Abstract
본 발명은 생물분해성 형상기억 중합체 조성물과 제조 물품, 제조 방법 및 이들의 이용방법에 관한 것으로, 특히 이러한 조성물은 적어도 하나의 하드 세그먼트와 적어도 하나의 소프트 세그먼트를 포함한다. 상기 하드 세그먼트의 Ttrans는 -30에서 270℃ 사이이고, 하드나 소프트한 세그먼트중 적어도 하나는 가교 결합할 수 있는 작용기을 포함할 수 있으며, 그 세그먼트는 상호투과할 수 있는 망상조직이나 반투과할 수 있는 망상조직의 형성 및 소프트 세그먼트의 Ttrans이하에서 물리적인 작용에 의해 결합될 수 있다. 물체들은 하드 세그먼트의 Ttrans이상의 온도에서 주어진 형상으로 형성될 수 있으며, 소프트 세그먼트의 Ttrans이하의 온도로 냉각될 수 있다. 만약 물체가 나중에 이차 형상으로 형성된다면, 물체는 소프트 세그먼트의 Ttrans이상, 그리고 하드 세그먼트의 Ttrans이하로 물체를 가열함으로써 원형상으로 되돌릴 수 있다.
Description
형상기억이란 일방향(one-way) 효과인 기계적인 변형(도1)이나 이방향(two-way) 효과인 냉각 및 가열(도2)에 의한 변형 후에 원래의 형상을 기억하는 물질의 능력을 말한다.
이러한 현상은 구조 위상 변환(structural phase transformation)에 근거한다.
이들 특징들을 갖고 있는 것으로 알려진 첫번째 물질로 TiNi(니티놀, Nitinol), CuZnAl과 FeNiAl 합금을 포함하는 형상기억 금속 합금(shape memory metal alloys, SMAs)이 있다. 이들 물질의 구조 위상 변환은 마텐자이트계(martensitic) 변환으로 알려져 있다. 이들 물질은 인공혈관(vascular stents), 의료용 도자철선(guidewire), 치열 교정용 철선, 진동 댐퍼(damper), 파이프 연결, 전기 커넥터(connector), 자동 온도 조절 장치(thermostats), 발동기(actuators), 안경테와 브래지어 아래철선을 포함하는 다용한 용처가 알려져 있다. 이들 물질은 아직 널리 이용되지는 않았는데, 부분적으로는 그들이 상대적으로 비싸기 때문이다.
형상기억 중합체(shape memory polymers, SMPs)는 SMAs의 사용을 대체하거나 또는 늘리는 방향으로 발전되어 왔는데, 부분적으로 이들 중합체가 가볍고 형상 회복 능력이 높으며, 조작이 쉽고 SMAs에 비하여 경제적이기 때문이다. 문헌상에서, SMPs는 일반적으로 하드 세그먼트(hard segment)와 소프트 세그먼트(soft segment)를 갖고 있는 선형 블록 공중합체(linear block co-polymers)로 분리된 위상을 특징으로 한다고 되어있다. 하드 세그먼트는 일반적으로 제한된 용융점(melting point)을 갖고 있는 결정체이며, 소프트 세그먼트는 일반적으로 제한된 유리전이온도(glass transition temperature)를 갖는 비결정체이다. 그러나 몇몇 실시예에서 하드 세그먼트는 비결정체이며 용융점보다는 오히려 유리전이온도를 갖는다. 다른 실시예에서 소프트 세그먼트는 결정체이며 유리전이온도보다는 오히려 용융점을 갖는다. 소프트 세그먼트의 용융점이나 유리전이온도는 하드 세그먼트의 용융점이나 유리전이온도보다는 대체로 낮다.
SMP가 하드 세그먼트의 용융점이나 유리전이온도 이상에서 가열될 때 일정한 물질이 형상화될 수 있다. 이 원형상(original shape)은 하드 세그먼트의 용융점이나 유리전이온도 이하에서 SMP를 냉각함으로써 기억될 수 있다. 이와 같이 형상화된 SMP가 소프트 세그먼트의 용융점이나 유리전이온도 이하로 냉각될 때 형상이변형되고, 새로운 임시(temporary) 형상이 고정된다. 상기 원형상은 소프트 세그먼트의 용융점이나 유리전이온도 이상, 하드 세그먼트의 용융점이나 유리전이온도 이하로 물질을 가열하면 회복된다. 임시 형상을 고정시키는(setting) 또 다른 방법으로, 물질을 소프트 세그먼트의 용융점이나 유리전이온도 보다 낮은 온도에서 소프트 세그먼트에 의해 흡수되는 스트레스와 스트레인(strain)에 의해 물질이 변형되도록 하는 방법이 있다. 물질이 소프트 세그먼트의 용융점이나 유리전이온도 이상에서, 그러나 하드 세그먼트의 용융점 또는 유리전이온도 이하에서 가열될때, 스트레스와 스트레인이 풀리게 되며 물질은 원형상으로 되돌아온다. 온도가 상승됨으로 일어나는 원형상으로의 회복을 열적 형상기억 효과(thermal shape memory effect)라 한다. 물질의 형상기억능을 설명하는 특징들로는 원형상으로의 형상 회복 및 임시 형상의 형상 고정(fixity)이 있다.
일본 의료기 공급 회사(Japen Medical Supply Co. Ltd.)의 EPA 0 326 426에는 락타이드(lactide)나 글리코라이드(glycolide)의 상동중합체(homopolymer)로 이루어진 생물분해성 형상기억 중합체로 성형된 물품이 공지되어 있다. 이들 중합체는 오직 온도 증가를 함수로 하여 형상이 변하며, 두 중합체 모두 10% 미만정도의 신장만으로도 파손이 되어(elongations at break) 매우 부서지기 쉽기때문에 응용에 매우 제한적이다.형상를 기억할 수 있는 능력 이외에 SMPs의 몇가지 물리적인 특징들은 온도와 스트레스, 특히 소프트 세그먼트의 용융점, 유리전이온도 또는 다른 온도효과에서의 외부적인 변화에 반응하여 상당히 많이 바뀐다. 이러한 특징들로는 탄성계수, 경도(hardness), 유연성(flexiblity), 증기 투과성, 감쇠(damping), 굴절 지수, 유전(dielectric) 상수가 있다. SMP의 탄성계수(해당 스트레인에 대한 몸체(body)에서의 스트레스 비)는 소프트 세그먼트의 용융점이나 유리전이온도 이상에서 가열될때 200 정도의 계수비로 변할 수 있다. 또한 물질의 경도는 소프트 세그먼트의 용융점이나 유리전이온도에서 또는 그 이상에 있을때 극적으로 변한다. 물질이 소프트 세그먼트의 용융점이나 유리전이온도 이상으로 가열될 때, 감쇄능력(damping ability)은 기존의 고무 제품 보다 다섯배까지 높아질 수 있다. 물질은 수많은 열적 순환 이후 원래의 주형 형상으로 쉽게 회복될 수 있으며, 하드 세그먼트의 용융점 이상으로 가열될 수 있고 새로운 원형상을 고정하도록 다시 형상화되어 냉각될 수 있다.
기존의 형상기억 중합체는 일반적으로 폴리우레탄(polyurethane)으로 절편되며(segmented) 방향족 일부(moieties)를 포함하는 하드 세그먼트을 갖고 있다. 예를 들어 Hayashi의 U.S. 특허 No. 5,145,935에는 이작용기(difunctional) 디이오시시아나이트(diiiosicyanate), 이작용기 폴리올(polyol) 그리고 이작용기 사슬 증량제(chain extender)로 중합된 폴리우레탄 엘라스토머(elastomer)로 만들어지는 형상기억 폴리우레탄 엘라스토머 성형 물품이 제시되어 있다.
공지된 SMPs의 하드 세그먼트와 소프트 세그먼트를 만들때 이용된 중합체의 예로는 다양한 폴리에테르(polyethers), 폴리아크릴레이트(polyacrylates), 폴리아마이드(polyamides), 폴리실록산(polysiloxanes), 폴리우레탄(polyurethanes), 폴리에테르 아마이드(polyether amides), 폴리우레탄/우레아(polyurethane/ureas)가 있다. 예를 들어 Ward 등의 U.S. 특허 NO. 5,5506,300; Hayashi의 U.S. 특허 No. 5,145,935; Bitler 등의 U.S. 특허 No. 5,665,822; Gorden의 " 형상기억 폴리우레탄의 응용"[Proceedings of the First International Conference on Shape Memory and Superelastic Technologies, SMST International Committee, pp.115-19(1994)]을 보라.
비록 이들 중합체의 많은 용도가 알려져 있더라도, 이들의 의학적인 응용은 신체에 이식되지 않거나 남지 않는 장치로 한정되어 있다. 그러므로 의학적인 응용을 위하여 사용되는 중합체는 형상기억 중합체를 갖지만 생물분해성 것이 바람직하다. 생물분해성 형상기억 중합체에 대한 많은 다른 응용으로는 기저귀 또는 의료용 천(drape) 안감을 제조하는 데 이용하거나, 폐기시 문제가 되는 곳에서 음식이나 다른 물질을 포장할때 이용하고 있다. 형상기억 중합체에 생물분해성 물질을 접목시켜 형상기억 중합체와 그들의 응용품에 필수적인 구조적이고 다른 물리화학적 특징들을 보유한 상업적으로 이용할 수 있는 폴리우레탄 물질은 명확하지 않다. 더우기 공지된 폴리우레탄 형상기억 중합체의 성분들은 생물학적으로 적합(biocompatible)하지 않은 생각되는 방향족 그룹을 포함하고 있다.그러므로 본 발명은 생물분해성 형상기억 중합체를 제공하는 것을 목적으로 한다.
본 발명의 다른 목적은 기존의 형상기억 중합체의 것과는 다른 물리화학적 특징들과 화학 구조를 갖는 형상기억 중합체를 제공하는 것이다.
이 출원은 일반적으로 형상기억 중합체의 분야, 특히 생물분해성 형상기억 중합체 분야에 해당된다.
이하에서는 생물분해성 형상기억 중합체의 조성물, 이들의 제조 물품과 제조 방법 및 이의 이용 방법을 설명하도록 한다. 중합체 조성물은 하나 또는 그 이상의 하드 세그먼트와 하나 또는 그 이상의 소프트 세그먼트를 포함한다. 여기서 상기 조성물은 생물학적으로 적합하며(biocompatible) 세그먼트들 중 적어도 하나는 생물분해성이 있거나, 세크먼트들 중 적어도 하나는 생물분해성 결합에 의해 다른 세그먼트에 결합된다.
하드 세그먼트의 용융점이나 유리전이온도 또는 다른 열적 효과(이후 Ttrans로 표기함)는 소프트 세그먼트의 Ttrans보다 적어도 10℃, 바람직하게는 20℃가 높다. 바람직하게는 하드 세그먼트의 Ttrans는 -30℃에서 270℃이고, 보다 바람직하게는 30℃에서 150℃사이이며, 가장 바람직하게는 30 내지 100℃ 사이이다. 하드 세그먼트: 소프트 세그먼트의 중량비는 약 5:95에서 95:5 사이이며, 바람직하게는 20:80에서 80:20이다. 형상기억 중합체는 적어도 하나의 물리적인 가교결합(하드 세그먼트의 물리 작용)을 갖거나 또는 하드 세그먼트 대신에 공유적인 가교결합을 갖고 있다. 형상기억 중합체는 상호 투과성이 있는 망상조직 또는 상호 반투과성이 있는 망상조직일 수 있다.
고체에서 액체 상태로의 변화(용융점 또는 유리전이온도)뿐 아니라 하드 세그먼트와 소프트 세그먼트는 고체에서 고체 상태 변화를 겪을 것이며, 매우 조직화된(highly organized) 수소결합에 근거한 다중전해질(polyelectrolyte) 세그먼트나 초분자(supramolecular) 효과를 포함하는 이온간의 상호작용을 겪을 수 있다.
결정체나 비결정체이며, 본 명세서에서 제한된 범위내의 Ttrans를 갖는 임의의 중합체가 하드 세그먼트와 소프트 세그먼트를 형성하기 위해 이용될 수 있다. 대표적인 생물분해성 중합체로는 폴리하이드록산(polyhydroxy acids), 폴리알카노에이트(polyalkanoates), 폴리언하이드라이드(polyanhydrides), 폴리포스파젠 (polyphosphazenes), 폴리에테르에스테르(polyetheresters), 폴리에스테라마이드 (polyesteramides), 폴리에스테르(polyesters), 폴리오쏘에스테르 (polyorthoesters)가 있다. 생물분해성 결합의 예로 에스테르(ester), 아마이드(amide), 언하이드라이드(anhydride), 카보네이트(carbonate) 그리고 오쏘에스테르(orthoester) 결합이 있다.
형상기억 중합체 조성물로부터 예를 들어 삽입 주조(injection molding), 취입성형(blowing), 압축성형(extrusion) 그리고 레이저 절제(laser ablation)를 이용하여 물품들을 만들어질 수 있다. 형상를 기억하는 물체를 만들기 위해, 물체는 하드 세그먼트의 Ttrans이상의 온도에서 형성될 수 있으며, 소프트 세그먼트의 Ttrans이하의 온도에서 냉각될 수 있다. 만약 이어서 물체가 이차 형상으로 만들어진다면, 물체는 소프트 세그먼트의 Ttrans이상 그리고 하드 세그먼트의 Ttrans이하로 물체를 가열함으로써 그것의 원형상으로 회복될 수 있다.
열경화성수지(thermoset) 중합체는 예를 들어 압축성형에 의해 거대단량체
(macromonomer)를 미리 형상화하고, 예를 들어 거대단량체에 반응성이 있는 그룹을 광경화(photocuring)함으로써 열경화성수지 중합체의 Ttrans이상의 온도에서 원형상을 고정하여 만들시킬 수 있다.

이하에서는 생물분해성 형상기억 중합체 조성물, 이들의 제조 물품 그리고 제조 방법과 그들의 이용방법을 설명하도록 한다.
정의
본 명세서에서 "생물분해성(biodegradable)"이라는 용어는 필수 구조적인 원상태(integrity)를 유지하면서도, 수분에서 3년, 바람직하기로는 1년 미만의 기간이 지나서 생리적 환경과의 상호작용으로 대사(metabolizable)되거나 배설(excretable)이 되는 성분들로 기계적인 분해에 의해 재흡수되고 (bioresorbable) 또는 분해되고(degrade) 또는 깨지는(break down), 또는 이들 과정의 조합들을 겪을 수 있는 물질을 말한다. 본 명세서에서 중합체에 대해 언급할 경우, "분해하다(degrade)"라는 용어는 중합체 사슬의 절단(cleavage)을 말하는 것으로 분자량은 분해 후 남게되는 저중합체(oligomer) 수준과 중합체의 입자들(particles)로 대략 일정하게 유지된다. 형상기억 중합체의 조성물
열가소성(thermoplastic) 형상기억 물질들은 하드 세그먼트의 Ttrans이상에서 원하는 형상화/성형(shaped/molded)되며, 중합체가 기계적인 변형을 겪고 중합체에 스트레스가 생길 수 있는 형상 회복 온도 이하의 온도로 냉각된다. 변형된 중합체의 원형상은 그들의 형상 회복 온도보다 더 높은 온도로 그들을 가열함으로써 회복된다. 이 온도 이상에서, 중합체의 스트레인은 경감되어 중합체가 그 원형상으로 회복되도록 한다. 대조적으로, 열경화성수지 형상기억 물질들은 열경화성수지 중합체를 형성하는데 사용되는 거대단량체가 중합되기 전에 원하는 형상으로 형상화/성형된다. 형상이 고정된 후, 거대단량체는 중합된다.
중합체 조성물은 열, 광선, 초음파, 자기장 또는 전기장같은 자극을 가함으로 써 고정된 변형을 갖게 되는 형상 회복 온도 이하의 온도에서 바람직하게는 적어도 1% 정도 압축할 수 있고 또는 본래 두께중 적어도 5배 정도 확대될 수 있다. 몇몇 실시예에서는 물질들은 98%(실험 실시예와 비교해서)의 회복율을 보여준다.
상당한 스트레스가 가해질때, 결과적으로 형상 회복 온도보다 더 낮은 온도에서 기계적인 변형이 일어나며, 스트레인은 소프트 세그먼트나 비결정체 부분에서 유지되며, 거대한(bulky) 형상 변화는 중합체의 탄력성에 의한 스트레인의 부분적인 유리 후에도 계속된다. 만약 유리전이온도보다 낮은 온도에서 분자 사슬의 정해진 배열에 영향을 줌으로써 분자 사슬의 배열이 끊어진다면(disturbed), 부피 크기의 증가와 자유부피 용량(free volume content)의 감소로 인해 분자 사슬의 재배열이 일어난다고 생각된다. 원형상은 기억된 형상으로 회복되는 중합체의 형상와 사슬 구조의 엄격한 조절에 따른 온도의 상승으로 인한 하드 세그먼트 집합체들(aggregates)의 수축에 의해 회복된다.
고체에서 액체 상태(용융점 또는 유리전이온도)로의 변화뿐 아니라 하드나 소프트 세그먼트는 다중전해질 세그먼트(polyelectrolyte segments), 또는 다중전해질 세그먼트 및 이온, 또는 다중음이온(polyanions) 세그먼트 및 다중양이온 (polycation)와 관련된 이온의 상호작용, 또는 매우 조직화된 수소결합에 기초한 초분자(supramolecular) 효과와 관련된 상호작용을 겪을 수 있다. SM 중합체는 또한 고체 상태에서 고체 상태 변화를 겪을 수 있다(예: 조직(morphology)상의 변화). 고체 상태에서 고체 상태 변화는 예를 들어, 폴리스틸렌블록뷰타디엔 (poly(styrene-block-butadiene))과 같이 당업자에게 잘 알려져 있다.
다양한 변화들이 형상기억 중합체를 이용해서 형성된 물체의 구조에서 일어날 수 있다. 만약 물체들이 삼차원적인 물체라면 형상에서의 변화는 이차원적일 수 있다. 만약 물체들이 실질적으로 섬유같은 이차원적인 물체라면, 형상에서의 변화는 길이방향을 따라 일어나는 일차원일 수 있다. 물질의 열과 전기적 전도율은 또한 온도 변화에 대한 반응으로 변화할 수 있다.
조성물의 수분 투과성은 특히 중합체가 박막(thin film, 예를 들어 약 10㎛ 미만) 형태일 때 가변될 수 있다. 원상태에서 어떤 중합체 조성물은 충분한 투과성을 가져 수증기 분자가 중합체 필름을 투과할 수 있으나, 물 분자는 중합체 필름을 침투할만큼 충분히 크지 않다. 결과적으로 생긴 물질들은 실온 이하 온도에서 낮은 수증기 투과성과 실온 이상의 온도에서 높은 수증기 투과성을 갖는다.
온도와는 다른 자극이 형상 변화를 유도하는데 이용될 수 있다. 아래 구체적으로 참조문헌에서 설명된 것처럼, 광 활성화 또는 중합체간의 결합을 바꾸는 이온같은 작용제에 노출됨으로 형상 변화가 유도될 수 있다.

중합체 세그먼트
세그먼트는 주로 저중합체(oligomers)이다. 본 명세서에서, "저중합체 (oligomer)"란 용어는 15,000 달톤 정도의 분자량을 갖는 선형 사슬 분자를 말한다.
중합체는 원하는 유리전이온도(만약 적어도 하나의 세그먼트가 비결정체일 경우) 또는 용융점(만약 적어도 하나의 세그먼트가 결정체일 경우)을 기초로 하고, 차례로 사용 환경을 고려하여 희망하는 응용을 기초로 하여 선택된다. 바람직하게는 중합체 블록의 수평균(number average) 분자량은 400보다 크며, 바람직하게는 500 내지 15,000 사이의 범위이다.중합체가 갑자기 유연해지고 변형되는 변화(transition) 온도는 단량체 조성과 단량체 종류를 변화시킴으로써 제어될 수 있으며, 희망하는 온도에서 형상기억 효과를 조정할 수 있다.
중합체의 열적 특성들은 예를 들어 동역학(dynamic mechanical) 온도분석법(thermoanalysis)이나 차등 스캐닝 칼로리메트리(differential scanning cololimetry, DSC) 연구에 의해 탐지될 수 있다. 뿐만 아니라 용융점은 표준 용융점 장치(standard melting point apparatus)를 이용해서 측정될 수 있다.
1. 열경화성수지나 열가소성의 중합체
비록 열가소성의 중합체가 성형이 용이하기 때문에 우선적으로 선택된다해도 중합체는 열경화성수지나 열가소성의 중합체일 수 있다.
바람직하게는 중합체나 중합체성 블록의 결정성(crystallinity) 정도는 3 내지 80% 사이이며, 보다 바람직하게는 3 내지 60% 사이이다. 모든 소프트 세그먼트가 비결정체이면서 결정성 정도가 80%보다 더 클때, 결과적으로 생긴 중합체 조성물은 빈약한 형상기억 특징을 갖는다.
Ttrans이하의 중합체의 장력계수는 일반적으로 50 MPa 내지 2 GPa(기가파스칼)사이이다. 반면 Ttrans이상의 중합체의 장력계수는 일반적으로 1 내지 500 MPa 사이이다. 바람직하게는 Ttrans이상과 이하의 장력계수의 비는 20 또는 그 이상이다. 비율이 높을수록 결과적으로 생긴 중합체 조성물의 형상기억이 더 좋다.
중합체 세그먼트는 비록 합성 중합체가 더 바람직하다해도 천연물이거나 합성물일 수 있다. 비록 의학적인 응용에 생물분해성의 생체적합성이 있는 중합체인 최종 SMP 조성물이 특히 바람직하다 할지라도 중합체 세그먼트는 생물분해성이 있을 수도 또는 생물분해성이 없을 수도 있다. 일반적으로 이들 물질들은 가수분해나 생리적 조건하에서 물이나 효소에 노출되거나 표면 부식, 대량 침식 또는 그것 모두에 의해 분해된다. 의학적인 응용에 주로 이용되는 비생물분해성(non-biodegradable) 중합체는 자연적으로 생기는 아미노산에 존재하는 것들 외의 다른 방향족 그룹을 포함하지 않는 것이 바람직 하다.
대표적인 천연 중합체 세그먼트나 중합체들에는 제인(zein), 변형된 제인(modified zein), 카세인(casein), 젤라틴(gelatin), 글루텐(gluten), 혈청 알부민(serum albumin)과 콜라겐(collagen) 같은 단백질과 키틴(chitin), 폴리(3-하이드록시알카노에이트)(poly(3-hydroxyalkanoate)s), 특히 폴리(베타-하이드록시뷰티레이트)(poly(β-hydroxybutyrate)), 폴리(3-하이드록소타노에이트)(poly(3-hydroxyoctanoate))와 폴리(3-하이드록시패티 액시드)(poly(3-hydroxyfatty acids))뿐만 아니라 알지네이트(alginate), 셀롤로오스(cellose), 덱스트란 (dextran), 플루란(pullulane) 그리고 폴리하이알루론산(polyhyaluronic acid)같은 다당류가 있다.
대표적인 생물분해성 천연 중합체 세그먼트나 중합체에는 알지네이트, 덱스트란, 셀룰로오스, 콜라겐같은 다당류와 이들의 화학 유도체(치환, 알킬기 (alkyl), 알킬렌(alkylene), 하이드록실화(hydroxylation), 산화와 같은 화학 그룹의 첨가, 그리고 당업자에 의해 가능한 다른 변형), 그리고 알부민(albumin), 제인, 공중합체와 같은 단백질, 또한 하나의 합성 중합체 또는 합성중합체의 조합과 같은 이들의 혼합물(brends) 같은 것이 있다.

또한 중합체의 부드러운 표면이 침식됨에 따라 외부 표면에 노출된 카르복실(carboxyl) 그룹을 갖고 있는 폴리락타이드-코-글리코라이드 (poly (lactide-co-glycoride)s), 폴리언하이드라이드(polyanhyderide)와 폴리오쏘에스테르(polyorthoester) 같이 빠른 생물침식성(bioerodible) 중합체가 이용될 수 있다. 게다가, 폴리언하이드라이드와 폴리에스테르 같은 불안정한(labile) 결합을 갖고 있는 중합체는 그들의 가수분해 반응성으로 잘 알려져있다. 그들의 가수분해속도는 일반적으로 중합체 뼈대에서의 간단한 변화와 그들의 연쇄(sequence) 구조에 의해 바뀔수 있다.
폴리아세틸렌과 폴리피롤(polypyrrole)같은 다양한 중합체는 전도할 수 있는 중합체이다. 이들 물질들은 전기적 전도가 중요한 곳에 사용할때 특히 바람직하다. 이 물질들의 사용예로 세포 성장이 촉진되는 조직 공학(tissue engineering) 및 다른 생물의학적 응용을 들 수 있다. 이들 물질들은 온도의 증가 없이도 SMAs 보다 더 잘 열을 흡수할 수 있기 때문에 컴퓨터 과학분야에서 특별한 용처를 갖는다. 형상기억 중합체로 행동하는 것은 예를 들어 신경조직같은 조직의 성장을 촉진하기 위한 조직 공학분야에서 유용하다.
2. 하이드로겔
중합체는 일반적으로 물 중량의 약 90%까지를 흡수할 수 있는 하이드로겔의 형태로 있을 수 있으며, 선택적으로 다가(multivalent) 이온이나 중합체와 이온적으로 가교결합될 수 있다. 소프트 세그먼트간의 이온 가교결합은 변형될때 소프트 세그먼트간의 이온 가교결합을 깸으로써 변형될 수 있는 구조를 유지하는데 이용될 수 있다. 중합체는 또한 물이나 수용액과 다른 용매에서 겔의 형상으로 있을 것이다. 이들 중합체에서, 임시 형상는 소프트 세그먼트간의 친수성의 상호작용에 의해 고정될 수 있다.

이트(poly(ethylene terephthalate)), 폴리비닐아세테이트(poly(vinyl acetate)), 및 이들의 공중합체와 혼합물로부터 형성될 수 있다. 예를 들어 아크릴산(acylic acid)같은 몇가지 중합체의 세그먼트는 중합체가 수화되고 하이드로겔이 형성될때만 엘라스토머이다. 예를 들어 메타크릴산(methacrylic acid)같은 다른 중합체의 세그먼트는 결정체이며 중합체가 수화되지 않을 때에도 용융될 수 있다. 희망하는 응용과 이용 조건에 따라 다른 타입의 중합체 블록이 이용될 수 있다.
예를 들어 형상기억은 하이드로겔 상태에서만 아크릴산 공중합체에서 관찰되는데, 아크릴산 단위는 실질적으로 수화되며 매우 낮은 유리전이온도를 갖는 연한 엘라스토머처럼 작용하기 때문이다. 건조 중합체는 형상기억 중합체가 아니다. 건조될때, 아크릴산 단위는 유리전이온도 이상에서도 견고한 플라스틱으로 작용하며, 가열시 기계적인 특징들에 있어 급박한 변화는 보이지 않는다. 대조적으로 소프트 세그먼트같은 메틸 아크릴레이트 중합체 세그먼트를 포함하는 공중합체는 건조할때만 형상기억 특징들을 보인다.
3. 온도 상승시 겔을 형성할 수 있는 중합체
예를 들어 폴리에틸렌 옥사이드-코-프로필렌(poly(ethylene oxide-co-propylene oxide)(PLURONICSTM)같은 어떤 중합체는 체온보다 낮은 온도에서 물에 녹으며 체온보다 높은 온도에서 하이드로겔이 된다. 형상기억 중합체에서 세그먼트처럼 이들
중합체의 결합(incorporation)은 기존의 형상기억 중합체의 것과 반대방법으로 온도 변화에 반응할 수 있는 능력을 그들에게 제공한다. 이들 물질들은 그들의 형상기억 온도 이상에서 가열되기보다는 오히려 그들의 형상 회복 온도 이하에서 냉각될때 그들의 형상를 복원한다. 이러한 효과를 역전된 온도 형상기억 효과라 부른다. 이들 중합체 세그먼트를 포함하는 형상기억 중합체 조성물은 중합체가 액체상태로 사출되고, 원위치(in situ)에서 의도된 형상를 복원하기 위해 냉각될 수있는 다양한 생물의학적 응용에 유용하다. 역전된 온도 형상기억 효과는 두 개의 다른 세그먼트를 Tmisc보다 더 낮은 온도에서 섞일 수 있지만(miscible) 더 높은 온도에서는 섞일 수 없는 중합체로 결합시킴으로써 얻을 수 있다. 더 높은 온도에서 위상 분리는 임시 형상를 안정화시킨다.

II.
중합체 세그먼트의 조립
형상기억 중합체는 한개 이상의 하드 세그먼트와 한개 이상의 소프트 세그먼트를 포함하며, 여기서 세그먼트중 적어도 하나는 생물분해성이 있거나 세그먼트중 적어도 하나는 생물분해성 결합에 의해 다른 세그먼트와 결합된다. 대표적인 생물분해성 결합으로는 에스테르-, 아미드-, 언하이드라이드-, 카보네이트- 또는 오쏘에스테르(orthoester) 결합이 있다.
1. 중합체 구조
형상기억 효과는 중합체의 형태(morphology)에 기초를 두고 있다. 열가소성의 엘라스토머와 관련하여, 물체의 원형상은 하드 세그먼트에 의해 생기는 물리적인 가교결합으로 인해 고정된다. 열경화성수지 중합체에 있어서, 소프트 세그먼트는 하드 세그먼트를 갖는 대신 공유적으로 가교결합된다. 원형상은 가교결합 과정에 의해 만들어진다.
종래의 절편으로 된(segmented) 폴리우레탄 SMPs과는 대조적으로, 본 명세서에서 설명하고 있는 조성물의 세그먼트는 선형일 필요는 없다. 세그먼트는 덴드리머의 측면 그룹에 부분적으로 융합되거나 부착된다.
A. 열가소성과 열경화성수지 중합체
중합체는 덴드릭 구조를 포함하며, 그것을 혼합한 선형의 이중블록, 삼중블록, 사중블록 또는 다중블록의 공중합체, 가지(branch)나 그래프트(graft) 중합체, 수수지상결정(樹枝狀結晶, dendritic) 구조를 포함하는 열가소성의 엘라스토머 (elastomer) 및 이들의 혼합 형태로 있을 수 있다. 도 3은 하드 세그먼트와 소프트 세그먼트를 형성하는 적당한 종류의 열가소성의 물질의 몇몇 조합을 나타낸 것이다. 열가소성의 형상기억 중합체 조성물은 또한 한개 이상의 이중블록, 삼중블록, 사중블록, 또는 다중블록을 갖는 하나 이상의 상동(homo-) 또는 공(co-)중합체, 가지나 그라프트(graft) 중합체의 혼합물일 수 있다. 이들 중합체 타입은 당업자에게 잘 알려져 있다.
본 명세서에서 "분해성 열경화성수지(degradable thermoset)"란 용어는 i) 쪼개질 수 있는 결합을 갖는 하나의 소프트 세그먼트만을 포함하는 열경화성수지의 SMPs와, ii) 적어도 하나의 소프트 세그먼트는 분해될 수 있거나 또는 다른 소프트 세그먼트가 쪼개질 수 있는 결합으로 연결된 한개 이상의 소프트 세그먼트를 포함한 열경화성수지를 말한다. 또한 분해성 열경화성수지 중합체는 250℃ 내지 -40℃ 사이의 Tm을 갖는 공유적으로 가교결합된 결정체의 소프트 세그먼트를 포함하거나 또는 200℃ 내지 0℃ 사이의 Tm을 갖는 공유적으로 가교결합된 결정체의 소프트 세그먼트를 포함할 수 있다.형상기억능을 갖는 열경화성수지 중합체는 4종류의 다른 타입이 있다. 이들은 중합체 망상조직, 반투과성 망상조직, 상호투과성 망상조직 그리고 혼합된 상호투과성 망상조직를 포함한다.
i. 중합체 망상조직
중합체 망상조직은 공유적으로 가교결합하는 거대단량체, 예를 들어 탄소간 이중결합처럼 중합할 수 있는 말단기를 갖는 중합체로 만들어진다. 중합 과정은 빛이나 열에 민감한 개시제(initiator)를 이용하거나 또는 개시제가 없이 자외선으로 경화함으로써 유도될 수 있다. 형상기억 중합체 망상조직은 한번 이상의 온도 변화에 따른 한개 이상의 소프트 세그먼트를 가교결합시킴으로써 만들어진다.
생물의학적 응용에 대해 구체적으로 말하자면, 가교결합은 광가교결합제 (photocrosslinker)를 이용하여 수행하며 화학적 개시제를 필요로 하지 않는다. 광가교결합제는 독성이 있을 수 있는 개시제 분자의 필요성을 유리하게 제거한다. 도 4는 약 65%의 수율을 생성하는 바람직한 광가교결합제의 합성을 위한 반응 순서에 대한 것이다.
ii. 상호투과성이 있는 망상조직
상호투과성이 있는 망상조직(interpenetrating networks, "IPN")은 두개의 성분이 가교결합되지만 망상조직 상호간에는 가교결합되지 않는 망상조직으로 정의한다. 원형상은 가장 높은 가교결합 밀도와 가장 높은 기계적 강도를 갖는 망상조직으로 결정된다. 물질은 두 망상조직의 다른 소프트 세그먼트에 상응하는 적어도 두개의 Ttrans를 가진다.
iii. 혼합된 상호투과성이 있는 망상조직
혼합된 IPN은 적어도 하나의 물리적으로 가교결합된 중합체 망상조직(열가소성의 중합체)과 어떠한 물리적인 방법에 의해서도 분리될 수 없는 적어도 하나의 공유적으로 가교결합된 중합체 망상조직(열경화성수지의 중합체)을 포함하고 있다. 원형상은 공유적으로 가교결합된 망상조직에 의해 고정된다(set). 임시 형상는 소프트 세그먼트의 Ttrans와 열가소성의 엘라스토머 성분의 하드 세그먼트의 Ttrans에 상응한다.
특히 바람직한 혼합된 상호투과성이 있는 망상조직은 예를 들어 탄소간 이중결합의 광중합화에 의해 열가소성의 중합체의 존재하에서 반응성 거대단량체를 중합함으로써 만들어진다. 이 경우에 열가소성의 중합체에 대한 열경화성수지 중합체의 중량비는 바람직하게는 5:95 내지 95:5 사이이며 보다 바람직하게는 20:80 내지 80:20 사이이다.
iv. 반투과성이 있는 망상조직
반투과성이 있는 망상조직("semi-IPN")은 한 성분은 가교결합된 중합체(중합체 망상조직)이고 다른 성분은 비가교결합된 중합체(상동중합체나 공중합체)인 두개의 독립적인 성분으로 정의한다. 여기서 성분들은 물리적 방법으로 분리될 수 없다. 세미(semi)-IPN은 소프트 세그먼트와 상동중합체나 공중합체 성분에 상응하는 적어도 한번의 온도 변화를 갖는다. 가교결합된 중합체는 반투과성이 있는 망상조직 조성물의 중량으로 인해 바람직하게는 약 10 내지 90%사이에서 구성된다.
v. 중합체 혼합
바람직한 실시예에서, 본 명세서에서 기술된 형상기억 중합체 조성물은 생물분해성 중합체 혼합물로 형성된다. 본 명세서에서 "생물분해성 중합체 혼합물 (biodegradable polymer blend)"은 적어도 하나의 생물분해성 중합체를 갖는 혼합물이다.
형상기억 중합체는 열가소성 중합체의 물리적인 혼합물로써 존재할 수 있다. 일실시예에서, 형상기억 중합체 조성물은 두 개의 열가소성 중합체를 상호작용시키거나 혼합시킴으로써 만들어질 수 있다. 중합체는 반결정성(semicrystalline)
상동중합체(homopolymer), 반결정성 공중합체, 선형 사슬을 갖는 열가소성의 엘라스토머, 임의의 수지상결정(dendritic) 구조 성분이나 측쇄(side chain)를 갖는 열가소성의 엘라스토머, 및 가지를 갖는(branched) 공중합체, 그리고 이들의 조성물이 임의로 혼합된 형태일 수 있다.
예를 들어, 상대적으로 높은 Ttrans를 갖는 하드 세그먼트와 상대적으로 낮은 Ttrans를 갖는 소프트 세그먼트를 포함하는 첫번째 다중블록 공중합체는 상대적으로 낮은 Ttrans를 갖는 하드 세그먼트와 두번째 다중블록 공중합체와 동일한 소프트 세그먼트를 갖는 두번째 다중블록 공중합체와 섞이거나(mixed) 혼합(blended)될 수 있다. 상기 두 다중블록 공중합체의 소프트 세그먼트는 동일하므로, 이 중합체들은 소프트 세그먼트가 용융될때 서로 섞일 수 있다. 이 경우에 세개의 전이온도가 있는데, 이는 첫번째 하드 세그먼트, 두번째 하드 세그먼트, 그리고 소프트 세그먼트의 것이다. 따라서 이들 물질들은 두개의 다른 형상를 기억할 수 있다. 이들 중합체의 기계적인 특징들은 두 중합체의 중량비를 바꿈으로써 조정될 수 있다.
세그먼트들 중 적어도 한 개가 다른 다중블록 공중합체의 세그먼트 중 적어도 한개와 혼합될 수 있는, 적어도 두 개의 다중블록공중합체의 다른 종류가 제도될 수 있다. 만약 두개의 다른 세그먼트가 섞여서 함께 하나의 계(domain)을 형성한다면, 이 계의 열적 전이는 두 세그먼트의 중량 하량에 의존한다. 기억된 형상의 최대수는 혼합물의 온도 전이 수로부터 유래한다.
형상기억 혼합물은 혼합 성분 혼자인 것보다 더 나은 형상기억능을 가질 것이다. 형상기억 혼합물은 적어도 하나의 다중블록(multiblock) 공중합체와 적어도 하나의 상동중합체 또는 공중합체로 구성되어있다. 대체로 디(di-), 트리(tri-), 테트라(tetra-) 블록 공중합체가 다중블록 공중합체 대신에 이용될 수 있다.
형상기억 혼합물은 산업적인 응용에 매우 유용한데, 이는 기계적, 열적, 그리고 형상기억능의 폭넓은 범위가 단지 둘 또는 세 개의 기초 중합체로부터 이들을 서로 중량비율로 혼합함으로써 얻어질 수 있기 때문이다. 쌍정 나사 압출기(twin screw extruder)는 성분을 혼합하고 혼합물을 처리하는 데 이용될 수 있는 표준 처리 장치의 일례이다.
III. SMPs를 만드는 방법
위에서 언급한 중합체들은 상업적으로 이용할 수 있거나 또는 일반 화학을 이용하여 합성될 수 있다. 당업자는 공지된 화학을 이용하여 이들 중합체를 쉽게 제조할 수 있다. 아래의 실시예 1과 2는 SMPs를 만드는데 이용된 실험 과정을 설명하고 있다.
IV. SMP 조성물을 형성하는 방법
조성물은 적당한 조건, 예를 들어 하드 세그먼트의 Ttrans이상의 온도에서 일차 형상으로 형성될 수 있으며, 소프트 세그먼트의 Ttrans이하에서 냉각될 수 있다. 표준 기술들로는 압출과 사출성형이 있다. 선택적으로, 물체들은 이차 형상으로 재형성(re-formed)될 수 있다. 열이나 다른 적절한 일련의 조건을 적용하면 물체는 원형상으로 회복한다.
열경화성수지 중합체는 중합하기전(pre-polymerized) 물질(거대단량체)을 압출하고, 예를 들어 상기 단량체의 반응 그룹을 광경화함으로써 열경화성수지 중합체의 Ttrans이상의 온도에서 원형상을 고정하여 제조될 수 있다. 임시 형상는 물질을 변형한 후 Ttrans이하로 물질을 냉각함으로써 고정된다. 도 5는 광유도된 (photoinduced) 형상기억 효과를 그린 것이다.
또한 가교결합은 거대단량체의 용액에서 일어날 수 있다. 용매는 나중 단계에서 형성된 겔로부터 제거된다.
열가소성 중합체로 만들어진 이 조성물들은 박판(sheet)으로 압출되거나 예를 들어 섬유를 형성하도록 사출성형에 의해 형성될 수 있다. 조성물들은 또한 고체 물체을 형성하기 위해 당업자에게 알려진 다른 방법, 예를 들어 레이저 절제, 미세기계가공, 열선의 이용 및 CAD/CAM 공정에 의해 형성될 수 있다. 이들 공정들은 열경화성수지 중합체를 형성하는데 바람직하다.
V.
치료, 진단 그리고 예방의 응용
임의의 다양한 치료, 예방 및/또는 진단 약품은 환자에 투여후 국부적으로 또는 체계적으로 혼합 약품을 전달할 수 있는 중합체 조성물 내에 혼입될 수 있다.
1. 치료, 진단 그리고 예방의 응용
임의의 다양한 치료제들은 환자에게 투여된 후 혼합한 약품을 국부적으로 또는 체계적으로 전달하기 위해 입자 내에 혼입(incorporated)될 수 있다. 그 예로 치료, 예방, 또는 진단활성을 갖는 합성 무기화합물과 유기화합물 또는 분자, 단백질과 펩티드, 다당류와 다른 당류, 지질 그리고 핵산 분자가 있다. 핵산분자에는 유전자, 플라스미드 디엔에이(plasmid DNA), 네이키드 디엔에이(naked DNA), 전사(transcription)를 저해하는 보조(complementary) 디엔에이에 결합된 안티센스(antisense) 분자, 리보자임(ribozyme) 및 리보자임 안내 시퀀스(ribozyme guide sequence)가 있다. 혼입된 약품은 혈관에 작용하는(vasoactive) 약품, 신경활성(neuroactive) 약품, 호르몬, 성장인자, 사이토카인(cytokine), 마취제, 스테로이드(steroid), 항혈액응고제, 항염증제, 면역조절약품, 세포파괴(cytotoxic)약품, 예방제, 항생제, 항바이러스, 안티센스, 항원, 및 항체같은 다양한 생물학적 활성을 갖는다. 예를 들어, 단백질은 적당한 반응을 유도하기 위해 주사로 투여되어야 할 항체나 항원일 수 있다. 단백질은 100개 이상의 아미노산 잔기로 구성된 것으로; 펩티드는 100개 미만의 아미노산 잔기를 갖는 것으로 한정된다. 만약 달리 언급하지 않는다면, 단백질이란 용어는 단백질과 펩티드 둘 다를 말한다. 또한 헤파린같은 다당류가 투여될 수도 있다. 예를 들어 몰당 10 내지 500,000 그램 사이인 넓은 분자량을 갖는 화합물이 캡슐에 넣어질 수 있다.
사용될 수 있는 가상의 약품들로는 양전자 방출 x선 단층 촬영법(positron emission tomography, PET), 컴퓨터를 사용한 x선 단층 촬영법(computer assited tomography, CAT), 단일 광자 방출 컴퓨터 x선 단층 촬영법, x-ray, 형광투시법(fluoroscopy), 자기 공명 영상법(MRI), 초음파 약품에 이용되는 상업적으로 이용가능성이 있는 약품이 있다.
VI. 물
품, 장치와 코팅
SMP 조성물은 생물의학적(biomedical)으로 그리고 다른 응용에서 이용될때 수많은 제조 물품을 만드는데 이용될 수 있다.
1.생물의학적 응용을 위한 물품과 장치
중합체 조성물은 생물의학적 응용에 사용되는 제조 물품을 만들때 이용될 수 있다. 예를 들어, 봉합선, 치열 교정(orthodontic) 물질, 뼈 나선부(bone screw), 손톱, 의치(plates), 도뇨관(catheters), 관(tubes), 피막(films), 스텐츠 (stents), 치열 교정기(orthopedic braces), 부목(splints), 깁스 테입(tape for preparing casts)과 조직공학에 사용되는 뼈대(scaffolds), 콘택트 렌즈, 약 전달 기구, 이식조직(implant), 열 감지기를 만들 수 있다.
SMP 조성물은 주로 생체적합성(biocompatible) 중합체와 대부분의 응용에서 생물분해성 중합체로부터 만들어지는 것이 바람직하다. 생물분해성 중합체는 중합체의 조성과 가교결합에 의존하여 제어되는 속도로 분해된다. 분해성 중합체 조직(implant)은 이식후 회복(implant retrieval)의 필요를 없애며 동시에 치료제를 전달용으로 이용될 수 있다.
물질들은 부하지지력(load-bearing capacity)과 제어된 분해를 필요로하는 많은 응용에 이용될 수 있다.
중합체 조성물은 기계적인 기능을 제공하기 위해 체내에 이식될 수 있는 이식조직의 형상으로 형성될 수 있다. 이러한 이식물의 예로 막대, 핀, 나사, 의치와 해부판(anatomical shapes)이 있다.
조성물 중 특히 바람직한 사용으로는 삽입을 용이하게 하기 충분히 견고하나, 체온에 도달하면 치료에 적합하면서도 환자가 보다 편안하게 느끼도록 부드럽게 되어 두번째 형상을 형성하는 봉합선이 있다.
다른 바람직한 이용으로 도뇨관(catheters) 분야가 있다. 도뇨관은 삽입이 용이하도록 체온 부근에서는 견고하지만, 체온으로 가열된 후에는 환자가 편안함을 느끼드록 유연해질 수 있다.
바람직한 실시예에서, 이식을 위한 물품은 방향족 그룹을 포함하지 않는 형상기억 중합체로부터 제조된다.중합체 조성물은 주입기(fillers), 강화물질, 방사영상(radioimaging) 물질, 첨가제(excipients) 또는 특히 이식응용에 필요한 다른 물질과 결합될 수 있다. 주입기(fillers)의 예로는 U.S.특허 No.5,108,755에 기재된 칼슘-나트륨-메타포스페이트(calcium-sodium-metaphosphate)를 포함하고 있다. 당업자는 조성물에 포함되는 이들 물질의 적절한 양을 쉽게 결정할 수 있다.
상기 물품들은 위에서 설명한 것처럼, 다양한 치료와 진단에 적용할 수 있다.
2.비 의학적 응용
생물 의학적 응용과는 다른 형상기억 중합체 조성물에 대한 많은 응용들이 있다.
생물분해성 중합체에 대한 비의학적 타입의 응용예로는 일회용 기저귀와 포장물질 같이 폐기시 문제가 되는 항목을 포함된다.
3.분해가 억제되는 코팅
형상기억 중합체는 분해속도가 다양하도록 설계될 수 있다. 예를 들어, 일실시예에서는 가수분해 분해성 중합체는 물이 거대 중합체(bulk polymer)의 가수분해에 의해 깨질 수 있는 결합에 일시적으로 도달하지 못하도록 소수성의 SMP 코팅을 적용함으로써 선택적으로 보호될 수 있다. 그리고나서 코팅의 보호적 특성은 외부 자극의 적용을 원할 때 변형될 수 있는데, 이는 코팅의 확산 특성이 물이나 다른 수용액이 코팅을 침투하여 분해 과정을 개시하도록 변형되기 때문이다. 만약 가수분해속도가 물의 확산속도보다 상대적으로 높다면, 코팅을 침투하는 물의 확산 속도가 분해속도를 결정한다. 다른 실시예에서는 빽빽히 가교결합된 소프트 세그먼트로 이루어진 소수성 코팅이 물이나 수용액에 대한 확산 장벽(barrier)으로써 이용될 수 있다. 소프트 세그먼트는 자극이 가해짐으로 쪼개질 수 있는 결합에 의해 적어도 부분적으로라도 가교결합되어야한다. 물의 확산 속도는 가교결합 밀도를 낮춤으로써 증가될 수 있다.
VII.
사용방법
어떤 제조 물품은 만약 정상적인 사용에 모순된 방법으로 작용하지만 않는다면 지정된 형상를 유지하도록 디자인된다. 예를 들어, 차의 범퍼(bumper)는 충격을 받지 않는다면 지정된 형상를 유지할 것이다. 이들 제조 물품은 예를들어 열 처리에 의해 손상을 입으면 지정된 형상으로 이용되거나 수리가 된다.
다른 제조 물품의 경우 일차 형상는 처음 이용할 때 지정되고, 이차 형상는 차후에 이용시 지정되어 사용되도록 고안된다. 이들의 예로는 체온에서 또는 체온 이상에서 장치를 가열하는 것과 같은 외부자극을 가할 경우 이차 형상를 형성할 수 있는 생물 의학 장치가 있다.
또한 다른 제조 물품은 이들 형상이 반응이나 조절시 의료 장치의 열감지기 같이 온도에 따라 변형되도록 디자인될 수 있다.
본 발명은 다음의 비제한적인 실시예들을 참조하여 보다 잘 이해될 수 있다.
실시예 1: 코폴리에스터우레탄(copolyesterurethane) 형상기억 중합체
열 형상기억 효과를 나타내는 생체적합성(biocompatible)이 있고 생물분해성이 있는 다중블록-공중합체 군을 합성하였다. 이들 중합체는 실온과 체온 사이의 열 전이 온도 Ttrans를 갖는 결정성 하드 세그먼트(Tm)와 소프트 세그먼트로 이루어져 있다. 종래의 단편으로 된(segmented) 폴리우레탄과는 대조적으로, 하드 세그먼트는 올리고에스테르(oligoester)나 올리고에테르에스테르(oligoetherester)이지만 어떤 방향족 성분도 포함하지 않았다.
임시 형상를 프로그램하고 다중블록-공중합체의 영구적인 형상를 회복시키는 메카니즘은 도 6에 나타나있다. 물질의 영구적인 형상는 Ttrans이상에서 중합체를 녹이고 냉각함으로써 고정된다(도 6- 맨위). 그리고나서, 중합체는 임시 형상를 이루게되며(도 6-오른쪽), Ttrans이하에서 냉각함으로써 고정되었다(도 6-아래쪽). 부하제거(unloading) 후 영구적인 형상는 Ttrans이상으로 재가열함으로써 회복되었다.
양말단에 작용기를 갖는 저중합체(oligomer)인 텔레켈릭(telechelics)의 합성.
텔레켈릭 매크로디올(telechelic macrodiol)은 대기중 질소 가스하에서 트랜스에스테르화(tranesterization) 촉매로써 디엔부틸티녹사이드(di(n-butyl)tinoxide)를 갖는 고리 단량체의 개환(ring opening) 중합화에 의하여 합성되었다.
하드 세그먼트
알파,오메가-디하이드록시[올리고(에틸렌글리콜글리콜레이트)에틸렌올리고(에틸렌글리콜글리콜레이트)]-(PDS 1200과 PDS 1300)(α,ω-dihydroxy[oligo(ethylene glycol glycolate)ethylene oligo(ethylene glycol glycolate)]-(PDS 1200 and PDS 1300))은 다음과 같이 만들어졌다. 단량체 피-디옥산-2-온(p-dioxne-2-one)은 사용전에 상기 저중합체(oligomer)의 증류(열적 해중합(depolymerization))함으로써 얻어졌다. 단량체 57그램(0.63몰), 에틸렌 글리콜(etylene glycol) 0.673그램(10.9mmol), 디(엔-부틸)티녹사이드(di-n-butyl tinoxide) 0.192그램 (0.773mmol)을 24시간동안 80℃로 가열했다. 반응 종결은(평형) GPC에 의해 측정되었다. 생성물은 뜨거운 1,2-디클로로에탄(1,2dichloroethane)에서 용해되고 실리카겔(silica gel)이 충진된 Buechner-funnel을 통해 뜨거운 상태로 걸러졌다. 헥산(hexane)에서 침전하고 6시간동안 진공상태로 건조하여 생성물을 얻었다.
소프트 세그먼트
i. 결정체
다른 Mn을 갖는 폴리(입실론-카프로락톤)-디올(poly(ε-caprolactone)-diol)은 예를 들어 알드리치社와 폴리사이언스社로부터 구입할 수 있다. 본 실시예에서는 PCL-2000이 사용되었다.
ii. 비결정체
알파,오메가-디하이드록시[올리고(엘-락테이트-코-글리코레이트)에틸렌 올리고(엘-락테이트-코-글리코레이트)]-(약칭하여:PLG2000-15)(α, ω-oihydroxy[oligo
(L-lactate-co-glycolate) ethylene oligo(L-lactate-co-glycorate)]는 다음과 같이 만들어졌다. 1000ml 이목(two-neck) 둥근바닥 플라스크에, 엘,엘-디락타이드(L-L-dilactide) 300그램(2.08몰), 디글리콜리드 (diglycolide) 45그램(0.34몰)과 에틸렌 글리콜(ethylene glycol) 4.94그램(0.8몰)을 넣고 녹이기 위해 40℃에서 가열하고 저어주었다. 디(엔-부틸)티녹사이드 (di(n-butyl)tinoxide) 0.614그램(2.5mmol)을 첨가했다. 7시간이 지난후 반응은 GPC로 측정했을때 평형에 도달했다. 반응 혼합물은 1,2-디클로로에탄(1,2-dichloroethane)에서 용해되고 실리카겔 컬럼(column)에서 분리되었다. 헥산에서 침전하고 6시간동안 진공상태로 건조하여 생성물을 얻었다.
텔레켈릭의 특징
매크로디올(macrodiol)의 분자량 Mn과 온도 특성은 아래 표 1에서처럼 측정되었다.

열가소성의 엘라스토머의 합성(다중블록 공중합체)
0.4nm 크기의 분자 지브(sieve)로 충진된 삭렉스(soxleth) 추출장치가 연결되어 있고 이목 둥근바닥 1000ml 플라스크에 아래 표 2에서 설명하고 있는 두개의 다른 매크로디올을 1,2-디클로로에탄 80ml에서 용해시켰다. 혼합물은 용매의 아제오트로픽(azeotropic) 추출에 의해 건조하도록 환류(reflux)되었다. 새로 증류된 트리메틸헥산-1,6-디이소시아나이드(trimethylhexane-1,6-diisocyanide)는 주사기로 첨가되며, 반응 혼합물은 적어도 10일동안 80℃로 가열되었다. 일정한 시간간격으로, 혼합물의 샘플을 취하여 GPC로 중합체의 분자량을 측정하였다. 반응의 마지막에, 생성물은 헥산에서 중합체를 침전시켜 얻었고 1,2-디클로로에탄에서 반복적으로 녹이고 헥산에서 침전시켜 분리했다.
다중블록 공중합체는 중합체의 다음 두가지 타입으로부터 제조되었다.
(i) PDC 중합체는 폴리(입실론-카프로락톤)을 포함한다. 소프트 세그먼트에 대한 Ttrans는 용융점이다.
(ii) PDC 중합체는 알파, 입실론-디하이드록시[올리고(엘-락테이트-코-글리코레이트)에틸렌 올리고(엘-락테이트-코-글리코레이트)]을 포함한다. 소프트 세그먼트에 대한 Ttrans는 유리 전이 지점이다.

열가소성 엘라스토머의 특징
조성물을 결정짓는 물리적, 기계적 그리고 분해 특성은 아래 표 3-9에 나와있다.
새로운 물질의 가수분해작용은 37℃에서 pH 7의 완충용액에서 시험되었다. 중합체는 완전히 분해되며 그들의 분해 속도는 쉽게 가수분해성 에스테르 결합의 농도에 의해 조정될 수 있음을 보여준다. 37℃에서 상대 질량 손실값은 mr=m(t0)/m(t) %이며, 37℃에서 상대 분자량의 손실은 Mr=Mw(t)/Mw(t0) %이다.
두가지 다른 다중블록-공중합체의 독성은 달걀 시험을 이용해서 조사했다. 혈관은 일정하게 발달했으며 그들의 조건은 중합체 샘플에 의해 영향을 받지 않았다.

도 7은 열분해(thermolytic) 순환 수의 함수로 다중블록 공중합체에 대해 실행한 장력 실험의 결과를 보여준다. 열순환적으로 처리된 중합체의 평균 형상 고정율(fixity rate)과 스트레인 회복율의 의존도의 순화수의 관계는 아래 표 10과 11에 각각 나타내었다. 중합체는 높은 형상 고정성을 가지며, 단지 두번의 순환후 평형상태에 도달했다.

폴리(입실론-카프로락톤)디메타크릴레이트과 열경화성수지의 범위는 그들의 기계적 특성과 형상기억 특성으로 평가되었다.
거대단량체의 합성
폴리(입실론-카프로락톤)디메타크릴레이트(PCLDMAs)는 다음과 같이 만들어졌다. 건조상태의 THF 200ml에 Mn=2'000g/mol(20.0g, 10mmol)인 폴리(입실론-카프로락톤)디올과 트리에틸아민(triethylamine)(5.3mL, 38mmol)이 용해된 용액에 메타크릴로일(methacryloyl) 클로라이드(3.7mL, 38mmol)를 0℃에서 방울로 첨가했다. 용액을 3일동안 0℃에서 저어주고 침전된 염을 여과시켰다. 압력을 감소시킨 상태로 실온에서 혼합물을 농축시킨 후, 에틸 아세테이트 200mL을 첨가하고 수용액을 다시 여과하여 헥산, 에틸 에테르와 메탄올(methanol)을 18:1:1로 섞은 혼합물을 10배 이상으로 침전시켰다. 무색의 침전물이 모집되었으며, 디클로로에탄 200mL에 녹여 다시 침전시키고, 압력을 감소한 상태에서 실온에서 주의하여 건조시켰다.
열경화성수지의 합성
거대단량체(단량체 혼합물)는 그 용융점(Tm) 보다 10℃ 이상으로 가열되고 두개의 유리판(25 mm x 75 mm)과 0.6 mm 두께의 테프론 스페이서로 이루어진 주형에 채웠다. 높은 동질성을 얻기위해, 주형은 다른 시간동안에 Tm에서 저장되었다.
광경화과정은 15분동안 Tm에서 가열된 판에서 실행되었다. 열 램프 헤드와 샘플간의 거리는 5cm였다. 실온에서 냉각한 후, 샘플을 추출하고 하루동안 100배 과잉 디클로로메탄으로 팽창시켜서 주의깊게 세척되었다. 마지막으로, 샘플은 감압상태로 실온에서 건조되었다.
거대단량체와 열경화성수지의 특성
아래 표 12는 상대적인 아크릴화 정도(Da)(%)에 따라 만들어진 폴리(입실론-카프로락톤)디메타크릴레이트를 열거한 것이다. 다음 PCLDMA 번호는 1H-NMR과 GPC를 이용하여 측정했을때 합성에 사용된 폴리(입실론-카프로락톤)디올의 분자량Mn이다.

실온에서 폴리(입실론-카프로락톤) 열경화성수지 C1에서 C7의 장력 특성은 아래 표 13에 나와있다. 여기서 E는 탄성 계수(Young율)이며, εS는 신장율(elongation)이고, σS는 산출점에서의 스트레스이고, σmax는 최대스트레스이며, εmax는 σmax에서의 신장율이고, εR은 파괴시(at break)의 신장율이며, σR은 파괴시의 스트레스를 뜻한다. 아래의 표 14는 70℃에서 같은 폴리(입실론-카프로락톤) 열경화성수지의 장력 특성을 보여준다.

형상기억 특성
열경화성수지는 표 15에서 나타난 열기계적인 특성을 갖는 것으로 정해졌다. 수평균분자량(Mn)은 거대단량체의 것이다. 낮은 한계온도 T1은 0℃이며, 높은 한계온도 Th는 70℃이다. 임시 형상에서 신장은 50%이다. Rr(2)는 이차 순환의 스트레인 회복율이며, Rr,tot는 5번 순환 이후 총 스트레인 회복율이고, Rf는 평균 스트레인 고정율이다.

물품, 장치와 코팅
1.생물의학적 응용을 위한 물품과 장치
중합체 조성물은 생물의학적 응용에 사용되는 제조 물품을 만들때 이용될 수 있다. 예를 들어, 봉합선, 치열 교정(orthodontic) 물질, 뼈 나선부(bone screw), 손톱, 의치(plates), 도뇨관(catheters), 관(tubes), 피막(films), 스텐츠 (stents), 치열 교정기(orthopedic braces), 부목(splints), 깁스 테입과 조직공학에 사용되는 뼈대(scaffolds), 콘택트 렌즈, 약 전달 장치, 이식조직(implant), 열 감지기를 만들 수 있다. 또한 체내에 이식될 수 있는 임플란트의 형상으로 형성될 수 있다. 그러한 임플란트의 예로 막대, 핀, 나사, 접시와 해부판이 있다. 그리고 삽입을 용이하게 하기위해 그러나 체온상태에서 충분히 견고하며, 치료동안 환자에게 좀더 편리한 이차 형상를 만들고 유연하게하는 조성물을 갖는 봉합선을 만드는데 사용할 수 있다. 다른 이용으로 카테테르의 영역이 있다. 중합체 조성물은 주입기, 강화물질, 방사영상 물질, 첨가제(excipients) 또는 특히 이식에 필요한 다른 물질에 결합될 수 있다.
2.비 의학적 응용
비의학적 타입의 응용의 예로 일회용 기저귀과 포장물질같은 처리가 문제가 되는 항목을 포함하고 있다.
3.분해가 억제되는 코팅
가수분해에 의해 분해할 수 있는 중합체는 물이 부피가 큰 중합체의 가수분해에 의해 쪼갤 수 있는 결합에 일시적으로 도달하지 못하도록 소수성의 SMP 코팅을 적용함으로써 선택적으로 보호될 수 있다.
Claims (28)
- (1) 하드 및 소프트 세그먼트, 또는(2) 공유적으로 또는 이온적으로 가교결합된 적어도 하나의 소프트 세그먼트 (soft segment), 또는(3) 중합체 혼합물을 포함하여 이루어지는 형상기억 중합체 조성물이되,이 형상기억 중합체는 분해성과 생체적합성이 있으며,상기 중합체 혼합물은 적어도 하나의 디(di-), 트리(tri-), 테트라(tetra-), 또는 다중 블록 공중합체와 적어도 하나의 상동- 또는 공중합체를 포함하고,상기 중합체의 원형상은 온도의 변화 또는 빛과 같은 다른 자극을 가함으로써 회복되도록 되어 있는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서, 상기 조성물은a) -40 내지 270℃ 사이의 Ttrans를 갖는 적어도 하나의 하드 세그먼트, 및b) 상기 하드 세그먼트의 것보다 적어도 10℃ 낮은 Ttrans를 갖고, 적어도 하나의 하드 세그먼트와 연결되어 있는 적어도 하나의 소프트 세그먼트를 포함하여 이루어지되,상기 하드 또는 소프트 세그먼트 중 적어도 하나는 생물분해성 부분을 포함하거나, 상기 하드 세그먼트 중 적어도 하나는 상기 소프트 세그먼트 중 적어도 하나에 분해성 결합으로 연결되어 있는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 2 항에 있어서,상기 하드 세그먼트의 Ttrans는 30 내지 150℃ 사이인 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 3 항에 있어서,상기 하드 세그먼트의 Ttrans는 30 내지 100℃ 사이인 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 2 항에 있어서,상기 소프트 세그먼트의 Ttrans는 하드 세그먼트의 것보다 적어도 20℃ 이하인 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 2 항에 있어서,상기 하드 및 소프트 세그먼트 중 적어도 하나는 열가소성(thermoplastic) 중합체인 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 2 항에 있어서,상기 하드 세그먼트는 고리부분(cyclic moities)을 포함하는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 2 항에 있어서,상기 하드 및 소프트 세그먼트의 중량비는 약 5:95 내지 95:5 사이인 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 형상기억 중합체는 그래프트(graft) 중합체, 선형 중합체(linear polymer), 덴드리머(dendrimer) 중합체로 이루어지는 군으로부터 선택되는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 폴리하이드록산, 폴리(에테르 에스테르), 폴리오쏘에스테르, 폴리(아미노산), 합성 폴리(아미노산), 폴리언하이드라이드, 폴리카보네이트, 폴리(하이드록시알카노에이트), 및 폴리(입실론-카프로락톤)으로 이루어진 군에서 선택되는 분해성 부분을 포함하는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 에스테르 그룹, 카보네이트 그룹, 아마이드 그룹, 언하이드라이드 그룹, 오쏘에스테르 그룹으로 이루어진 군에서 선택되는 생물분해성 결합을 포함하는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 완전히 생물분해될 수 있는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서, 상기 조성물은250 내지 -40℃ 사이의 Tm을 갖는 공유적으로 가교결합된 결정성 소프트 세그먼트 또는 250 내지 -60℃ 사이의 Ttrans를 갖는 공유적으로 가교결합된 소프트 세그먼트를 포함하는 분해성 열경화성수지 중합체를 포함하여 이루어진 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 13 항에 있어서,상기 분해성 열경화성수지 중합체는 200 내지 0℃ 사이의 Tm을 갖는 공유적으로 가교결합된 결정성 소프트 세그먼트 또는 200 내지 0℃ 사이의 Ttrans를 갖는 공유적으로 가교결합된 소프트 세그먼트를 포함하여 이루어진 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서, 상기 조성물은a) -40 내지 270℃ 사이의 Ttrans를 갖는 적어도 하나의 제1 세그먼트, 및b) 적어도 하나의 제1 세그먼트에 결합되어 있는 적어도 하나의 제2 세그먼트를 포함하여 이루어지고, 상기 제2 세그먼트가 용융점 또는 유리전이온도를 갖지 않도록 물리적 가교결합을 형성할 수 있는 충분한 힘의 이온상호작용을 포함하며,상기 제1 또는 제2 세그먼트 중 적어도 하나는 분해성 부분을 포함하거나, 또는 제1 세그먼트 중 적어도 하나가 생물분해성이 있는 결합을 통하여 상기 제2 세그먼트 중 적어도 하나와 결합되어 있는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 15 항에 있어서,상기 이온상호작용은 다중전해질 세그먼트(polyelectrolyte segments), 또는 다중전해질 세그먼트 및 이온, 또는 다중음이온(polyanions) 세그먼트 및 다중양이온(polycation), 또는 매우 조직화된 수소결합에 기초한 초분자(supramolecular) 효과를 포함하게 되는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 역전된 온도효과(inverse temperature effect)를 가지며, 이 경우 상기 조성물은 형상회복 온도 이하로 냉각될 때 그 형상를 회복하게 되는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 빛에 반응하여 형상이 변화되는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서,상기 중합체는 중합체 혼합물(blend)인 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 19 항에 있어서,상기 중합체 혼합물은 중합체의 물리적인 혼합물, 서로 다른 Ttrans를 갖는 하드 세그먼트와 서로 같은 Ttrans를 갖는 소프트 세그먼트를 포함하는 중합체 혼합물, 제1 공중합체 세그먼트 중 적어도 하나는 제2 공중합체의 세그먼트 중 적어도 하나와 섞일 수 있도록(miscible) 된 다중 블록 공중합체의 혼합물, 및 적어도 하나의 다중블록 공중합체와 적어도 하나의 상동(homo-)중합체 또는 공(co)중합체의 혼합물로로 이루어진 군으로부터 선택되는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항에 있어서, 상기 조성물은상기 형상기억 중합체의 분해를 변경시키는(altdering) 코팅을 포함하고 있는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항 내지 제 21 항 중 어느 항에 따른 분해성 형상기억 중합체 조성물을 포함하고 있는 것을 특징으로 하는 물품(article).
- 제 22 항에 있어서, 상기 물품에는치료, 진단, 및 예방제로 이루어진 군으로부터 선택되는 약제(agent)가 혼입(incorporating)되어 있는 것을 특징으로 하는 물품.
- 제 22 항에 있어서,상기 물품은 이식할 수 있고(implantable), 상기 생물분해성 형상기억 중합체는 생체적합성(biocompatibe)이 있는 것을 특징으로 하는 물품.
- 제 24 항에 있어서,상기 형상기억 중합체는 방향족 그룹을 포함하지 않는 것을 특징으로 하는 물품.
- 제 22 항에 있어서,상기 물품은 스텐츠(stents), 도뇨관(catheter), 보철물(prosthetic), 이식조직(grafts), 나사(screws), 핀(pins), 펌프(pumps), 망사(meshes)로 이루어진 군으로부터 선택되는 의료 장치인 것을 특징으로 하는 물품.
- 제 13 항 또는 제 14 항에 있어서,상기 중합체는 중합체 망상조직, 반투과성 망상조직, 상호투과성 망상조직 및 혼합된 상호투과성 망상조직으로 이루어진 군으로부터 선택되는 망상조직(network)을 포함하는 것을 특징으로 하는 분해성 형상기억 중합체 조성물.
- 제 1 항 내지 제 21 항 중 어느 한 항에 따른 형성기억 중합체 조성물로부터 제조된 물품을 포함하는, 봉합선, 치열교정(orthodontic) 물질, 뼈 나선부(bone screw), 손톱, 의치(plates), 도뇨관(catheters), 관(tubes), 피막(films), 스텐츠 (stents), 치열 교정기(orthopedic braces), 부목(splints), 깁스 테입, 콘택트 렌즈, 이식조직(implants), 열감지기, 조직공학에 사용되는 뼈대(scaffolds), 및 약 전달 기구로 이루어진 군으로부터 선택되는 물품의 제조방법.
