Description
Foreliggende oppfinnelse omhandler sammensetninger for topisk påføring på hudoverflaten, spesielt hudvasksamrnensetninger som tilføres til vannfuktet hud og deretter blir renset av med vann. Spesielt omhandler den foreliggende oppfinnelsen hudvasksammensetningen innbefattende en a-hydroksysyre og andre syrer som er definert heri som en aktiv bestanddel formulert i en mild og ikke-irriterende detergentbase.
Forbindelser som generelt er kjent innen området for kosmetisk og terapeutisk hudbehandling som a-hydroksysyrer og andre syrer som definert heri innbefatter forbindelser slik som salicylsyre, melkesyre og glykolsyre. Disse forbindelsene innbefatter aktive ingredienser fra et antall kommersielt tilgjengelige produkter som er påført huden for å oppnå en fordelaktig effekt med hensyn til utseende og hudkondisjon, spesielt glatthet og smidighet. Salicylsyre har f.eks. vist seg å ha keratolytisk og komedolytisk aktivitet så vel som anti-bakteriell effekt og har blitt brukt ved behandling og forebygging av mild til moderat acne i mer enn 100 år. Melkesyre har en fordelaktig effekt på hudhydratisering og er en bestanddel av flere hudtonisk- og fuktighetsgivende produkter.
a-hydroksysyrer og andre syrer som defineres heri kan, samtidig som de gir en fordelaktig effekt på huden, forårsake lokal irritasjon ved påføring på sensitive områder av huden, spesielt ansiktet. Videre kan lokal irritasjon bli forverret av bærersystemet hvori den aktive ingrediensen er formulert. F.eks. kan hudvasksamnrensetninger og spesielt ansiktvasksammensetninger, som er generelt formulert med en såpefri detergentbase for effektiv rensing og skumming, gi en irritasjonseffekt på grunn av bruken av overflateaktive stoffer ofte brukt i slike detergentsystemer.
Et videre problem forbundet med formulering av sammensetninger inneholdende a-hydroksysyrer og andre syrer som defineres heri fremkommer på grunn av ønskeligheten av å formulere sammensetningen med en sur pH med hvilket den mest virkningsfulle fri syreform av syren vil dominere. For hudvasksammensetninger er dette problemet forverret ved problemene ved å oppnå et detergentbasesystem ved tilstrekkelig lav pH. En hudvasksammensetning som angis å inneholde et mildt, men likevel effektivt, rensemiddel for acnebehandling og inneholder 2,0 vekt-% salicylsyre i en detergentbase innbefattende en blanding av anioniske og amfotære overflateaktive stoffer er kommersielt tilgjengelig. Analyse av dette produktet viser at det har en pH på 5. Det følger videre med hensyntagen til at pKa for salicylsyre som har verdien 2,97 at den frie syreformen av hydroksysyren er ikke dominerende i dette produktet og at dets effektivitet tilsvarende ikke er fullt optimalisert.
Det er et formål med den foreliggende oppfinnelsen å tilveiebringe en hudvasksammensetning inneholdende en a-hydroksysyre og andre syrer som defineres her og en detergentbase som har god rensende og skummende egenskaper, er mild og ikke-irriterende og hvor sammensetningens pH nærmer seg pKa for syren slik at effektiviteten til syren økes. Dette formålet er oppnådd i henhold til den foreliggende oppfinnelsen som er basert på det uventede resultatet at en detergentbase inneholdende en blanding av et ikke-ionisk surfaktant i form av alkylglykosid på et amfotaert surfaktant som ikke bare har de ønskede rensende og skummende karakteristika og er mild og ikke-irriterende ved anvendelse på huden, men også muliggjør blandingen å bli formulert ved lav pH.
Foreliggende oppfinnelse angår således hudvasksammensetning, kjennetegnet ved at den innbefatter fra 0,1 til 10 vekt-% av en a-hydroksysyre, salicylsyre, maleinsyre eller pyrodruesyre, og en detergentbase bestående av en blanding av et ikke-ionisk alkylpolyglukosid surfaktant og et amfotaert surfaktant hvori pH på den nevnte sammensetningen er i området 3,0 til 4,5.
Passende syrer for inkorporering inn i denne oppfinnelsens hudvasksamnrensetninger inkluderer salicylsyre, melkesyre, sitronsyre, glykolsyre, malonsyre, maleinsyre, pyrodruesyre og hydroksy-oktanolsyre. Foretrukne syrer er salicylsyre, melkesyre og glykolsyre, spesielt salicylsyre og melkesyre. Passende konsentrasjon av salicylsyre forekommende i sammensetninger i denne oppfinnelsen vil være i området 0,2 til 5,0 vekt-%, mest passende i området 1,0 til 3,0 vekt-%. En foretrukken konsentrasjon av salicylsyre er 2,0 vekt-%. Passende konsentrasjon av melkesyre forekommende i sammensetninger i denne oppfinnelsen vil være i området 0,1 til 5,0 vekt-%, mest passende i området 0,5 til 2,5 vekt-%. En foretrukken konsentrasjon av melkesyre er 1 vekt-%. Passende konsentrasjon av glykolsyre vil forekomme i området 2,0 til 10,0 vekt-%.
I det følgende vil termen alkylpolyglykosid surfaktant bli brukt for et ikke-ionisk surfaktant som stammer fra vanlige naturlige organiske monomerenheter som finnes i stivelse, fett og sukker, og mest foretrukket, stammer fra D-glukosemonomerenheter. Alkylpolyglukosider som stammer fra D-glukose er acetal forbindelser hvor alkylresiduet har en karbonkjedelengde fra 8 til 16 karbonatomer og graden av glukosidasjon (eller polymerisasjon), dvs. gjennomsnittlig antall av glukoseenheter pr. alkylradikal, er mellom 1,1 og 6. En rekke av passende alkylpolyglukosider er kommersielt tilgjengelige enten individuelt eller som miksturer eller blandinger. Sammensetninger i henhold til denne oppfinnelsen vil generelt inneholde miksturer eller blandinger av forskjellige alkylpolyglukosider. Foretrukne alkylpolyglukosider for bruk i den foreliggende oppfinnelsen inkluderer dekylglukosid og laurylglukosid og miksturer av disse. Alkylpolyglykosid overflateaktive stoffer innbefatter generelt opp til 20% w/w av hudvasksammensetningen, passende fra 2,0 til 15,0 vekt-% og foretrukket fra 4,0 til 10,0 vekt-% av sammensetningen. Det er åpenbart at mengden av alkylpolyglukosid i noen grad bestemmes av beskaffenheten og mengden av amfotaert surfaktant tilstedeværende i blandingen.
I prinsippet kan et hvilket som helst amfotært surfaktant, som er akseptabelt for topisk påføring av huden, innbefattes i detergentbasen sammen med det alkylpolyglukosid overflateaktive stoffet, men, i lys av deres iboende mildhet og gode skummeegenskaper, vil det foretrukne amfotære overflateaktive stoffet tilhøre klassen av forbindelser som går under navnet betainer. Strukturelt inneholder betainforbindelser en karboksylatfunksjonell gruppe og en kvaternær nitrogenfunksjon adskilt med en metylendel. Disse inkluderer n-alkylbetainer slik som cetylbetain og behenylbetain, og n-alkylamidobetainer slik som kokoamidopropylbetain. Detergentbasens innhold av komponenter av typen amfotære overflateaktive stoffer i sammensetninger i den foreliggende oppfinnelse kan være en enkel forbindelse eller en mikstur av blandinger av to eller flere substanser. Et foretrukket amfotært surfaktant er kokoamidopropylbetain. Amfotære overflateaktive stoffer vil generelt innbefatte opp til 10 vekt-% av hudvasksammensetningen, passende fra 2,0 til 8,0 vekt-% og foretrukket fra 2,5 til 6,0 vekt-% av hudvasksammensetningen. Mengden av amfotært surfaktant vil til en viss grad bli bestemt av detergentbasens komponent av alkylpolyglukosid surfaktant.
Det er typisk at detergentbasen bestående av miksturen av ikke-ionisk alkylpolyglukosid surfaktant og amfotært surfaktant vil innbefatte opp til 30 vekt-% av hudvasksammensetningen. Passende vil detergentbasen innbefatte fra 5,0 til 20,0 vekt-% og mest passende fra 8,0 til 18,0 vekt-% av hudvasksammensetningen.
Kontroll av pH innen definerte grenser er essentielt ved den foreliggende oppfinnelsen. En pH i det ønskede området 3,0 til 4,5 er delvis oppnådd ved de iboende egenskapene av syren og spesifikke overflateaktive stoffer og kvantiteter av disse som utgjør detergentbasen og delvis, hvor dette kreves, ved bruk av et passende nøytraliseirngsmiddel for syren. Et hvilket som helst topisk akseptabelt nøytraliseringsmiddel som er kompatibelt med de andre forbindelsene i blandingen kan bli brukt. Det har blitt funnet at nøytraliseirngsmidlet trometamin er spesielt passende for hudvasksamnrensetninger bestående av a-hydroksysyrer eller andre syrer som defineres heri. Andre passende nøytraliseirngsmidler innbefatter natriumhydroksyd eller trietanolamin. Mengden av nøytraliseringsmiddel vil bli bestemt av syre/baseegenskapene av de andre ingrediensene som utgjør blandingen og den valgte pH for blandingen.
Hudvasksammensetningen i denne oppfinnelsen kan i tillegg også inneholde topisk akseptable hudkondisjonerings- og beroligende midler som f.eks. i form av anti-inflammasjonsmidler og vitaminer eller vitaminderivater, typisk ved lave konsentrasjoner som f.eks. i området 0,01 til 2,0 vekt-% av hele sammensetningen. Eksempler på topisk akseptable anti-inflammasjonsmidler inkluderer allantoin og bisabolol. Et foretrukket vitaminderivat er vitamin E-acetat som har anti-inflammasjonsegenskaper.
I tillegg vil sammensetninger ifølge foreliggende oppfinnelse passende inneholde farmasøytisk eller kosmetisk akseptable additiver eller tilsetninger som er vanlige innenfor området av topisk medisin og kosmetikk, innbefattende f.eks. tykningsmidler, fuktighetsgivere, gjenfettingsmidler, konserveringsmidler, kondisjoneirngsmidler, chelateringsmidler, fargestoffer, duftstoffer, UV-filtre og/eller emulsjonsgivere. Additivene eller tilsetningene som brukes i hvilken som helst av de gitte sammensetninger vil være kompatible både med hverandre og de essensielle bestanddelene av sammensetningen slik at det ikke er noen interaksjon som vil forringe den aktive ingrediensens funksjon. Alle additiver eller tilsetninger må selvfølgelig være ikke-toksiske og tilstrekkelig rene for å gjøre dem passende for menneskelig bruk.
Egnede fortykningsmidler inkluderer polymere ikke-ioniske overflateaktive stoffer med høy molekylvekt bestående av en langkjedet (C[2 til Qg) polyetylenglykolfettsyre eller fettsyreresidu. Eksempler omfatter PEG 200 hydrogenert glycerylpalmitat, PEG 55 propylenglykololeat, PEG 150 distearat og PEG 200 glyceryltallovat. Passende tykningsmidler med lav molekylvekt inkluderer kokamid DEA, lauret-3 og glycerylmonolaurat. Et fortykningsmiddel omfattende et polyuretanresin, propylenglykol og vann solgt under betegnelsen "Acrysol 44" har også god ytelse i hudvasksammensetrungens detergentbase. Et fortykningsmiddel vil passende innbefatte opp til 10,0 vekt-% av en sammensetning, mest passende fra 2,0 til 5,0 vekt-%. Foretrukne fuktighetsgivere inkluderer glycerin, propylenglykol, sorbitol og polyetylenglykol. En fuktighetsgiver kan innbefatte opp til 15 vekt-% av sammensetningen, mer generelt fra 2,0 til 6,0 vekt-% av sammensetningen. Passende gjenfettingsmidler innbefatter generelt 0,5 til 5,0 vekt-% av sammensetningen, foretrukket 0,75 til 2,0 vekt-% inkludert polyetylenglykol 7 og glycerylkokoat. Passende konserveringsmidler omfatter generelt 0,01 til 1,00 vekt-% av sammensetningen og passende 0,10 til 0,30 vekt-%, inkludert fenoksyetanol og metyldibromoglutaronitril og blandinger av disse. Passende kondisjoneringsmidler innbefatter generelt 0,1 til 5,0 vekt-% av sammensetningen og passende 1,0 til 3,5 vekt-% inkludert hydroksycetyl hydroksyetyldimoniumklorid og polykvaternium 39. Passende cheleringsmiddel innbefattende generelt opp til 1,0 vekt-% av sammensetningen og passende 0,1 til 0,3 vekt-% inkludert etylendiamintetraeddiksyre (EDTA), hydroetylendiamintrieddiksyre (HEEDTA), dietylentriaminpentaeddiksyre (DPT A) og
cykloheksandiamintetraeddiksyre (CTDA).
Den gjenværende delen/resten i sammensetningen er typisk vann og/eller et annet ikke-alkoholholdig løsemiddel slik at dette utgjør 100 vekt-% av sammensetningen. Det foretrukne løsemiddel er vann som vil generelt innbefatte mer enn 50 vekt-% av hudvasksammensetningen. Andre passende ikke-alkoholholdige løsemidler som kan anvendes for å lette oppløsning av oc-hydroksysyren inkluderer glykoler slik som propylenglykol og makrogol.
Oppfinnelsens hudvasksamnrensetninger kan bli fremstilt ved metoder som er godt kjente i teknikken og lett tilgjengelige for fagfolk. Generelt blir syren og de overflateaktive stoffene som utgjør detergentbasen sammen med eventuelle additiver løst i vann, pH på den resulterende blandingen blir sjekket og justert hvis hensiktsmessig og viskositeten av sammensetningen blir satt til det ønskede nivået ved tilsetning av fortykningsmidler. Den foreliggende oppfinnelsen omfatter videre en fremgangsmåte for fremstilling av en hudvasksammensetning som definert i det foregående innbefattende blandingen av a-hydroksysyren eller andre syrer som defineres heri med detergentbasen i et vandig løsemiddelsystem og tilpasset pH som krevet slik at sammensetningens pH er i området 3,0 til 4,5.
Foreliggende oppfinnelse angår også en fremgangsmåte for fremstilling av en sammensetning ifølge oppfinnelsen, kjennetegnet ved at den innbefatter blanding av a-hydroksysyre, salicylsyre, maleinsyre eller pyrodruesyre med detergentbasen i et vandig løsemiddelsystem og justering av pH om nødvendig.
I tillegg angår oppfinnelsen anvendelse av en sammensetning ifølge opprinnelsen for fremstilling av et medikament for behandling av og/eller profylakse av acne.
Oppfinnelsen angår også anvendelse av en sammensetning ifølge oppfinnelsen som en kosmetisk behandling for forbedring av utseendet og kondisjonen av menneskelig hud.
De følgende eksempler beskriver og demonstrerer videre sammensetninger som omfattes av oppfinnelsen. Eksemplene har bare til hensikt å illustrere, og ikke begrense, området for oppfinnelsen.
EKSEMPEL 1 - Hudvasksammensetning inneholdende melkesyre
En sammensetning omfattende følgende ingredienser ble fremstilt. Den ferdige sammensetningen hadde en pH på ca. 3,5.
EKSEMPEL 2 - Hudvasksammensetning inneholdende salicylsyre
En sammensetning omfattende de følgende ingredienser ble fremstilt. Den ferdige sammensetningen var et klart produkt med en pH på ca. 4,5.
EKSEMPEL 3 - Hudvasksammensetning inneholdende salicylsyre
Sammensetningen i eksempel 2, inneholdende i tillegg innholdsstoffer som er indikert nedenunder, ble fremstilt. Det resulterende klare produktet hadde en pH på 4,5.

Claims (21)

Hudvasksammensetning, karakterisert ved at den innbefatter fra 0,1 til 10 vekt-% av en a-hydroksysyre, salicylsyre, maleinsyre eller pyrodruesyre, og en detergentbase bestående av en blanding av et ikke-ionisk alkylpolyglukosid surfaktant og et amfotært surfaktant hvori pH på den nevnte sammensetningen er i området 3,0 til 4,5.
