Description
Foreliggende oppfinnelse angår en ny fremgangsmåte for fremstilling av e-fenylglycidinsyre-(2R,3R) med formelen:
eventuelt i form av et salt eller en ester.
Produktene med den generelle formel (I) kan benyttes for å fremstille taksol under de betingelser som er beskrevet i artikler av J-N. Denis et al., "J. Org. Chem.", 51, 46-50
(1986); "J. Amer. Chem. Soc.", 110, 5917-5919 (1988) eller derivater av taksol som beskrevet i EP 253 738.
Det er kjent, for eksempel i henhold til J-N. Denis et al., "J. Org. Chem.", 5_1> 46-50 (1986), å fremstille produkter med den generelle formel (I) ved asymmetrisk epoksydering, katalysert med titan [T. Katsuki og K.B. Sharpless, "J. Amer. Chem. Soc", 102» 5974-5976 (1980 ); J.G. Hill et al., "J. Org. Chem.", 48, 3607 (1983)] av cis-cinnaminalkohol fulgt av oksydasjon og forestring av den oppnådde epoksy-alkohol. Imidlertid er utbyttene ikke tilfredsstillende, enantiomeroverskuddet er generelt under 80$ og arbeidsveien er relativt lang.
I henhold til K. Earada, "J. Org. Chem.", 31, 1407-1410
(1966) er det kjent å separere isomerene av trans-g<->fenylglycidinsyre ved precipitering av et salt med optisk aktiv a—metylbenzylamin.
I henhold til K. Harada og Y. Nakajima, "Bull. Chem. Soc. Japan", 47 (11) 2911-2912 (1974) er det kjent å separere isomerene av cis-P-fenylglycidinsyre ved precipitering av et salt med optisk aktiv efedrin.
I EP 0 342 903 er det beskrevet en separering av en enantiomer av p<->(4-metoksyfenyl)glycidinsyre ut fra den tilsvarende diastereoisomer ved hjelp av optisk aktiv a—metylbenzylamin.
For at disse fremgangsmåter skal kunne gjennomføres er det imidlertid nødvendig på forhånd å separere cis- og trans-isomerene av p<->fenylglycidinsyre.
Det er i mellomtiden funnet, og det er gjenstand for foreliggende oppfinnelse, at p<->fenylglycidinsyre-(2R,3R) kan oppnås fra en blanding av cis- og trans-isomerene.
I henhold til dette angår foreliggende oppfinnelse en fremgangsmåte for fremstilling av cis-p<->fenyl-glycidinsyre-(2R,3R) med formel (I), eventuelt i form av et salt eller en ester, og denne fremgangsmåte karakteriseres ved at man selektivt, i et hensiktsmessig oppløsningsmiddel, valgt blant vann og organiske oppløsningsmidler valgt blant alifatiske C1_4-alkoholer, etere og ketoner, eventuelt i blanding med vann.
Generelt gjennomføres precipiteringen av saltet av g<->fenylglycidinsyre-(2R, 3R ) med (+ )-cx-metylbenzylamin-(R ) ved tilsetning av aceton til en vandig eller organisk, fortrinnsvis etanolisk, oppløsning av blandingen av salter av de optiske isomerer av cis- og trans-p<->fenylglycidinsyre med (+)-a-metylbenzylamin.
Når man arbeider i organisk medium, er det spesielt fordelaktig å sette aceton til den organiske oppløsning under tilbakeløp for derefter å precipitere det ønskede salt ved avkjøling.
Når man arbeider i vandig medium er det spesielt fordelaktig å tilsette aceton inntil man oppnår en blanding som inneholder 25 til 50$ vann og 75 til 50$ aceton. Fortrinnsvis arbeider man ved og de beste resultater oppnås når krystalli-ser ingsoppløsningen inneholder ca. 35$ vann og 65$ aceton.
Blandingen av de optiske isomerer av cis- og trans-<p>—fenyl-glycidinsyrene med (+ )-a-metylbenzylamin-(R ) kan oppnås: ved innvirkning av (+)-a-metylbenzylamin-(R) på en
blanding av cis- og trans-p-fenylglycidinsyre fremstilt in
situ, eller
- ved innvirkning av et salt av (+)-a-metylbenzylamin-(R) som hydrokloridet, på en blanding av alkalisalter som kaliumsaltene, av cis- og trans-fenylglycidinsyrene.
Blandingen av alkalisalter av cis- og trans-p<->fenylglycidinsyrene kan oppnås ved forsepning av de tilsvarende estere ved hjelp av en mineralbase. Det er spesielt fordelaktig å benytte etanolisk kalium og å arbeide ved en temperatur nær 20° C. Det er ikke nødvendig å isolere estrene før inn-virkningen av mineralbasen.
Blandingen av cis- og trans-p<->fenylglycidinsyrene kan oppnås in situ ved surgjøring av en vandig oppløsning av blandingen av de tilsvarende alkalimetallsalter.
Blandingen av estrene av cis- og trans-p-fenylglycidinsyrene kan oppnås ved innvirkning av et alkylhalogenacetat som alkylklor- eller -bromacetat, på benzaldehyd i henhold til den metode som er beskrevet av F.V/. Bacheler og R.K. Bansal, "J. Org. Chem.", 34 (11 ) 3600-3604 (1969 ). Det er spesielt fordelaktig å benytte t-butylkloracetat som tillater å oppnå en praktisk talt ekvimolekylær blanding av cis- og trans-isomerene.
Ved å erstatte t-butylkloracetat med t-butylbromacetat og ved fortrinnsvis å arbeide ved en temperatur nær 0°C, er det mulig å oppnå en blanding der cis-isomer-andelen er nær 75$. Ved innvirkning av en mineralbase og fortrinnsvis etanolisk kalium, på esteren som fremstilt in situ som angitt ovenfor, er det mulig å precipitere blandingen av alkalisaltene av cis- og trans-e-fenylglycidinsyrene (kaliumsaltene) i forbindelse med et alkalihalogenid (kaliumklorid eller —bromid) som, efter isolering, kan behandles med en vandig oppløsning av et salt av (+ )-a-metylbenzylamin-(R) som antydet ovenfor.
"Uansett hvilken fremgangsmåte man benytter for å fremstille blandingen av cis- og transsyreestrene er det mulig å oppnå en blanding inneholdende 80$ cis-ester og 20$ trans-ester ved krystallisering fra trans-isomeren i cis-isomeren som spiller rollen som oppløsningsmiddel.
p-fenylglycidinsyre-(2R,3R) som oppnås ifølge oppfinnelsen er spesielt brukbar for fremstilling av taksanderivater med den generelle formel:
der R betyr et hydrogenatom eller en acetylrest og R^ betyr en fenyl- eller t-butoksyrest.
De følgende eksempler skal illustrere oppfinnelsen uten å begrense den.
Eksempel 1
a) Til en 2-liters beholder settes 106 g eller 1 mol benzaldehyd, 150,7 g eller 1 mol t-butylkloracetat og 450
cm<5> t-butanol. Man tilsetter så, i løpet av 2 timer og 40
minutter og ved en temperatur mellom 18 og 24° C, en oppløsning av 112,5 g eller 1 mol kalium-t-butylat i 850 cm<5> t-butanol. Efter 18 timers omrøring ved en temperatur nær 20°C blir t-butanolen fordampet under redusert trykk. Den oppnådde rest tas opp med 1000 cm<5> vann. Man ekstra-herer med 3 x 300 cm<5> metylenklorid. De forenede organiske faser tørkes over magnesiumsulfat. Efter filtrering og konsentrering under redusert trykk oppnås 210 g av en olje hvis molare sammensetning, bestemt ved proton-NMR, er som følger:
b) Ved å arbeide som ovenfor, men ved å erstatte t-butylkloracetat med 195 g eller 1 mol t-butylbromacetat oppnås 213 g av en olje hvis sammensetning, bestemt på samme måte, er som følger.:
Til en reaktor settes 800 cm<5> etanol og 298 g av en 80 $-ig t-butyl-cis-p<->fenylglycidat og 20$ t-butyl-trans-p<->fenylglycidat, oppnådd efter krystallisering ved 4°C i flere dager og separering av trans-isomeren i cis- og trans-isomer-blandingen som oppnådd ovenfor under a) og b).
Den etanoliske avkjøles til en temperatur nær 0°C og behandles så i 3 timer med 134 g eller 2,03 mol 85 $-ig kaliumhydroksyd i oppløsning i 600 cm<5> etanol. Efter 18 timers omrøring ble reaksjonsblandingen filtrert. Det oppnådde faststoff vaskes med 200 cm<5> kold etanol og tørkes så til konstant vekt. Man oppnår på denne måte 185 g av en faststoff hvis sammensetning, bestemt ved proton-NMR, er som følger:
Til en reaktor settes:
185 g kalium-p<->fenylglycidat, oppnådd som tidligere,
625 cm<5> av en is/vann-blanding og
770 cm<5> etyleter.
Blandingen avkjøles til 0°C. Ved denne temperatur tilsettes, i løpet av 2 timer, 185 cm<5> 5N saltsyre. Efter ferdig tilsetning blir de organiske og vandige faser separert ved dekantering. Den vandige fase ekstraheres med 2 x 100 cm<3 >eter. De organiske faser tørkes over natriumsulfat. Efter filtrering blir de organiske faser under sterk omrøring behandlet med 125 cm<5> eller 0,95 mol (+ )-a-metylbenzylamin-(R). Det dannede precipitat separeres ved filtrering, vaskes med 200 cm<5> kold etyleter og tørkes. Man oppnår på denne måte 194 g av et hvitt pulver hvis proton-NMR-analyse viser at det består av
81$ cis-p-fenylglycidat av (+)-a-metylbenzylamin-(R) og 19$ trans-p<->fenylglycidat av (+)-cx-metylbenzylamin-(R ).
Til en reaktor settes 193 g av saltet som oppnådd ovenfor og 800 cm<5> etanol. Blandingen oppvarmes til tilbakeløp. Man oppnår på denne måte en farveløs homogen oppløsning som behandles under tilbakeløp med 1600 cm<5> aceton. Man lar den homogene oppløsning avkjøles til 45° C og tilsetter så noen krystaller av p<->fenylglycidat-(2R,3R) av (+)-a-metylbenzylamin-(R). Efter at temperaturen har nådd 42°C, danner det seg et rikelig precipitat. 2 timer efter slutten av acetontilset-ningen ligger temperaturen nær 25°C. Precipitatet separeres ved filtrering, skylles med aceton og tørkes til konstant vekt. Man oppnår på denne måte 36 g eller 0,126 mol praktisk talt rent p-fenylglycidat-(2R,3R) av (+)-a-metylbenzylamin-(R).
Det enantiomere overskudd, målt efter derivatisering til metylester fulgt av en kiral HPLC-analyse, er 97$.
Eksempel 2
Til en reaktor settes 28,53 g eller 0,1 mol 3-fenylglycidat-(2R,3R) av (+)-a-metylbenzylamin-(R), oppnådd i eksempel 1, samt 200 cm<5> diklormetan. Til den heterogene blanding settes det i løpet av 20 minutter og ved 22° C, 50 cm<5> 2N kaliumhydroksyd. Efter ferdig tilsetning separeres de to flytende faser ved dekantering. Den organiske fase vaskes med vann. De vandige faser forenes og konsentreres til tørr tilstand. Man oppnår på denne måte 19,9 g kalium-e-fenylglycidat-(2R,3R) hvis optiske dreining [a]p = -2,8 (c = 7,4; vann).
Eksempel 3
Til en 6-liters kolbe settes 523 cm<5> eller 3,97 mol (+)-a-metylbenzylamin-(R) og 760 cm<5> etanol. Reaksjonsblandingen avkjøles eksternt ved hjelp av en is/aceton-blanding. Man tilsetter, mens temperaturen holdes under 10°C, 143 cm<5> 2,78N saltsyre. Reaksjonsblandingen blir uklar og farveløs.
Til en 10-liters kolbe settes 3,8 1 etanol og 768 g av et produkt med følgende sammensetning:
Til den således oppnådde heterogene blanding, oppvarmet til 35 til 40°C, settes den ovenfor fremstilte (+)-a-metylbenzylamin-(R)-hydrokloridoppløsning som fremstilt ovenfor. Reaksjonsblandingen holdes ved 50°C i 2 timer. Kaliumkloridet som precipiteres separeres ved filtrering og vaskes med kokende metanol. Filtratet konsentreres til en vekt på 2445 g og settes så hen i 18 timer ved omgivelsestemperatur. De opptredende krystaller separeres ved filtrering, vaskes med 6 x 200 cm<5> etanol og tørkes til konstant vekt. Man oppnår på denne måte 530 g krystaller hvis proton-NMR viser at de består av cis-g<->fenylglycidatet av (+)-a-metylbenzylamin-(R) i ren form og hvis enantiomeroverskudd er 7$.
Man anbringer 525 g av det således oppnådde salt i en reaktor inneholdende 3,15 1 etanol. Blandingen oppvarmes til tilbakeløp og blir praktisk talt homogen. Man tilsetter så 6,3 1 kokende aceton og noen krystaller (2R,3R)-salt. Man lar det hele langsomt avkjøles til omgivelsestemperatur. De opptredende krystaller separeres ved filtrering, vaskes med 6 x 200 cm<5> aceton og tørkes til konstant vekt.
Man oppnår på denne måte 217 g g<->fenylglycidat-(2R,3R) av ( +)-a-metylbenzylamin-(R) hvis enantiomeroverskudd er 98,4$.
Eksempel 4
a) Til en reaktor på 170 1 av rustfritt stål oppløses, under nitrogenatmosfære, ved 30°C, 17,6 kg kalium-t-butylat i
en blanding av 60 1 t-butanol og 65 1 tetrahydrofuran. Man oppnår på denne måte 134 1 av en oppløsning A.
Til en reaktor på 250 1, settes under nitrogen og ved 20°C, 28,720 kg t-butyl-bromacetat og 16,2 kg benzaldehyd i 125 1 t-butanol. Man avkjøler til 0°C og tilsetter derefter, i løpet av 3 timer, oppløsning A, mens temperaturen holdes ved 0°C. Temperaturen holdes ved 0°C i ytterligere 2 til 3 timer.
Til den oppnådde oppløsning, holdt ved 0°C, settes, i løpet av 1 time og 20 minutter, en oppløsning av 12,480 kg kaliumpastiller i 50 1 absolutt etanol. Man holder temperaturen ved 20° C i 20 timer under sterk omrøring ved 150 omdr./min. Man tilsetter så 24 1 av et dismutert vann og oppvarmer til 50° C i 1 time og 30 minutter og holder det hele i 10 minutter ved denne temperatur. Det hele avkjøles til -10°C i 6 timer og filtreres så ved et nitrogentrykk på 2 bar. Filterkaken vaskes med 3 x 30 1 og så med 20 1 metyl-tert-butyleter: etanol 1:1 og tørkes så ved 0,13 kPa ved 30°C. Man oppnår på denne måte 45,58 kg av et produkt som, i henhold til proton-NMR, inneholder 57 vekt-$ av en blanding av kaliumsaltene av cis-p<->fenylglycidinsyre ( 73%) og trans-p<->fenylglycidinsyre (27$) samt 43 vekt-$ kaliumbromid.
b) Til en reaktor på 100 1 settes, under nitrogen, 31,86 kg ( +)-a-metylbenzylamin-(R) og 12 kg is. Det hele avkjøles
eksternt til -20°C og derefter tilsettes i løpet av 2 timer, 23,64 1 10,9N konsentrert saltsyre, mens temperaturen i blandingen holdes mellom 20 og 25°C. Man oppnår på denne måte 65 1 av en farveløs, uklar væske.
Til en 250 1 reaktor settes, under nitrogen, 87,3 kg av en blanding av kaliumsaltene av cis- og trans-p<->fenylglycidin-syren og kaliumbromid som oppnås under de ovenfor beskrevne betingelser, samt 200 1 destillert vann. Man oppvarmer til 45°C akkurat til oppløsning og avkjøler så til 30°C. I løpet av 10 minutter tilsettes 20 1 (+)-a-metylbenzylamin-(R)-hydrokloridoppløsning. Man understøtter krystalliseringen med 13 g cis-(R,R)-saltet. Krystalliseringen er umiddelbar. Man tilsetter så, i løpet av 15 minutter, resten av (+)-a-metylbenzylamin-(R)-hydrokloridoppløsningen.
Efter avkjøling i 1 time ved 22°C tilsettes 17,5 kg natriumklorid og det hele omrøres i 3 timer. Temperaturen er 17° C. Man filtrerer under et nitrogentrykk på 2 bar. Man oppnår 263 1 filtrat og 60 1 filterkake. Filterkaken vaskes med 2 x 40 1 av en vandig oppløsning av natriumklorid på 340 g/l og derefter 25 1 natriumkloridoppløsning. Man oppnår 118 1 vaskevann og 48 1 filterkake. Filterkaken behandles på filteret med 50 1 vann ved 0°C il time. Man oppnår 55 1 filtrat og 45 1 filterkake som tørkes til konstant vekt. Man oppnår på denne måte 34,1 kg salt av cis-p<->fenylglycidinsyre med (+ )-a-metylbenzylamin-(R) i praktisk talt ren tilstand (dampfasevæskekromatografi med derivatisering, proton-NMR ved 200 MHz, potensiometrisk titrering).
Til en emaljert reaktor på 250 1 settes 51,2 kg vann og 36,4 1 aceton og derefter 33,7 kg salt av cis-g<->fenylglycidinsyre med ( + )-a-metylbenzylamin-(R) med en titer på 95$ og inneholdende 53,2$ 2R,3R-produkt, det vil si 60,4 mol.
Reaksjonsblandingen oppvarmes til tilbakeløp ved 63" C og avkjøles langsomt efter ymping ved tilsetning av 58 g rent 2R,3R-salt.
Efter noen timer ved en temperatur nær 20 °C separeres krystallene ved filtrering, vaskes med 3 x 15 1 vannraceton i volumforholdet 36:64 og derefter med 3 x 15 1 aceton. Efter tørking til konstant vekt ved 5 mbar og 30°C, oppnås 12,5 kg salt av cis-g<->fenylglycidinsyre-(2R,3R) med a-metylbenzylamin-(R) i ren form i henhold til kiral HPLC-analyse.
Utbyttet er 72$.
Claims (5)
1.
Fremgangsmåte for fremstilling av cis-p-fenylglycidinsyre-(2R,3R) med formelen:
eventuelt i form av et salt eller en ester, karakterisert ved at man selektivt, i et hensiktsmessig oppløsningsmiddel, valgt blant vann og organiske oppløsningsmidler valgt blant alifatiske C1_4-alkoholer, etere og ketoner, eventuelt i blanding med vann, krystalli-serer saltet av <p>—fenylglycidinsyre-(2R,3R) med (+)-a-metylbenzylamin-(R), i en oppløsning av en blanding av salter av (+)-a-metylbenzylamin-(R) med e-fenylglycidin-(2R,3R)-, (3-fenylglycidin(2S,3S)-, e-fenylglycidin-(2R,3S)-og p<->fenylglycidin-(2S,3R)-syre og derefter isolerer det oppnådde produkt og eventuelt omdanner det til alkalimetall-salt eller en ester.

Fremgangsmåte ifølge krav 1, karakterisert ved at man tilsetter aceton til en vandig, organisk eller hydroorganisk oppløsning av blandingen som angitt i krav 1 og precipiterer saltet av e-fenylglycidinsyre-(2R,3R) med ( + )-cx-metylbenzylamin-(R) og isolerer det ved filtrering.

Fremgangsmåte ifølge et av kravene 1 til 2, karakterisert ved at man benytter en blanding bestående 80% cis-isomer av saltet av e-fenylglycidinsyre med (+)-a-metylbenzylamin-(R) og ca. 20% av trans-isomeren av saltet av p<->fenylglycidinsyre med (+ )-a-metylbenzylamin-(R ).

Fremgangsmåte ifølge et av kravene 1 til 3, karakterisert ved at blandingen av salter av (+)-«-metylbenzylamin-(R) med cis- og trans-isomerene av <p>—fenyl-glycidinsyre oppnås ved innvirkning av (+ )-a-metylbenzylamin på en blanding av cis- og trans-isomerene av p<->fenylglycidinsyre, fremstilt in situ.
