Description
Denne oppfinnelse vedrører en fremgangsmåte for fremstilling av nye høyredreiende spiro-hydantoin-forbindelser som er spesielt verdifulle med hensyn til deres evne til effektiv regulering av visse kroniske komplikasjoner som oppstår fra diabetes mellitus (f.eks. diabetisk cataracta og nevropati).
I de senere år er det utført forskjellige forsøk av
en rekke forskere på det området som består av organisk medisinsk kjemi på å oppnå nye og bedre orale antidiabetiske midler. I
de fleste tilfeller har disse forsøk medført synteser og testing av forskjellige hittil nye og utilgjengelige organiske forbindelser, spesielt på sulfonylurinstoffområdet, ved anstrengelser for å bestemme deres evne til å nedsette nivået av blodsukker (dvs. glukose) i en vesentlig stor grad ved oral administrering.
Ved forskningen efter nyere og enda mer effektive antidiabetiske midler er imidlertid lite kjent om virkningen av andre organiske forbindelser til å forhindre eller stanse visse kroniske komplikasjoner av diabetes, så som diabetisk cataracta, nevropati og retinopati, etc. Ikke desto mindre så åpenbarer K. Sestanj et
al. i US-patent 3.821.383 at visse aldose-reduktase-inhibitorer,
så som 1,3-diokso-lH-benz[d,e]-isokinolin-2(3H)-eddiksyre og visse nær beslektede derivater derav, er nyttige for disse formål, selv om disse spesielle forbindelser ikke er kjent for å være hypoglykemiske i og for seg. Disse spesielle aldose-reduktase-inhibitorer virker alle ved å inhibere aktiviteten til enzymet aldose-reduktase, som primært er ansvarlig for reduksjonen av aldoser (så som glukose og galaktose) til de tilsvarende polyoler (så som sorbitol og galaktitol) i menneskelegemet. På denne måte kan uønskede oppsamlinger av galaktitol i linsene til galaktosemiske personer og sorbitol i linsene, periferiske nerve-strenger og nyrene til forskjellige diabetiske personer hindres eller på annen måte reduseres efter som omstendighetene er. Som et resultat av dette er disse forbindelseravgjort av verdi som aldose-reduktase-inhibitorer ved regulering av visse kroniske diabetiske komplikasjoner, innbefattet slike som er av okular natur, siden det allerede er kjent i industrien at nærvær av polyoler i øyelinsene uavvendelig fører til cataracta-dannelse sammen med et ledsagende tap av linseklarhet.
I samsvar med foreliggende oppfinnelse er det heller overraskende blitt funnet at visse nye høyredreiende spiro-hydantoinforbindelser er ytterst nyttige når de anvendes i terapi som aldose-reduktase-inhibitorer ved regulering av kroniske komplikasjoner som oppstår hos en diabetisk person. Disse nye høyredreiende isomerer er alle langt mer aktive i denne henseende enn de tilsvarende dl-forbindelser hvorfra de er avledet, på tross av det faktum at de samme dl-, d- og l-former alle er funnet å være i alt vesentlig ekvipotente som antikonvulsive midler.
De nye forbindelser fremstilt i henhold til denne oppfinnelse er alle valgt fra gruppen bestående av de høyre-dreiende former av asymmetriske spiro-hydantoiner med formelen:
og alkali- og jordalkalisaltene derav, hvor Y er oksygen eller svovel. Forbindelsene som fremstilles ifølge denne oppfinnelse er således henholdsvis d-6-fluor-spiro-[kroman-4,4<1->imidazolidin]-2',5'-dion og d-6<1->fluor-spiro-[imidazolidin-4,4'-tiokroman]-2,5-dion. Disse forbindelser er begge ytterst kraftige med hensyn til deres aldose-reduktase-inhiberende aktivitet, i tillegg til å vaere like effektive til å nedsette sorbitol-mengdene i sittebensnerven og linsene til diabetiske personer og galaktitol-mengdene i linsene til galaktosemiske personer i en meget betydelig høy grad.
I henhold til den fremgangsmåte som anvendes ifølge oppfinnelsen for å oppnå de nye høyredreiende forbindelser, blir et tilsvarende racemisk eller dl-spiro-hydantoin som har den samme strukturforme 1 som tidligere angitt for den optisk aktive forbindelse, bragt i kontakt med minst én ekvimolar mengde av 1-brucin eller et lignende optisk aktivt alkaloid, så som cinchonidin, i et egnet reaksjonsinert, organisk løsningsmiddel, fortrinnsvis valgt fra den gruppe som består av lavere alkanoler. De resulterende diastereoisomere salter separeres så ved hjelp av fraksjonert krystallisering og det minst løselige salt blir omdannet til det ønskede optisk aktive spiro-hydantoin ved hjelp av spaltning med syre på konvensjonell måte.
I henhold til en mer detaljert betraktning av den forannevnte fremgangsmåte for spaltning i henhold til denne oppfinnelse, så blir det første trinn som fører til dannelse av diastereo-isomerene, fortrinnsvis utført i et lavere-alkanol-løsningsmiddel med fra ett til tre karbonatomer ved anvendelse av 1-brucin som det valgte spaltningsmiddel. I praksis er det vanligvis foretrukket å anvende ekvimolare mengder av racemisk forbindelse og spaltningsmiddel for å nedsette omkostningene til et minimum og oppnå maksimal renhet for produktet, men det kan anvendes et svakt overskudd av alkaloid uten påviselig påvirkning av resul-tatet av saltdannelsestrinnet eller av naturen til det endelige oppnådde produkt. Det er unødvendig å si at tiden ikke er kritisk i denne forbindelse og den er nødvendigvis avhengig av naturen til utgangsmaterialene, deres konsentrasjon i løsning og de virkelige temperaturer som anvendes. Efter avslutning av saltdannelsestrinnet separeres vanligvis den ønskede diastereoisomer fra blandingen ved fraksjonert krystallisering og til dette er det vanligvis nødvendig med en periode på fra ca. 2 til ca. 24 timer innen et krystalliserings-temperaturområde på fra ca. -20 til ca. 60°C
for de nærværende formål. Diastereoisomeren blir så ytterligere renset ved hjelp av omkrystallisering, fortrinnsvis ved anvendelse av samme type alkanol-løsningsmiddel som tidligere ble anvendt ved saltdannelsestrinnet, inntil det er oppnådd full optisk renhet, dvs. som bevist ved et konstant smeltepunkt og konstant optisk rotasjon for den forannevnte diastereoisomer.
Omdannelse av de således oppnådde alkaloidale salter
til de ønskede optisk aktive hydantoiner blir så utført på lett måte ved hjelp av spaltning med syre, og fortrinnsvis ved anvendelse av standardteknikkene ved sur hydrolyse. Saltet kan f.eks. behand-les i et vandig medium med en mineralsyre så som svovelsyre, saltsyre, bromhydrogensyre eller jodhydrogensyre eller med en organisk syre så som en lavere alkansyre så som eddiksyre eller en halogenert lavere alkansyre, f.eks. Ø-klorpropionsyre eller trikloreddiksyre. I praksis er det mest bekvemt å anvende en fortynnet vandig syre for nærværende formål, og svovelsyre og saltsyre foretrekkes vanligvis som de sure komponenter i denne
forbindelse. For å lette hydrolysetrinnet ytterligere anvendes det fortrinnsvis et egnet vannublandbart organisk løsningsmiddel så som en lavere alkylalkansyreester som etylacetat i forbindelse med det forannevnte fortynnede vandige sure medium, hvorved den ønskede optisk aktive spiro-hydantoinforbindelse (dvs. den høyre-dreiende isomer) bekvemt ekstraheres inn i det organiske skikt og derefter isoleres ved konvensjonelle midler.
De dl-spiro-hydantoinforbindelser som anvendes som substrater ved spaltningsprosessen ved foreliggende oppfinnelse, syntetiseres alle lett ved først å kondensere en passende karbonyl-ring-forbindelse, så som det tilsvarende 4-kromanon eller tio-kroman-4-on, med formelen:
hvor Y er som tidligere angitt, med et alkalimetallcyanid (f.eks. natriumcyanid eller kaliumcyanid) og ammoniumkarbonat for å danne det ønskede endelige spiro-hydantoinprodukt (dvs. racemisk forbindelse) med den strukturformel som tidligere er angitt. Denne spesielle omsetning blir vanligvis utført i nærvær av et reaksjonsinert polart organisk løsningsmiddelmedium i hvilket både reaktantene og reagensene er gjensidig blandbare. Foretrukne organiske løsnings-midler for anvendelse i denne forbindelse innbefatter cykliske etere så som dioksan og tetrahydrofuran, lavere alkylenglykoler, så som etylenglykol og trimetylenglykol, vannblandbare lavere alkanoler, så som metanol, etanol og isopropanol, og også N,N-di(lavere alkyl)lavere alkanamider, så som N,N-dimetylformamid, N,N-dietylformamid og N,N-dimetylacetamid, etc. Omsetningen blir vanligvis utført ved en temperatur som ligger i området fra ca. 20 opptil ca. 120°C i en periode på fra ca. 2 timer til ca. 4 dager. Selv om mengden av reaktanter og reagenser som anvendes ved omsetningen, kan varieres i en viss utstrekning, så er det foretrukket å anvende i det minste et svakt molart overskudd av alkalimetallcyanid-reagenset i forhold til karbonylring-utgangsforbindelsen for å oppnå maksimalt utbytte. Efter full-føring av omsetningen isoleres det ønskede produkt lett på en konvensjonell måte, f.eks. ved først å fortynne reaksjonsblandingen med vann (om nødvendig kokende) og så avkjøle den resulterende vandige løsning til romtemperatur, fulgt av surgjøring for å
oppnå den spesielle dl-spiro-hydantoinforbindelse i form av en lett utvinnbar utfelling.
De utgangsmaterialer som er nødvendige for fremstilling av de forannevnte dl-spiro-hydantoinforbindelser er for det meste kjente forbindelser som lett kan syntetiseres av fagfolk i industrien ved å gå ut fra vanlige kjemiske reagenser og anvende konvensjonelle organiske syntesemetoder. F.eks. er 6-fluor-tiokroman-4-on en kjent forbindelse, mens 6-fluor-4-kromanon lett oppnås ved kondensering av 0-(p-fluorfenoksy)propionsyre i nærvær av polyfosforsyre. Den sistnevnte organiske syre, anvendt som utgangsmateriale i denne forbindelse, er opprinnelig utledet fra en kommersielt tilgjengelig forbindelse.
De alkali- og jordalkalibaser som anvendes som reagenser ved denne oppfinnelse for å fremstille de forannevnte farmasøytisk godtagbare alkali- og jordalkalisalter, er slike som danner ikke-toksiske salter med de her beskrevne sure høyredreiende spiro-hydantoinforbindelser, så som f.eks. d-6-fluor-spiro-[kroman-4,4'-imidazolidin]-2<1>,5<1->dion. Disse spesielle ikke-toksiske alkali- og jordalkalisalter har en slik natur at deres kationer sies å være i alt vesentlig ikke-toksiske av karakter over det vide doseringsområdet hvorved de administreres. Eksempler på slike kationer innbefatter de av natrium, kalium, kalsium og magnesium etc. Disse salter kan lett fremstilles ved ganske enkelt å behandle de forannevnte d-spiro-hydantoinforbindelser med en vandig løsning av de ønskede farmakologisk godtagbare kation, og så inndampe den resulterende løsning til tørrhet mens den fortrinnsvis er satt under redusert trykk. De kan alternativt også fremstilles ved å blande sammen lavere alkanoliske løsninger av nevnte sure forbindelser og det ønskede alkali- eller jord-alkalimetallalkoksyd, og så inndampe den resulterende løsning til tørrhet på samme måte som før. I begge tilfeller må det anvendes støkiometriske mengder av reagenser for å ha sikkerhet for at omsetningen fullføres og at det oppnås maksimalt utbytte med hensyn til det ønskede endelige produkt.
Som tidligere angitt er alle de høyre-dreiende spiro-hydantoinforbindelser fremstilt i henhold til denne oppfinnelse lett tillempelige til terapeutisk anvendelse som aldose-reduktase-inhibitorer for regulering av kroniske diabetiske komplikasjoner, med hensyn på deres evne til å redusere sorbitol-mengdene i linser hos diabetiske personer i.en statistisk betydelig grad. For eksempel er det blitt funnet at d-6-fluor-spiro-[kroman-4,4'-imidazolidin]-2',5'-dion, som er et typisk og foretrukket middel, konsekvent regulerer (dvs. inhiberer) dannelsen av sorbitol-mengder i diabetiske rotter i en betydelig høy grad ved oral administrering med dosemengder som ligger i området fra 0,25
til 5,0 mg/kg, uten å vise noen vesentlige tegn på toksiske bivirkninger. Den andre forbindelsen samt saltene fremstilt i henhold til denne oppfinnelse gir også lignende resultater. De her beskrevne forbindelser fremstilt i henhold til oppfinnelsen kan videre administreres ved enten oral eller parenteral administrering, f or nærværende formål, uten å forårsake noen betydelige uheldige farmakologiske bivirkninger i den person hvortil de administreres. Disse forbindelser blir vanligvis administrert i doser innen området fra ca. 0,25 til ca. 5,0 mg pr. kg kropps-vekt pr.-dag, selv om det nødvendigvis vil forekomme variasjoner avhengig av vekten og tilstanden til den person som blir behandlet og den spesielle administrasjonsmåte som velges.
I forbindelse med anvendelsen av de høyredreiende spiro-hydantoinforbindelser fremstilt i henhold til denne oppfinnelse ved behandling av diabetiske personer, så skal det bemerkes at disse forbindelser kan administreres enten alene eller i kombinasjon med farmasøytisk godtagbare bærere ved hvilken som helst av de måter som tidligere er angitt.
Aktiviteten til forbindelsene fremstilt i henhold til foreliggende oppfinnelse, som midler for å regulere kroniske diabetiske komplikasjoner, bestemmes ved deres evne til å tilfreds-stille én eller flere av de følgende standardbiologiske og/eller farmakologiske tester, henholdsvis (1) måling av deres evne til å inhibere enzymaktiviteten i isolert aldose-reduktase, (2) måling av deres evne til å redusere eller inhibere sorbitol-oppsamling i sittebensnerven til akutt streptozotociniserte (dvs. diabetiske) rotter, (3) måling av deres evne til å reversere allerede forhøyede sorbitol-mengder i sittebensnerven og linsene til kronisk streptozotocin-indusertediabetiske rotter, (4) måling av deres evne til å forhindre eller inhibere galaktitoldannelse i linsene til akutt galaktosemiske rotter og (5) måling av deres evne til å utsette cataracta-dannelse og redusere graden av linse-uklarhet hos kronisk galaktosemiske rotter.
Preparat A
En blanding bestående av 3,5 g (0,019 mol) med j3-(p-fluorfenoksy)-propionsyre [Finger et al., journal og the American Chemical Society, Vol.81, s. 94 (1959)] og 40 g polyfosforsyre ble oppvarmet på et dampbad i en periode på 10 minutter og ble så hellet inn i 300 ml isvann. Den resulterende vandige blanding ble deretter ekstrahert med tre separate porsjoner av etylacetat, og de samlede organiske sjikt ble deretter vasket med fortynnet vandig natriumbikarbonatløsning og så med vann, fulgt av tørking over vannfritt magnesiumsulfat. Etter fjerning av tørkemidlet ved hjelp av filtrering og løsningsmidlet ved hjelp av inndampning under redusert trykk, ble det til sist oppnådd en rest som deretter ble omkrystallisert fra etanol for å gi 2,93 g (93%) med rent 6-fluor-4-kromanon, sm.p. 114-116°C.

Preparat B
En blanding bestående av 397 g (2,39 mol) med 6-fluor-4-kromanon (fremstilt som beskrevet under preparat A), 233 g (3,58 mol) kaliumcyanid og 917 g (9,56 mol) pulverisert ammoniumkarbonat i 3000 ml med 50 %ig vandig etanol ble oppvarmet ved 65°C i en periode på tilnærmet 63 timer. Reaksjonsblandingen ble så avkjølt til romtemperatur (■^25°C), fortynnet med 2000 ml vann og deretter surgjort med 6N saltsyre. De således oppnådde lysegule krystaller ble deretter oppsamlet ved hjelp av sugefiltrering, vasket godt med vann og deretter oppløst i 2N vandig natrium-hydroksydløsning. Ekstrahering av sistnevnte løsning med tre 1000 ml<1>s porsjoner etylacetat, fulgt av surgjøring av den basiske vandige fase med 6N saltsyre ga så lysegule krystaller som igjen ble vasket med vann og lufttørket til konstant vekt. Etter om-kr,ystallisering fra kokende metanol (opprinnelig volum på 9 liter ble redusert til 5 liter), ble det oppnådd rent dl-6-fluor-spiro-[kroman-4,4'-imidazolidin ]-2',51-dion (sm.p. 239-241°C) medet utbytte på 276 g (44%). En ytterligere mengde (82 g) av krystaller oppnådd fra filtratet økte utbyttet av rent materiale til 64%.
Preparat C
Fremgangsmåten beskrevet under preparat B ble gjentatt, bortsett fra at 191 g (1,05 mol) med 6-fluortiokroman-4-on [Chemical Abstracts, Vol. 70, s. 47335x (1969)], 102 g (1,57 mol) kaliumcyanid og 391 g (4,08 mol) med pulverisert ammoniumkarbonat ble .omsatt i 1000 ml med 50 %ig vandig etanol ved 65°C (ved anvendelse av et oljebad) i en periode på tilnærmet 66 timer. Reak-sjonsblandingen ble så hellet inn i 1500 ml vann og kokt i 15 minutter for å ødelegge overskudd av ammoniumkarbonat. Etter av-kjøling til romtemperatur ble den surgjort med konsentrert saltsyre og så opparbeidet på samme måte som beskrevet for den tilsvarende blanding under preparat B. På denne måte ble det til sist oppnådd 224 g (85%) med rent dl-6 '-f luor-spiro-[imidazolidin-4,4 ' - tiokroman ]-2,5-dion (sm.p. 200-202°C) uten at det var nødvendig med noe omkrystalliseringstrinn.
Eksempel I
En løsning bestående av 120 g (0,508 mol) med dl-6-fluor-spiro- [kroman-4 ,4 ' -rimidazolidin ]-2 1 , 5 ' -dion (sm.p. 239-241°C ) og 23 7 g (0,508 mol) med 1-brucin-tetrahydrat oppløst i 1,8 liter med kokende etanol ble hensatt for å avkjøles sakte, og de utfelte krystaller (A) ble deretter oppsamlet ved hjelp av sugefiltrering, og det resulterende filtrat (B) ble deretter tatt vare på. Krystallene (A) bestod av 1-brucin-saltet av d-6-fluor-spiro-[kroman-4,4<1->imidazolidin]-2',5<1->dion, isolert som etanolatet, sm.p. 114-118°C (spalt.) etter omkrystallisering fra etanol.
Analyse Beregnet for ciiH9FN2°3'C23<H>26<N>2°4<-C>2<H>5OH:
C, 63,88; H, 6,12; N, 8,28. Funnet: C, 63,60; H,6,07; N, 8,22.
Etter ytterligere omkrystallisering av de ovennevnte krystaller (A) fra etanol (1,5 liter), ble den forannevnte diastereoisomer behandlet med 1,0 liter etylacetat og 1,0 liter med IN vandig saltsyre. Det fraskilte organiske sjikt ble så oppsamlet, tørket over vannfritt magnesiumsulfat, filtrert og deretter konsentrert i vakuum for å gi et gjenværende fast materiale. Dette sistnevnte residuum ble så krystallisert fra 1,0 liter etanol for å gi 45 g med urenset produkt, nemlig den høyredreiende isomer av 6-fluor-spiro-[kroman-4,4'-imidazolidin]-2<1>,5<1->dion. Omkrystallisering av det sistnevnte materiale fra 300 ml etanol ga så 37 g (62%) med rent d-6-fluor-spiro-[kroman-4,4<1->imidazolidin]-2',5'-dion, sm.p. 241-243°C, [ a]^ 5°+ 54,0° (c = 1 i metanol).

N, 11,86. Funnet: C, 55,59; H, 3,88; N, 11,52.
Det opprinnelige filtrat (b) ble så behandlet med 75 ml
av 10 %ig vandig saltsyre og de utfelte krystaller ble deretter oppsamlet på vanlig måte for å gi 1-brucin-saltet av 1-6-fluor-spiro- [kroman-4,41-imidazolidin]-2',51-dion, isolert som mono-hydroklorid-dihydratet, sm.p. 172-174°C.
Analyse Beregnet for C1:LHgFN203 •C23H26°4 *HCl" 2H2°:
C, 58,07; H, 5,73; N, 7,97. Funnet: C, 58,05; H, 5,79; N, 7,98.
Denne spesielle diastereoisomer (sm.p. 172-174°c) ble så behandlet med 1,0 liter etylacetat og 600 ml med lo %ig vandig svovelsyre og det resulterende organiske sjikt ble deretter fra-skilt, tørket over vannfritt magnesiumsulfat, filtrert og deretter konsentrert i vakuum for å gi 41 g av den urensede 1-isomer. Omkrystallisering av det sistnevnte materiale fra 400 ml etanol

Eksempel li
En løsning bestående av 2,52 g (0,01 mol) med dl-6<1->fluor-spiro- [imidazolidin-4,4'-tiokroman]-2,5-dion (sm.p. 200-202°C) og 4,3 g (0,01 mol) med 1-brucin-dihydrat oppløst i 125 ml med kokende etanol ble hensatt for å avkjøles sakte, og de utfelte krystaller (A) ble deretter oppsamlet ved hjelp av sugefiltrering og det resulterende filtrat (B) ble deretter tatt vare på. De ovennevnte krystaller (A) ble så ristet med 100 ml etylacetat og 200 ml med 3N vandig saltsyre for å omdanne den forannevnte diastereoisomer til det tilsvarende optisk aktive hydantoin. Det fraskilte organiske sjikt oppnådd på denne måte ble så oppsamlet, tørket over vannfritt magnesiumsulfat og filtrert,
og det resulterende filtrat ble deretter konsentrert i vakuum for
å gi et fint gjenværende materiale. Omkrystallisering av det sistnevnte materiale fra 20 ml etanol ga så 230 mg (18%) av rent d-6 1 - fluor-spiro- [imidazolidin-4,41-tiokroman]-2,5-dion, sm.p. 224-226°C, [a]25 + 71,8° (c i metanol).

C, 62,40; H, 5,97; N, 8,09. Funnet: C,62,21; H, 5,94; N, 8,09.
Denne spesielle diastereoisomer (sm.p. 120-124°C) ble så ristet med 100 ml etylacetat og 200 ml med IN vandig saltsyre, og det resulterende organiske sjikt ble deretter separert, tørket over vannfritt magnesiumsulfat, filtrert og etterpå inndampet til tørrhet mens det ble holdt under redusert trykk. Eksempel III
Natriumsaltet av d-6-fluor-spiro-[kroman-4,4<1->imidazolidin ]-2 ', 5 ' -dion (produktet fra eksempel I) fremstilles ved å oppløse nevnte forbindelse i vann inneholdende en ekvivalent mengde i mol av natriumhydroksyd og så fryse-tørke blandingen. på denne måte oppnås det ønskede metallsalt av hydantoin i form av et amorft pulver som er lett løselig i vann.
på samme måte blir også kalium- og litiumsaltene fremstilt, og likeledes alle alkalimetallsaltene av d-6'-fluor-spiro-[imidazolidin-4,4'-tiokroman]-2,5-dion (produktet fra eksempel II).
Eksempel IV
Kalsiumsaltet av d-6-fluor-spiro- [imidazolidin-4,41 - tiokroman ]-2,5-dion (produktet fra eksempel II) blir fremstilt ved oppløsning av nevnte forbindelse i vann inneholdende en ekvivalent mol-mengde av kalsiumhydroksyd og så fryse-tørke blandingen. Det tilsvarende magnesiumsalt blir også fremstilt på denne måte, og også alle de andre jordalkalimetallsaltene, ikke bare av denne spesielle forbindelse, men også av d-6'-fluor-spiro-[kroman-4,41 - imidazolidin]-2',5'-dion (produktet fra eksempel I).
De følgende spiro-hydantoinforbindelser av henholdsvis preparatene B og C og fra eksemplene I-II ble testet med hensyn til deres evne til å redusere eller inhibere aldose-reduktase-enzymaktivitet ved fremgangsmåten til S. Hayman et al., så som beskrevet i Journal of Biological chemistry, Vol. 240, s. 877
(1965) og som modifisert av K. Sestanj et al., i US-patentskrift nr. 3.821.383. I hvert tilfelle var det anvendte substrat parsielt renset aldose-reduktase-enzym oppnådd fra kalvelinser. De oppnådde resultater er for hver forbindelse uttrykt nedenfor som prosent inhibering av enzym-aktivitet ved de forskjellige testede konsentrasjonsnivåer:
De følgende spiro-hydantoinforbindelser fra henholdsvis preparatene B og C og eksemplene I-II ble testet med hensyn til deres evne til å redusere eller inhibere sorbitol-oppsamling i sittebensnerven til streptozotociniserte (d.v.s. diabetiske)
rotter ved fremgangsmåten i alt vesentlig beskrevet i US-patentskrift nr. 3.821.383. Ved denne undersøkelse ble mengden av sorbitol-oppsamling i sittebensnerven målt 2 7 timer etter indusering
av diabetes. Forbindelsene ble administrert oralt med de angitte dosis-mengder 4, 8 og 24 timer etter administrasjonen av streptozotocin. Claims (1)
- Analogifremgangsmåte for fremstilling av den terapeutisk aktive, høyredreiende isomer av en asymmetrisk spiro-hydantoin-forbindelse med formelen:hvor Y er oksygen eller svovel og dens alkali- og jordalkalisalter,karakterisert ved å(l) bringe det tilsvarende racemiske eller dl-spiro-hydantoin som har samme strukturformel i kontakt med minst én ekvimolar mengde av et optisk aktivt alkaloid, fortrinnsvis 1-brucin, i et reaksjonsinert organisk løsningsmiddel, (2) separere det resulterende par av diastereoisomere salter ved fraksjonert krystallisering og (3) deretter omdanne det mindre løselige salt til den tilsvarende optisk aktive spiro-hydantoin-forbindelse via syre-hydrolyse, og eventuelt fremstilles alkali- og jordalkalisaltene på kjent måte.
