Description
4重量%のキトサンおよび2重量%の硫酸バリウムを含む酸性水溶液(12.5%酢酸)を調製した。溶液を球形の鋳型に配置し、次に、鋳型内で、−30℃で16時間凍結させた。凍結した本体を鋳型から取り出し、3日間凍結乾燥させて水を除去した。凍結乾燥した本体を、10%水酸化アンモニウム溶液中で1時間中和させ、次に、水酸化アンモニウムがなくなるまで脱イオン水によってすすいだ。本体を16時間真空乾燥させた。本体は胸部組織に近い海綿状のコンシステンシーを有し、乳腺腫瘤摘出の空洞の周囲の胸部組織を支持するのに十分な圧縮強度を有していた。それは、67%のキトサンおよび33%の硫酸バリウムを含んでいた。表面の多孔性のSEM写真(30X)を図6に、中心部の多孔性のSEM写真(30X)を図7に示す。インプラントは胸部組織とほぼ等しい海綿状のコンシステンシーを有していた。キトサンの量を増加させることにより、インプラントをより硬くすることができた。
一定量のアルギン酸ナトリウム(0.5〜4%(重量))を水に溶解させ、ペースト状の粘性の液体もしくはゲルを形成し、空気または他の生体適合性の気体を混合物に導入する。混合物を所望のインプラント形状の鋳型に配置し、次に所望の形状で凍結乾燥または空気乾燥させる。アルギン酸ナトリウムからなる成形インプラント構造を塩化カルシウム溶液(0.5〜約4%(重量))に入れ、ここで、アルギン酸ナトリウムの少なくとも一部を、アルギン酸カルシウムに変え、これが析出する。析出した多孔質構造のインプラントを、組織を除去した体の空洞に導入する。インプラントは、十分な期間、当該部位に残り、体の空洞内の組織の内方成長(in-growth)を促進する足場として機能する。デンプン、例えば微細に分割された微粒子形態のトウモロコシデンプンを、アルギン酸ナトリウム−水の混合物に組み込み、アルギン酸カルシウムが形成されたときに、それをデンプン粒子の周囲に析出させて、アルギン酸ナトリウムがアルギン酸カルシウムに変換する間の収縮を最小化することができる。デンプンは、体液の存在下、体の空洞内で迅速に分解する。インプラントの表面上のアルギネートは、分解して、取り込まれているデンプン粒子を、進展性の多孔性を提供する分解に対して開放する。デンプン対アルギネートの重量比は約15:1〜約1:1である。
この例は、30gの塩(NaCl)顆粒を約30mlの3%(重量)アルギン酸ナトリウム水溶液と混合したことを除いて、例2と同様である。次いで、球形の鋳型に配置した溶液を4時間凍結させた。凍結したインプラントを鋳型から取り出し、2%(重量)の塩化カルシウム溶液中に配置し、これにより、アルギン酸カルシウムのゲルを形成し、取り込まれた塩の顆粒の少なくともいくらかを溶解させて多孔質構造を形成した。インプラントは、胸部組織とほぼ等しい海綿状のコンシステンシーを有していた。溶液中のアルギン酸ナトリウムの量を増加させるか、塩の量を低下させるか、または塩の顆粒のサイズを低下させることにより、インプラントをより硬くすることができた。

体の空洞(body cavity)のためのインプラントであって、当該インプラントの有意な生体吸収の前における組織の内方成長(in-growth)を確実にするのに十分な多孔性を有する生体吸収性材料から形成される本体と、当該インプラントの外縁の画像化を容易にする放射線不透過性造影剤と、当該インプラントの外縁から内側に離れており、当該インプラントと放射線源との間の相対的な方位合わせを容易にする内部方位合わせマーカーとを含むインプラント。

Claims (6)
- 組織が除去された空洞(cavity)に適したインプラントの形成方法であって、a.可溶性のアルギネートと放射線不透過性造影剤を、水とともに混合し;b.ガスまたは孔形成剤をアルギネート−水混合物中に組み込み;
c.アルギネート−水混合物中を、ガスまたは孔形成剤とともに鋳型中に移し、当該混合物を所望の形状の固体物本体に成形し;
d.前記本体から水を除去し;
e.可溶性のアルギネートの少なくとも一部を、可溶性がより低いアルギネートに変える、
上記工程を含み、
更に当該インプラントの外縁から内側に離れている方位合わせマーカーを当該本体に組み込むことを含む方法。 - 複数の放射線不透過性要素(element)を、平面内にある(lying in a plane)方位で組み込むことを含む、請求項1に記載の方法。
- 凍結乾燥または空気乾燥により当該混合物から水を除去する、請求項1に記載の方法。
- 可溶性アルギネートがアルギン酸ナトリウムである、請求項1に記載の方法。
- 可溶性がより低いアルギネートがアルギン酸カルシウムである、請求項1に記載の方法。
- 空洞の中に収まり、多孔性インプラントの周囲の空洞をライニングしている組織に沿うような、多孔性インプラントの大きさ付けとおよび形状付けを含む、請求項1に記載の方法。
