Abstract
1. Способ синтеза мультиспецифического антитела, где проводят реакцию первого фрагмента антитела, полученного из первого исходного антитела, обладающего первой моноспецифичностью и свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, полученным из второго исходного антитела, обладающего второй моноспецифичностью и свободной сульфгидрильной группой, для получения мультиспецифического антитела, и где первая моноспецифичность отличается от второй моноспецифичности.2. Способ синтеза аналога антитела, где проводят реакцию первого фрагмента антитела, обладающего свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, обладающим свободной сульфгидрильной группой, для получения аналога антитела, и где первый фрагмент антитела и второй фрагмент антитела получены из одного исходного антитела.3. Способ синтеза панели мультиспецифических антител, где проводят реакцию первого фрагмента антитела, полученного из первого исходного антитела, обладающего первой моноспецифичностью и свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер попарно с каждым из двух или более дополнительных фрагментов антител, полученных из одного или нескольких исходных антител с моноспецифичность
Claims (23)
1. Способ синтеза мультиспецифического антитела, где проводят реакцию первого фрагмента антитела, полученного из первого исходного антитела, обладающего первой моноспецифичностью и свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, полученным из второго исходного антитела, обладающего второй моноспецифичностью и свободной сульфгидрильной группой, для получения мультиспецифического антитела, и где первая моноспецифичность отличается от второй моноспецифичности.
2. Способ синтеза аналога антитела, где проводят реакцию первого фрагмента антитела, обладающего свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, обладающим свободной сульфгидрильной группой, для получения аналога антитела, и где первый фрагмент антитела и второй фрагмент антитела получены из одного исходного антитела.
3. Способ синтеза панели мультиспецифических антител, где проводят реакцию первого фрагмента антитела, полученного из первого исходного антитела, обладающего первой моноспецифичностью и свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер попарно с каждым из двух или более дополнительных фрагментов антител, полученных из одного или нескольких исходных антител с моноспецифичностью, отличной от первого фрагмента антитела, каждый из которых имеет свободную сульфгидрильную группу, для получения панели мультиспецифических антител.
4. Способ синтеза панели аналогов антител, где проводят реакцию первого фрагмента антитела, обладающего свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и где проводят реакцию группы фрагмент антитела-кросслинкер попарно с каждым из двух или более дополнительных фрагментов антител, каждый из которых имеет свободную сульфгидрильную группу, для получения панели аналогов антител, где каждый из фрагментов антител получен из одного и того же исходного антитела.
5. Мультиспецифическое антитело, синтезированное способом, включающим
(i) реакцию первого фрагмента антитела, полученного из первого исходного антитела, обладающего первой моноспецифичностью и свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и
(ii) реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, полученным из второго исходного антитела, обладающего второй моноспецифичностью и свободной сульфгидрильной группой, для получения мультиспецифического антитела,
и где первая моноспецифичность отличается от второй моноспецифичности.
6. Способ лечения злокачественной опухоли, включающий введение терапевтически эффективного количества мультиспецифического антитела по п.5 индивиду, нуждающемуся в лечении.
7. Аналог антитела, синтезированный способом, включающим
(i) реакцию первого фрагмента антитела, обладающего свободной сульфгидрильной группой, с тио-реактивным кросслинкером для получения группы фрагмент антитела-кросслинкер, и
(ii) реакцию группы фрагмент антитела-кросслинкер со вторым фрагментом антитела, обладающим свободной сульфгидрильной группой, для получения аналога антитела,
и где первый фрагмент антитела и второй фрагмент антитела получены из одного исходного антитела.
8. Антитело по п.7, где модифицированный кросслинкер представляет собой бис-малеимидоацетилацетат (BMata).
9. Антитело по п.7, где проводят дополнительную реакцию антитела, содержащего модифицированный кросслинкер, со средством, содержащим функциональную группу.
10. Антитело по п.9, где средство выбрано из полиэтиленгликоля (PEG), альбуминсвязывающего пептида (ABP), флуоресцентной метки, средства для радиовизуализации, цитотоксического средства и миРНК.
11. Способ по п.10, где средство представляет собой PEG, и PEG выбран из PEG с mw 2000 (2K), PEG с mw 12000 (12K) и PEG с mw 20000 (20K).
12. Аналог антитела по п.7, где аналог антитела обладает одним или несколькими видами биологической активности, неотличимыми от активности исходного антитела.
13. Аналог антитела по п.12, где биологическая активность представляет собой пролиферацию клеток.
14. Аналог антитела по п.13, где аналог антитела представляет собой антагонист Her2-экспрессирующих клеток, и исходное антитело представляет собой трастузумаб.
15. Аналог антитела по п.14, где аналог антитела выбран из бис-Fab 1324, бис-Fab 1328 и бис-Fab 1329.
16. Аналог антитела по п.7, где аналог антитела обладает одним или несколькими видами биологической активности, отличными от активности исходного антитела.
17. Аналог антитела по п.16, где биологическая активность аналога антитела является антагонистической, и биологическая активность исходного антитела является агонистической.
18. Аналог антитела по п.16, где биологическая активность аналога антитела является агонистической, и биологическая активность исходного антитела является антагонистической.
19. Аналог антитела по п.18, где биологическая активность представляет собой пролиферацию клеток.
20. Аналог антитела по п.19, где аналог антитела представляет собой агонист пролиферации Her2-экспрессирующих клеток, и исходное антитело представляет собой трастузумаб.
21. Аналог антитела по п.20, где аналог антитела выбран из бис-Fab 1188, бис-Fab 1321, бис-Fab 1322, бис-Fab 1323 и бис-Fab 1325.
22. Композиция, содержащая одно или несколько мультиспецифических антител, выбранных из бис-Fab 1187, бис-Fab 1189, бис-Fab 1190, бис-Fab 1191, бис-Fab 1192, бис-Fab 1193, бис-Fab 1299, бис-Fab 1300, бис-Fab 1301, бис-Fab 1302, бис-Fab1303, бис-Fab 1304, бис-Fab 1305, бис-Fab 1306 и бис-Fab 1307.
23. Композиция, содержащая один или несколько аналогов антител, выбранных из бис-Fab 1188, бис-Fab 1204, бис-Fab 1321, бис-Fab 1322, бис-Fab 1323, бис-Fab 1324, бис-Fab 1325, бис-Fab 1326, бис-Fab 1327, бис-Fab 1328, бис-Fab 1329, бис-Fab 1400 и бис-Fab 1401.
