Oxazafosforin-4-thioalkaanzuurverbindingen en farmaceutisch preparaat met antineopiastische eigenschappen
De uitvinding heeft betrekking op oxazafosforin-4-thioalkaanzuurverbindingen met antineopiastische 5 eigenschappen.
Dergelijke verbindingen zijn bekend uit een artikel van Hirano et al. in Tetrahedron Letters, 10 (1979), 883-886. In dit artikel is de bereiding van verschillende geactiveerde cyclofosfamidederivaten met functionele groepen beschreven. Hieronder bevinden zich cyclofosfamidederivaten met op de 4-plaats van het cyclofosfamide een carbonzuurakyleenthiogroep. Voorts wordt in het artikel een publicatie aangekondigd in 10 Makromol. Chemie (180 (4; 1979), blz. 1125-1131), waarin de immobilisatie van de beschreven verbindingen op polymere dragers, alsmede hun bevestiging aan tumorcel-specifieke immunoglobulines zullen worden beschreven. In Cancer.Res. 40 (7; 1980), 2263-2267, 40 (10; 1980), 3704-3708 en 40 (11, 1980), 4216-4220 is de anti-tumor werking nader toegelicht.
Derivaten van 4-hydroxioxazafosforine, die door hydrolyse in de actieve metaboliet 15 4-hydroxioxazafosforine kunnen worden omgezet, zijn geschikt als anti-tumorgeneesmiddel. Oxazafosforin-4-thioalkaancarbonzuurverbindingen zijn hier voorbeelden van.
Ten opzichte van de bekende, verwante handelsproducten cyclofosfamide en ifosfamide, waarvan de actieve metaboliet eveneens 4-hydroxioxazafosforine is, leveren oxazafosforin-4-thioalkaancarbonzuur-verbindingen geen verbetering van de therapeutische werking op, terwijl wel een toegenomen toxiciteit 20 waargenomen werd. Een mogeiijke verklaring hiervoor is dat de bij de hydrolyse vrijkomende mercaptoal-kaancarbonzuren toxicologisch ongunstig zijn.
Opmerking verdient het nog dat de actieve metaboliet 4-hydroxioaxazafosforine zelf extreem instabiel en derhalve ongeschikt is om voor toepassing als geneesmiddel in aanmerking te komen.
Gevonden is nu dat bepaalde oxazafosforin-4-thioalkaansulfonzuurverbindingen geschikt zijn als 25 anti-tumorgeneesmiddelen, een uitstekende thermische stabiliteit bezitten en een lagere toxiciteit bezitten dan het commercieel verkrijgbare cyclofosfamide. Bovendien hebben de mercaptoalkaansulfonzuren die worden verkregen bij de hydrolytische omzetting van oxazafosforin-4-thioalkaansulfonzuren volgens de uitvinding tot de actieve 4-hydroxioxazafosforines een uroprotectieve werking, die bij de bekende carbonzuurverbindingen ontbreekt.
30 De uitvinding wordt gekenmerkt, doordat de oxazafosforin-4-thioalkaanzuurverbindingen oxazafosforin-4-thioalkaansulfonzuren en neutrale zouten daarvan met de algemene formule 1 van het formuleblad zijn, waarin R.,, R2en R3 gelijk of verschillend kunnen zijn en waterstof, methyl, ethyl, 2-chloorethyl of 2-methaansulfonyloxiethyl voorstellen en waarbij ten minste twee van deze groepen 2-chloorethyl en/of 35 2-methaansulfonyloxiethyl zijn; R4 waterstof of methyl is; x een rechte of vertakte C2_e alkyleengroep is, die aan het koolstofatoom op de 2-, 3-, 4- of 5-plaats van de alkyleenketen een mercaptogroep kan dragen; Y+ een waterstofkation, een alkalimetaal of aardalkalimetaalkation, het guadinium-, morfolinium-, of 40 cyclohexylammoniumkation, of een kation dat is afgeleid van een amine met de formule NR5R6R7 voorstelt, waarbij de groepen R5-R7 gelijk of verschillend zijn en waterstof, C.,-C2 alkylgroepen of oxiethylgroepen voorstellen, dan wel waarin Y+ het ethyleendiammoniumkation H3N®-CH2CH2-NH3+ of het piperazoniumka-tion is; en z 1 is, indien Y+ een éénbasisch kation is, of 2 is, indien Y+ een tweebasisch kation of het kation van een 45 verbinding met twee éénbasische kationen is.
Opgemerkt wordt dat in de Europese octrooiaanvrage 2495 de farmaceutische combinatie van een cytostaticum op basis van een oxazafosforine zoals cyclofosfamide en sufosfamide en het natriumzout van 2-mercaptoethaan sulfonzuur is beschreven. Door de combinatie wordt de urotoxische nevenwerking van het cytostaticum praktisch voorkomen.
50 Vanwege hun goede toegankelijkheid en goede eigenschappen verdienen daarbij die verbindingen met de formule 1 de voorkeur, waarin Y® het guanidinium-, morfalinium- of cyclohexylammoniumkation of een kation dat is afgeleid van een amine met de formule NR5R6R7 is, waarin Rs - R7 gelijk of verschillend zijn en waterstof, C1-C2-alkylgroepen of oxiethylgroepen voorstellen, dan wel waarin Y® het ethyleendiammoniumkation H3të-CH2CH2-f5H3 of het piperazoniumkation is, en z is 1, indien Y® een eenbasisch kation of 2 is, 55 indien Y® een tweebasisch kation of het anion van een verbinding met twee éénbasische kationen is.
Andere gunstige eigenschappen hebben ook de kalium- en aardalkalimetaalzouten van de zuren met de formule 1 en ook deze verdienen derhalve de voorkeur.

Onder de met stikstofbasen geneutraliseerde zuren met de formule 1 verdienen bijzondere voorkeur de ammoniumzouten, waarin Y® het NH4®-kation is, de cyclohexylammoniumzouten, waarin Y® het C6H11NH3®-kation is, resp, de guanidiniumzouten, waarin Y® het l5H2=C(NH2)-kation is.
Van de onderhavige zouten is bijzonder gemakkelijk toegankelijk en dus een voorkeursverbinding het 5 cis-natrium-2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2,-oxazafosforin-4-yl]-thioëthaansulfonaat en het overeenkomstige ammonium-, cyclohexylammonium- en kaliumzout. Een bijzondere voorkeur gaat hierbij uit naar het 2-[2-(bis-(2-chloorethyl)-amino)-2-oxotetrahydro-2H-1,3,2,-oxazafosforin-4-yl]-thioëthaansulfonzuur-cyclohexylammoniumzout. Deze verbinding is zeer goed kristalliseerbaar en heeft een zeer lage toxiciteit.
10 De telling van de plaats van de mercaptogroep in de alkyleenketen X begint met het C-atoom direct naast de sulfonzuurgroep.
Van bijzonder belang zijn die verbindingen met de formule 1, waarin R, en R2 de 2-chloorethylgroep, R3 en R4 waterstof voorstellen, X een ethyleengroep en daarbij z = 1 en Y® = NH4®, |3h3-® <$) H of ®NH2=C(NH2)2, of X = (CH2)3, 15 z = 1 en daarbij Y = NH2=C(NH2)2 is.
De onderhavige oxazafosforine-4-thio-alkaansulfonzuren en hun neutrale zouten met de algemene formule 1 kunnen volgens de uitvinding bijvoorbeeld worden bereid door een 4-hydroxi- of 4-C,-4-alkoxioxazafosforine met de formule 2 waarin R,, R2, R3 en R4 dezelfde betekenis als bij formule 1 bezitten, en Z waterstof of C^-alkyl is, met een verbinding met de formule 3 20 HS-X-S03®, Y® waarin X en Y® de boven weergegeven betekenis bezitten, om te zetten en, indien Y® waterstof is, de verkregen oxazafosforine-4-thioalkaansulfonzuren met een base met een van de andere betekenissen van 25 Y® te neutraliseren.
De reactie wordt bij temperaturen tussen -60 en +80°C, bij voorkeur bij -30 tot +40°C uitgevoerd, d.w.z. eventueel onder koelen, bij kamertemperatuur of onder verwarmen. De omzetting kan bij aanwezigheid van een zure katalysator zoals een anorganisch of organisch zuur, zoals speciaal trichloorazijnzuur of een lewiszuur als A1C13, ZnC12 of TiC14 worden uitgevoerd.
30 Het kation Y® van een sulfonaat volgens formule 1 kan tegen een ander kation worden uitgewisseld bijvoorbeeld met een geschikt beladen ionenuitwisselaar. Deze uitwisseling is in die gevallen gunstig, waar een zout met een bepaald kation Y® moeilijk volgens het eerder weergegeven procédé kan worden bereid. Het gewenste zout kan zo uit een ander goed te bereiden zout met hoge opbrengsten worden gewonnen.
De nieuwe verbindingen met formule 1 worden door gebruikelijke opwerkingsmethoden voor dit soort 35 producten, speciaal door kristallisatie, neerslaan of chromatografisch zuiveren, speciaal over Sephadex (Pharmacia Fine Chemicals Ine.) geïsoleerd. De structuurbevestiging vindt plaats door het smeltpunt, dunneiaagchromatografie, elementenanalyse of IR- en NMR-spectraalanalyse.
De bij de onderhavige werkwijze als uitgangsmateriaal gebruikte verbindingen zijn bekend, kunnen kristallijn of als ruw product worden toegepast en kunnen als volgt op een bekende wijze worden gesynthe-40 tiseerd: 4-hydroxi-oxazofosforinen worden door een reductie van 4-hydroxiperoxiderivaten verkregen (zie b.v. J. Am. Chem. Soc. 95, 985-986 (1973) en Duits Offenlegungsschrift 2.317.178).
4-alkoxi-oxazafosforinen vormen zich bij een zure katalyse uit de hydroxiderivaten in de passende alcohol. De thiolen worden door een omzetting van het overeenkomstige natriumbroomalkaansulfonaat met 45 thioureum in het thiouroniumzout en een splitsing daarvan met ammoniak alsmede omzetting in het gewenste zout van het mercaptoalkaansulfonaat gewonnen.
Indien Y® een alkalimetaalkation is, gaat het speciaal om het natrium- of kaliumzout; indien Y® een aardalkalimetaalkation is, gaat het speciaal om het neutrale calcium- of magnesiumzout; indien Y® een cyclohexylammoniumkation is gaat het om het kation 06ΗιηΝΗ3®.
50 Onder de onderhavige oxazafosforinederivaten met de formule 1 worden alle vier mogelijke stereo-ïsomeren, d.w.z. de beide racemische cis-isomeren (2RS, 4RS) en de beide racemische trans-isomeren (2RS, 4RS) en de vier gescheiden optisch actieve cis-isomeren (2R, 4S en 2S,4S) alsmede de trans-isomeren (2R, 4R en 2S, 4S) alsmede mengsels daarvan verstaan. De cisArans-mengsels kunnen op een bekende wijze, bij voorkeur door een gefractioneerde kristallisatie worden gescheiden. Optisch actieve 55 verbindingen kunnen volgens een gebruikelijke methode van racemaatsplitsing, bijvoorbeeld door een gefractioneerde kristallisatie, van de diastereoïsomere zouten van het racemische sulfonzuur met de formule 1 met optisch actieve basen of eventueel door een toepassing van optisch actieve uitgangsmaterialen met 3 192741 de formule 2 bij de synthese worden verkregen.
Over het algemeen ontstaan bij de synthese cis/trans-mengsels. Daaruit verkrijgt men bij goed kristalliserende verbindingen cis- resp. trans-vorm, speciaal de cis-vorm, kristallijn. Voert men echter de omzetting in watervrije oplosmiddelen of in oplosmiddelen met een gering watergehalte uit, dan verkrijgt men uitsluitend 5 of overwegend één enkele vorm, speciaal de cis-vorm. Zo kan men de zuivere cis-vorm van een niet of slecht kristalliserende verbinding volgens formule 1 bijvoorbeeld bereiden door een acetonoplossing van de verbinding met formule 2 aan een waterige oplossing van een verbinding met de formule 3 bij temperaturen tussen -30 en +20°C toe te voegen en na afloop van de omzetting enige malen neer te slaan.
De uitgangsverbindingen met de formule 2 kunnen als racemische cis- en trans-isomeren (bereiding zie 10 boven) als optisch actieve cis- en trans-vorm en als mengsels daarvan worden toegepast. Optisch actieve verbindingen met de formule 2 verkrijgt men bijvoorbeeld uit de zouten van de optisch actieve sulfonzuren (bereiding zie boven), door ze tot de overeenkomstige optisch actieve 4-dioxioxazafosforinen met de formule 2 te hydrolyseren (in water bijvoorbeeld tussen 0 en 50°C bij voorkeur 5-30°C) en het vrijkomende thiol met de formule 3 tot disulfide te oxideren (bijvoorbeeld met jood of waterstofperoxide in water). Een verdere 15 methode gaat uit van een gesplitst optisch actief cyclofosfamidederivaat volgens voorbeeld 16 resp. 33 van de niet-voorgepubliceerde Nederlandse octrooiaanvrage 8104093 en hydrolyseert deze verbinding tot optisch actief 4-hydroxi-cyclofosfamide. Bij een verdere werkwijze wordt b.v. optisch actief cyclofosfamide (Offenlegungsschrift 2.944.106) volgens een bekend procédé (Peter et al, Cancer Treatment Reports 60, 429 (1976)) in optisch actief 4-hydroxicyclofosfamide omgezet.
20 Een verdere werkwijze voor het bereiden van oxazafosforine-4-thio-alkaansulfonzuren en hun neutrale zouten vindt plaats door een verbinding met de formule 2, waarin R,, R2, R3 en R4 de boven weergegeven betekenis bezitten, en Z waterstof of 0,-0,,-alkyl is, met een verbinding met de formule 3 om te zetten, waarin Y® van een optisch actieve base is afgeleid, die niet met een reeds gegeven definitie van Y overeenkomt, en na deze omzetting deze andere optisch actieve base tegen waterstof of een base volgens 25 de weergegeven definitie van Y uit te wisselen.
De onderhavige verbindingen kunnen bij het bestrijden van kanker en voor een immunosuppressie worden gebruikt. Ze bezitten een sterke antitumor-activiteit. Ze munten uit door een hoge activiteit bij parenterale en orale toediening, alsmede door geringe algemeen giftige verschijnselen. Ze bezitten in vivo een hoge carcinotoxische selectiviteit en in vitro een hoge cytotoxische specificiteit. Voor het vaststellen van 30 de carinotoxische activiteiten werden de stoffen bij dierexperimenten op een reeks tumoren met verschillende chemoresistentie onderzocht. Voor de beoordeling werd met behulp van een probietanalyse uit het verband tussen de logaritme van de doses en de frequentie van genezen en overlevende proefdieren de gemiddelde curatief werkzame doses (DC 50 in mg/kg) bepaald. Als vergelijkingsstof, die aan de onderhavige producten qua chemische opbouw verwant is, fungeerde de handelsverbinding cyclofosfamide.
35 Bij lymfatische leukemie L5222 bij de rat (stam BD IX) lag de gemiddelde curatieve dosis, bij een eenmalige intraveneuze toediening op de 5e dag na enting met de leukemie, voor de onderhavige producten evenals door cyclofosfamide bij 1,5 mg/kg.
Bij het chemosensitieve Yoshida-ascites-carcinosarcoom (lijn AH 13) van de rat (stam Sprague-Dawley) lag deze DC 50 voor de onderhavige producten evenals voor de vergelijkingsstof cyclofosfamide bij 1 40 mg/kg.
Op analoge wijze werd de toxische activiteit onderzocht, waarbij uit een verband tussen de logaritme van de doses en de overlijdensfrequentie de gemiddelde letale dosis (DL 50 in mg/kg) werd bepaald.
Voor de onderhavige producten lag deze DL 50 bij eenmalige intraveneuze toediening omstreeks 300 mg/kg. De ter vergelijking bepaalde gemiddelde letale dosis van cyclofosfamide bij een eenmalige 45 intraveneuze gift lag bij 244 mg/kg. De onderhavige producten bezitten dus een ca. 20% geringere acute giftigheid resp. bij een gelijke curatieve activiteit een overeenkomstig hogere therapeutische breedte.
Voor een onderzoek op de cytotoxische specificiteit in vitro werden telkens 3 x 107 steriel gewonnen cellen van het chemoresistente Yoshida-ascites-carcinosarcoom (lijn AH 13, stam ASTA) met oplopende concentraties van de onderhavige producten, 2 uren bij 37°C geïncubeerd en na enige malen uitwassen in 50 de buikholte van onbehandelde proefdieren geïmplanteerd. Uit het verband tussen de logaritme van de concentraties en de frequentie van zich ontwikkelende tumoren werd met behulp van de probietanalyse de gemiddelde cytotoxische dosis (CE 50 in mg/dm3) bepaald. Onder deze proefvoorwaarden lag de CE 50 van de onderhavige producten tussen de 3 en 5 mg/dm3.
Omdat cyclofosfamide als eerste inactieve transportvormverbinding geen cytotoxische activiteit bezit, 55 werd bij deze proeven ais vergelijkingsstof de actieve primaire metaboliet 4-hydroxicyclofosfamide toegepast, die in het lichaam door een enzymatische activering, hoofdzakelijk in de lever, wordt gevormd. Voor deze vergelijkingsstof ligt de gemiddelde cytotoxische concentratie eveneens bij 5 mg/dm3.

De onderhavige verbindingen zijn voor het bereiden van farmaceutische preparaten en bereidingsvormen geschikt. Deze farmaceutica resp. geneesmiddelen bevatten als actieve stof een of meer van de onderhavige verbindingen, eventueel gemengd met andere farmacologisch of farmaceutisch actieve stoffen. De bereiding van deze geneesmiddelen vindt op een bekende wijze plaats, waarbij de bekende en gebruikelijke 5 farmaceutische hulpstoffen alsmede verdere gebruikelijke drager- en verdunningsmiddelen kunnen worden gebruikt.
Voor het bereiden van oplossingen komen bijvoorbeeld water of fysiologisch geschikte organische oplosmiddelen in aanmerking.
Bij de bereiding van de preparaten kunnen bekende en gebruikelijke oploshulpmiddelen of emulgatoren 10 worden gebruikt.
Bovendien is een toeslag van conserveermiddelen, stabilisatoren, bufferstoffen, zoals calciumwaterstof-fosfaat, colloïdaal aluminiumhydroxide, smaakcorrigentia, antioxidantia en complexvormers (bijvoorbeeld ethyleendiaminotetra-azijnzuur) mogelijk. Eventueel kan ter stabiliteit van het actieve stof molecule met de fysiologisch geschikte zuren of buffers op een pH van 3-7 worden ingesteld. Over het algemeen geniet een 15 zoveel mogelijk neutraal tot zwak zuur milieu (tot pH 5) de voorkeur.
De farmaceutische en gallenische hantering van de onderhavige verbindingen vindt volgens gebruikelijke standaardmethoden (Sucker, Fuchs, Speiser, Pharmazeutïsche Technologie, Thieme. Verlag, Stuttgart (1978)) plaats. B.v. worden actieve stof en hulp- resp. dragerstoffen door roeren of homogeniseren goed gemengd, waarbij over het algemeen bij temperaturen tussen 20 en 80°C, bij voorkeur 20-50°C, in het 20 bijzonder bij kamertemperatuur, wordt gewerkt.
De toediening van de actieve stoffen of geneesmiddelen kan op de huid of slijmvliezen of in het lichaam plaats vinden derhalve oraal, enteraal, pulmonaal, rectaal, nasaal, vaginaal, linguaal, intraveneus, intra-arterieel, intracardiaal, intramusculair, intraperitoneaal, intracutaan, subcutaan, intrapleuraal, intrathekaal, intracavitair.
25 In het bijzonder is ook een toevoeging van andere geneesmiddelen, vooral van uroprotectoren zoals het natriumzout van 2-mercapto-ethaansulfonzuur (zie Europese octrooiaanvrage 2495) of het dinatriumzout van het overeenkomstige disulfide met de formule H03S-CH2CH2SSCH2CH2-S03H, maar ook van andere systemisch of regionaal detoxificerende actieve stoffen mogelijk resp. gunstig.
De onderhavige verbindingen tonen bij intraveneuze, intraperitoneaie of orale toediening bij verschillende 30 experimentele tumoren bij de rat en de muis een goede cytostatische en curatieve activiteit.
Voorbeelden voor dit soort indicaties zijn: leukemieën, lymfogranulomatose, lymfosarcoom, reticulosar-coom, plasmocytoom, M. Waldenstrom; tumoren met uitgezaaide groei, speciaal ovarieel-, mamma-, bronchiaalcarcinoom, neuroblastoom, teelbaltumoren, niercarcinomen, pancreascarcinomen, seminoom, Ewing-sarcoom; post-operatieve hulpbehandelingen, speciaal bij chemosensibele tumoren, die vermoedelijk 35 de lokale grenzen reeds overschreden en ondanks radicale operatie een slechte prognose hebben.
Voorts komen de onderhavige uitvindingen in tegenstelling tot de bekende middelen cyclofosfamide en ifosfamide in het bijzonder ook voor de volgende toepassingen in aanmerking: 1) voor een regionale perfusie in de extremiteiten en de grote lichaamsholten, 2) voor een in vitro-behandeling van het bottenmerg bij een extracorporale behandeling van het bottenmerg 40 van leukemiepatiënten; 3) voor pretherapeutische sensibilisatieproeven op tumoren in vitro.
De onderhavige verbindingen werden bijvoorbeeld aan de rat 5 dagen na intraperitoneaie implantatie van 105 cellen van leukemie L5222 met verschillende doses intraveneus, intraperitoneaal of oraal toegediend en afhankelijk van de dosis een curatief effect bereikt. Als heling werd het recidief en metastasevrij overleven 45 van de tumordragende dieren na 90 dagen beschouwd. Uit de met de verschillende doses verkregen frequenties van genezingen werd met de probietanalyse volgens R. Fisher als gemiddelde curatieve dosis (DC 50) die dosis berekend, waarmede 50% van de tumordragende dieren kan worden genezen.
De onderhavige verbindingen werden ook 1 dag na intraperitoneaie implantatie van 106 cellen van Yoshida-ascites-sarcoom AH13 met verschillende doses intraveneus, intraperitoneaal of oraal toegediend en 50 afhankelijk van de dosis een curatief effect bereikt. Ook hier wordt het curatieve effect als recidief en metastasevrij overleven van de tumordragende dieren gedurende meer dan 90 dagen gedefinieerd. Op overeenkomstige wijze wordt met de probietanalyse volgens R. Fisher als gemiddelde curatieve dosis (DC 50) die dosis berekend, waarbij 50% van de tumordragende dieren kan worden genezen,
Voorts werden de onderhavige verbindingen een- of meermalen (viermaal) op achtereenvolgende dagen 55 na een intraperitoneaie implantatie van 106 cellen van muizenleukemie L1210 met verschillende doses intraveneus, intraperitoneaal of oraal toegediend en een cytostatisch effect bereikt.
De cytostatische activiteit wordt gemeten als een verlenging van de mediane overlevingstijd van de 5 192741 tumordieren en wordt als dosisafhankelijke procentuele verlenging van de overlevingstijd, vergeleken met een onbehandelde controlegroep, uitgedrukt.
De gemiddelde curatieve dosis ligt bij de rattentumoren onafhankelijk van de toedieningsvorm tussen 0,1 en 10 mg/kg. Met dezelfde doses kan een verlenging van de mediane overlevingstijd van 100% bij 5 muizenleukemie L1210 worden bereikt (zie N. Broek: Pharmakologische Grundlagen der Krebs-
Chemotherapie in: A. Georgii (Hrsg), Verhandlungen der Deutschen Krebsgesellschaft Band 1, blz. 15-42, Gustav Fischer Verlag, Stuttgart (1978)).
Deze curatieve en cytostatische activiteit is met het effect van het bekende geneesmiddel cyclofosfamide (EdnoxanR) of ifosfamide (HoloxanR) vergelijkbaar. De laagste, reeds curatief of cytostatisch werkzame 10 dosis bij de boven weergegeven dierproeven is bijvoorbeeld 0,01 mg/kg oraal 0,01 mg/kg intraperitoneaal 0,01 mg/kg intraveneus.
Als algemeen dosistraject voor een curatief en cytostatisch effect (dierproeven zie boven) komt b.v. in 15 aanmerking: 0,01-100 mg/kg oraal, liefst 0,1-10,0 mg/kg, 0,01-100 mg/kg intraperitoneaal, liefst 0,1-10 mg/kg, 0,01-100 mg/kg intraveneus, liefst 0,1-10 mg/kg.
Indicaties voor de onderhavige verbindingen zijn bijvoorbeeld maligne ziekten van mens en dier.
De farmaceutische preparaten bevatten over het algemeen tussen 1 mg en 1 g, liefst 10-300 mg van de 20 onderhavige actieve componenten.
Toedieningsvormen zijn tabletten, capsules, pillen, dragees, ouweltjes, zalven, geleien, crèmes of in vloeibare vorm plaatsvinden. Als vloeibare toepassingsvormen komen bijvoorbeeld in aanmerking: olievormige of alcoholische resp. waterige oplossingen alsmede suspensies en emulsies. Voorkeurstoepassingsvormen zijn tabletten, die tussen 10 en 200 mg of oplossingen, die tussen 0,1-5% actieve stof 25 bevatten.
De enkelvoudige dosis van de onderhavige actieve componenten kan bijvoorbeeld liggen a) bij orale geneesmiddelen tussen 1 en 100 mg/kg, liefst 10-60 mg per kg, b) bij parenterale geneesmiddelen (bijvoorbeeld intraveneus of intramusculair) tussen 1 en 100 mg/kg, liefst 10-60 mg/kg, 30 c) bij geneesmiddelen voor rectale of vaginale toediening tussen 1 en 100 mg/kg, liefst tussen 10 en 60 mg/kg, d) bij geneesmiddelen voor lokale toediening op de huid of slijmvliezen (b.v. als oplossingen, lotions, emulsies of zalven) tussen 1 en 100 mg/kg, liefst 10-60 mg/kg.
(De doses zijn telkens berekend op de vrije basis) 35 Zo kunnen één- tot driemaal daags 1-10 tabletten met 10-300 mg actieve stof of bijvoorbeeld bij intraveneuze injectie één- tot tweemaal daags één of meer ampullen met 1-10 cm3 inhoud en 10-250 mg actieve stof worden aanbevolen. Bij orale toediening is de minimale dagelijkse dosis bijvoorbeeld 200; de maximale dagelijkse dosis bij orale toediening liefst niet meer dan 5000 mg. Ook een langdurige infusie van passende doses gedurende 12 en meer uren kan in sommige gevallen worden aanbevolen.
40 Voor de behandeling van honden en katten ligt de enkelvoudige orale dosis over het algemeen tussen 10 en 60 mg/kg lichaamsgewicht; de parenterale dosis ongeveer tussen 10 en 60 mg/kg lichaamsgewicht.
Voor de behandeling van paarden en vee ligt de afzonderlijke orale dosis over het algemeen tussen 10 en 60 mg/kg; de parenterale enkelvoudige dosis ongeveer tussen 10 en 60 mg/kg lichaamsgewicht.
De weergegeven doseringen betreffen ook een toepassing van de onderhavige verbindingen voor een 45 immunosuppressie.
De acute giftigheid van de onderhavige verbindingen bij de muis (uitgedrukt als LD 50 in mg/kg volgens Proc. Soc. Exper. Biol. Med. 57, 261 (1944)) bedraagt bij orale toediening tussen 100 en 1000 mg/kg, en soms zelfs meer dan 1000 mg/kg.
De geneesmiddelen kunnen bij mensen, dieren en bij de veeteelt op zichzelf of gemengd met andere 50 farmacologisch actieve stoffen worden gebruikt.
De volgende voorbeelden lichten de uitvinding nader toe zonder deze te beperken.
Voorbeeld I
55 2-[2-(bis-(2-chloorethyl)amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur cyclo-hexylammoniumzout.
5,6 g (20 mmol) 4-hydroxicyclofosfamide (= 2-(bis-(2-chloorethyl)-amino)-4-hydroxi-tetrahydro-2H-1,3,2- 192741 6 oxazafosforin-2-oxide) en 4,8 g (20 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat werden in 100 cm3 gedestilleerd water opgelost, hieraan enig trichloorazijnzuur toegevoegd en 3 dagen bij 0°C in de koelkast bewaard. Hierna werd het oplosmiddel in een hoog vacuüm verwijderd, het residu in aceton opgenomen, opnieuw ingedampt, uit aceton gekristalliseerd en uit isopropanol herkristalliseerd. Opbrengst 5 7,2 g (72%); Smp. 149 -151 °C (cisvorm).
Voorbeeld II
2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazofosforin-4-yl-thio]-ethaansulfonzuur 10 cyclohexylammoniumzout.
2,9 g (10 mmol) 2-(bis-(2-chloorethyl)-amino)-4-methoxi-tetrahydro-2H-1,3,2-oxazafosforin-2-oxide en 2,4 g (10 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat werden samen met enig trichloorazijnzuur in 10 cm3 dimethylformamide opgelost en 20 uren bij -25°C bewaard. Na nog 3 uren staan bij 0°C werd met diethylether tot een beginnende troebeling gemengd, aangewreven, het kristallisaat na 20 uren staan bij 0°C 15 afgezogen, gewassen en gedroogd. Opbrengst 4,7 g (94% van de theoretische), Smp, 145°C (ontl.). Herkristalliseren uit ethanol diether; Smp. 149-151 °C (cis-vorm).
Voorbeeld III
20 2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur cyclohexylammoniumzout.
1.4 g (5 mmol) 4-hydroxicyclofosfamide en 1,2 g (5 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat werden samen met een spoor trichloorazijnzuur in 25 cm3 methanol opgelost en 1 nacht bij -25°C in een ijskast bewaard. Vervolgens werd de reactieoplossing tot 5 cm3 geconcentreerd, 25 ether tot beginnende troebeling toegevoegd en aangewreven. Het kristallisaat werd na 20 uren staan bij 0°C afgezogen, gewassen en gedroogd. Opbrengst 2,1 g (84% van de theoretische); Smp. 143-145°C (cis-vorm).
Voorbeeld IV 30 2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur natriumzout.
2.5 g (5 mmol) cyclohexylammonium-2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl]-thioëthaansulfonaat werden in 25 cm3 zuurstofvrij water opgelost en over een met 35 natriumionen beladen kationenuitwisselenden kolom van de firma Merck geleid. Het eluaat werd onder stikstof opgevangen, gevriesdroogd en het vaste residu boven fosforpentoxide in vacuo gedroogd.
Opbrengst 1,9 g (91%); Smp. 78-83°C (ontl.); Rf-waarde 0,61 (loopmiddel: ethylacetaat/isopropanol/l n azijnzuur (5:3:2 7v).
40 Voorbeeld V
2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur ammoniumzout.
2.8 g (10 mmol) 4-hydroxicyclofosfamide en 1,6 g (10 mmol) ammonium-2-mercaptoëthaansulfonaat 45 werden samen met een katalytische hoeveelheid trichloorazijnzuur in 50 cm3 water opgelost en 3 dagen bij 0°C in een ijskast bewaard. Hierna werd het water in een hoog vacuüm afgedestilleerd, het residu tweemaal in aceton opgenomen en in vacuo geconcentreerd. Herkristalliseren uit aceton/ether leverde 3,9 g (93%) met Smp. 131-133°C (cis-vorm).
50 Voorbeeld VI
2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]- ethaansulfonzuur ammoniumzout.
2.8 g (10 mmol) 4-hydroxicyclofosfamide en 1,6 g (10 mmol) ammonium-2-mercaptoëthaansulfonaat 55 werden met een katalytische hoeveelheid trichloorazijnzuur in 10 cm3 dimethylformamide opgelost en 20 uren in een ijskast bij -25°C bewaard. Na nog 5 uren staan bij 0°C werd met ether tot beginnende troebeling gemengd en aangewreven. Het kristallisaat werd na 1 dag bij 0°C afgezogen, gewassen, gedroogd en uit 7 192741 n-propanol herkristalliseerd. Opbrengst 3,2 g (77%) van de cis-vorm; Smp. 132°C.
Voorbeeld VII
5 3-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazofosforin-4-yl-thio]-propaansulfonzuur natriumzout.
5,4 g (20 mmol) 4-hydroxicyclofosfamide en 5,2 g (20 mmol) cyclohexylammonium-3-mercaptopropaansulfonaat werden in 100 cm3 water opgelost, met enig trichloorazijnzuur gemengd en 1 dag bij 0°C bewaard. 1,6 g (5 mmol) 2-(bis-)2-chloorethyl)-amino)-4-ethoxitetrahydro-2H-1,3,2-oxazafosforin-2-oxide en 0,8 g (5 20 mmoi) natrium-2-mercaptoëthaansulfonaat in 10 cm3 dimethylformamide werden met een spoor trichloorazijnzuur gemengd en 1 nacht bij -25°C bewaard. Vervolgens werd aan de reactieoplossing 20 cm3 ether toegevoegd. Het residu werd na 20 uren staan bij 0°C afgezogen, gewassen en gedroogd. Opbrengst 1,5 g (71%); Smp. 145 - 150°C (ontl.); Rf-waarde 0,56.

0,8 g (3,0 mmol) 4-hydroxicyclofosfamide in 3 cm3 water werd onder koelen met ijswater met 420 mg (3,0 mmol) 2-mercaptoëthaansulfonzuur gemengd. Na 1 uur werd in hoog vacuüm geconcentreerd en 30 gekristalliseerd; Opbrengst 1,1 g (92%) met Smp. 75-78°C.

Het eluaat werd vervolgens gevriesdroogd en uit dimethylformamide/chloroform herkristalliseerd. De reactieoplossing werd 1 nacht bij -25°C bewaard. Het kristallijne materiaal werd afgezogen en uit methanol/aceton herkristalliseerd. Opbrengst 530 mg (71% van de theoretische) met Smp. 133-134°C (cis-vorm).

55 540 mg (2 mmol) 4-hydroxicyclofosfamide en 300 mg (2 mmol) lithium-2-mercaptoëthaansulfonaat werden samen met een spoor trichloorazijnzuur in 7 cm3 water opgelost en 20 uren bij 0°C bewaard. Het reactiemengsel werd in vacuo ingedampt, het residu in aceton opgenomen en gefiltreerd. Vervolgens werd 192741 8 de oplossing geconcentreerd, het residu in ethanol opgelost, geconcentreerd, opnieuw in ethanol opgenomen, met ether neergeslagen en het neerslag afgezogen, gewassen en gedroogd. Opbrengst 500 mg (61%), Rf-waarde 0,56 (toopmiddel: ethylacetaat/isopropanol/1 n azijnzuur = 5:3:2).

Neutraal 2-[2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur 10 magnesiumzout.
1.4 g (5 mmol 4-hydroxicyclofosfamide en 750 mg (2,5 mmol) magnesium-di-2-mercaptoëthaansulfonaat werden samen met enig trichloorazijnzuur in 15 cm3 water opgelost en na 3 dagen bij 0°C in vacuo drooggedampt. Het residu werd in ethanol opgelost, dit geconcentreerd, opnieuw in ethanol opgenomen, met ether neergeslagen, afgezogen, gewassen en gedroogd. Opbrengst 1,3 g (63%) met Smp. 100-115°C

1.4 g (5 mmol) 4-hydroxicyclofosfamide en 800 mg (2,5 mmol) calcium-di-2-mercaptoëthaansulfonaat werden samen met enig trichloorazijnzuur in 15 cm3 water opgelost, na 3 dagen bij 0°C in vacuo ingedampt, 25 tweemaal met ethanol gemengd, wederom ingedampt, in ethanol opgelost en met ether neergeslagen, afgezogen, gewassen en gedroogd. Opbrengst 1,3 g (62%) met Smp. 110-115°C (ontl.); Rf-waarde 0,56.

30 2-t2-(bis-(2-chloorethyl)-amino-6-methyl-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-ethaansulfonzuur cyclohexylammoniumzout.
1,45 g (5 mmol) 2-(bis-(2-chloorethyl)-amino-4-hydroxi-6-methyl-tetrahydro-2H-1,3,2-oxazafosforin-2-oxide en 1,2 g (5 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat werden in 5 cm3 water en 5 cm3 aceton opgelost, met trichloorazijnzuur aangezuurd en 20 uren bij 0°C bewaard. Vervolgens werd gefiltreerd, het 35 filtraat in vacuo voorzichtig ingedampt, het residu tweemaal in aceton opgenomen en geconcentreerd.
Hierna werd het residu in aceton opgelost, met ether neergeslagen, met ether gewassen en gedroogd. Opbrengst 1,4 g (56%) met Smp. 120-125°C (ontl.); Rf-waarde 0,61.

277 mg (1 mmol) 4-hydroxicyclofosfamide en 192 mg (1 mmol) natrium-3-mercapto-2-methyl-propaansulfonaat werden in 4 cm3 water opgelost, met een spoor trichloorazijnzuur gemengd en 20 uren bij 45 0°C bewaard. Na concentreren in vacuo werd het residu tweemaal in watervrije ethanol opgelost en opnieuw ingedampt en in alcohol/ether neergeslagen. Opbrengst 420 mg (86%); Rf-waarde 0,61.

Vervolgens werd het reactiemengsel in vacuo voorzichtig ingedampt, het residu tweemaal in watervrije 55 ethanol opgelost en wederom geconcentreerd. 5 277 mg (1 mmol) 3-(2-chloorethyl)-2-(2-chloorethylamino)-hydroxi-tetrahydro-2H-1,3,2-oxazafosforin-2- oxide en 240 mg (1 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat werden in 4 cm3 water opgelost, met een spoor trichloorazijnzuur gemengd en 20 uren bij 0°C bewaard. 340 mg (1 mmol) 3-(2-chloorethyl)-2-(bis-chloorethyl)-amino)-4-hydroxi-tretrahydro-2H-1,3,2-oxazafosforin-2-oxide en 240 mg (1 mmol) cyclohexylammonium-2-mercaptoëthaansulfonaat in 2 cm3 dimethylformamide werden met een spoor trichloorazijnzuur gemengd en 2 dagen bij -25°C bewaard. Vervolgens werd het reactiemengsel met een 20-voudige hoeveelheid ether neergeslagen. Het residu werd 20 gewassen en gedroogd. Opbrengst 400 mg (71%); Smp. 102 -107°C (ontl.); Rf-waarde 0,63.

3-[2-(bis-)2-chloorethyl)-amino-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-2-mercaptopropaansul fo nzuur cyclohexylaminezout.
5 1,39 g (5 mmol) 4-hydroxicyclofosfamide en 1,44 g (5 mmol) 2,3-dimercaptopropaansulfonzuur- cyclohexylaminezout werden in 10 cm3 ethanol opgelost, met trichloorazijnzuur aangezuurd en 2 dagen bij 0°C bewaard. Hierna werd het ether neergeslagen, na 20 uren gedecanteerd en de achterblijvende olie in een hoog vacuüm gedroogd. Deze olie stolde. Opbrengst 1,8 g (77%) met Smp. vanaf 70°C (ontl.).

2- [2-(bis-(2-chloorethyl)-amino)-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]ethaansulfonzuur cis-natriumzout.
2,8 g (10 mmol) 4-hydroxicyclofosfamide in 10 cm3 aceton werden met 1,3 g (8 mmol) natrium-2-15 mercaptoëthaansulfonaat in 2 cm3 water gemengd en 3 uren bij 0°C bewaard. Vervolgens werd het reactiemengsel in vacuo bij 25°C ingedampt. Het residu werd in watervrije aceton opgenomen in de koude met watervrije ether tot beginnende troebeling gemengd. Bij het concentreren van de oplossing in vacuo vormde zich een neerslag. Opbrengst 1,5 g (44%) vain het cis-isomeer; Smp. 83-85°C. Door her-kristailiseren in aceton verkreeg men een in aceton slecht oplosbaar product.

3- [2-(bis-(2-chloorethyl)-amino-2-oxo-tetrahydro-2H-1,3,2-oxazafosforin-4-yl-thio]-propaansulfonzuur guanidiniumzout.
25 14 g (50 mmol) 4-hydroxicyclofosfamide en 11 g (50 mmol) 3-mercaptopropaansulfonzuur guanidinezout werden in 100 cm3 ethanol opgelost, met trichloorazijnzuur gemengd en bij 0°C bewaard. Na 20 uren voegde men 50 cm3 ether toe. Het kristallisaat werd na nog 20 uren afgezogen, gewassen en gedroogd. Opbrengst 18,5 g (78%); Smp. 128-132°C (ontl.).

Preparaten, die als actieve stof 50 mg resp. 200 mg stof 23 volgens volgend overzicht bevatten: 35 verbinding volgens voorbeeld XXIII 50 mg 200 mg mannitol 230 mg 380 mg water voor injectiedoeleinden tot 2 cm3 tot 4 cm3 40 De stof volgens voorbeeld XXIII en mannitol werden telkens onder begassen met stikstof en bescherming tegen licht in zoveel water opgelost, dat een oplossing van 2 cm3 resp. 4 cm3 ontstond. Bij de bereiding en de verdere verwerking werd zo te werk gegaan, dat de temperatuur van de oplossing niet boven 5°C kwam. De oplossingen werden op een bekende wijze steriel gefiltreerd, onder aseptische voorwaarden in bruine injectiekolfjes van 10 cm3 voor 2 cm3 resp. 4 cm3 gedoseerd, van vriesdroogstoppen voorzien en in een 45 vriesdrooginstallatie gelyofiliseerd. Vervolgens werd deze installatie met droge stikstof belucht en de ampulflesjes in de installatie gesloten.
Het restant watergehalte van de kolfjes mag niet boven 0,5% liggen.
Voor de bereiding van een direct toepasbare injectieoplossing wordt de inhoud van de kolfjes met 50 mg actieve stof in 5 cm3, met 200 mg actieve stof met 10 cm3 water voor injectiedoeleinden opgelost.

11 192741 stof volgens voorbeeld XXXIII 50 mg 200 mg 5 mannitol 245 mg 430 mg water voor injectiedoeleinden tot 2 cm3 tot 4 cm3 De stof van voorbeeld XXXIII en mannitol werden telkens onder begassen met stikstof en beschermen 10 tegen licht in zoveel water opgelost, dat een oplossing van 2 cm3 resp. 4 cm3 ontstond. Bij de bereiding en de verdere verwerking werd zo te werk gegaan, dat de temperatuur van de oplossing de 5°C niet overschreed. De oplossingen werden op een bekende wijze steriel gefiltreerd, onder aseptlsche voorwaarden in bruine injectieflesjes van 10 cm3 voor 2 cm3 resp. 4 cm3 gedoseerd, van vriesdroopstoppen voorzien en in een vriesdrooginstallatie geiyofiliseerd. Vervolgens werd de installatie met droge stikstof belucht en de 15 ampullen in de installatie gesloten. Het restant watergehalte van de kolfinhoud mag niet boven 0,5% zijn gelegen. Voor de bereiding van direct toedienbare injectieoplossingen wordt de inhoud van de kolfjes met 50 mg actieve stof in 5 cm3 en die met 200 mg actieve stof in 10 cm3 water voor injectiedoeleinden opgelost. 20

Oxazafosoforin-4-thioalkaanzuurverbindingen met antineoplastische eigenschappen, met het kenmerk, dat het oxazafosforin-4-thioalkaansulfonzuren en neutrale zouten daarvan met de algemene formule 1 van 25 het formuleblad zijn, waarin R,, R2 en R3 gelijk of verschillend kunnen zijn en waterstof, methyl, ethyl, 2-chloorethyl of 2-methaansulfonyloxiethyl voorstellen en waarbij ten minste twee van deze groepen 2-chloorethyl en/of 2-methaansulfonyloxiethyl zijn: R4 waterstof of methyl is; 30. een rechte of vertakte C2_6 alkyleengroep is, die aan het koolstofatoom op de 2-, 3-, 4- of 5-plaats van de alkyleenketen een mercaptogroep kan dragen; Y+ een waterstofkation, een alkalimetaal- of aardalkalimetaalkation, het guadinium-, morfolinium-, of cyclohexylammoniumkation, of een kation dat is afgeleid van een amine met de formule NR5R6R7 voorstelt, waarbij de groepen R5-R7 gelijk of verschillend zijn en waterstof, 0,-02 alkylgroepen of 35 oxiethylgroepen voorstellen, dan wel waarin Y+ het ethyleendiammoniumkation H3të-CH2CH2-NH3+ of het piperazoniumkation is; en z 1 is, indien Y+ een éénbasisch kation is, of 2 is indien Y+ een tweebasisch kation of het kation van een verbinding met twee éénbasische kationen is. 2. 2-[2-(bis-(2-chloorethyl)-amino)-2-oxotetrahydro-2H-1,3,2-oxazafosforin-4-yl]-thioëthaansulfonzuur 40 cyclohexylammoniumzout.
