Description
본 발명은 적어도 하나의 알칼로이드를 포함하는 제형에 관한 것이다. 특별히 한정되지 않으나 더 구체적으로는 적어도 하나의 알칼로이드 및, 적어도 하나의 전자 전달제 유도체를 포함하는 제형에 관한 것이다.
본 명세서에서 인용하거나 검토하는 문헌, 사실 또는 기사 등은 이들 문헌, 사실 또는 기사가 우선일자 현재 공지된 지식의 일부이거나 또는 본 명세서에서 다룬 문제점을 해결하기 위한 시도에 관련하여 공지된 사실로서 이들의 인용 또는 검토에 별도의 허가를 받지 않는다.
알칼로이드
알칼로이드는 오래전부터 약제에 사용해왔다. 이 화합물은 처음에 식물로부터 추출하였으며 약물 및 독물 등 인체에 대해 생리학적 작용을 하는 질소계 화합물을 포함한다. 상세한 설명 및 청구범위에서 사용한 "알칼로이드" 라는 용어는 1차, 2차 또는 3차 아민 치환체를 포함하는 모든 천연 및 합성 화합물을 포함한다. 아민은 하나 또는 두개의 고리에 결합되기도 하며 비환식 구조도 포함한다. 예를 들면,
* 3차 아민:
* 고리 3개의 공통 부분인 질소 원자를 함유한 지환식 구조를 가진 것 (예, 모르핀, 아트로핀, 퀴닌 등);
* 질소가 단일 고리에 결합되어 알킬화된 환식 구조를 가진 것 (예, 니코틴, 펜스피라이드 등);
* 질소와 결합한 환식 구조를 갖지 않은 것 (예, 플루라제판);
* 질소가 지환식 구조에 결합하거나(예, 콘린, 펜딜린) 또는 직쇄 구조에 결합한 2차 아민(예, 에핀프린);
* 1차 아민 (예, 에피드린);
* 피리딘 (예, 니코틴);
* 메타미딘 유도체;
* 퀴놀린 (예, 신코닌); 및
* 구아니딘 (예, 아르기닌)
대부분의 알칼로이드는 수용성이 아니며 유기 용매에 용해한다. 그러나, 알칼로이드는 모두 염기성이며 산과 결합하여 적어도 부분적으로 수용성인 결정성 염을 형성한다. 전형적으로, 알칼로이드는 염 형태로 경구 투여하거나 혹은 정맥 주사로 투여한다. 알칼로이드는 염류의 친수성이 경피전달을 제한하기 때문에 통상 경피적으로 투여하지는 않는다. 모르핀 및 아트로핀은 임상적으로 유용한 알칼로이드이며 경피 투여하지 않는다. 또한 상기 중 일부는 림프계를 통해 작용하는 것으로 생각되며 따라서 알칼로이드의 경구 전달을 개선하는데 바람직하다.
국소 투여
약물을 직접 인체의 일부에 도포하는 것을 국소 투여라 하며 경피식 투여(피부 도포) 및 구강 투여(구강 내면에 도포)를 포함한다.
피부는 인체에서 가장 큰 기관이며 외부의 화학, 물리 및 병리적 위해로부터 내부 기관을 보호하는 역할을 한다. 정상 피부는 상피, 내피 및 피하조직이라는 3가지 층으로 분리되어 있다. 상피의 바깥 각질층은 미생물 침입 및 증식을 방해하는 강도, 유연성, 고 전기 임피던스 및 건조함 등의 성질을 가진다. 각질층은 또한 경피적 약물 흡수를 차단하는 기본 차단막이 된다.
경피 전달법은 순수한 상태의 약물 또는 통상, 피부 전달율을 향상시키기 위한 물질을 포함하는 제형 형태로서 적용되는 약물을 도포하는 방법을 포함한다. 전통적인 경피 전달계는 연고, 크림, 습포제 및 경고제(또는 고약) 등으로서 피부에 접착하여 유효성분을 전달하는 것이었다. 최근에는 기술 발달로, 경고제를 "패치" 형태로 만들어서 피부에 대한 접착력 및 전달율 제어를 더욱 개선하였다.
경피 전달은, 느린 정맥 주입으로 달성되는 것과 비슷한 혈중 농도를 얻으면서 불편함은 없는 것; 경구 투여에 비해 더 우수한 흡수 및 대사 제어; 약물 특히, 짧은 반감기를 가진 약물의 효과 지속성; 간 초회 통과(hepatic first pass)시 우회로 인해 적은 약량으로 동등한 효능을 발휘하는 것; 과소 투약량 또는 과대 투약량의 위험 감소; 또한 약량 형태의 단순화를 통한 환자에 대한 순응도의 향상 등을 포함하여 다수의 장점을 제공할 수 있는 것으로 알려져 있다.
계통적 약물치료에 유리한 혈중 농도에 도달할 정도의 속도로 모든 약물을 경피 투여할 수 있는 것은 아니다. 가령 유사한 분자량 및 크기를 가진 약물이라도 서로 상이한 속도로 피부를 통과하여 흡수되기도 한다. 피부를 통한 약물 흡수력을 개선하기 위한 피부 강화제 및 다양한 제형 기술이 개발되었다. 그러나, 약물 투과력의 증가시 피부에서 가장 중요한 보호층에 손상을 입히는 결과를 가져오기 때문에 장기간 사용시의 위험에 대한 우려도 커지고 있다.
경피 치료력 개선을 위해 현재 이용되고 있는 방법들은 보편적으로 성공을 거두지 못하였기 때문에 아직 개선의 대상으로 남아있다. 특히 알칼로이드 전달을 위한 경피 전달계가 요구된다.
또한 구강 전달시, 약물을 경구 투여할 때 문제를 일으킬 수 있는 간에 의한 대사를 피하게 되므로 이 방법에 대한 관심이 날로 증가하고 있다. 일반적으로, 상기 약물은 혀 밑에 놓일 약용 정제형으로 제조된다. 입의 안쪽 가장자리는 각질층 피부의 각질층과 다르기 때문에 약물을 구강 전달 형태로 투여하는 것이 그리 어렵지는 않다. 그러나 이 투여 방법은 전달 속도가 매우 느리고 유효 성분이 구강 점막을 통과할 수 없을 경우 무용해지므로 보편적으로 사용되지 않는다. 과거에는 약물의 국소 투여를 개선하려는 노력이 있었다. 예를 들어, 국제 특허 출원 PCT/AU03/00998 에서는 약제학적으로 수용가능한 화합물의 인산염 유도체 착물, 예컨대, 라우릴아미노디프로피온산 토코페릴 포스페이트를 포함하는 약제물용 담체를 개시하였다. 또한, PCT/AU03/00998 은 토코페릴 포스페이트를 양쪽성 계면활성제, 양이온성 계면활성제, 질소 작용기를 가진 아미노산 및 이것이 농축된 단백질로 구성된 군에서 선택된 착화제에 착물화시키는 것에 대해 개시하였다. 이 담체는 테스토스테론, 에스트로겐, 아트로핀 및 모르핀의 국소 투여를 개선하는 것으로 공지되 어 있다. 그러나, 모르핀 및 아트로핀에 있어서는 피부 투과력 향상이 요구된다.

경구 투여하는 약물은 많지만, 유용하면서도 장벽을 통과하지 못하여 사용할 수 없는 약물도 다수이다. 지방 같은 물질은 장을 통과하여 효과적으로 전달되지만 토코페롤 같이 장을 거의 통과하지 못하는 물질도 많다. 따라서, 알칼로이드의 경구 투여 능력을 개선할 수 있는 시스템이 요구된다.
알칼로이드 화합물을 전자 전달제의 인산염 유도체에 직접 착화시킬 경우 투여력이 현저히 개선된다는 사실을 확인했다. 예를 들면, 모르핀 투여 능력은 이것을 토코페릴 포스페이트에 직접 착화시켰을 때 향상되었다.
본 발명에 따르면, 적어도 하나의 알칼로이드와 하나 이상의 전자 전달제의 적어도 하나의 인산염 유도체를 반응시켜 얻은 생성물을 포함하는 알칼로이드 제형을 제공한다.
바람직하게, 전자 전달제의 인산염 유도체는 토코페롤의 적어도 하나의 인산염 유도체를 포함하는 군으로부터 선택한다.
알칼로이드 제형은 국소 또는 경구 투여하는 것이 바람직하다.

바람직하게, 하나 이상의 전자 전달제의 적어도 하나의 인산염 유도체는 모노-토코페릴 포스페이트, 디-토코페릴 포스페이트 및 그 혼합물로 구성된 군에서 선택된다. 더 바람직하게, 하나 이상의 전자 전달제의 적어도 하나의 인산염 유도체는 모노-토코페릴 포스페이트 및 디-토코페릴 포스페이트의 혼합물이다.
경우에 따라, 높은 수용해도 같은 부가적인 성질이 바람직한 포스파티드 등의 포스페이트 유도체를 이용할 필요가 있다. 포스파티딜 유도체는 유기 인산염의 아미노 알킬 유도체이다. 이 유도체는 R1R2N(CH2)nOH 의 구조를 가진 아민으로 제조할 수 있으며 여기서 n 은 1 내지 6 의 정수이고 R1 및 R2 는 3 이하의 탄소를 가진 짧은 알킬 사슬이다. 알칼로이드 제형은 보충제, 장관 영양제, 비경구 약형, 좌제, 비강 전달형, 패치 및 크림을 포함한 피부 전달형, 구강 전달형 등의 각종 투약 형태를 통해 인간이나 동물에게 투여된다. 바람직하게, 본 발명에 따른 경구 알칼로이드 제형은 장용성 코팅을 포함한다. 장용성 코팅은 위속의 산성 환경으로부터 착물을 보호한다. 경구 제형은 정제, 분말, 씹는 정제, 캡슐, 경구 현탁액, 현탁액, 유제나 유동제, 어린이용 제형, 장관 영양제, 건강식품 및 기능성 식품 형태를 취하기도 한다.
투약 형태는 또한 전분이나 고분자성 결합제, 감미제, 착색제, 유화제, 코팅물 등과 같이 상기 투약 형태의 조제에 관레적으로 사용되는 첨가제를 포함하기도 한다. 안정도 혹은 전달력 개선 등의 부가적인 성질이 유용한 경우, 또다른 적절한 첨가제로서 전자 전달제의 인산염 유도체 착물을 이용하기도 한다. "인산염 유도체 착물" 이란 용어는 국제 특허 출원 PCT/AU01/01476 에서 기술한 바와 같이, 양쪽성 계면활성제, 양이온성 계면활성제, 질소 작용기를 가진 아미노산 및 아미노산 농축 단백질로 구성된 군으로부터 선택된 적어도 하나의 착화제와 전자 전달제의 적어도 하나의 유도체를 반응시켜 얻은 생성물을 말한다. 이러한 첨가제를 사용할 경우, 제형 안에 과량의 전자 전달제가 존재함을 확실히 아는 것이 중요하다. 이 분야의 지식이 있다면 다른 적절한 첨가제도 쉽게 예상할 수 있다.
도 1은 돼지의 심박수에 대한 각종 아트로핀 제형의 영향을 도시한다. 상기 데이타는 10분간의 누적 평균이며 공분산 분석(covariate analyses)을 이용하여 기저치(적용 1시간 전의 평균)에 대해 보정했다.

본 발명의 다양한 구현/측면을 다음의 비제한적인 실시예에 따라 상세히 기술한다.
실시예 1
이 실시예는 본 발명에 따른 제형내의 아트로핀을 돼지에 경피 전달한 것에 대해 연구한다. 이 실험에서는 겔 형태로 도포했을 때 돼지의 심박수에 대한 아트로핀의 피부 투과에 대한 영향을 연구 검토했다.
방법과 재료
아트로핀(20mg/kg)을 다음과 같은 시험용 베이스 크림으로 제형화 했다. 하기와 같이 특정한 성분 이외에도, 상기 크림은 모두 다음 성분들을 포함했다: 12% 울트레즈-10 카르보머 - 3% 용액, 0.25% 트리에탄올아민, 0.1% 수르사이드 DMDMH 및 최고 100% 까지의 탈이온수.
아르토핀과 조합된 조성물(F), (H) 및 (J)는 본 발명에 따른 제형을 생성한다. 조성물(B), (D) 및 (E)는 종래의 방법에 따른 제형을 생성하며 조성물(A), (C) 및 (I)은 부형제의 영향을 나타낸다.

본 실험에서 10마리의 수컷 크로스브레드(crossbred)(대형 백색 x 란드라스) 돼지 (초기 평균 무게 51.5kg 및 최종 평균 무게 61.0kg)를 사용했다. 연구 4일 전, 14마리 돼지의 무게를 잰 후 새 환경 적응기간 동안 실험시설에 있는 개별 우리(1.75m x 0.65m)에 나누어 넣었다. 이 기간 동안, 돼지의 등에 있는 털을 동물가위(Oster-U.S.A.)로 깎고 이어서 사람이 사용하는 전기면도기(Philishave HQ 5041, Philips Aust Pty Ltd)로 표준 면도하여 제거하였다. 탄성 벨트를 돼지 가슴둘레에 설치하여 돼지가 심박수 모니터 장착에 익숙해지도록 했다. 실험 시작시 환경 및 정규 관리에 가장 잘 적응한 10마리의 돼지를 선택하고 인접한 우리를 비운 상태에서 수용했다. 이 물리적인 격리는 동일한 주파수에서 작동하는 모든 심박수 모니터에서 방출하는 신호 간의 충돌 가능성을 피하기 위한 것이었다. 상기 10마리의 돼지를 5마리씩 두개의 그룹(짝홀수)로 나누어 교대 날짜별로 실험에 이용했다. 연속 이틀의 치료일 동안 실험을 반복했다. 각 반복 실험시 10마리의 돼지를 임의로 10개의 처리그룹 중 하나에 할당했다. 따라서 각 돼지를 다섯가지 경우에서 취한 데이타에 이용하였고 또한 각 처리는 5회씩 적용했다.
각 측정일에는 오전 8시에 5마리의 실험 돼지의 무게를 재고 심박수 모니터를 장착하고 1분간 심박수를 기록하는 것으로 측정을 개시했다. 인체용 심박수 모니터(Polar Sport Tester PE4000 - Polar Electro Finland)를 사용하여 심박수 데이타를 수득했다. 센서 및 트랜스미터가 내장된 체스트 벨트를 돼지의 앞다리 바로 뒷편의 가슴둘레에 걸쳐 설치했다. 상기 벨트는 우수한 심박수 신호를 확보하기 위해 초음파 겔(Virbac Aust Pty Ltd)이 센서 접촉 면적에 도포된 자유 코팅층을 가 진다. 폭 100mm 의 탄성 및 벨크로 재질로 구성된 제2 벨트를 상기 돼지의 트랜스미터 벨트 위로 다시 돼지의 가슴둘레를 따라 설치했다. 이 벨트는 트렌스미터의 물리적 손상을 방지했으며, 기록하는 동안 모니터 기록 유닛(손목시계와 유사한)을 보관하기 위한 포켓을 포함했다. 돼지 등판의 한 부분을 전기면도기로 면도했다. 이 부분 안에서 템플레이트 모형 영구 마커로 직사각형의 처리 면적 172.5cm2 (75 x 230mm)의 윤곽을 그렸다. 그 뒤 먹이를 100g/kg 체중0 .75 (예: 55kg 돼지 = 2020g/일) 으로 공급했다. 심박수 기록 개시 뒤 적어도 1시간 후에 처리 작업을 시작했다. 보호용 고무 장갑을 낀 세명의 스탭이 각각 실험용 제형을 5ml 주사기로 공급했다. 도포한 제형을 문질러 바르면서 헤어 드라이어를 이용해 더운 공기를 처리 영역에 공급했다. 8 내지 10분후 피부 표면이 끈적해지자 문지름 작업을 중단했다. 투명 드레싱(10 x 12cm)(Tegaderm - 3M Health Care U.S.A.) 3개를 처리 영역에 부착했다. 처리작업 후 돼지를 기록 기간의 나머지 6 내지 7시간 동안 가만히 두었다. 처리작업에 사용한 주사기와 장갑은 작업 전후의 무게를 재어 돼지에 대한 실제 도포량을 정확히 계산할 수 있게 했다. 기록 기간이 끝나면, 심박수 모니터 및 투명 드레싱을 벗기고 처리 영역을 소량의 액상 세척제와 함께 온수로 씻어냈다.

검토 및 결론
상기 데이타에서, 아트로핀의 경피적 도포시 돼지의 심박수가 증가하고 도포 뒤 약 60분 후에 최고치에 도달한다는 것을 알 수 있다. 또한 베이스 크림 단독 사용시 심박수는 증가하지 않고 조제물의 영향은 아트로핀 자체에 따른 것임을 시사한다.
토코페릴 포스페이트/디-토코페릴 포스페이트 혼합물을 함유하는 제형 G는 아트로핀에 관한 가장 우수한 전달계를 제공했다. 심박수는 증가한 상태로 다른 제형보다 더 오랜 기간동안 지속되었다. 이것은 표 1에서 확인할 수 있는데, "기저치와의 차이" 항목 아래의 0 내지 60분 및 60 내지 120분의 데이타 수치가 G 에 대해 최고값을 나타낸다. 표 1은 라우릴이미노디프로피오네이트-토코페릴 포스페이트 함 유 조성물이 유사한 농도의 아트로핀을 함유한 경우보다 제형 G가 더욱 효과적이며 그 효과가 일관되고 있음을 입증한다.
표 2의 데이타 평가는 로그 피크율, 로그 피크시간 및, 중요한 평가로서, 로그 상승기울기 및 로그 하강기울기에 관련하여 제형 G 대 제형 H 의 일관적인 효능 증가를 나타낸다.
또한, 본 발명에 따른 제형은 염증을 일으키지 않았으므로 피부에 대한 자극 없이 장기간 피부에 접촉시킬 수 있는 것으로 확인되었다.
실시예 2
이 실시예는 돼지에 대한 모르핀의 경피 전달 효과를 연구했다. 돼지 피부는 인간의 피부와 유사한 성질이 있어 약물의 피부전달을 연구하기에 최상의 모델이 된다.
이 연구는 돼지에게 모르핀을 경피 전달한 후 열(62℃)에 대한 꼬리 위축 반응의 지연을 측정함으로써 진통 수준을 평가하기 위해 고안되었다.
위축 실험 데이타는, 고정 모델로서 처리 및 시간, 또한 임의 모델로서 돼지, 반복 실험횟수 및 제로(0)시간에서의 위축 시간을 이용하여 REML(잔차 최대 가능성)으로 분석했다. 데이타는 초기에 그대로 분석했으나 6시간째는 다소 변형된 데이타가 되었기 때문에 로그 변환하여 분석했다. 양쪽 모두 기본적으로 동일하게 해석했다.

전반적으로, 조제물 AGM 로 처리한 돼지의 위축 시간은 다른 조제물로 처리한 돼지보다 더 길었다 (AG, AH, AGM 및 AHM 로 처리한 돼지 각각에 대해 2.63, 2.88, 4.82 및 3.17초, 표 4 참조). 흥미로운 것은 상기 반응이 처리뒤 6시간 후에 가장 크다는 것으로서(도 5 참조), 이는 특히 대조부인 AG 와 비교할 때 그 지속적인 효과를 시사한다. 이에 관하여, AGM 처리된 돼지의 6시간후 위축 시간이 AG 에 대해 133% 로 더 컸다. 또한 대조부인 AH 와 비교할 때 AHM 이 처리뒤 2시간 후의 위축 시간이 더 길었지만 이 효과는 지속되지 않은 것으로 나타났다. 즉 AHM 은 AGM 에서 얻은 효과 지속의 결과는 없었다.
결과적으로, 실험 데이타를 통해 본 발명에 따른 제형(AGM)에 함유된 모르핀의 경피 전달이 1시간 내지 6시간후, 열처리에 의한 꼬리 위축 반응의 지연으로 측정했을 때 신속하고 지속적인 진통 효과를 제공하는 사실이 입증되었다. 또한, 본 발명에 따른 제형은 염증을 일으키지 않았으므로 자극 없이 장기간 피부 접촉이 가능할 것으로 생각된다.

실시예 3
이 실시예는 래트에 모르핀을 경피 전달할 때 토코페릴 포스페이트 착물을 사용한 대조부와 비교하여 본 발명에 따른 각종 제형의 효과를 연구한다.
방법
동물: 의식있는 스프라그 다울리 래트(Sprague Dawley rats)(~280g) 각 그룹당 6마리.
경피 전달 제형 조제물: 모르핀 HCl, 글락소 오스트레일리아 Pty Ltd. (카탈로그 번호: 22284). 모르핀 유리 염기를 탄산칼륨을 첨가하여 HCl 수용액으로부터 얻었다. 이 공정은 모나세 유니버시티에서 완료했다 (모르핀 HCl 은 크림으로 사용할 수 없으므로 유리 염기 형태로 사용했다).
모르핀(10mg/kg)을 표 5에서와 같은 각각의 제형에 부가했다. 모르핀의 효과 는, 모르핀 작용시 발바닥 두드림(pat)이 그치는데 걸린 시간의 지연을 확인하여 열에 대한 래트의 지연 반응을 통해 측정했다.

대조부로 사용한 베이스 겔은 토코페릴 포스페이트를 제외하고 다른 모든 성분을 함유했다. 바이탈 ET 는 이 실험에서 사용하지 않고 바이탈 ET 및 본 발명의 제형간 성분을 비교하기 위해 수록한 것이다.
시험 방법:
소형 실험 동물에 대한 진통 수준을 신속하고 효과적으로 스크리닝하기 위한 평판형 진통제 시험기(analgesiometer)를 사용한다. 이 장치는 열원(적외선광에서 방출한 ~45℃ 의 열원)을 동물의 뒷발에 가하는데 이용하며 발을 움추리는데 걸린 시간(발 움추림 잠복기)를 측정한다. 열판은 정밀도 0.1℃ 의 내장형 디지탈 온도 계 및 정밀도 0.1초의 타이머를 이용하여 일정한 표면 온도를 제공한다. 동물을 열판 위에 놓고 열판을 둘러싼 아크릴 우리에 가두어 발-핥기 반응을 관측한다. 발-핥기 반응 전의 시간 증가가 진통 작용을 나타낸다.
래트의 등쪽 후부 영역의 피부에 경피 패치 부착하기 24시간 전 털 제거용 크림을 (마취상태에서) 발랐다. 의식이 있는 스프라그 다울리 래트(~400mg)에게 체중 1kg 당 10mg 의 모르핀 HCl 의 약량으로 모르핀을 투여했다. 제형에는 10% w/w 모르핀 HCl이 함유되어 있다. 따라서 0.2kg 짜리 래트에 대해 도포한 제형의 양은 20mg 이며 2mg의 모르핀 HCl 을 함유한 것이다. 아침에 1회 도포한 뒤 다양한 시점에서 진통작용을 측정했다. 약물/비히클에 노출된 피부 영역을 Tegaderm 패치로 덮었다. 모든 동물에 대해 모르핀 투여 전후에 진통제 실험을 했다.
결과:
도 6 은 각각의 제형에 대해 얻은 결과를 나타낸다. 이 결과는 약량 의존 방식으로 반응 시간의 증가를 보여주며 이것이 진통작용을 가리킨다. 모르핀은 함유하나 TPM 은 함유하지 않는 겔의 대조 실험은 경피적 경로에 TMP 가 필수 불가결함을 보여준다. 이 결과를 대조값에 비교한 움추림 시간의 변화로 나타내며, 대조값은 불완전 제형(예, 모르핀이나 TPM 을 함유하지 않은 제형)으로 처리한 래트에서 얻은 값이며 제로 시간 값은 완전 제형(TPM-01/M)으로 처리한 래트에서 얻은 것이다.
결론:
이 연구에서 사용한 제형은 표 5에서 열거한 바와 같은 TP/T2P 혼합물 (또는 TPM), 모르핀 HCl 및 기타의 부형제를 포함한다. 상기 제형은 라우릴디아미노프로피온산을 함유하지 않았다.
도 6 은 뚜렷한 약량-반응 및 지속 효과를 보여준다. 두종류의 대조부와 비교할 때 (예, 모르핀과 TP/T2P 혼합물을 함유하지 않고 염기성 부형제만 함유한 대조용 겔, 또한 부형제와 모르핀을 함유하지만 TP/T2P 는 함유하지 않은 대조용 겔), TP/T2P 혼합물과 함께 제형화한 모르핀이 가장 큰 전달 효과를 나타내었다.
본 명세서 및 특허청구범위에서의 "포함하는" 및 "포함하는" 이란 표현 형식은 본 발명을 한정하기 위한 것이 아니며 다양한 변형과 부가를 포함한다.
본 발명의 수정 및 개선은 당해 분야의 지식이 있다면 용이하게 예상할 수 있으며 본 발명의 범위에 포함된다.
상기와 같이 본 발명은 알칼로이드를 포함하는 제형, 특히 알칼로이드 및 전자 전달제 유도체를 포함하는 제형에 관한 것으로서 이에 의해 모르핀 투여 능력이 현저히 향상된다.
Claims (24)
- 적어도 하나의 알칼로이드 및 하나 이상의 전자 전달제의 적어도 하나의 인산염 유도체를 반응시켜 얻은 생성물을 포함하되,상기 알칼로이드는, 고리 3개의 공통 부분인 질소 원자를 함유한 지환식 구조를 가진 것; 질소가 단일 고리에 결합되어 알킬화된 환식 구조를 가진 것; 또는 질소와 결합한 환식 구조를 갖지 않은 것인 3차 아민류로 구성된 군에서 선택되며,상기 전자전달제는 에난시오머 및 라세미체 형태의 알파, 베타, 감마 및 텔타 토콜(tocol)을 포함하는 히드록시 크로만; 비타민 K1 및 유비퀴논의 환원체 형태인 퀴놀; 레티놀 함유 히드록시 카로테노이드; 칼시페놀 및 아스코르빈산 및 그의 혼합물로 구성된 군에서 선택되는 것을 특징으로 하는 알칼로이드 제형.
