Abstract
Nye diselenobis-benzosyreamider av primære hetero-cykliske aminer av den generelle formelen (I). og R, som er like eller forskjellige, står for hydrogen, halogen, C_-alkyl, C]L_4-alkoksy, tri-fluormetyl, nltro eller, begge står sammen for. metylendi oksy,. n er null eller et tall fra 1 til 4, og rer en mettet eller umettet heerocyklisk rest med ett eller to heteroatomer i den cykliske kjernen,. valgt fra gruppen bestående av nitrogen, svovel og oksygen, nevnte rester er usubstituerte eller substituerte med en eller to substituenter, som er like eller forskjellige, valgt fra halogen, C_alkyl, C_-alkoksy, nitro eller hydroksy, er beskrevet.Det er videre angitt fremgangsmåte for fremstilling av forbindelsene, og det er beskrevet farmasøytiske preparater innbefattende forbindelsene.
Description
Foreliggende oppfinnelse vedrører analogifremgangsmåte for fremstilling av terapeutisk virksomme diselenobis-benzosyreamider. Disse forbindelsene kan anvendes i preparater som spesielt er gunstige for behandling av sykdommer som er forårsaket av celleskade på grunn av forøket dannelse av aktive oksygenmetabolitter, så som leverskader, hjerteinfarkt, infeksjoner, psoriasis og strålingsskader.
Forbindelsene som fremstilles ifølge oppfinnelsen tilsvarer den generelle formelen (I)
hvori
n er null eller 1, og
R<3> er en furyl-, pyridyl-, tienyl-, tiazolyl- eller isoksa-zolyl-gruppe som kan være substituert med en eller to metylgrupper.
Forbindelsene med formel (I) fremstilles ifølge oppfinnelsen ved at man
a) omsetter en benzisoselenazolon med generell formel (II)
hvor
R<3> og n har samme betydning som i formel (I), med en ekvimolar mengde av et mekaptan med formelen R'SH, hvor R' eksempelvis er etyl, i et egnet organisk oppløsningsmiddel ved romtemperatur til en forbindelse med generell formel
(III)
hvor n og R<3> har samme betydning som angitt ved formel (I), og som som omvandles ved tilsetning av et amin med formelen R''NH2» f°r eksempel metylamin, til diselenobis-benzosyreamider med formel (I),
eller
b) omvandler et benzisoselenazolon med generell formel (II), hvor n og R<3> har samme betydning som angitt ved formel (I), i
et egnet organisk oppløsningsmiddel ved romtemperatur i nærvær .av ekvimolare mengder av en ditiol, til diselenobis-benzosyreamider med formel (I).
Forbindelsene fremstilt ifølge oppfinnelsen viser egenskaper svarende til glutationperoksidase og kan derfor erstatte dette enzymet og kan, svarende til reaksjonen mellom dette enzymet og en merkaptan, forhindre og forebygge den skadelige aktiviteten av aktive oksygenmetabolitter.
Glutation(GSH)-peroksidase (Px) som er avhengig av nærværet av elementet selen eller forbindelser innbefattende selen, katalyserer reduksjonen av HgOg og organiske hydroperoksyder.
Det selenholdige enzymet beskytter cellene mot peroksydasjon og spiller derfor en viktig rolle i modulasjonen av ara-chidonsyremetabolismen (CC. Reddy, E.J. Massaro, Fundam. and Appl. Toxicology (3), 9-10 (1983), side 431-436 og L. Flohé i "Free Radicals in Biology", bind V, redigert av W.Å. Pryor 1982 Academic Press, side 223-254).
Glutation-peroksidase spiller en viktig rolle i alle de sykdommene som gir en celleskade av det affiserte vevet og endelig en nekrose, på grunn av den forøkede dannelsen av aktive oksygenmetabolitter i form av peroksyder (f.eks. lipidperoksidaser og hydrogenperoksyd). Dette såkalte "oksydative stresset" observeres f.eks. ved leversykdommer-indusert av betennelsesreaksjoner eller autoimmunologiske reaksjoner, av alkohol eller av legemidler, men også i andre typer sykdommer som f.eks. ved hjerteinfarkt. Det er kjent at etter et hjerteinfarkt trenger leukocytter inn i det skadede vevet, og at avlivelsen av cellevev er forbundet med en forøket dannelse av de ovenfor nevnte aktive oksygenmetabolittene. Dette gir til slutt en forøket dekomponering av vev.
I slike tilfeller er det naturlige eksisterende beskytt-elsessystemet mot slik skade, bestående av forskjellige enzymer som dekomponerer peroksyder og aktivt oksygen, overbelastet. Enzymer for dette formålet er f.eks. super-oksyd-dismutase, katalase, og spesielt glutation-redoks-systemet med dets enzymkomponent glutation-peroksidase. Dette sistnevnte enzymet er av spesiell betydning fordi det kan detoksifisere vev for organiske peroksyder, så vel som hydrogenperoksyd. Det er kjent at dette enzymsystemet er viktigst for den korrekte funksjonen av leveren (Wendel et al.: Biochemical Pharmacology, bind 31, side 3601 (1982); Remmer: Deutsches Årzteblatt - Årztliche Mitteilungen 79, brochure 26, side 42 (1982)). Omfanget av den eksperimentelt frembragte leverskade avhenger av akkurat dette enzymsystemet, dvs. av innholdet av glutation i leveren på den ene siden, og av aktiviteten av enzymet glutationperoksidase på den annen side. Under en generell betennelsestilstand er denne beskyttelsesmekanismen for leveren sterkt redusert (Bragt et al., Agents ånd Actions, Supp. 17, side 214
(1980)). Følgelig opplever leveren et sterkt forøket "oksida-tivt stress".
De reaktive oksygenmetabolittene er en meget viktig faktor som formidlere av betennelsestilstander. De utgjør tydeligvis en viktig faktor både i leucotaxis, permeabiliteten av blodkar, i skade av forbindende vev i immunkompleks/komple-ment-induserte effekter så vel som i skader som opptrer ved gjentagende intrusjon i ischemiske områder (L. Flohé et al., i "The Pharmacology of Inflammation" red. I.: L. Bonta et al., "Eandbook of Inflammation", bind 5, Elsevier, Amsterdam, under fremstilling).
Skader som opptrer etter ioniserende stråling er også forårsaket av dannelsen av radikaler og aktive oksygenmeta-bol itte.r.
Følgelig er en mulighet som kjemisk cytobeskyttelse for-bedring av aktiviteten av glutation/glutation-peroksydase-systemet H. Rink i "Glutathione", Proceedings of the 16th Conference of the German Society of Biological Chemistry 1973, redigert av Flohé, Benohr, Sies, Walter og Wendel, side 206).
Målingen av glutationperoksydase-lignende aktivitet utføres ved fremgangsmåten ifølge A. Wendel (A. Wendel, "Methods in Enzymology", bind 77, side 325-333 (1981)). Ved denne undersøkelsen bestemmes omvandlingen av kosubstratet nikotin-amid-adenin-dinukleotid-fosfat. Det reduserende midlet i denne reaksjonen er glutation. Det er overraskende funnet at forbindelsene av formel (I) gir en glutation-peroksidaselignende aktivitet.
Glutation- peroksidaselignende aktivitet
Ved in vitro forsøk ble katalysen av nedbrytningen av peroksidase undersøkt. Det er bestemt at forbindelsene fremstilt ifølge foreliggende oppfinnelse er i stand til å substituere glutation-peroksidase.
Den katalytiske aktiviteten uttrykkes som mengden av glutation-peroksidase. Som referanseprodukt benyttes det nylig beskrevne produktet "Ebselen" = 2-fenyl-l,2-benzisoselenazol-3(2H)-on (A. Wendel, M. Fansel, H. Sagayhi, G. Tiegs, R. Otter, Biochem. Pharmac. 33, 3241, 1984). Aktiviteten av "Ebselen" settes som 100$, og aktiviteten av forbindelsene fremstilt ifølge foreliggende oppfinnelse relateres med aktiviteten av "Ebselen".
"Ebselen" er undersøkt i en konsentrasjon på 30 jjmol og dimetylformamid (DMF) er anvendt som oppløseliggjørende middel. Diselenidene fremstilt ifølge oppfinnelsen er undersøkt i en konsentrasjon på 15 pmol ved anvendelse av DMF som oppløseliggjørende middel, idet det er tilstede to atomer selen pr. mol i diselenidene fremstilt ifølge oppfinnelsen.
Som en sammenligning av forbindelsene fremstilt ifølge oppfinnelsen med forbindelser ifølge teknikkens stand, skal det vises til den nedenstående tabellen.
Forbindelse 1 er fremstilt ifølge foreliggende oppfinnelse. Forbindelsene 2 og 3 omfattes av EP 0 100 899, og forbindelsene 4 og 5 omfattes av EP 0 044 453.
Grunnlaget for tabellen er, som ovenfor, aktiviteten av "Ebselen" satt til 100%.
Forbindelsene av formel (I) fremstilles ved at benzisoselen-azoloner av den generelle formelen (II), hvori R<3> og n har betydningene angitt i formel (I), omsettes med ekvimolare mengder av en merkaptan så som etylmerkaptan i et egnet organisk oppløsningsmiddel ved romtemperatur, slik at man får mellomforbindelsene av formel (III), som vist ved følgende ligning:
Forbindelsene av formel (III) omvandles lett, ved tilsats av aminer så som metylamin, til forbindelsene av formel (I), hvilket fremgår av følgende ligning:
En annen fremgangsmåte tar utgangspunkt i benzisoselenazo-loner av den generelle formel (II). Oppløst i et egnet organisk oppløsningsmiddel omsettes de ved romtemperatur med ekvimolare mengder av en ditiol, så som ditioerythritt slik at man får forbindelsene av formel (I).
Utgangsmaterialene benzisozelenazolonene av formel (II) fremstilles som beskrevet i DE-OS 3027073, DE-OS 3027074, DE-OS 3027075 og DE-OS 3236284.
Fremstillingen av forbindelsene med formel (I) er illustrert ytterligere i de følgende eksemplene. De angitte smelte-punktene er bestemt i en "Biichi 510"-apparatur. De er angitt 1 °C, og de angitte data er ikke korrigerte.
Eksempel 1
2 g (0,0071 mol) av 2-furfuryl-l,2-benzisoselenazol-3(2H)-on oppløses i ca. 50 ml metanol. Til oppløsningen tilsettes 0,56 ml etylmerkaptan. Blandingen omrøres ved romtemperatur. Etter 30 minutter utfelles en hvit forbindelse. Dette bunnfallet oppløses i 15 ml dimetylformamid. 5 ml av en 33$ oppløsning av metylamin tilsettes og omrøring fortsettes ved romtemperatur over natten. Den utfelte hvite forbindelsen separeres videre ved tilsats av eter, separeres ved filtrering med undertrykk og tørkes.
Utbytte: 1,5 g (74,7# av teoretisk) smp. 220°C.

Denne forbindelsen fremstilles som beskrevet i eksempel 1 ved å omsette: 2 g av 2-(2-pyridyl)-l,2-benzisoselenazol-3(2H)-on 0,53 ml av etylmerkaptan
5 ml av 33$ metylamin
Utbytte: 1 g (50,2$ av teoretisk) smp. 105°C.
Eksempel 3
Denne forbindelsen fremstilles som beskrevet i eksempel 1 ved å omsette: lg av 2-(2-pyridylmetyl)-l,2-benzisoselenazol-3(2H)-on 0,26 ml av etylmerkaptan
3 ml av 33Sé metylamin
Utbytte: 0,6 g (60$ av teoretisk) smp.: 192-196<0>C.

Utbytte: 1 g (50$ av teoretisk) smp.: 225"C.

Denne forbindelsen fremstilles som beskrevet i eksempel ved å omsette: 2 g av 2-(3-pyridyl)-l,2-benzisoselenazol-3(2H)-on 0,74 ml av etylmerkaptan
5 ml av 33$ metylamin
Utbytte: 1,19 g (36,1$ av teoretisk) smp.: 248°C

Utbytte: 0,53 g (27,1$ av teoretisk) smp.: 175°C.
Eksempel 7
?±?lÉi§ÉlÉ522i§l£^I§ll5=metylisoksaz 0,5 g (0,00179 mol) av 2-[3-(5-metylisoksazolyl)]-1,2-benzisoselenazol-3(2H)-on oppløses i 30 ml metanol. 0,276 g (0,00179 mol) ditioerythritt oppløses i 10 ml metanol, og denne oppløsningen tilsettes til den ovenfor nevnte metanol-oppløsningen av den andre forbindelsen. Blandingen omrøres ved romtemperatur i 3 timer og deretter avdampes oppløsnings-midlet. Resten rekrystalliseres fra etanol.
Utbytte: 0,4 g (34,7$ av teoretisk) smp. 245°C.

- Analogifremgangsmåte for fremstilling av terapeutisk virksomme diselenobis-benzosyreamider med generell formel (I) hvor n er null eller 1, og R<3> er en furyl—, pyridyl—, tienyl—, tiazolyl— eller isoksa-zolyl-gruppe som kan være substituert med en eller to metylgrupper,karakterisert ved at man a) omsetter en benzisoselenazolon med generell formel (II) hvorR<3> og n har samme betydning som i formel (I), med en ekvimolar mengde av et mekaptan med formelen R'SH, hvor R' eksempelvis er etyl, i et egnet organisk oppløsningsmiddel ved romtemperatur til en forbindelse med generell formel (III) hvor n og R<3> har samme betydning som angitt ved formel (I), og som omvandles ved tilsetning av et amin med formelen R''NH2> for eksempel metylamin, til diselenobis-benzosyreamider med formel (I), eller b) omvandler et benzisoselenazolon med generell formel (II), hvor n og R<3> har samme betydning som angitt ved formel (I), i et egnet organisk oppløsningsmiddel ved romtemperatur i nærvær av ekvimolare mengder av en ditiol, til diselenobis-benzosyreamider med formel (I).
