、固定化試薬が固定化されたクロマトグラフ媒体と毛管で繋がるように配置することができる。
本発明で用いられる検出試薬は、被検出物質と特異的に結合する物質、例えば抗体であり、標識物質により標識化される。イムノクロマトグラフ法における検出試薬の標識には、一般に酵素等も使用されるが、被検出物質の存在を目視で判定するのに適していることから、本発明の標識物質としては不溶性担体が用いられる。本発明においては、検出試薬を不溶性担体に感作することにより標識化した検出試薬を調製する。
本発明で用いられる標識物質としての不溶性担体には、金、銀、白金のようなコロイド状金属粒子、酸化鉄のようなコロイド状金属酸化物粒子、硫黄などのコロイド状非金属粒子及び合成高分子よりなるラテックス粒子、その他を用いることができる。
これらの不溶性担体に関しては、ラテックス粒子又はコロイド状金属粒子のいずれにおいても、その表面が負に荷電していることが知られている(例えば、特開平5-133956号公報参照)。例えば、コロイド状金属粒子では、その製造過程で添加される還元剤由来のアニオンがその表面に吸着しており、相互の凝集が妨げられて分散した状態を保っている。そして、この状態のコロイド状金属粒子に、表面電荷を中和しない程度の低濃度の界面活性剤を添加すると、粒子が鎖状に数個程度凝集することが知られている(特開2006-58781号公報)。このように、本発明のイムノクロマトグラフ法において、移動相を構成する展開液に酸素原子含有極性基を有するビニル系水溶性ポリマー及び非イオン性界面活性剤を添加すると、クロマトグラフ媒体上の反応部位に捕捉された不溶性担体が数個程度凝集することにより、反応部位で観察される陽性シグナルの増幅が見られると推測される。特に、コロイド状金属粒子においては、凝集によって反応部位に蓄積する粒子の数が増加することにより、目視判定されるシグナル量が増加するのみならず、粒子の光吸収スペクトル特性が変化することにより、反応部位においてより明瞭な陽性シグナルを得ることが可能となると推測される。このような利点を有することから、コロイド状貴金属粒子、特にコロイド状金粒子は、本発明の標識物質として好ましいものである。
コロイド状金属粒子として、例えばコロイド状金粒子を用いる場合には、市販のものを用いてもよい。あるいは、常法、例えば塩化金酸をクエン酸ナトリウムで還元する方法によりコロイド状金粒子を調製することができる。
本発明の方法により検出される被検出物質としては、それに特異的に結合する物質が存在するものであれば特に限定されず、蛋白質、ペプチド、核酸、糖(特に糖タンパク質の糖部分、糖脂質の糖部分等)、複合糖質などを例示することができる。本発明において「特異的に結合する」とは、生体分子が持つ親和力に基づいて結合することを意味する。このような親和力に基づく結合としては、抗原と抗体との結合、糖とレクチンとの結合、ホルモンと受容体との結合、酵素と阻害剤との結合、相補的核酸同士及び核酸と核酸結合蛋白質との結合などが挙げられる。従って、被検出物質が抗原性を有する場合、被検出物質に特異的に結合する物質としてはポリクローナル抗体又はモノクローナル抗体を例示することができる。また、被検出物質が糖の場合、被検出物質に特異的に結合する物質としてはレクチンタンパク質を例示することもできる。具体的な被検出物質としては、例えば、癌胎児性抗原(CEA)、HER2タンパク、前立腺特異抗原(PSA)、CA19-9、α-フェトプロテイン(AFP)、免疫抑制酸性タンパク(IPA)、CA15-3、CA125、エストロゲンレセプター、プロゲステロンレセプター、便潜血、トロポニンI、トロポニンT、CK-MB、CRP、ヒト絨毛性ゴナドトロピン(HCG)、黄体形成ホルモン(LH)、卵胞刺激ホルモン(FSH)、梅毒抗体、インフルエンザウイルス、ヒトヘモグロビン、クラミジア抗原、A群β溶連菌抗原、HBs抗体、HBs抗原、ロタウイルス、アデノウイルス、アルブミン、糖化アルブミン等が挙げられるが、これらに限定されるものではない。中でも非イオン性界面活性剤により可溶化される抗原が好ましく、ウイルス核タンパク質のように自己集合体を形成する抗原がより好ましい。
実際のイムノクロマトグラフ法の実施において、反応部位に対する試料の適用は、試料液をクロマトグラフ媒体上で展開させ、クロマトグラフ的に移動させることによって行うことができる。この場合に、試料液の展開は、移動相を構成する展開液の展開と同時に行ってもよいし、あるいは試料液を先行して展開し、その後に展開液を展開してもよい。
本発明で用いられる展開液は、イムノクロマトグラフ法において移動相を構成する液体であり、固定相であるクロマトグラフ媒体上を、被検出物質を含む試料及び標識化した検出試薬と共に移動する。
また、本発明の展開液は展開液としてだけでなく、試料希釈液としても使用することができ、試料の希釈に用いた希釈液をそのままイムノクロマトグラフ法における展開液として使用することができる。
本発明で用いられる展開液は、酸素原子含有極性基を有するビニル系水溶性ポリマーを含有する点に特徴を有する。この酸素原子含有極性基を有するビニル系水溶性ポリマーとしては、ビニルアルコール、ビニルメチルエーテル、(メタ)アクリル酸、ヒドロキシアルキル(メタ)アクリレート、(メタ)アクリルアミド、ジメチル(メタ)アクリルアミド、ビニルピロリドン等の酸素原子含有極性基を有する水溶性ビニル系モノマー、好ましくは酸素原子及び窒素原子含有極性基を有する水溶性ビニル系モノマー、より好ましくは酸素原子含有極性基を有する非イオン性水溶性ビニル系モノマー、さらに好ましくは酸素原子及び窒素原子含有極性基を有する非イオン性水溶性ビニル系モノマーの二重結合が開裂した構造単位を有するポリマーを挙げることができ、特にビニルピロリドンの二重結合が開裂した構造単位を有するポリマーが好ましい。
これらのビニル系水溶性ポリマーは、本発明の効果が損なわれない程度に、酢酸ビニル、アルキル(メタ)アクリレートなどの他のビニル系モノマーが、例えば50モル%以下、好ましくは30モル%以下、特に好ましくは15モル%以下の割合で共重合されたものであってもよい。
本発明で用いられる展開液は、上記酸素原子含有極性基を有するビニル系水溶性ポリマーに加え、非イオン性界面活性剤をさらに含有する。非イオン性界面活性剤としては、HLB値が10~18、さらに好ましくはHLB値が13~18のポリオキシエチレン系界面活性剤を好ましく用いることができる。ポリオキシエチレン系界面活性剤の好ましい例としては、ポリオキシエチレンアルキルエーテル、ポリオキシエチレンソルビタン脂肪酸エステル(商品名「Tween」シリーズ)、ポリオキシエチレンp-t-オクチルフェニルエーテル(商品名「Triton」シリーズ)、ポリオキシエチレンp-t-ノニルフェニルエーテル(商品名「TritonN」シリーズ)等を挙げることができ、より具体的には、「Tween」シリーズでは、特にTween20(商品名、HLB値16.7)、Tween40(商品名、HLB値15.6)、Tween60(商品名、HLB値15.0)、Tween80(商品名、HLB値14.9)、「Triton」シリーズでは、特にTriton X-100(商品名、HLB値13.5)、Nonidet P-40(商品名、HLB値13.1)、TritonX-102(商品名、HLB値14.6)、TritonX-165(商品名、HLB値15.8)、TritonX-405(商品名、HLB値17.9)、「TritonN」シリーズでは、特にTritonN-101(商品名、HLB値13.5)、TritonN-111(商品名、HLB値13.8)、TritonN-150(商品名、HLB値15.0)等を挙げることができる。非イオン性界面活性剤は、単独でも2種以上を混合して用いることもできる。上記した非イオン性界面活性剤の含有量は、特に限定されないが、組成物全体の重量に対し0.01~10重量%の範囲であり、好ましくは0.05~5重量%である。
25×2.5cmのニトロセルロース膜(ミリポア社製:HF120)に、抗体塗布機(BioDot社製)を用いて、5重量%のイソプロピルアルコールを含むリン酸緩衝液(pH7.4)で1.0mg/mLの濃度になるように希釈した抗インフルエンザBモノクローナル抗体を塗布し、50℃で30分間乾燥させた。乾燥後、該ニトロセルロース膜を、0.5重量%のカゼイン(和光純薬工業社製)を含むリン酸緩衝液(pH7.4)200mLに30℃で30分間浸潰し、ブロッキングを行った。ブロッキング後、0.05重量%のTween20を含有する洗浄液で洗浄し、室温で一晩乾燥させ、クロマトグラフ媒体上へ反応部位を作製した。
金コロイド懸濁液(田中貴金属工業社製:平均粒子径40nm)0.5mLに、リン酸緩衝(pH7.4)で0.1mg/mLの濃度になるように希釈した抗インフルエンザBモノクローナル抗体を0.1mL加え、室温で10分間静置した。次いで、10重量%の牛血清アルブミンを含むリン酸緩衝液(pH7.4)を0.1mL加え、十分撹絆した後、8000×gで15分間遠心分離を行った。上清を除去した後、1重量%の牛血清アルブミンを含むリン酸緩衝液(pH7.4)0.1mLを加え、標識物質溶液とした。
上記作製した標識物質溶液をグラスファイバー製パッドに均一になるように添加した後、真空乾燥機にて乾燥させ、検出試薬保持部材とした。次いで、バッキングシートから成る基材に、上記調製したクロマトグラフ媒体、検出試薬保持部材、試料を添加する部分に用いるサンプルパッド、及び展開した試料、不溶性担体を吸収するための吸収パッドを貼り合わせた。最後に、裁断機で幅が5mmとなるように裁断し、クロマトグラフ媒体を作製した。
上記作製したクロマトグラフ媒体を用いて、以下の方法で試料中のインフルエンザBウイルスの存在の有無を測定した。即ち、0.5重量%Tween20、0.6重量%ポリビニルピロリドン(PVP)K-90(平均分子量36万)、1.0重量%牛血清アルブミンと150mM塩化ナトリウムを含むトリス緩衝溶液(pH8.0)から成る展開液を陰性検体試料とし、ここに不活化処理したインフルエンザBウイルスを加えたものを陽性検体試料とし、各々150μLをクロマトグラフ媒体のサンプルパッド上に載せて展開させ、15分後に目視判定をした。反応部位におけるテストラインの赤い線を非常に明確に確認できるものを「+++」、赤い線を明確に確認できるものを「++」、赤い線を確認できるものを「+」、赤い線は確認できるが、非常に色が薄いものを「±」、赤い線を確認できないものを「-」とした。表1に結果を示す。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはTween20を含まないものを用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。PVP K-90量を2倍にしても同様の結果が得られた。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはPVP K-90を含まないものを用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。Tween20量を2倍にしても同様の結果が得られた。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはTween20及びPVP K-90を含まないものを用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはPVP K-90に代えて0.6重量%カルボキシメチルセルロースナトリウム(CMC・Na)(フルカ社製:viscosity 400-1000 mPa.s 2% in H2O,25℃)を用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはPVP K-90に代えて0.6重量%ポリエチレングリコール(PEG)(和光純薬工業社製、平均分子量2万)を、Tween20に代えて0.3重量%Triton X-100を用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。
実施例1で作製したクロマトグラフ媒体を用いて、展開液としてはTween20に代えて0.3重量%コール酸ナトリウムを用いたことを除いては、実施例1と同様に測定した。表1に結果を示す。

上記作製したクロマトグラフ媒体を用いて、以下の方法で試料中のヒトヘモグロビンの存在の有無を測定した。即ち、0.05重量%Tween20及び0.3重量%Triton X-100、0.6重量%ポリビニルピロリドン(PVP)K-90(平均分子量36万)、1.0重量%牛血清アルブミンと150mM塩化ナトリウムを含むトリス緩衝溶液(pH8.0)から成る展開液を陰性検体試料とし、ここにヒトヘモグロビンを加えたものを陽性検体試料とし、各々150μLをクロマトグラフ媒体のサンプルパッド上に載せて展開させ、15分後に目視判定をした。反応部位におけるテストラインの赤い線を非常に明確に確認できるものを「+++」、赤い線を明確に確認できるものを「++」、赤い線を確認できるものを「+」、赤い線は確認できるが、非常に色が薄いものを「±」、赤い線を確認できないものを「-」とした。表5に結果を示す。
Claims (8)
- 酸素原子及び窒素原子含有極性基を有するビニル系水溶性ポリマー及びHLB値が13~18である非イオン性界面活性剤を含む、不溶性担体で標識化された検出試薬を用いるイムノクロマトグラフ法における展開液。
- 前記不溶性担体が、コロイド状金属粒子である、請求項1に記載の展開液。
- 前記コロイド状金属粒子が、コロイド状金粒子である、請求項2に記載の展開液。
- 移動相を構成する展開液として請求項1~3のいずれかに記載の展開液を使用することを特徴とする、イムノクロマトグラフ法。
- イムノクロマトグラフ法におけるクロマトグラフ媒体、及び請求項1~3のいずれかに記載の展開液を含んでなるイムノクロマトグラフ法による検出キット。
- 酸素原子含有極性基を有するビニル系水溶性ポリマー及び非イオン性界面活性剤を含む、イムノクロマトグラフ法における試料希釈液。
- 前記酸素原子含有極性基を有するビニル系水溶性ポリマーが、酸素原子及び窒素原子含有極性基を有するビニル系水溶性ポリマーである、請求項9に記載の試料希釈液。
- 非イオン性界面活性剤が、HLB値13~18である非イオン性界面活性剤である、請求項9又は10に記載の試料希釈液。
