Claims (1)
- 【特許請求の範囲】 1.以下の(a)〜(e)からなる群より選択される、澱粉顆粒結合型ならびに可溶 型として植物細胞内に存在する蛋白質をコードする核酸分子: (a)配列番号:2に示すアミノ酸配列を有する蛋白質をコードする核酸分子、 (b)配列番号:1に示すヌクレオチド配列のコード領域を含む核酸分子、 (c)(a)または(b)に記載の核酸分子とハイブリダイズする核酸分子、 (d)(a)または(b)に記載の核酸分子の配列と比較して、その配列が遺伝子 コードにより縮重している、核酸分子、および (e)(a)または(d)に記載の核酸分子の断片、誘導体、または対立遺伝子変 異体。 2.請求項1記載の核酸分子を含むベクター。 3.核酸分子が原核細胞および真核細胞において転写を確実にする調節要素と 結合している、請求項2記載のベクター。 4.請求項1記載の核酸分子または請求項2もしくは3記載のベクターによっ て遺伝的に修飾された、宿主細胞。 5.トランスジェニック植物細胞である、請求項4記載の宿主細胞。 6.請求項5記載の植物細胞を含む植物体。 7.請求項5記載の植物細胞または請求項6記載の植物体から得られる澱粉。 8.請求項4記載の宿主細胞を、蛋白質を発現可能な条件下で培養し、蛋白質 を細胞および/または培養培地から単離することを含む、澱粉顆粒結合型ならび に可溶型として植物細胞内に存在する蛋白質の産生方法。 9.請求項1記載の核酸分子によってコードされる、または請求項8記載の方 法によって得られる蛋白質。 10.請求項9記載の蛋白質を特異的に認識する抗体。 11.請求項1記載の核酸分子と特異的にハイブリダイズする、長さが少なくと も15ヌクレオチドの核酸分子。 12.請求項1記載のDNA分子の転写物と相補的なアンチセンスRNAをコードする DNA分子。 13.請求項1記載のDNA分子の転写物を特異的に開裂するリボザイム活性を有 す るRNAをコードするDNA分子。 14.植物細胞における発現時に、共抑制効果によって、請求項1記載の核酸分 子の発現を減少させるRNAをコードするDNA分子。 15.請求項12〜14のいずれか一項に記載のDNA分子を含むベクター。 16.DNA分子が、植物細胞における転写を確実にするDNA調節要素と結合してい る、請求項15記載のベクター。 17.請求項12〜14のいずれか一項に記載のDNA分子または請求項15もしくは16 記載のベクターを含む宿主細胞。 18.植物細胞において転写を確実にするDNA調節要素と共に、請求項12〜14の いずれか一項に記載のDNA分子を含む、トランスジェニック植物細胞。 19.澱粉生合成または修飾に関与する少なくともさらに一つの酵素の活性が非 形質転換植物と比較して減少している、請求項18記載のトランスジェニック植物 細胞。 20.分枝酵素の活性が減少している、請求項19記載のトランスジェニック植物 細胞。 21.澱粉顆粒結合型澱粉シンターゼアイソタイプI(GBSS I)の活性が減少し ている、請求項20記載のトランスジェニック植物細胞。 22.請求項18〜21のいずれか一項に記載の植物細胞を再生させることによって 得られるトランスジェニック植物体。 23.請求項18〜21のいずれか一項に記載の植物細胞または請求項22記載の植物 体から得られる澱粉。 24.請求項12〜14のいずれか一項に記載のDNA分子の転写によって得られるRNA 分子。 25.内因性の形で細胞において合成される請求項10記載の蛋白質の量が細胞内 で減少していることを特徴とする修飾澱粉を合成するトランスジェニック植物細 胞の作製方法。 26.細胞内における請求項10記載の蛋白質の量の減少がアンチセンス効果によ って引き起こされることを特徴とする、請求項25記載の方法。 27.細胞内における請求項10記載の蛋白質の量の減少がリボザイム効果によっ て引き起こされることを特徴とする、請求項25記載の方法。 28.細胞内における請求項10記載の蛋白質の量の減少が共抑制効果によって引 き起こされることを特徴とする、請求項25記載の方法。 29.澱粉生合成および/または修飾に関与する少なくともさらに一つの酵素の 酵素活性が減少している、請求項25〜28のいずれか一項に記載の方法。 30.酵素が分枝酵素である、請求項29記載の方法。 31.酵素が澱粉顆粒結合型澱粉シンターゼのアイソタイプI(GBSS I)である 、請求項29記載の方法。 32.請求項25〜31のいずれか一項に記載の方法によって得られる植物細胞。 33.請求項32記載の植物細胞を再生させることによって得られるトランスジェ ニック植物体。 34.請求項32記載の植物細胞または請求項33記載の植物体から得られる澱粉。 35.ジャガイモ由来であることを特徴とする、請求項34記載の澱粉。 36.請求項5記載の植物細胞を含む請求項6記載の植物体の繁殖材料。 37.請求項18〜21のいずれか一項に記載の植物細胞または請求項32記載の植物 細胞を含む、請求項22または32記載の植物体の繁殖材料。 38.ジャガイモ植物である、請求項22または33記載のトランスジェニック植物 体。 39.請求項38記載のジャガイモ植物体の塊茎。 40.野生型植物の塊茎と比較して低温甘味化の減少を示す、請求項39記載の塊 茎。
