Claims (1)
- 【特許請求の範囲】 1.力を加えるとつぶれる壁、直径が約0.25ミクロンから約6.0ミクロ ンの孔を有し力が加えられたときに外側へ凸状構成に突出するように十分な可と う性を有する可とう性の多孔性膜で少なくとも部分的に覆われた容器の開口部、 及び膜の孔を通過するときに患者の肺に吸入されうる粒子のエアロゾルを形成で きることを特徴とする薬学的に活性な薬物と担体とを含む製剤を備える、肺への 薬物のエアロゾル化輸送に使用するための使い捨て容器。 2.開口部が、開口部自体から破壊可能なシールへ至る開放流路を形成し、該 破壊可能なシールを越えた領域が、可とう性の多孔性膜で覆われた領域である、 請求項1記載の容器。 3.孔が、直径0.25ミクロンから6.0ミクロンの小さな端部の開口部と 小さな端部の直径の2倍から10倍の大きな端部の開口部とを含む断面構成を有 する、請求項1記載の容器。 4.相互接続された複数の使い捨て容器を備える、請求項1に記載の使い捨て パッケージ。 5.流路を通じて直径が約0.25ミクロンから約6.0ミクロンの範囲の孔 を有する可とう性の使い捨て多孔性膜の表面の上方に空気を引き込むこと、及び 製剤から流路を通じて引き込まれた空気の方向へ凸状構成に膜を外側に突出させ るために、薬学的に活性な薬物と担体とを含む製剤を膜の孔を通過させることを 含む、エアロゾルを生成する方法。 6.可とう性膜が、流路内に生成された空気流の境界層を越えて外側に突出し 、空気が、ほぼ直線的な流路を介して、突出した膜の上方から流路の端部へと引 き込まれる、請求項5記載の方法。 7.流路内の空気流を測定し吸気流量及び吸気体積を決定すること、並びに吸 気流量及び吸気体積のリアルタイム値に基づいて製剤を膜の孔を通過させるため の開始点を判定することをさらに含む、請求項5記載の方法。 8.約0.10リットル/秒から約2.0リットル/秒の範囲の吸気流量及び 約0.15リットルから約1.5リットルの範囲の吸気体積で行われる通過がほ ぼ同じ吸気流量及び吸気体積で反復的に開始されるように、吸入段階、測定段階 、判定段階、及び輸送段階を繰り返すことをさらに含む、請求項7記載の方法。 9.測定が機械的に行われる、請求項7記載の方法。 10.測定が電子的に行われる、請求項7記載の方法。 11.エアロゾル化粒子に接触させた空気を積極的に加熱することによって該 粒子にエネルギーを加えること、及び患者の肺に粒子を吸入させることをさらに 含む、請求項5記載の方法。 12.粒子が形成されたときに粒子中の担体の50%以上が粒子が患者に到達 する前に蒸発するような量だけエネルギーが加えられ、患者が吸入を行う前に加 熱された加熱材料内を空気を通過させることにより空気を積極的に加熱すること によって該エネルギーが加えられる、請求項11に記載の方法。 13.粒子が患者に到達する前に実質的に乾燥する程度に担体が蒸発するよう な量だけエネルギーが加えられ、該担体が水、エタノール、及びそれらの混合物 からなる群より選択され、さらに多孔性膜が約0.1mm2から約1cm2の領域 に10個から10,000個の孔を含む、請求項11記載の方法。 14.乾燥空気を得るために空気から水蒸気を吸収するデシケータを通じて流 路内に空気を吸入すること、エアロゾル化粒子が形成され流路内に引き込まれる ように薬学的に活性な薬物と水担体とを含む製剤を多孔性膜の孔を通過させるこ と、並びにエアロゾル化粒子及び乾燥空気を患者の肺に引き込むために吸入を続 けることを含む、薬物輸送の方法。 15.デシケータが、P2O5、Mg(ClO4)、KOH、H2SO4、NaO H、CaO、CaCl2、ZnCl2、及びCaSO4からなる群より選択される ものである、請求項14に記載の方法。 16.吸入中の患者に至る流路に空気を吸入すること、エアロゾル化粒子が形 成され流路内に引き込まれるように薬学的に活性な薬物と水担体とを含む製剤を 多孔性膜の孔を通過させることを含む薬物輸送の方法であって、輸送に関するパ ラメータが、 (a)毎秒0.1リットルから毎秒2.0リットルの範囲内の吸気流量、 (b)0.15リットルから2.0リットルの範囲内の吸気体積、 (c)孔の直径が0.5ミクロンから6ミクロンの範囲内の多孔性膜を使用す ること、 (d)薬物が0.01%から12.5%の範囲で担体中に存在する製剤を使用 すること、 (e)製剤10μl当たり20ジュールから100ジュールの量のエネルギー を加えることにより吸入された空気を加熱することによって、製剤に熱を加える こと、 (f)575キロヘルツから15,000キロヘルツの範囲内の周波数で振動 を生じさせること、 (g)100psiから500psiの範囲の圧力を加えることによって製剤 を多孔性膜を通過させることと、 (h)吸入された空気に周囲の湿度の測定に基づきエネルギーを加えること、 及び (i)最初の輸送に関して(a)〜(h)のいくつかを考慮し、その後、輸送 が再現可能になるように(a)〜(h)のいくつかに関して実質的に同じ点で製 剤を多孔性膜を通過させること、からなる群より選択される、方法。 17.流路を通じて空気を引き込むこと、液体担体と薬学的に活性な薬物とを 含む製剤の粒子をエアロゾル化するように、液体製剤を流路内の空気中へと多孔 性膜の孔を通過させること、及び液体担体を蒸発させ直径が0.5ミクロンから 12.0ミクロンの範囲の粒子を得るのに十分な程度に空気を加熱することを含 む、直径が0.5ミクロンから12.0ミクロンのエアロゾル化量の粒子を供給 する方法。 18.加熱が、流路内に引き込まれた空気の一部を加熱要素へ送ることによっ て行われる、請求項17記載の方法。 19.加熱が、流路内に引き込まれた空気の一部を予め加熱された要素を通じ て引き込むことによって行われ、該要素が、流路を通じて引き込まれた空気の温 度を一定に維持するように加熱されている、請求項17記載の方法。 20.空気が流路を通じて引き込まれる間だけ加熱要素が活動化される、請求 項18記載の方法。 21.加熱要素が、空気が流路を通じて引き込まれる50ミリ秒から500ミ リ秒前に活動化され、製剤が孔を通過した後に非活動化される、請求項18記載 の方法。 22.加熱が、液体担体を所定の量だけ蒸発させ、かつ周囲の温度及び湿度に はかかわらず一定の粒径が得られるように、周囲の温度及び湿度に基づいて変化 する、請求項17記載の方法。 23.周囲の湿度が湿度計によって測定される、請求項22記載の方法。 24.内部に空気を吸入することができる第1の開口部と、患者が空気を引き 込むための第2の開口部とを有する流路、作動時に製剤に物理的力を加える機構 、及び流路内に吸入された空気にエネルギーを加える空気加熱装置を備える、総 重量が1キログラム以下の手持ち内蔵式装置である、薬物輸送装置。 25.製剤に物理的力を加える機構が、ピストンと振動装置とからなる群より 選択される、請求項24記載の薬物輸送装置。 26.空気加熱装置によって加えるべきエネルギーの量を求めるために使用さ れる情報を供給する、周囲の湿度を測定するための湿度計をさらに備える、請求 項24記載の薬物輸送装置。 27.力を加えるとつぶれる壁、容器からの出口経路を覆い、直径が約0.2 5ミクロンから約6.0ミクロンの範囲の孔を有し、剛性であり、出口経路から 上向きに突出する凸状構成を有する多孔性膜、及び膜の孔を通過するときに患者 の肺に吸入されうる粒子のエアロゾルを形成できることを特徴とする薬学的に活 性な薬物と担体とを含む製剤を備える、肺に薬物をエアロゾル化輸送する際に使 用するための使い捨て容器。 28.出口経路が、破壊可能なシールに至る開放流路に至り、該破壊可能なシ ールを越えた領域が、多孔性膜で覆われた領域である、請求項27記載の容器。 29.膜の厚さが5ミクロンから200ミクロンである、請求項27記載の容 器。 30.相互接続された複数の使い捨て容器を備える、請求項27に記載の使い 捨てパッケージ。
