Description
本發明係由ε-N-(γ-麩胺醯基)離胺酸或其鹽所構成之育毛劑,及含ε-N-(γ-麩胺醯基)離胺酸或其鹽之育毛用組成物。進而係關於含有特定胺基酸之育毛用組成物。
脫毛症係由於遺傳因素、男性荷爾蒙的影響、毛囊血流減少及老化等身體因素,再加上壓力增加及生活型態改變等複雜交錯的原因而發生。
以往的育毛劑例如以十五酸甘油酯、腺核苷及米諾地爾等為主成分之育毛劑,主要係借重其於營養補給、血流促進、頭皮改善、細胞賦活化效果等。
十五酸甘油酯已知對能量代謝基礎低下之毛囊可做為有效率的能量供給物質,而促進毛髮生長(FRAGRANCE JOURNAL,No.80,1986,p.109-114)。但卻無法斷言其具有令人滿意的育毛效果。
已知腺核苷及米諾地爾(Minoxidil)係作用於毛乳頭細胞而促進其細胞生長因子的產生,具有使毛囊細胞活化的效果(參照J.Invest.Dermatol.,117,2001,p.1594-1600;FRAGRANCE JOURNAL,No.6,2005,p.13-18以及FRAGRANCE JOURNAL,No.3,2001,p.39-46)。但卻無法斷言其具有令人滿意的育毛效果。
另一方面,已知有含ε-N-(γ-麩胺醯基)離胺酸或其鹽之化粧料(特開平5-301814號公報)。但其使用目的為“美膚效果”。
以往的育毛劑及含其之有效成分並未具有充分地令人滿意的育毛效果,而深切企望具新穎作用機制之新穎育毛用組成物。
本發明的課題係提供具新穎作用機制之新穎育毛用組成物。
本發明之發明者們專心檢討結果,發現ε-N-(γ-麩胺醯基)離胺酸或其鹽,可藉由使毛乳頭細胞及人類角質細胞增殖而具有育毛效果,遂完成本發明。另外,發現本發明之育毛劑藉由併用特定的胺基酸,可使人類角質細胞增殖,因而具有更佳之育毛效果。
亦即,本發明係包含以下之實施方式。
〔1〕一種育毛劑,其係由ε-N-(γ-麩胺醯基)離胺酸或其鹽所構成。
〔2〕一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽。
〔3〕一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種以上選自該胺基酸第一群之胺基酸或其鹽,胺基酸第一群:麩胺醯胺、白胺酸。
〔4〕一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種以上選自該胺基酸第一群之胺基酸或其鹽,以及1種或2種以上選自該胺基酸第二群之胺基酸或其鹽,胺基酸第一群:麩胺醯胺、白胺酸,胺基酸第二群:離胺酸、異白胺酸、天門冬胺酸、精胺酸、麩胺酸。
〔5〕如前述〔3〕或〔4〕之育毛用組成物,其中胺基酸第一群(B成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(B/A)為0.015~700。
〔6〕如前述〔4〕或〔5〕之育毛用組成物,其中胺基酸第二群(C成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(C/A)為0.0005~200。
〔7〕一種為進行育毛,ε-N-(γ-麩胺醯基)離胺酸或其鹽之使用。
〔8〕一種育毛方法,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽之組成物塗佈於皮膚。
〔9〕一種育毛方法,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種以上選自該胺基酸第一群之胺基酸或其鹽之組成物塗佈於皮膚,胺基酸第一群:麩胺醯胺、白胺酸。
〔10〕一種育毛方法,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,1種或2種以上選自該胺基酸第一群之胺基酸或其鹽,以及1種或2種以上選自該胺基酸第二群之胺基酸或其鹽之組成物塗佈於皮膚,胺基酸第一群:麩胺醯胺、白胺酸,胺基酸第二群:離胺酸、異白胺酸、天門冬胺酸、精胺酸、麩胺酸。
〔11〕如前述〔9〕或〔10〕之育毛方法,其中胺基酸第一群(B成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(B/A)為0.015~700。
〔12〕如前述〔10〕或〔11〕之育毛方法,其中胺基酸第二群(C成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(C/A)為0.0005~200。
於本說明書中,「A成分之質量」係指麩胺醯基離胺酸之質量,使用麩胺醯基離胺酸鹽為A成分時,可適宜地換算再使用。
相同地,於本說明書中,「B成分之質量」以及「C成分之質量」係指做為B成分以及C成分時所含胺基酸之游離體之質量,使用其鹽時,可適宜地換算再使用。
根據本發明,可提供促進包含毛母質細胞、毛乳頭細胞及毛囊細胞等增殖之育毛用組成物。
針對本發明之育毛用組成物以下依序加以說明。
本發明所使用之ε-N-(γ-麩胺醯基)離胺酸(以下略稱為麩胺醯基離胺酸),係具有下述構造式(1)之構造之二胜肽(異胜肽)。
關於獲得麩胺醯基離胺酸之方法,可使用記載於Biochemistry 2卷740-745頁(1963)或特開昭51-105014號公報之方法等。
以如上所述方式所得之麩胺醯基離胺酸,係無味白色粉末,難溶或不溶於甲醇、乙醇、丙酮等各種有機溶劑中,但可溶解於鹼性水或酸性水等中。
前述麩胺醯基離胺酸可以鹽類的形式使用。藉由以鹽類的形式使用麩胺醯基離胺酸,不僅前述之鹼性水或酸性水,亦可溶解於中性水中。
相關鹽類的型態,可舉出鉀、鈉等鹼金屬鹽,鈣、鎂等鹼土類金屬鹽,銨、三乙醇銨等有機銨鹽,離胺酸、精胺酸等鹽基性胺基酸鹽,及鹽酸、硫酸等礦酸鹽,醋酸、檸檬酸等有機酸鹽等。考慮溶解性、安全性等問題時,以鹼金屬鹽、鹼土類金屬鹽、鹽基性胺基酸鹽為佳,鹼金屬鹽、鹼土類金屬鹽更佳,鹼金屬鹽特佳。
以下將ε-N-(γ-麩胺醯基)離胺酸或其鹽記載為A成分。
併用ε-N-(γ-麩胺醯基)離胺酸或其鹽,及1種或2種以上選自胺基酸第一群之胺基酸或其鹽時,可不阻礙細胞正常的增殖,而使麩胺醯基離胺酸單獨的細胞增殖效果增強。
以下將胺基酸第一群記載為B成分。胺基酸第一群係麩胺醯胺或白胺酸。這些胺基酸可為游離體或鹽類之狀態。可使用左旋體、右旋體任一種光學異構體,亦可使用消旋性體(DL體)。可僅使用1種,或可混合2種以上而使用。
使用組成物時,並未限制本發明之B成分之混合量,但以相對於A成分之質量B成分之質量(B/A)為0.015~700為佳。較0.015為小時,會有細胞增殖率不如預期為高的情形出現,較700為大時,反而會造成阻礙細胞增殖率。以可有效增強細胞增殖率之觀點,下限值以0.1為佳,0.2更佳,2更為佳,4進而更佳,4.8則特佳。而以不成為阻礙細胞增殖之主要因素之觀點,上限值以300為佳,250更佳,125更為佳,60進而更佳,35則特佳。
特別以不阻礙細胞正常的增殖,而可使麩胺醯基離胺酸單獨的效果增強之觀點,以合併使用麩胺醯胺或其鹽,及白胺酸或其鹽為佳。此時麩胺醯胺或其鹽,及白胺酸或其鹽之混合比例,以質量比1:2~250:1為佳,3:1~60:1更佳,5:1~25:1最佳。
進而,併用1種或2種以上選自胺基酸第二群之胺基酸或其鹽時,可使細胞增殖效果更增強。以下將胺基酸第二群記載為C成分。胺基酸第二群係離胺酸、異白胺酸、天門冬胺酸、精胺酸、麩胺酸。這些胺基酸可為游離體或鹽類之狀態。可使用左旋體、右旋體任一種光學異構體,亦可使用消旋性體(DL體)。可僅使用1種,或可混合2種以上而使用。特別自具有顯著細胞增殖率之觀點,以離胺酸、異白胺酸為佳,離胺酸特佳。
並未限制本發明之C成分之混合量,但以相對於A成分之質量C成分之質量(C/A)為0.0005~200為佳。較0.0005為小時,無法對細胞增殖效果產生影響,較200為大時,會造成阻礙細胞增殖率。以可使細胞賦活化之觀點,下限值以0.001為佳,0.01更佳,0.015更為佳,0.03進而更佳,0.07則特佳。而以不阻礙細胞增殖率之觀點,上限值以160為佳,120更佳,60更為佳,30進而更佳,15則特佳。
本發明之育毛劑,係可藉由使其溶解於溶劑,而以育毛用組成物方式而使用。所使用之溶劑具體可使用乙醇、異丙醇等低碳醇類;乙二醇、丙烯乙二醇、丁烯乙二醇等多價醇;水等。所使用之溶劑量可因應育毛用組成物之組成而適宜地加以選擇。
育毛用組成物中A成分之含量以0.0001~10質量%之範圍為佳。未達0.0001質量%時會出現缺乏育毛效果之情形,超過10質量%時,增加效果並未如實質上期望,且會因為析出A成分而使毛孔阻塞。自可有效發揮育毛效果之觀點,下限值以0.0005質量%為佳,0.0007質量%更佳,0.001質量%特佳。自不會阻塞毛孔且可發揮效果之觀點,上限值以5質量%為佳,2質量%更佳,0.1質量%特佳。
可於本發明之育毛用組成物中,以不阻礙本發明效果的程度,適宜地混合使用一般用於醫藥品、化妝品等之各種成分。具體而言可混合使用水性成分、油性成分、粉末成分、界面活性劑、保濕劑、增黏劑、顏色劑、香料、抗氧化劑、PH調整劑、螯合劑、防腐劑或防止紫外線劑、抗發炎劑、美白劑、抗老化劑、血流促進劑、防止白髮劑等。
本發明之育毛用組成物之型態並無特別限制,可舉出具體例為液狀、膏狀、凝膠狀、泡沫狀、乳霜狀、固狀等,但自可更佳發揮育毛效果之觀點而言,以液狀、凝膠狀為佳。
本發明之育毛效果的試驗方法,係如記載於「FRAGRANCE JOURNAL,臨時增刊No.13,1994,p.105-106」般,採用測定毛乳頭細胞及毛母質細胞之細胞增殖率之試驗方法。特別本發明之試驗方法,係利用毛乳頭細胞及人類角質細胞(毛母質細胞之代替品),於可簡便且迅速地試驗育毛效果之觀點,可謂係一極為適用之試驗方法。特別人類角質細胞增殖試驗(使用不含胺基酸培養液)此一試驗方法,係使完全不存在做為營養源之胺基酸,為極為適合能夠觀察處於衰退期頭皮之細胞增殖情形的簡便方法之-種試驗方法。
以下藉由實施例詳細說明本發明,但本發明並未因為下述之實施例而有所限定。
說明實驗方法。
(1)使用增殖用培養液PCGM:Papilla Cell Growth Medium(購自東洋紡,TPGM-250),於96孔滴定盤種入細胞密度為1.0*104
cells/well之毛乳頭細胞。
(2)於37℃,5%二氧化碳(CO2
)環境下培養1天後,移除培養液,再以用於細胞增殖試驗評價用之培養液,即含1%FCS(牛胎兒血清)之DMEM(Dulbecco’s Modified Eagle’s Medium)培養液進行洗淨。其後,將各種試驗成分溶解於相同之培養液,添加成為各種濃度(μmol/l)。
(3)接著,於37℃,5%二氧化碳(CO2
)環境下再培養2天後,以MTT法測定毛乳頭細胞之增殖率。
(4)將未加入各種育毛成分,僅含有培養液者定為100,計算出相對評價值,以其為增殖率。
(1)使用添加了增殖添加劑HuMedia-KG(購自KURABO,KK-6150)之增殖用無血清培養液HuMedia-KB2(購自KURABO,KK-2350S),於96孔滴定盤種入細胞密度為2.0*104
cells/well之人類角質細胞。
(2)於37℃,5%二氧化碳(CO2
)環境下培養1天後,移除培養液,再以用於細胞增殖試驗評價用之培養液,即MCDB153培養液進行洗淨。其後,將各種試驗成分溶解於相同之培養液,添加成為各種濃度(μmol/l)。
(3)接著,於37℃,5%二氧化碳(CO2
)環境下再培養2天後,以MTT法測定人類角質細胞之增殖率。(4)將未加入各種育毛成分,僅含有培養液者定為100,計算出相對評價值,以其為增殖率。
(1)使用添加了增殖添加劑HuMedia-KG(購自KURABO,KK-6150)之增殖用無血清培養液HuMedia-KB2(購自KURABO,KK-2350S),於96孔滴定盤種入細胞密度為2.0*104

)環境下培養1天後,移除培養液,再以用於細胞增殖試驗評價用之培養液,即去除全部胺基酸之MCDB153培養液(不含胺基酸培養液)進行洗淨。其後,溶解於相同之評價用培養液,添加如表2所示之各種育毛用組成物。
(3)接著,於37℃,5%二氧化碳(CO2
)環境下再培養2天後,以MTT法測定人類角質細胞之增殖率。
(4)將未加入各種胺基酸以及各種育毛成分,僅含有培養液者定為100,計算出相對評價值,以其為增殖率。
於表1所示針對化合物於各種濃度,進行人類角質細胞增殖試驗及毛乳頭細胞增殖試驗。
自毛乳頭細胞及人類角質細胞增殖之觀點,可明顯得知麩胺醯基離胺酸較周知的育毛成分之腺核苷及米諾地爾,具有相同或是超過之增殖效果。
調製實施例2-14之組成物,進行人類角質細胞增殖試驗(不含胺基酸培養液)。
本發明育毛劑之麩胺醯基離胺酸,係藉由添加胺基酸第一群,進而增大人類角質細胞之增殖效果。加之,可明顯得知藉由添加胺基酸第二群,進而增大人類角質細胞之增殖效果。
以下表3所示組成,製作育毛乳液。
本發明遂達成可提供具新穎作用機制之育毛劑以及育毛用組成物。
Claims (11)
- 一種育毛劑,其係由ε-N-(γ-麩胺醯基)離胺酸或其鹽所構成。
- 一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽。
- 一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種選自下述胺基酸第一群之胺基酸或其鹽,胺基酸第一群:麩胺醯胺、白胺酸。
- 一種育毛用組成物,其係含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種選自下述胺基酸第一群之胺基酸或其鹽,以及1種或2種以上選自下述胺基酸第二群之胺基酸或其鹽,胺基酸第一群:麩胺醯胺、白胺酸,胺基酸第二群:離胺酸、異白胺酸、天門冬胺酸、精胺酸、麩胺酸。
- 如申請專利範圍第3或4項之育毛用組成物,其中胺基酸第一群(B成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(B/A)為0.015~700。
- 如申請專利範圍第5項之育毛用組成物,其中胺基酸第二群(C成分)相對於ε-N-(γ-麩胺醯基)離 胺酸或其鹽(A成分)之質量之質量比(C/A)為0.0005~200。
- 一種用途,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽之組成物使用於製造塗佈於皮膚的育毛用組成物。
- 一種用途,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽,以及1種或2種選自下述胺基酸第一群之胺基酸或其鹽之組成物使用於製造塗佈於皮膚的育毛用組成物;胺基酸第一群:麩胺醯胺、白胺酸。
- 一種用途,其係將含有ε-N-(γ-麩胺醯基)離胺酸或其鹽、1種或2種選自下述胺基酸第一群之胺基酸或其鹽、以及1種或2種以上選自下述胺基酸第二群之胺基酸或其鹽之組成物使用於製造塗佈於皮膚的育毛用組成物;胺基酸第一群:麩胺醯胺、白胺酸,胺基酸第二群:離胺酸、異白胺酸、天門冬胺酸、精胺酸、麩胺酸。
- 如申請專利範圍第8或9項之用途,其中胺基酸第一群(B成分)相對於ε-N-(γ-麩胺醯基)離胺酸或其鹽(A成分)之質量之質量比(B/A)為0.015~700。
- 如申請專利範圍第10項之用途,其中胺基酸第二群(C成分)相對於ε-N-(γ-麩胺醯基)離胺酸或 其鹽(A成分)之質量之質量比(C/A)為0.0005~200。
