Abstract
La presente invención se refiere a composiciones y métodos para la producción heteróloga de isoprenoides derivados de acetil-CoA en una célula hospedera. En algunas modalidades, la célula hospedera es genéticamente modificada para comprender una secuencia de nucleótidos heteróloga que codifica una acetaldehído deshidrogenasa, acetilante (ADA, E.C. 1.2.1.10) y una vía de MEV que comprende una HMG-CoA reductasa que usa NADH. En algunas modalidades, la célula hospedera es genéticamente modificada para comprender una secuencia de nucleótidos heteróloga que codifica una ADA y una vía de MEV que comprende una acetoacetil-CoA sintasa. En algunas modalidades, la célula hospedera genéticamente modificada además comprende una o más secuencias de nucleótidos heterólogas que codifican una fosfoctolasa y una fosfotransacetilasa. En algunas modalidades, la célula hospedera genéticamente modificada además comprende una alteración funcional de la derivación de PDH nativa. Las composiciones y métodos descritos en el presente documento proveen una vía eficiente en energía pero equilibrada en redox para la producción heteróloga de isoprenoides derivados de acetil-CoA.
