Description
Claims (1)
- 【特許請求の範囲】 1. ポリペプチドの露出された標的を保護することにより当該ポリペプチドの インビボでの機能を改良する方法であって a)有機化学合成により製造したリガンドを有するグループ特異的吸着剤との相互 作用によりポリペプチドを固定化し、 b)生体適合性ポリマーを活性化させ、 c)活性化した生体適合性ポリマーを固定化ポリペプチドの外側の部位とコンジュ ゲーションさせ、その後 d)吸着剤からコンジュゲートを溶出させる ことを特徴とする前記方法。 2. ポリペプチドが凝固活性を持ちまたは凝固機能を補佐するものであること を特徴とする、請求項1に記載の方法。 3. ポリペプチドが第VIII因子、フォン・ウィルブランド因子もしくはそれら の組合せまたは第IX因子の1つであることを特徴とする請求項2に記載の方法。 4. ポリペプチドが完全長の第VIII因子からの欠失誘導体であって凝固活性を 有することを特徴とする請求項3に記載の方法。 5. 第VIII因子の欠失誘導体が欠失誘導体リコンビナント第VIIISQ因子(r-VII ISQ)であることを特徴とする請求項4に記載の方法。 6. 第VIII因子の比活性が、固定化前は少なくとも総タンパク質1mg当たり2,0 00IUであることを特徴とする請求項3から5のいずれか1つに記載の方法。 7. 第VIII因子の比活性が固定化前は総タンパク質1mg当たり5,000から20,000 IUの範囲にあることを特徴とする請求項6に記載の方法。 8. 第VIII因子の活性が固定化前は少なくとも2,000IU/mlであることを特徴と する請求項3から7のいずれか1つの請求項に記載の方法。 9. 第VIII因子の活性が固定化前は5,000から50,000IU/mlの範囲にあることを 特徴とする請求項8に記載の方法。 10. 生体適合性ポリマーがモノアルキルでキャッピングされたポリアルキレン オキシドのホモポリマー、共重合体又はブロック共重合体からなる群より選ばれ たものであることを特徴とする請求項1から9のいずれか1つの請求項に記載の 方法。 11. モノアルキルでキャッピングされたポリアルキレンオキ シドがポリエチレングリコール・ホモポリマーとポリプロピレングリコール・ホ モポリマーからなる群より選ばれたものであることを特徴とする請求項10に記載 の方法。 12. モノアルキルでキャッピングされたポリアルキレンオキシドがモノメトキ シ・ポリエチレングリコール(mPEG)であることを特徴とする請求項11に記載の方 法。 13. リガンドが陰イオン若しくは陽イオン交換基、スルフヒドリル基、固定化 金属アフィニティークロマトグラフィー(IMAC)基または直鎖の若しくは分枝した 、飽和もしくは不飽和の炭化水素鎖若しくは芳香族基からなる群より選ばれたも のである請求項1から12のいずれかに記載の方法。 14. 陰イオン交換基が第4アミノメチル、第4アミノエチル及びトリメチルア ミノエチルまたはそれらの混合物からなる群より選ばれたものである請求項13に 記載の方法。 15. 活性化モノメトキシ・ポリエチレングリコール(mPEG)からなる群から選ば れた試薬によりポリペプチドのアミノ基に共有結合が形成されることを特徴とす る請求項1から14のいずれかに記載の方法。 16. 活性化されたモノアルキル-キャッピングポリアルキレ ンオキシドがmPEG-トレシレートとmPEGコハク酸混合無水物からなる群から選ば れたものであることを特徴とする請求項15に記載の方法。 17. 請求項1から16のいずれか1の請求項に記載の方法により得られた生体適 合性ポリマーとポリペプチドのコンジュゲート。 18. コンジュゲーション後に維持されるポリペプチドの活性が当該コンジュゲ ーション前の活性の少なくとも30%であることを特徴とする請求項17に記載のコ ンジュゲート。 19. コンジュゲーション後に維持されるポリペプチドの活性が当該コンジュゲ ーション前の活性の少なくとも50%であることを特徴とする請求項18に記載のコ ンジュゲート。 20. 請求項3から16のいずれか1つの請求項に記載の第VIII因子、フォン・ウ ィルブランド因子、それらの組合せまたは第IX因子と生体適合性ポリマーとのコ ンジュゲートであって、薬剤として使用されるコンジュゲート。 21. 請求項20に記載の欠失誘導体リコンビナント第VIIISQ因子(r-VIIISQ)と生 体適合性ポリマーのコンジュゲートであって、コンジュゲーション程度がモノア ルキルでキャッピングさ れたPEG/r-VIIISQ分子比が2から15の範囲にあるコンジュゲート。 22. 請求項3から16のいずれか1つの請求項に記載の第VIII因子、フォン・ウ ィルブランド因子、それらの組合せまたは第IX因子と生体適合性ポリマーとのコ ンジュゲートの血友病A、フォン・ウィルブランド病または血友病Bの治療用薬剤 の生産への使用。 23. 請求項22に記載の第VIII因子、フォン・ウィルブランド因子、それらの組 合せまたは第IX因子と生体適合性ポリマーとのコンジュゲートの皮下、筋肉内ま たは皮内投与への使用。 24. 請求項22に記載の第VIII因子、フォン・ウィルブランド因子、それらの組 合せまたは第IX因子と生体適合性ポリマーとのコンジュゲートの静脈内投与への 使用。 25. 請求項3から16のいずれか1つの請求項に記載の第VIII因子と生体適合性 ポリマーのコンジュゲートの皮下、筋肉内、皮内または静脈内投与による血友病 Aの治療法。
