Claims (39)
1. Стабильный фармацевтический состав, содержащий фармацевтически приемлемую соль терапевтического средства, имеющего низкую растворимость в воде, и одно или несколько средств для приготовления состава, при этом фармацевтически приемлемая соль является моновалентной или дивалентной солью, и одно или несколько средств для приготовления состава включают комплексообразующий агент, солюбилизатор и/или буферное средство; при этом соль терапевтического средства находится в составе в растворе.
2. Состав по п.1, в котором фармацевтически приемлемой солью терапевтического средства является соль пазопаниба.
3. Состав по п.2, в котором фармацевтически приемлемая соль является моновалентной или дивалентной галоидной солью пазопаниба.
4. Состав по п.3, в котором моновалентная или дивалентная соль является хлоридной солью.
5. Состав по п.4, в котором моновалентная соль стабильна в составе вплоть до концентрации примерно 60 мг/мл.
6. Состав по п.4, в котором дивалентная соль стабильна в составе вплоть до концентрации примерно 70 мг/мл.
7. Состав по п.4, в котором моновалентная соль является лиофилизированной моновалентной солью терапевтического средства.
8. Состав по п.7, в котором моновалентную соль подвергают лиофилизации с использованием диметилсульфоксида (ДМСО), трифторэтанола (TFE) или смеси трифторэтанол-вода.
9. Состав по п.1, в котором комплексообразующим агентом является циклодекстрин, выбранный из группы, состоящей из: 2-гидроксипропил-β-циклодекстрина, метил-β-циклодекстрина, случайным образом метилированного β-циклодекстрина, этилированного β-циклодекстрина, триацетил-β-циклодекстрина, перацетилированнного β-циклодекстрина, карбоксиметил-β-циклодекстрина, гидроксиэтил-β-циклодекстрина, 2-гидрокси-3-(триметиламмоний)пропил-β-циклодекстрина, гликозил-β-циклодекстрина, мальтозил-β-циклодекстрина, сульфобутилового эфира β-циклодекстрина, разветвленного β-циклодекстрина, гидроксипропил-γ-циклодекстрина, случайным образом метилированного γ-циклодекстрина, триметил-γ-циклодекстрина и любого их сочетания(ий).
10. Состав по п.1, в котором солюбилизатором является поли(винилпирролидон) (ПВП).
11. Состав по п.1, в котором буферным средством является гистидин-HCl.
12. Способ получения стабильного фармацевтического состава в виде раствора фармацевтически приемлемой соли терапевтического средства, имеющего низкую растворимость в воде, в котором соль является моновалентной солью, при этом способ включает (a) получение органического раствора соли в органическом растворителе; (b) лиофилизацию органического раствора с получением при этом лиофилизированной соли терапевтического средства; (c) растворение солюбилизатора и буферного средства в воде с получением при этом раствора; (d) растворение комплексообразующего агента в растворе с получением при этом раствора с низкой вязкостью; и (e) добавление лиофилизированной соли к раствору и перемешивание для растворения моновалентной соли в растворе при температуре, равной или выше чем примерно температура окружающей среды, с получением при этом стабильного фармацевтического состава в виде раствора.
13. Способ по п.12, в котором способ дополнительно включает добавление и растворение примерно в 2 раза большего количества комплексообразующего агента после стадии (e).
14. Способ по п.12, в котором органическим растворителем является диметилсульфоксид (ДМСО), трифторэтанол (TFE) или смесь трифторэтанол-вода.
15. Способ по п.14, в котором лиофилизация в ДМСО превращает одну кристаллическую форму соли терапевтического средства в другую кристаллическую форму.
16. Способ по п.14, в котором лиофилизация в ДМСО превращает форму A кристаллической фазы соли терапевтического средства в вещество, содержащее, по меньшей мере, 70% формы G кристаллической фазы, как определено с использованием порошковой рентгеновской дифракции.
17. Способ по п.14, в котором лиофилизация в TEE превращает форму A кристаллической фазы соли терапевтического средства в частично или полностью аморфную фазу.
18. Способ по п.12, в котором pH корректируют во время приготовления стабильного фармацевтического состава в виде раствора.
19. Способ по п.12, в котором pH не корректируют во время приготовления стабильного фармацевтического состава в виде раствора.
20. Способ по п.12, в котором солюбилизатором является поли(винилпирролидон) (ПВП).

23. Способ по п.12, в котором фармацевтически приемлемой солью терапевтического средства является соль пазопаниба.
24. Способ по п.12, в котором моновалентная соль является галоидом.
25. Способ по п. 12, в котором галоид является хлоридом.
26. Способ по п.12, в котором лиофилизированную соль растворяют в растворе на стадии (e) при температуре примерно от 37°C до примерно 50°C.
27. Способ по п.13, в котором способ дополнительно включает непрерывное перемешивание, по меньшей мере, солюбилизатора, буферного средства, комплексообразующего агента и лиофилизированной соли по мере добавления воды при температуре, равной или выше, чем примерно температура окружающей среды.
28. Способ по п.27, в котором pH состава корректируют до примерно 6-7, используя основание.
29. Способ лечения и/или ослабления глазного заболевания или расстройства заднего сегмента глаза, при этом способ включает доставку стабильного фармацевтического состава фармацевтически приемлемой соли терапевтического средства, имеющего низкую растворимость в воде, и одного или нескольких средств для приготовления состава из устройства для интравитреальной доставки, содержащего камеру емкости, соединенную с пористой структурой, при этом состав находится в емкости устройства, и контролируемое высвобождение состава из емкости через пористую структуру увеличивает время полужизни терапевтического средства в стекловидном теле;
при этом фармацевтически приемлемая соль является моновалентной или дивалентной солью, и одно или несколько средств для приготовления состава включают комплексообразующий агент, солюбилизатор и буферное средство; при этом соль терапевтического средства находится в составе в растворе.
30. Способ по п.29, в котором заболевание или расстройство выбрано из диабетической ретинопатии, возрастной макулярной дегенерации (ВМД), патологической хороидальной неоваскуляризации (CNV), патологической неоваскуляризации сетчатки, увеита, окклюзии вены сетчатки, травмы глаза, вызванного хирургической операцией отека, индуцированной хирургической операцией неоваскуляризации, кистозного макулярного отека, ишемии глаза, ретинопатии недоношенных, болезни Коутса, серповидно-клеточной ретинопатии и неоваскулярной глаукомы.
31. Способ по п.29, в котором камера емкости является повторно заполняемой, и ее повторно заполняют составом после введения устройства в глаз.
32. Способ по п. 31, в котором камеру емкости повторно заполняют составом после того, как устройство находилось в глазу в течение 30-90 суток или вплоть до 6 месяцев.
33. Способ по п.29, в котором солюбилизатором является поли(винилпирролидон) (ПВП).

Способ по п. 29, в котором комплексообразующим агентом является циклодекстрин, выбранный из группы, состоящей из: 2-гидроксипропил-β-циклодекстрина, метил-β-циклодекстрина, случайным образом метилированного β-циклодекстрина, этилированного β-циклодекстрина, триацетил-β-циклодекстрина, перацетилированнного β-циклодекстрина, карбоксиметил-β-циклодекстрина, гидроксиэтил-β-циклодекстрина, 2-гидрокси-3-(триметиламмоний)пропил-β-циклодекстрина, гликозил-β-циклодекстрина, мальтозил-β-циклодекстрина, сульфобутилового эфира β-циклодекстрина, разветвленного β-циклодекстрина, гидроксипропил-γ-циклодекстрина, случайным образом метилированного γ-циклодекстрина, триметил-γ-циклодекстрина и любого их сочетания(ий).
36. Способ по п.29, в котором фармацевтически приемлемой солью терапевтического средства является соль пазопаниба.
37. Способ по п.29, в котором моновалентная соль является галоидом.
38. Способ по п.37, в котором галоид является хлоридом.
