Claims (67)
1. Очищенный полипептид PtIP-83, имеющий по меньшей мере 80% идентичность последовательности по всей длине аминокислотной последовательности, любой из:
(a) PtIP-83Fa представленной на Фигурах 2a-2j;
(b) PtIP-83Ca представленной на Фигурах 2a-2j;
(c) PtIP-83Cb представленной на Фигурах 2a-2j;
(d) PtIP-83Cc представленной на Фигурах 2a-2j;
(e) PtIP-83Cd представленной на Фигурах 2a-2j;
(f) PtIP-83Ce представленной на Фигурах 2a-2j;
(g) PtIP-83Cf представленной на Фигурах 2a-2j;
(h) PtIP-83Cg представленной на Фигурах 5a-5e; и
(i) PtIP-83Da представленной на Фигурах 5a-5e;
где полипептид PtIP-83 обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
2. Очищенный полипептид PtIP-83 по п.1, где полипептид PtIP-83 имеет по меньшей мере 90% идентичность последовательности по всей длине аминокислотной последовательности, любой из:
(a) PtIP-83Fa представленной на Фигурах 2a-2j;
(b) PtIP-83Ca представленной на Фигурах 2a-2j;
(c) PtIP-83Cb представленной на Фигурах 2a-2j;
(d) PtIP-83Cc представленной на Фигурах 2a-2j;
(e) PtIP-83Cd представленной на Фигурах 2a-2j;
(f) PtIP-83Ce представленной на Фигурах 2a-2j;
(g) PtIP-83Cf представленной на Фигурах 2a-2j;
(h) PtIP-83Cg представленной на Фигурах 5a-5e; и
(i) PtIP-83Da представленной на Фигурах 5a-5e.
3. Очищенный полипептид PtIP-83 по п.1, где полипептид PtIP-83 имеет по меньшей мере 95% идентичность последовательности по всей длине аминокислотной последовательности, любой из:
(a) PtIP-83Fa представленной на Фигурах 2a-2j;
(b) PtIP-83Ca представленной на Фигурах 2a-2j;
(c) PtIP-83Cb представленной на Фигурах 2a-2j;
(d) PtIP-83Cc представленной на Фигурах 2a-2j;
(e) PtIP-83Cd представленной на Фигурах 2a-2j;
(f) PtIP-83Ce представленной на Фигурах 2a-2j;
(g) PtIP-83Cf представленной на Фигурах 2a-2j;
(h) PtIP-83Cg представленной на Фигурах 5a-5e; и
(i) PtIP-83Da представленной на Фигурах 5a-5e.
4. Очищенный полипептид PtIP-83 по п.1, где полипептид PtIP-83 имеет по меньшей мере одну аминокислотную замену, делецию, вставку и/или добавление на N-конце или C-конце по сравнению с любой из:
(a) PtIP-83Fa представленной на Фигурах 2a-2j;
(b) PtIP-83Ca представленной на Фигурах 2a-2j;
(c) PtIP-83Cb представленной на Фигурах 2a-2j;
(d) PtIP-83Cc представленной на Фигурах 2a-2j;
(e) PtIP-83Cd представленной на Фигурах 2a-2j;
(f) PtIP-83Ce представленной на Фигурах 2a-2j;
(g) PtIP-83Cf представленной на Фигурах 2a-2j;
(h) PtIP-83Cg представленной на Фигурах 5a-5e; и
(i) PtIP-83Da представленной на Фигурах 5a-5e.
5. Очищенный полипептид PtIP-83 по п. 1, где полипептид PtIP-83 представляет собой не встречающуюся в природе аминокислотную последовательность.
6. Очищенный полипептид PtIP-83 по любому из пп. 1-5, где полипептид PtIP-83 специфически связывается с мембранной везикулой щеточной каемки, выделенной от Lepidoptera.
7. Очищенный полипептид PtIP-83 по любому из пп. 1-5, где полипептид PtIP-83 нарушает ионный баланс по сторонам мембранной везикулы щеточной каемки, выделенной от Lepidoptera.
8. Очищенный полипептид PtIP-83 по любому из пп. 1-3, где полипептид PtIP-83 получен из вида папоротника или примитивного растения отдела Lycophyta.
9. Очищенный полипептид PtIP-83 по п. 8, где папоротник относится к роду Lygodium.
10. Очищенный полипептид PtIP-83 по п. 8, где папоротник относится к роду Adiantaceae.
11. Очищенный полипептид PtIP-83 по п. 8, где папоротник относится к роду Polypodium.
12. Рекомбинантный полинуклеотид, кодирующий полипептид PtIP-83 по любому из пп. 1-11.
13. Рекомбинантный полинуклеотид по п. 12, где полинуклеотид представляет собой полинуклеотид, отличающийся от геномного.
14. Рекомбинантный полинуклеотид по п. 13, где полинуклеотид представляет собой кДНК.
15. Рекомбинантный полинуклеотид по п. 13, где полинуклеотид представляет собой синтетический полинуклеотид.
16. Рекомбинантный полинуклеотид по п. 13, где полинуклеотид имеет кодоны, оптимизированные для экспрессии в культуре, важной с точки зрения сельского хозяйства.
17. Трансгенное растение, содержащее полинуклеотид по любому из пп. 12-16, где трансгенное растение обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
18. ДНК-конструкция, содержащая полинуклеотид по любому из пп. 12-16, функционально связанный с гетерологичным регулятным элементом, где ДНК-конструкция придает инсектицидную активность против совки кукурузной (Helicoverpa zea).
19. Трансгенное растение, содержащая ДНК-конструкцию по п. 18, где трансгенное растение обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
20. Трансгенная растительная клетка, содержащая ДНК-конструкцию по п. 18, где трансгенная растительная клетка обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
21. Композиция для применения в сельском хозяйстве, содержащая инсектицидно эффективную дозу полипептида PtIP-83 по любому из пп. 1-11, где композиция для применения в сельском хозяйстве обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
22. Композиция для применения в сельском хозяйстве по п. 21, дополнительно содержащая по меньшей мере один приемлемый с точки зрения сельского хозяйства носитель.
23. Белок слияния, содержащий полипептид PtIP-83 по любому из пп. 1-11, где белок слияния обладает инсектицидной активностью против совки кукурузной (Helicoverpa zea).
24. Способ снижения популяции совки кукурузной (Helicoverpa zea), включающий приведение в контакт популяции совки кукурузной (Helicoverpa zea) с полипептидом PtIP-83 по любому из пп. 1-11.
25. Способ подавления роста или уничтожения совки кукурузной (Helicoverpa zea), включающий приведение в контакт совки кукурузной (Helicoverpa zea) с композицией, содержащей полипептид PtIP-83 по любому из пп. 1-11.
26. Способ снижения заражения совкой кукурузной (Helicoverpa zea) трансгенного растения и обеспечения управления устойчивостью совки кукурузной (Helicoverpa zea) насекомых, включающий экспрессию в растении полипептида PtIP-83 по любому из пп. 1-11.
27. Способ снижения популяции совки кукурузной (Helicoverpa zea), включающий приведение в контакт популяции совки кукурузной (Helicoverpa zea) с трансгенным растением по п. 19 или трансгенной растительной клеткой по п. 20.
28. Способ подавления роста или уничтожения совки кукурузной (Helicoverpa zea), включающий приведение в контакт совки кукурузной (Helicoverpa zea) с трансгенным растением по п. 19 или трансгенной растительной клеткой по п. 20.
29. Способ по любому из пп. 24-28, где совка кукурузная (Helicoverpa zea) или популяция совки кукурузной (Helicoverpa zea) устойчивы по меньшей мере к одному Bt-токсину.
30. Применение полипептида PtIP-83 по любому из пп. 1-11 для ингибирования роста или уничтожения совки кукурузной (Helicoverpa zea) или популяции совки кукурузной (Helicoverpa zea).
