Abstract
Se describe una proteína de fusión recombinante que comprende una porción de péptido de eritropoyetina humana enlazada a una porción de péptido de inmunoglobulina. La proteína de fusión tiene una media vida prolongada in vivo en comparación con eritropoyetina humana nativa que ocurre naturalmente o recombinante. En una modalidad de la invención, la proteína tiene una vida media in vivo de cuando menos tres veces superior a la eritropoyetina humana nativa. La proteína de fusión también exhibe bioactividad eritropoyética mejorada en comparación con eritropoyetina humana nativa. En una modalidad, la proteína de fusión comprende la secuencia de péptido completa de una molécula (EPO) de eritropoyetina humana y la secuencia de péptido de un fragmento Fc de IgG1 de inmunoglobina humana. El fragmento Fc en la proteína de fusión incluye la región de articulación, dominios CH2 y CH3 de IgG1 de inmunoglobulina humana. La molécula EPO puede estar enlazada directamente al fragmento Fc para evitar enlazadores de péptido extraños y reducir el riesgo de una respuesta inmunogénica cuando se administra in vivo.
