Claims (12)
- 1.以下の要素(a)〜(e): (a)安定化量の基質の存在下90℃で測定して至適pH4.0〜6.0、 (b)安定化量の基質の存在下pH5.5で測定して至適温度80〜120℃、 (c)本質的にカルシウムイオンから独立している、(d)安定化量の基質の存 在下で測定して90℃で1時間後100%、更に100℃1時間後少なくとも8 0%の残留活性、 (e)ピロコッカス(Pyrococcus)菌株の培養によって得ることので きる、 ことを特徴とするa−アミラーゼ。
- 2.以下の要素(a)〜(e): (a)基質の非存在下90℃で測定して至適pH5.2〜5.8、 (b)基質の非存在下pH5.5で測定して至適温度90〜105℃、 (c)本質的にカルシウムイオンから独立している、(d)安定化量の基質の存 在下で測定して100℃1時間後約70%の残留活性、 (e)ピロコッカス(Pyrococcus)菌株の培養によって得ることので きる、 ことを特徴とするa−アミラーゼ。
- 3.P.ポエセイ(woesei)又はP.フリオサス(furiosus)の 菌株を培養することにより得ることのできる、請求の範囲第1項又は第2項記載 のa−アミラーゼ。
- 4.P.ポエセイ(woesei),DSMNo.3773、又はP.フリオサ ス(furiosus),DSMNo.3638の菌株を培養することによって 得ることのできる請求の範囲第3項に記載のa−アミラーゼ。
- 5.ピロコッカス(Pyrococcus)のa−アミーゼ生産菌株を、炭素お よび窒素源および無機塩を含有する適当な栄養培地中で培養し、次いで目的酵素 を回収することを含んでなる請求の範囲第1〜5項のいずれかに記載のa−アミ ラーゼの調製方法。
- 6.P.ポエセイ(woesei)又はP.フリオサス(furiosus)を 培養する、請求の範囲第5項に記載の方法。
- 7.P.ポエセイ(woesei),DSMNo.3773、又はP.フリオサ ス(furiosus),DSMNo.3638を培養する、請求の範囲第5項 に記載の方法。
- 8.請求の範囲第1〜5項のいずれかに記載のa−アミラーゼの存在下、水性デ ンプンスラリーを酵素的液化に委ねることを含んでなるデンプン液化方法。
- 9.本質的にカルシウム塩をデンプンスラリーに添加することなくプロセスを行 う、請求の範囲第8項に記載のデンプン液化方法。
- 10.前記プロセスを、100〜140℃の範囲内の温度で120分までジェッ トクッキングにより行い、次いで所望により保持すべき温度を90〜100℃の 範囲の温度に約30〜120分減少させ、しかる後このように液化したデンプン は老化に対し安定であり、pHはプロセス中約4.0〜5.5に保持されている 、請求の範囲第8項又は第9項に記載のデンプン液化方法。
- 11.液化デンプンをしかる後、実質的に中間pH調節することなくグルコアミ ラーゼの存在下、酵素的糖化に委ねる請求の範囲第8〜10項のいずれかに記載 のデンプン液化方法。
- 12.更に酵母を用いたエタノール発酵を糖化と同時に行うか又はその後引続い て行う糖化を行う請求の範囲第11項記載のデンプン液化方法。
