Description
Foreliggende oppfinnelse vedrører fremstilling av pyridinderivater som er nyttige f.eks. som midler mot magesår.
Som pyridinderivater som har aktivitet mot magesår, er de som er beskrevet i NO-patent nr. 152216 og NO-patentsøknad nr. 823691 kjente.
Selv om disse kjente forbindelsene har en syresekresjons-inhiberende virkning er deres beskyttelsesvirkning for mage slimhinnen -utilstrekkelig, de kan følgelig ikke betraktes som tilfredsstillende midler mot magesår. Videre er disse forbindelsene beheftet med ulemper som f.eks. at deres fysikalsk-kjemiske egenskaper er ustabile, og at de lett dekomponerer.
Det antas at gastrointestinale sår induseres ved ubalanse mellom aggressive faktorer, f.eks. saltsyre, pepsin, og beskyttelsesfaktorer, f.eks. slimsekresjon og blodgjen-nomstrømming i slimhinnen. Følgelig ønsker man et legemiddel som både har en virkning ved at det inhiberer magesyre-sekresjonen og som samtidig forbedrer beskyttelsen av mages1imhinnen.
Formålet med foreliggende oppfinnelse er å fremstille et middel mot magesår som har utmerkede virkninger ved at det inhiberer magesyresekresjon, beskytter mageslimhinnen og forhindrer sårdannelse. Det er funnet at en viss type pyridinderivater oppfyller dette formålet, og det er utført studier som har ført fram til foreliggende oppfinnelse.
De nye pyridinderivatene som tilveiebringes ifølge oppfinnelsen har den generelle formelen:
hvor R<*> er hydrogen, metoksy eller trifluormetyl, R<2> og R<3> er uavhengig hydrogen eller metyl, R<4> er C2_5-fluorert alkyl, og n er 0 eller 1, eller farmakologisk akseptable salter derav. ;Ifølge oppfinnelsen fremstilles forbindelsene med formel (I) ved at en forbindelse med formelen: hvor Ri har den ovenfor angitte betydning, får reagere med en forbindelse av formel (III) ;hvor R<2>, R<3> og R<4> har de ovenfor angitte betydninger, en av X<*> og X<2> er SH og den andre er en avspaltbar gruppe og, om nødvendig, underkastes reaksjonsproduktet oksydasjon, og, om ønsket, omdannes en således erholdt forbindelse til et farmasøytisk akseptabelt salt derav.
Spesielt foretrukket er fremstillingen av følgende forbindelser: 2-[3-metyl-4-(2,2,2-trifluoretoksy)-pyrid-2-yl]metylsul-finylbenzimidazol, 2-[3-metyl-4-(2,2,3,3,3-pentafluorprop-oksy )pyrid-2-yl]metylsulfinylbenzimidazol og 2-[3-metyl-4-(2,2,3,3-tetrafluorpropoksy )pyrid-2-yl]metyl sulfinyl-benzimidazol, på grunnlag av disse forbindelsenes spesielle og foretrukne aktivitet, hvilket vil fremgå fra de senere angitte forsøksdata.
I formlene ovenfor kan C2-5 fluorerte alkylgrupper angitt ved R<4> eksemplifiseres ved 2,2,2-trifluoretyl, 2,2,3,3,3-pentafluorpropyl, 2,2,3,3-tetrafluorpropyl-l-(trifluormetyl)-2,2,2-trifluoretyl, 2,2,3,3,4,4,4-heptafluorbutyl og 2,2,3,3,4,4,5,5-oktafluorpentyl.
Eksempler på de avspaltbare gruppene X<1> og X<2> i formlene ovenfor er halogen, fortrinnsvis klor, brom eller jod, eller en reaktiv forestret hydroksygruppe, f.eks. en arylsul-fonyloksy, f.eks. fenylsulfonyloksy eller tosyloksy, eller en cl-4 alkylsulfonyloksy, f.eks. metansulfonyloksy, eller en organisk fosforyloksy, f.eks. difenylfosforyloksy, dibenzyl-fosforyloksy eller di-C1_4 alkylfosforyloksy (f.eks. dimetylfosforyloksy) o.l.
R<1> kan være plasssert ved 4- eller 5-posisjonen, fortrinnsvis ved 5-posisjonen.
Et sulfidderivat (I) (n = 0) kan fremstilles ved å la en forbindelse (II) reagere med en forbindelse (III). Det er hensiktsmessig å gjennomføre denne reaksjonen i nærvær av en base. Basen kan f.eks. være alkalimetallhydrid, f.eks. natriumhydrid og kaliumhydrid; alkalimetall, f.eks. metallisk natrium; natriumalkoholat, f.eks. natriummetoksyd eller natriumetoksyd; alkalimetallkarbonat, f.eks. kaliumkarboat eller natriumkarbonat; og organiske aminer, f.eks. trietylamin. Oppløsningsmidlet som benyttes i reaksjonen kan eksemplifiseres ved alkoholer, f.eks. metanol eller etanol, så vel som dimetylformamid. Mengden av en base som benyttes i reaksjonen er vanligvis et lite overskudd sammenlignet med den ekvivalente mengden, men den kan være et stort overskudd. Nærmere bestemt er den 1-10 ekvivalenter, fortrinnsvis 1-4 ekvivalenter. Reaksjonstemperaturen varierer vanligvis fra 0°C til kokepunket for oppløsningsmidlet som benyttes, fortrinnsvis fra 20°C til 80°C. Reaksjonstiden varierer fra 0,2 til 24 timer, fortrinnsvis fra 0,5 til 2 timer.
Et sulfinylderivat (I) (n = 1) kan fremstilles ved å utsette en forbindelse (I) (n = 0) for oksydasjon. Oksydasjonsmidlet som benyttes er f.eks. persyre, som m-klorperbenzosyre, pereddiksyre, trifluorpereddiksyre og permaleinsyre, eller natriumbromid eller natriumhypoklorit eller hydrogenperoksyd. Oppløsningsmidlet som benyttes i reaksjonen er f.eks. halogenert hydrokarbon som kloroform eller diklormetan, etere, f.eks. tetrahydrofuran og dioksan, amider, f.eks. dimetylformamid, alkoholer, f.eks. metanol, etanol, propanol og t-butanol eller vann, og disse oppløsningsmidlene kan benyttes enkeltvis eller i blanding. Oksydasjonsmidlet benyttes fortrinnsvis i tilnærmet ekvivalent mengde, eller et lite overskudd relativt forbindelsen (I) (n = 0). Nærmere bestemt er det 1-3 ekvivalenter, fortrinnsvis 1-1,5 ekvivalenter. Reaksjonstemperaturen varierer fra temperaturen under isavkjøling til rundt kokepunktet for oppløs-ningsmidlet som anvendes, vanligvis fra isavkjølingstempera-turen til romtemperatur, fortrinnsvis fra 0°C til ICC. Reaksjonstiden varierer vanligvis fra 0,1 til 24 timer, fortrinnsvis fra 0,1 til 4 timer.
Forbindelsen (I) fremstilt ved fremgangsmåten angitt ovenfor kan isoleres og renses ved konvensjonelle fremgangsmåter, f.eks. ved rekrystallisasjon og kromatografi.
Forbindelsen (I) kan overføres til farmakologisk akseptable salter derav ved i og for seg kjente fremgangsmåter, som eksempler på saltene kan nevnes hydrogenklorid, hydrogenbro-mid, hydrogeniodid, fosfat, nitrat, sulfat, acetat og citrat.
Blant forbindelsene (I) gir de som har n = 0 stabile salter, mens de med n = 1 kan eksistere som en vandig oppløsning som er ustabil.
Fremgangsmåten til fremstilling av utgangsmaterialet (III) beskrives på følgende måte.
En nitroforbindelse av formelen (IV) [hvor R<2> og R<3> hart de ovenfor angitte betydninger] får reagere med et alkoholderi-vat R<4>OE (IV) [hvor R<4> har den ovenfor angitte betydning] i nærvær av en base, slik at man får et alkoksyderivat av formel (VI) [hvor R<2>, R<3> og R<4> har de ovenfor angitte betydninger]. Basen kan eksemplifiseres ved alkalimetall, f.eks. litium, natrium og kalium; alkalimetallhydrid, f.eks. natriumhydrid og kaliumhydrid; alkoholat, f.eks. kalium-t-butoksyd og natriumpropoksyd; alkalimetallkarbonat eller hydrogenkarbonat, f.eks. kaliumkarbonat, litiumkarbonat, natriumkarbonat, kaliumhydrogenkarbonat og natriumhydrogenkarbonat; eller alkalihydroksyd, f.eks. natriumhydroksyd og kaliumhydroksyd. Oppløsningsmidlet som benyttes i reaksjonen er f.eks. ved siden av R<4>OH selv, etere som f.eks. tetrahydrofuran og dioksan, så vel som ketoner som aceton og metyletylketon, acetonitril, dimetylformamid og heksametyl-fosforsyre-triamid. Reaksjonstemperaturen velges fortrinnsvis innenfor området fra temperaturen for isavkjøling til temperaturer nær kokepunktet for oppløsningsmidlet som benyttes. Reaksjonstiden varierer vanligvis fra 1 til 48 timer.
Den slik fremstilte forbindelsen (VI) underkastes oppvarming (80 til 120°C) i nærvær av eddiksyreanhydrid alene eller sammen med en mineralsyre, f.eks. svovelsyre og perklorsyre, slik at man får et 2-acetoksymetylpyridinderivat av formel (VII) [hvor R<2>, R<3> og R<4> har de ovenfor angitte betydninger]. Reaksjonstiden varierer vanligvis fra 0,1 til 10 timer.
Deretter underkastes forbindelsen (VII) alkalihydrolyse, slik at man får et 2-hydroksymetylpyridinderivat av formelen (VIII) [hvor R<2>, R<3> og R<4> har de ovenfor angitte betydninger]. Alkali kan f.ekes. være natriumhydroksyd, kaliumhydroksyd, kaliumkarbonat og natriumkarbonat. Oppløsnings-midlet som benyttes i reaksjonen er f.eks. metanol, etanol og vann. Reaksjonstemperaturen varierer vanligvis fra 20°C til 60°C. Reaksjonstiden ligger innenfor området 0,1 til 2 timer.
Forbindelsen (VIII) underkastes videre reaksjon med et kloreringsmiddel som tionylklorid eller et forestringsmiddel, f.eks. et organisk sulfonsyreklorid som metansulfonylklorid eller p-toluensulfonylklorid, eller et uorganisk fosforsyre-klorid som f.eks. difenylfosforylklorid, slik at man får forbindelsen (III). Mengden av kloreringsmidlet som benyttes i reaksjonen er vanligvis et stort overskudd sammenlignet med mengden av forbindelsen (VIII). Oppløsningsmidlet som benyttes i reaksjonen er f.eks. kloroform, diklormetan og tetrakloretan. Reaksjonstemperaturen ligger vanligvis innenfor området fra 20°C til 80°C, og reaksjonstiden er 0,1 til 2 timer.
Mengden av det organiske sulfonsyrekloridet eller det organiske fosforsyrekloridet som benyttes i reaksjonen er vanligvis ekvivalent med et lite overskudd, og reaksjonen gjennomføres vanligvis i nærvær av en base. Basen er f.eks. en organisk base som trietylamin og tributylamin, eller en uorganisk base som natriumkarbonat, kaliumkarbonat og natriumhydrogenkarbonat. Mengden av base som benyttes i reaksjonen er vanligvis ekvivalent med et lite overskudd. Oppløsningsmidlet som benyttes ved reaksjonen er f.eks. kloroform, diklormetan, karbontetraklorid eller acetonitril. Reaksjonstemperaturen varierer vanligvis fra temperaturen under isavkjøling til en temperatur rundt kokepunktet for oppløsningsmidlet som benyttes. Reaksjonstiden varierer vanligvis fra få minutter til et par timer. Det er vanligvis foretrukket å benytte den fremstilte forbindelsen (III) umiddelbart i reaksjonen med en forbindelse (II).
Ved en reaksjon svarende til den ovenfor omtalte fremgangsmåte (I) overføres en forbindelse av formelen (IX) [hvor R<2> og R<3> har de ovenfor angitte betydninger] til en forbindelse av formelen (X) [hvor R<2>, R<3> og R<4> har de ovenfor angitte betydninger].
Deretter underkastes forbindelsen (X) metylering med dimetylsulfat, slik at man får en forbindelse av formel (XI)
[hvor R<2>, R<3> og R4 har de ovenfor angitte betydninger]. Reaksjonstiden kan vanligvis utføres uten oppløsningsmiddel. Reaksjonstemperaturern varierer fra 100°C til 120°C, og reaksjonstiden ligger innenfor området fra 0,1 til 4 timer.
Videre får forbindelsen (XI) reagere med en radialkilde som f.eks. ammoniumpersulfat, eller et hvilket som helst annet persulfat i metanol, slik at man får den ovenfor nevnte forbindelse (VIII). Reaksjonstemperaturen ligger innenfor området fra 20°C til 80°C, og reaksjonstiden varierer fra 0,5 til 4 timer.
De farmakologiske virkningene av forbindelsene ifølge foreliggende oppfinnelse "beskrives på følgende måte.
Som modeller for gastrointestinale sår har stress-induserte sår forårsaket av frihetsberøvelse og vann-neddykking, indometacin-induserte sår og etanol-induserte mageslim-hinnelesjoner vært benyttet. Som en modell for å etterligne magesår hos mennesker er imidlertid indometacin-indusert mage"antral"sår rapportert i "Gastroenterology" (Satoh et al. 81, side 719, 1981), denne ble betraktet som en verdifull eksperimentell modell. I det følgende gjengis data for anti-magesårvirkninger av forbindelsene (I) og noen av de andre representative kjente forbindelser, på magesårmodelien angitt i den ovenfor nevnte litteraturreferansen.
Ved fastsettelse av ulcus-indeks har man som nevnt målt arealet av lesjoner i antrum, mens man tidligere i noen tilfeller målte summen av lengden av lesjonene i korpus og i grenseområdet for korpus/antrum.
Ifølge beskrivelsen i Gastroenterology, 36, p 45, 1959, spesielt side 45, rapporteres det at størstedelen av magesår oppstår på eller innen 2,5 cm av den mindre krumming hos både menn og kvinner.
Ut fra rapporten er den eksperimentelle modell som er benyttet i foreliggende tilfelle mer lik det som er tilfellet hos mennesker i forbindelse med magesår enn den tidligere nevnte bestemmelse av summen av lengden av lesjnene i korpus og i grenseområdet for korpus/antrum.
Ved fastsettelse av ulcus-indeks er det her benyttet hannrotter.
Såvidt vites er det ingen rapporter i det hele tatt på magesår ved bruk av hunnrotter. Grunnen er at foringsmekan-ismen hos hunnrotter er forskjellig fra den hos hannrotter, og videre blir hunnrottens kjønnscyklus (4 dager) som beror på hunnlige kjønnshormoner, påvirket av lucerasjon. Det er i tillegg vist i J. Edward Berk, MD, D. Sc, ed., Bockus, Gastroenterology, vol. 2, side. 1013, 1985, spesielt side 1020, i høyre spalte som danner forbindelse med side 1021, venstre spalte, at hunnkjønn er dominerende i forhold til hunnkjønn når det gjelder ulcerasjon hos mennesker.
Av de ovenfor angitte grunner vil det fremgå at hannrptten bør benyttes når det gjelder nøyaktig fastsettelse av ulcus-indeks .
Eksperimentell fremgangsmåte:
7 uker gamle Sprague-Dawley hannrotter ble underkastet faste i 24 timer. Disse dyrene fikk administrert forsøksforbindel-sene inn i magen ved hjelp av et magerør. Etter 30 minutter ble det administrert indometacin, 30 mg/kg subkutant. I løpet av 30-90 minutter etter administreringen av indometacin hadde dissse dyrene fri tilgang til matpellets (Japan Clea, CE-2). 5 timer etter administreringen av indometacin ble 1 ml 1$ "Evans blått" injisert i dyrene via halevenen, deretter ble dyrene avlivet med karbondioksydgass. Magen ble fjernet sammen med den nedre delen av spiserøret og tolvfingertarmen. Spiserøret ble lukket, 10 ml 1$ formalin-oppløsning ble ført inn i magen fra tolvfingertarmen og deretter ble tolvfingertarmen lukket. Hele magen ble neddykket i 1% formalinoppløsning. 15 minutter senere ble magene åpnet langs den største krummingen. Sårområdene som fantes i den "antrale" mage sl imhinnen ble målt under et disseksjonsmikroskop med et okkular med firkantet nettinndel-ing (xlO). Den totale summen av de individuelle lesjonene på hvert dyr ble målt, og den gjennomsnittlige verdien pr. gruppe ble beregnet. Basert på forskjellen mellom den gjennomsnittlige verdien for hver gruppe og verdien for kontrollgruppen ble omfanget av inhiberingen bestemt. Forsøksforbindelsen på indometacin ble suspendert i en 55é gummi arabicum oppløsning, og administrert i et volum på
2 ml/mg.
Eksperimentelle resultater:
Som det fremgår av de ovenfor angitte data har forbindelsen med formel I en overlegen antimagesårvirkning som er 1,5-20 ganger høyere enn virkningen av kjente forbindelser. Videre viser forbindelsen (I) utmerket virkning ved at den inhiberer magesyresekresjon, beskytter mageslimhinnen og forhindrer sårdannelse.
Vedrørende toksisiteten av forbindelsen (I) forårsaket oral administrering av forbindelsen som ble anvendt i forsøket med ant i sårdannelse (forbindelsen med R<1> = E, R<2> = CH3, R<3> = E, R<4> = CE2CF2CF3, n = 1) på mus, selv i en dose på 2 000 mg/kg, ingen dødelig effekt, følgelig har forbindelsen (I) lav toksisitet.
Som beskrevet ovenfor har forbindelsen (I) antimagesårvirkning, den har en kontrollerende virkning på magesyresekre-sjonen og beskytter slimhinnen, videre har den lav toksisitet og er relativt stabil som en kjemisk forbindelse. Forbindelsen (I) kan følgelig benyttes til profylakse og behandling av sår i fordøyelseskanalen (f.eks. magesår, tolvfingertarmsår) og gastritt hos pattedyr (f.eks. mus, rotte, kanin, hund, katt og menneske).
Når forbindelsen (I) benyttes som et middel mot magesår for behandling av sår i fordøyelsessystemet hos pattedyr, kan den administreres oralt i doseringsformer som kapsler, tabletter, granulater etc. ved sammensetning med en farmakologisk akseptabel bærer, eksipiens, fortynningsmiddel osv. Den daglige dosen er 0,01-30 mg/kg, fortrinnsvis 0,1-3 mg/kg.
Forbindelsen (I) (n = 0) er nyttig som et utgangsmateriale for fremstilling av forbindelsen (I) (n = 1).
Fremgangsmåten til fremstilling av utgangsforbindelsen som skal anvendes ved fremgangsmåten ifølge foreliggende oppfinnelse, så vel som fremgangsmåtene til fremstilling av forbindelsen (I), er nærmere beskrevet i de følgende referanseeksemplene og arbeidseksemplene.
Referanseeksempel 1
I 2,2,3,3-tetrafluorpropanol (10 ml) ble det oppløst 2,3-dimetyl-4-nitropyridin-l-oksyd 82 g). Til oppløsningen ble det litt etter litt tilsatt kalium-t-butoksyd (1,6 g) ved romtemperatur. Blandingen ble deretter oppvarmet til 80-90°C i 22 timer. Reaksjonsoppløsningen ble fortynnet med vann, og deretter underkastet ekstraksjon i kloroform. Ekstraktet ble tørket over magnesiumsulfat og deretter konsentrert. Konsentratet ble kromatografert på en kolonne av silikagel (70 g). Eluering ble utført med metanol-kloroform (1:10), deretter ble det rekrystallisert fra etylacetat-heksan, slik at man fikk 2,6 g 2,3-dimetyl-4-(2,2,3,3-tetrafluorpropoksy)-pyridin-l-oksyd som fargeløse nåler, smeltepunkt 138-139°C.
Ved en fremgangsmåte svarende til den som er angitt ovenfor ble forbindelser (VI) fremstilt fra forbindelser av formel
(IV).
Referanseeksempel 2
En blanding av 2,3-dimetyl-4-nitropyridin-l-oksyd (2,0 g), metyletylketon (30 ml), 2,2,3,3,3-pentafluorpropanol (3,05 ml), vannfritt kaliumkarbonat (3,29 g) og heksametyl-fosforsyretriamid (2,07 g) ble oppvarmet til 70-80<>C i 4,5 dager under omrøring, deretter ble uoppløselig materiale filtrert fra. Filtratet ble konsentrert og det ble tilsatt vann. Blandingen ble underkastet ekstraksjon med etylacetat. Ekstraktoppløsningen ble tørket over magnesiumsulfat, deretter ble oppløsningsmidlet fjernet ved inndampning. Resten ble kromatografert på en kolonne av silikagel (50 g), eluert med kloroform-metanol (10:1), og rekrystallisert fra etylacetat-heksan, slik at man fikk 2,4 g 2,3-dimetyl-4-(2,2,3,3,3-pentafluorpropoksy)pyridin-l-oksyd som fargeløse nåler, smeltepunkt 148-149°C.
Ved en fremgangsmåte svarende til fremgangsmåten ovenfor ble forbindelser (VI) fremstilt fra utgangsforbindelsene (IV).
Referanseeksempel 3
Konsentrert svovelsyre (to dråper) ble tilsatt til en oppløsning av 2,3-dimetyl-4-(2,2,3,3-tetrafluorpropoksy)-pyridin-1-oksyd (2,6 g) i eddiksyre-anhydrid (8 ml). Blandingen ble omrørt ved 100°C i 4 timer, og deretter konsentrert. Resten ble oppløst i metanol (20 ml), og det ble tilsatt nat r iumhydr ok syd (1,2 g) oppløst i vann (5 ml). Blandingen ble omrørt ved romtemperatur i 30 minutter og ble deretter konsentrert. Til resten ble det tilsatt vann og blandingen ble underkastet ekstraksjon med etylacetat. Ekstraktet ble tørket over magnesiumsulfat, deretter ble oppløsningsmidlet fjernet ved inndamping. Resten ble kromatografert på en kolonne av silikagel (50 g), eluert med kloroform-metanol (10:1), og rekrystallisert fra isopropyleter, slik at man fikk 1,6 g 2-hydroksymetyl-3-metyl-4-(2,2,3,3-tetrafluorpropoksy)pyridin som gule krystaller, smeltepunkt 67-68°C.

Til en oppløsning av 3,5-dimetyl-4-nitropyridin-l-oksyd (2,0
g) i 2,2,3,3,3-pentafluorpropanol (10 g) ble det ved 0°C litt etter litt tilsatt kalium-t-butoksyd (2 g) i løpet av 15
minutter. Blandingen ble omrørt ved 60°C i 18 timer. Til reaksjonsblandingen ble det tilsatt kloroform, og den ble deretter filtrert med celitt. Filtratet ble kromatografert på en kolonne av silikagel (80 g), eluert med etylacetat-heksan (1:1), deretter med 20$ metanol-etylacetat, og rekrystallisert fra eterheksan, slik at man fikk 2,6 g 3,5-dimetyl-4-(2 ,2,3,3,3-pentafluorpropoksy)pyridin-1-oksyd som krystaller, smeltepunkt 89-91°C.
Ved en fremgangsmåte svarende til fremgangsmåten ovenfor ble forbindelser (X) fremstilt fra forbindelser (IX).
Referanseeksempel 5
En blanding av 3,5-dimetyl-4-(2,2,3,3,3-pentafluorpropoksy)-pyridin-l-oksyd (2,5 g) og dimetylsulfat (1 ml) ble oppvarmet til 120"C i 30 minutter, deretter ble blandingen tilsatt metanol (12,5 ml). Blandingen ble så ved 80°C dråpvis tilsatt amm on i umpe r su 1 f at (4,3 g) oppløst i vann (20 ml) metanol (10 ml) i løpet av et tidsrom på 30 minutter, og omrørt i ytterligere 30 minutter. Den resulterende oppløsningen ble konsentrert. Til resten ble det tilsatt is, det ble nøytralisert med natriumkarbonat, etterfulgt av ekstraksjon med kloroform. Ekstraktet ble tørket på natriumsulfat, etterfulgt av fjernelse av oppløsningsmidlet ved inndamping slik at man fikk 2,2 g 3,5-dimetyl-2-hydrok-symetyl-4-( 2,2,3,3,3-pentaf luorpropoksy )pyridin som et oljeformig stoff.
Ved en fremgangsmåte svarende til fremgangsmåten ovenfor ble forbindelser (VIII) fremstilt fra forbindelser av formel (X).
Eksempel 1
Til en oppløsning av 2-hydroksymetyl-3-metyl-4-(2,2,3,3,3-pentafluorpropoksy)pyridin (350 mg) i kloroform (10 ml) ble det tilsatt tionylklorid (0,2 ml). Blandingen ble oppvarmet under tilbakestrømming i 30 minutter, og ble deretter konsentrert. Resten ble oppløst i metanol (4 ml). Oppløsningen ble tilsatt til en blanding av 2-merkapto-benzimidazol (200 mg), 28% natriummetoksydoppløsning (1 ml) og metanol (6 ml), som ble oppvarmet under tilbakestrømming i 30 minutter. Fra den resulterende oppløsningen ble metanol fjernet ved inndamping. Til resten ble det tilsatt vann, og oppløsningen ble underkastet ekstraksjon med etylacetat. Ekstraktet ble vasket med en fortynnet natriumhydroksyd-oppløsning, etterfulgt av tørking på magnesiumsulfat. Fra den resulterende oppløsningen ble oppløsningsmidlet fjernet ved fordamping. Resten ble så kromatografert på en kolonne av silikagel (20 g), eluert med etylacetat-heksan (2:1), og deretter rekrystallisert fra etylacetat-heksan, slik at man fikk 370 mg 2-[3-metyl-4-(2,2,3,3,3-pentafluorpropoksy)pyrid-2-yl]metyltiobenzimidazol.1/2 hydrat i form av fargeløse plater, smeltepunkt 145-146°C.
Ved en fremgangsmåte svarende til den ovenfor angitte hie forbindelser (I) (n = 0) fremstilt ved å la forbindelser (II) reagere med forbindelser (III).
Eksempel 2
Til en oppløsning av 2-[3-metyl-4-(2,2,3,3,3-pentafluor-propoksy )pyrid-2-yl]metyltiobenzimidazol (2,2 g) i kloroform (20 ml) ble det under isavkjøling i løpet av et tidsrom på 30 minutter dråpvis tilsatt m-klorperbenzosyre (1,3 g) oppløst i kloroform (15 ml). Oppløsningen ble vasket med en mettet vandig oppløsning av natriumhydrogenkarbonat, deretter tørket over magnesiumsulfat og konsentrert. Resten ble kromatografert på en kolonne av silikagel (50 g), eluert med etylacetat og deretter rekrystallisert fra aceton-isopropyleter, slik at man fikk 1,78 g 2-[3-metyl-4-(2,2,3,3,3-pentafluorpropoksy)-pyrid-2-yl]metylsulfinylbenzimidazol som blekt gule prismer, smeltepunkt 161-163°C (dekomponering).
Ved en fremgangsmåte svarende til fremgangsmåten ovenfor ble forbindelser (I) (n = 1) fremstilt fra forbindelser (I) (n = 0).

Analogifremgangsmåte for fremstilling av terapeutisk aktivie pyridinderivater med formelen:
hvor R<1> er hydrogen, metoksy eller trifluormetyl, R<2> og R<3> er uavhengig hydrogen eller metyl, R<4> er C2- 5 fluorert alkyl, og n er 0 eller 1, eller et farmakologisk akseptabelt salt derav, karakterisert ved at man lar en forbindelse med formelen:
