Description
Pyrococcus woesel DSM 3773 wächst in einem Temperaturbereich von 80°C
bis 106°C, sowie einem pH-Bereich von 7,5 bis 4,5. Unter einer
kontinuierlichen H2/CO2-Atmosphäre mit Sacchariden wie z. B. Stärke,
Amylose, Amylopektin, Glykogen, verzweigten und linearen
Oligosacchariden (z. B. Maltotetraose, Maltotriose, Maltose und Glucose) und
Maltodextrin oder deren Gemische als C-Quelle, elementarem Schwefel als
Elektronenaktzeptor und 3% NaCl werden 250 U/l α-Amylase ins Medium
ausgeschieden. Bei nicht-kontinuierlicher Begasung können unter den
gleichen Bedingungen 40 U/l α-Amylase extrazellulär nachgewiesen
werden. Die Enzymsynthese des Organismus erfolgt zu allen Zeiten des
Wachstums, sie beginnt bereits in der frühen logarithmischen Phase und
erreicht in der stationären Phase ihr Maximum.
Als α-Amylase wurde ein Enzym definiert, daß in der Lage ist,
Glucosepolymere an den α-1,4-Bindungen zu spalten und daraus
Glucoseoligomere freizusetzen. DSM = Deutsche Sammlung von Mikro
organismen.
Das Enzym kann auf einfache Weise aus dem zellfreien Kulturüberstand
gewonnen werden. Es bindet an native und lösliche Stärke, die dann aus dem
Kulturüberstand durch Kälte oder Zugabe von organischen Lösungsmitteln
wie z. B. Ethanol ausgefällt werden kann. Dabei verliert das Enzym nicht an
Aktivität.
Die α-Amylase ist unter aeroben Bedingungen in Abwesenheit von
Metallionen in einem Temperaturbereich von 40°C bis 140°C, insbesondere
zwischen 60°C und 120°C, vorzugsweise zwischen 80°C und 110°C und
besonders bevorzugt zwischen 90°C und 105°C aktiv (siehe Abb. 1).
Die α-Amylase ist aktiv in einem pH-Bereich von 3,5 bis 8, insbesondere
zwischen pH 4,5 und 6,5, vorzugsweise zwischen pH 5 und 6 und besonders
bevorzugt bei einem pH-Wert von 5,2 bis 5,8 (siehe Abb. 2).
Das Enzym ist bei 70°C bis 100°C in Gegenwart von Polysacchariden wie
z. B. Stärke, Glykogen, verzweigte Glucosepolymere, Amylose, Amylopektin,
Maltodexrin und deren Gemische für mehr als 12 Stunden unter aeroben
Bedingungen in Abwesenheit von Metallionen stabil. Nach 60minütiger
Inkubation sind unter den gleichen Bedingungen bei 100°C 100%, bei 110°C
und 120°C 70%, 130°C 10% der Anfangsaktivität zu messen (siehe Abb. 3). Der
Verlauf der unter den gleichen Bedingungen gemessenen Aktivität über einen
Zeitraum von 12 h ist der Abb. 3 zu entnehmen.
Die Metallionenabhängigkeit wurde mit metallionenfreiem, vorgereinigtem
Enzym untersucht. Die unten aufgeführten Metalle wurden in
Konzentrationen bis zu 5 mM dem Enzym zugesetzt.
Molybdän-, Calcium- und Magnesiumionen beeinflussen die Enzymaktivität
nicht. Eine leichte Hemmung der Enzymaktivität ist in Gegenwart von
Cobalt-, Nickel- und Eisenionen festzustellen, Zink- und Kupferionen
hemmen das Enzym vollständig. EDTA und Jodessigsäure haben ebenfalls
keinen Einfluß auf die Enzymaktivität.
Native Stärke, lösliche Stärke, verzweigte Oligosaccharide und Amylose
werden von der α-Amylase hydrolysiert. Hauptprodukte der Hydrolyse
dieser Substrate sind größere Glucoseoligomere mit einem
Polymerisationsgrad von 6 und größer. Maltose, Maltotriose, Maltotetraose
und Maltopentaose werden nur in geringem Umfang freigesetzt, Glucose
entsteht nicht. Pullulan, Dextran und Maltose werden nicht vom Enzym
angegriffen. Die Fähigkeit dieses Enzyms native und lösliche Stärke bei
Temperaturen selbst über 100°C zu einem glucosefreien Saccharidgemisch
abzubauen und seine einfache Gewinnung machen es für einen industriellen
Einsatz z. B. in der Stärkeverzuckerung außerordentlich interessant.
Eine extrem thermostabile metallionenunabhägige α-Amylase aus einem
thermophilen Organismus ist in der Lage in einem Temperaturbereich von
40°C bis 140°C und einem pH-Bereich von 3,5 bis 8,0 Glucosepolymere wie
z. B. Stärke zu hydrolysieren. Das Enzym besitzt eine maximale Aktivität
bei pH 5,5 und einer Temperatur von 100°C. Die Produkte der
Stärkehydrolyse sind längerkettige Glucoseoligomere. Das extrazelluläre
Enzym kann auf einfache Weise durch Ausfällung aus dem zellfreien
Kulturüberstand gewonnen werden. Die Eigenschaften des Enzyms machen
es für eine industrielle Anwendung außerordentlich interessant.

- alpha-Amylase, erhältlich aus Kulturmedien von Pyrococcus woesei DSM 3773 und mit den folgenden Merkmalen:
- - Aktivitätsoptimum bei insbesondere 90 bis 105°C und
- - insbesondere pH 5,2 bis 5,8.
