Claims (1)
- 【特許請求の範囲】 1.フィターゼ活性を有するペプチドまたはたんぱく質をコードすることを特徴 とする精製単離したDNA配列。 2.請求の範囲1記載の精製単離したDNA配列で、さらに該配列が微生物に宙 来することを特徴とするDNA配列。 3.請求の範囲1記載の精製単離したDNA配列で、さらに該配列が菌類に由来 することを特徴とするDNA配列。 4.請求の範囲1記載の精製単離したDNA配列で、さらに該配列がアスペルギ ラス由来であることを特徴とするDNA配列。 5.請求の範囲1記載の精製単離したDNA配列で、さらに該配列がアスペルギ ラスフィカムまたはアスペルギラスニガー由来のものであることを特徴とするD NA配列。 6.請求の範囲1記載の精製単離したDNA配列で、さらに該配列が以下の特性 : a)アスペルギラス宿主中で発現した場合SDS−PAGEで85kDaの単一 バンドを与える、 b)脱グリコシル化後、約48〜56.5kDaの見かけの分子量を有する、 c)約100U/mgたんぱく質の比活性を有する、を示すフィターゼをコード することを特徴とするDNA配列。 7.精製単離したDNA配列で、以下に示す特性:a)第6図に示したDNA配 列に宙来するオリゴヌクレオチドプローブにハイブリダイズする、 b)第8図に示したcDNA配列に由来するオリゴヌクレオチドプローブにハイ ブリダイズする、 の少なくとも1つを示すことを特徴とするDNA配列。 8.請求の範囲1乃至7のいずれか1項記載のDNA配列が適当な発現ホスト中 フィターゼ活性を有するたんぱく質またはペプチドを発現させ得る調節領域に機 能的に結合していることを特徴とする発現構築物。 9.請求の範囲8記載の発現構築物で、さらに該調節領域がフィターゼ活性を有 する発現たんぱく質またはペプチドを分泌させる分泌リーダー配列を含むことを 特徴とする発現構築物。 10.請求の範囲9記載の発現構築物で、さらにフィターゼ活性を有するたんぱ く質またはペプチドを発現させるのにAGプロモーターを用いることを特徴とす る発現構築物。 11.請求の範囲10記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに同種のフィターゼリーダー配列を用 いることを特徴とする発現構築物。 12.請求の範囲10記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに18個のアミノ酸からなるAGリー ダー配列を用いることを特徴とする発現構築物。 13.請求の範囲10記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに24個のアミノ酸からなるAGリー ダー配列を用いることを特徴とする発現構築物。 14.請求の範囲9記載の発現構築物でフィターゼ活性を有するたんぱく質また はペプチドを発現させるために同種のフィターゼプロモーターを用いることを特 徴とする発現構築物。 15.請求の範囲14記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに同種のフィターゼリーダー配列を用 いることを特徴とする発現構築物。 16.請求の範囲14記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに18個のアミノ酸からなるAGリー ダー配列を用いることを特徴とする発現構築物。 17.請求の範囲14記載の発現構築物で、さらにフィターゼ活性を有する発現 たんぱく質またはペプチドを分泌させるのに24個のアミノ酸からなるAGリー ダー配列を用いることを特徴とする発現構築物。 18.請求の範囲8乃至17のいずれか1項記載の発現構築物を含むことを特徴 とする宿主細胞をトランスホームし得るベクター。 19.請求の範囲18記載のベクターで、さらに該ベクターがプラスミドである ことを特徴とするブクター。 20.請求の範囲18記載のベクターで、さらに該ベクターがpAF28−1、 pAF2−2S、pAF2−2、pAF2−3、pAF2−4、pAF2−6、 pAF2−7、p18FYT3、p24FYT3およびpFYT3からなる群か ら選ばれるプラスミドであることを特徴とするベクター。 21.請求の範囲18乃至20のいずれか1項記載のベクターでトランスホーム したことを特徴とする形質転換宿主細胞。 22.細菌、イースト類、カビ類からなる群から選ばれる請求の範囲21記載の 形質転換宿主細胞。 23.アスペルギラス、トリコデルマ、ペニシリウム、ムコール、バチルス、ク ルイベロミセスおよびサッカスミセスからなる群から選ばれる請求の範囲22記 載の形質転換宿主細胞。 24.アスペルギラスニガー、アスペルギラスフィカス、アスペルギラスアワモ リ、アスペルギラオリゼ、トリコデルマリーセイ、ムコールミエヘイ、クルイベ ロミセスラクチス、サッカロミセスセレジシエ、枯草菌およびバチリスリチェホ ホルミスからなる群から選ばれる請求の範囲22記載の形質転換宿主細胞。 25.フィターゼ活性を有するペプチドまたはたんぱく質の産生方法で、該フィ ターゼ活性を有するペプチドまたはたんぱく質の産生に適した条件下、請求の範 囲21乃至24のいずれか1項記載の形質転換宿主細胞を培養することを特徴と する方法。 26.請求の範囲25記載の方法で生産されるフィターゼ活性を有するペプチド またはたんぱく質。 25.以下に示す特性: a)アスペルギラス宿主において発現した場合にSDS−PAGEで85kDa の単一バンドを与える、 b)脱グリコシル化後約48〜56.5kDaの範囲の見かけの分子量を有する 、 c)約100U/mgたんぱく質の比活性を有する、を示すことを特徴とするフ ィターゼ。 28.請求の範囲26および27のいずれか1項記載のフィターゼ活性を有する ペプチドまたはたんぱく質を含むことを特徴とする動物用飼料。 29.フィチン酸塩をイノシトールおよび無機リン酸へ変換するための請求の範 囲26および27のいずれか1項記載のフィターゼ活性を有するペプチドまたは たんぱく質の使用。 30.請求の範囲26および27のいずれか1項記載のフィターゼを補った飼料 原料を含む飼料を動物に与えることを特徴とする動物の成長の促進方法。 31.飼料原料中に含まれるフィチン酸塩をイノシトールおよび無機リン酸に変 換させるのに有効な量の請求の範囲26および27のいずれか1項記載のフィタ ーゼを捕った飼料原料を含む飼料を動物に与えることを特徴とする動物排他物中 のフィチン酸塩レベルの減少方法。
