本発明による方法は、基本的な構成に於いて、FCS、FIDA等が実行可能な共焦点顕微鏡の光学系と光検出器とを組み合わせてなる光分析装置により実現可能である。ただし、本発明の方法に於いては、特に、複数の互いに異なる波長帯域の光の測定が実行されるので、図1(A)に模式的に例示されている如き、複数の互いに異なる波長帯域の光の測定が可能な装置が用いられる。
「発明の概要」の欄に記載されている如く、本発明の方法は、端的に述べれば、少なくとも二つの互いに発光波長の異なる発光部位を有する発光粒子に於いて、かかる少なくとも二つの発光部位からの光の強度比が顕微鏡の光検出領域内での発光粒子の位置に依らないという原理に基づき、走査分子計数法に於いて、少なくとも二つの発光部位からの光の強度比により、発光粒子の特徴付けをして、発光粒子の同定を行うというものである。換言すれば、本発明の一つの実施形態は、走査分子計数法を改良するものであるということができる。以下、走査分子計数法及び本発明の方法の原理について説明する。
FCS、FIDA等の分光分析技術は、従前の生化学的な分析技術に比して、必要な試料量が極めて少なく、且つ、迅速に検査が実行できる点で優れている。しかしながら、FCS、FIDA等の分光分析技術では、原理的に、発光粒子の濃度や特性は、蛍光強度のゆらぎに基づいて算定されるので、精度のよい測定結果を得るためには、試料溶液中の発光粒子の濃度又は数密度が、図9(A)に模式的に描かれているように、蛍光強度の計測中に常に一個程度の発光粒子が光検出領域CV内に存在するレベルであり、同図の右側に示されている如く、計測時間中に常に有意な光強度(フォトンカウント)が検出されることが要求される。もし発光粒子の濃度又は数密度がそれよりも低い場合、例えば、図9(B)に描かれているように、発光粒子がたまにしか光検出領域CV内へ進入しないレベルである場合には、同図の右側に例示されている如く、有意な光強度の信号(フォトンカウント)が、計測時間の一部にしか現れないこととなり、精度のよい光強度のゆらぎの算定が困難となる。また、計測中に常に一個程度の発光粒子が光検出領域内に存在するレベルよりも発光粒子の濃度が大幅に低い場合には、光強度のゆらぎの演算に於いて、バックグラウンドの影響を受けやすく、演算に十分な量の有意な光強度データを得るために計測時間が長くなる。
上記の走査分子計数法が実行される実際の実験系に於いて、光検出領域に於ける励起光の強度は、一様ではなく、通常、光検出領域の中心をピークとして領域の縁に行くほど低減し、また、光検出領域からピンホールを通過して光検出器に到達する間に於いても、光検出領域の全域からの光の強度が一様に光検出器に到達するとは限らない。即ち、或る発光粒子の発する光(又は光検出器で検出される光)の強度は、光検出領域に於ける発光粒子の位置によって変化することとなる(そのため、走査分子計数法では、典型的には、検出されたパルス状の信号の強度が発光粒子からの光として想定される有限の範囲内にあるときに、その信号が発光粒子からの光の信号であると判定される。)。従って、例えば、二種類の発光粒子を観測する場合、それらの発光粒子の量子収率又は発光能(同じ条件で発光される光の強度)が互いに異なっていても、両者をそれらの光の強度の絶対値から区別することは困難である。
図1(A)に例示の光分析装置1を用いた本発明の方法に従った走査分子計数法の実施形態に於いては、具体的には、(1)発光粒子を含む試料溶液の調製過程、(2)試料溶液の光強度の測定処理過程、及び(3)測定された光強度の分析処理過程とが実行される。図4は、フローチャートの形式にて表した本実施形態に於ける処理過程を示している。
本発明の方法に於いて観測対象となる粒子は、溶解された分子等の、試料溶液中にて分散し溶液中にてランダムに運動する粒子であれば、任意のものであってよく、例えば、タンパク質、ペプチド、核酸、脂質、糖鎖、アミノ酸若しくはこれらの凝集体などの生体分子、ウイルス、細胞、或いは、金属コロイド、その他の非生物学的粒子などであってよい(試料溶液は、典型的には水溶液であるが、これに限定されず、有機溶媒その他の任意の液体であってよい。)。また、観測対象となる粒子は、それ自体が発光する粒子であってもよく、或いは、発光標識(蛍光分子、りん光分子、化学・生物発光分子)が任意の態様にて付加された粒子であってよい。
本実施形態の走査分子計数法による光分析に於ける光強度の測定処理過程では、ミラー偏向器17を駆動して、試料溶液内での光検出領域の位置の移動(試料溶液内の走査)を行いながら、複数の波長帯域にて別々に、即ち、第一及び第二の発光部位(又は共通発光部位及び固有発光部位)の発光波長について別々に、且つ、同時に光強度の測定が為される(図4−ステップ100)。操作処理に於いて、典型的には、マイクロプレート9のウェル10に試料溶液を注入して顕微鏡のステージ上に載置した後、使用者がコンピュータ18に対して、測定の開始の指示を入力すると、コンピュータ18は、記憶装置(図示せず)に記憶されたプログラム(試料溶液内に於いて光検出領域の位置を移動するべく光路を変更する手順と、光検出領域の位置の移動中に光検出領域からの光を検出する手順)に従って、試料溶液内の光検出領域に於ける励起光の照射及び光強度の計測を開始する。計測が開始されると、まず、コンピュータ18のプログラムに従った処理動作の制御下、光源2から、試料溶液中の発光粒子の励起波長の光が出射される。特に、本実施形態に於いては、複数の波長帯域の光を検出するので、検出されるべき複数の波長帯域の光が発光粒子から放出されるよう光源2から出射される励起光の波長の選択が為される。なお、発光粒子上の各発光部位の励起光波長が互いに異なる場合には、複数の波長帯域のレーザー光が同時に出射されることとなる。一方、ミラー偏向器17は、コンピュータ18のプログラムに従った処理動作の制御下、ミラー7(ガルバノミラー)を駆動して、ウェル10内に於いて光検出領域の位置の移動を実行し、これと同時に光検出器16は、逐次的に検出された光を電気信号に変換してコンピュータ18へ送信し、コンピュータ18では、任意の態様にて、送信された信号から時系列の光強度データを生成して保存する。特に、本実施形態に於いては、複数の光検出器16の各々が、互いに異なる波長帯域の光を検出し、検出された互いに異なる波長帯域毎に時系列光強度データが生成される。また、典型的には、光検出器16は、一光子の到来を検出できる超高感度光検出器であるので、光の検出は、所定時間に亘って、逐次的に、所定の単位時間毎(BIN TIME)に、例えば、10μ秒毎に光検出器に到来するフォトンの数を計測する態様にて実行されるフォトンカウンティングであり、時系列の光強度のデータは、時系列のフォトンカウントデータであってよい。

Δt=(2Wo)2/6D …(2)
となるので、発光粒子がブラウン運動により移動する速度(拡散移動速度)Vdifは、概ね、
Vdif=2Wo/Δt==3D/Wo …(3)
となる。そこで、光検出領域の位置の移動速度は、かかるVdifを参照して、それよりも十分に早い値に設定されてよい。例えば、発光粒子の拡散係数が、D=2.0×10−10m2/s程度であると予想される場合には、Woが、0.62μm程度だとすると、Vdifは、1.0×10−3m/sとなるので、光検出領域の位置の移動速度は、その10倍以上の15mm/sなどと設定されてよい。なお、発光粒子の拡散係数が未知の場合には、光検出領域の位置の移動速度を種々設定して光強度の変化のプロファイルが、予想されるプロファイル(典型的には、励起光強度分布と略同様)となる条件を見つけるための予備実験を繰り返し実行して、好適な光検出領域の位置の移動速度が決定されてよい。
上記の処理により試料溶液中の発光粒子の発光部位別の時系列の光強度データが得られると、コンピュータ18に於いて、記憶装置に記憶されたプログラムに従った処理により、(i)各光強度データ上に於ける発光粒子からの光に対応する信号の検出、(ii)検出された信号に於いて発光部位別の時系列の光強度データ上で同時に発生した信号(同時発生パルス)の検出、(iii)同時発生パルスに於ける強度比の算出及び(iv)発光粒子の濃度算出等の各種分析が順に実行される。
時系列の光強度データに於いて、一つの発光粒子の光検出領域を通過する際の軌跡が、図5(B)に示されている如く略直線状である場合、その粒子に対応する信号に於ける光強度の変化は、(光学系により決定される)光検出領域の光強度分布を反映した略釣鐘状のプロファイルを有する。従って、適宜設定される閾値Ioを超える光強度が継続する時間幅Δτが所定の範囲にあるとき、その光強度のプロファイルを有する信号が一つの粒子が光検出領域を通過したことに対応すると判定され、一つの発光粒子の検出が為されるようになっていてよい。また、光検出領域の光強度分布が、ガウス分布:

であると仮定できるときには、有意な光強度のプロファイル(バックグラウンドでないと明らかに判断できるプロファイル)に対して式(4)をフィッティングして算出された強度A及び幅aが所定の範囲内にあるとき、その光強度のプロファイルが一つの粒子が光検出領域を通過したことに対応すると判定され、一つの発光粒子の検出が為されてよい。(強度A及び幅aが所定の範囲外にあるときには、ノイズ又は異物として分析に於いて無視されてよい。)
上記の如く複数の波長帯域の時系列光強度データ(即ち、発光部位別の時系列光強度データ)に於ける発光粒子に対応する信号(パルス信号)の検出が完了すると、検出された信号のうちで、複数の波長帯域の時系列光強度データに於いて同時に発生した信号が検出される(ステップ170)。かかる検出は、具体的には、例えば、図6(B)に例示されている如く、一方の波長帯域(波長I)の時系列光強度データに於ける発光粒子に対応するパルス信号のピーク(最大値)の時点tp1と他方の波長帯域(波長II)の時系列光強度データに於ける発光粒子に対応するパルス信号のピーク(最大値)の時点tp2との差ΔTが所定値を下回るとき、波長Iと波長IIに於いて同時に発生したパルス信号が発生したと判定する態様にて為されてよい。そして、かかる判定を時系列光強度データ上で順に繰り返すことにより、複数の波長帯域の時系列光強度データに於ける同時に発生したパルス信号(同時発生パルス信号)の全てが検出される。また、同時発生パルス信号の検出の別の態様として、一方の時系列光強度データに於ける信号の発生期間(始点〜終点)と他方の時系列光強度データに於ける信号の発生期間(始点〜終点)とが重複したときに、同時発生パルス信号が発生したと判定されてもよい。
上記の如く、同時発生パルスが検出されると、同時発生パルスの各々について、一方の波長帯域に於けるパルス信号の強度と他方の波長帯域に於けるパルス信号の強度との比率(第一の発光部位の発光波長のパルス信号の強度と第二の発光部位のパルス信号の強度との比率、或いは、共通発光部位の発光波長のパルス信号の強度と固有発光部位のパルス信号の強度との比率)が算出される。かかる比率は、例えば、各パルス信号のピークに於ける強度の比、或いは、各パルス信号の強度の積算値の比などであってよい。例えば、観測対象となる発光粒子に於いて共通発光部位と固有発光部位とが設定されている場合には、強度比率は、共通発光部位の発光波長帯域の信号のピーク強度Imax1と固有発光部位の発光波長帯域の信号のピーク強度Imax2とを用いて、

既に述べた如く、前記の信号強度(光強度)の比率は、発光粒子の光検出領域内での通過経路によらず、発光粒子の固有の値であると考えられるので、かかる信号強度の比率によって、発光粒子が特徴づけられ、その同定、即ち、発光粒子が予定された発光粒子であることの確認又はその種類の識別が為される。そして、信号強度の比率によって、又は、同定された結果によって、各信号を分類し、或いは、分類された信号毎にその数が計数されてよい。

により与えられる。また、参照溶液として、複数の異なる濃度の溶液が準備され、それぞれについて測定が実行されて、算出されたVtの平均値が光検出領域の通過した領域の総体積Vtとして採用されるようになっていてよい。そして、Vtが与えられると、粒子のカウンティング結果がnの試料溶液の発光粒子の濃度(数密度)cは、

により与えられる。なお、光検出領域の体積、光検出領域の通過した領域の総体積は、上記の方法によらず、任意の方法にて、例えば、FCS、FIDAを利用するなどして与えられるようになっていてよい。また、本実施形態の光分析装置に於いては、想定される光検出領域の移動パターンについて、種々の標準的な粒子についての濃度Cと粒子の数Nとの関係(式(6))の情報をコンピュータ18の記憶装置に予め記憶しておき、装置の使用者が光分析を実施する際に適宜記憶された関係の情報を利用できるようになっていてよい。特に、本発明に於いては、同時発生パルス信号に対応する発光粒子を観測するので、参照溶液として、観測される波長帯域の全てに発光波長を有する発光粒子の濃度が既知の溶液が準備され、光強度の測定により得られた時系列光強度データに於いて同時に発生した信号の検出及びカウンティングが実行され、そのカウント値と濃度とから光検出領域の通過した領域の総体積Vtが決定されてよい。
本発明の方法に従って、一本鎖のオリゴヌクレオチドと二本鎖のオリゴヌクレオチドとの識別が可能であることを検証した。

を満たすパルス信号のみを発光粒子(1T−A、2T−A)に対応する信号であると判定する一方、上記の条件を満たさないパルス信号はノイズとして無視した。
本発明の方法により、種々の鎖長の核酸分子が識別できることを検証した。
Claims (10)
- 共焦点顕微鏡又は多光子顕微鏡の光学系を用いて試料溶液中にて分散しランダムに運動する発光粒子からの光を検出し分析する方法であって、
第一の発光部位と該第一の発光部位と異なる発光波長を有する第二の発光部位とを有する発光粒子を含む試料溶液を調製する過程と、
前記光学系の光路を変更することにより前記試料溶液内に於いて前記光学系の光検出領域の位置を移動する過程と、
前記試料溶液内に於いて前記光検出領域の位置を移動させながら前記光検出領域からの前記第一の発光部位の光の強度と前記第二の発光部位の光の強度とを別々に且つ同時に測定して別々に光強度データを生成する過程と、
前記第一の発光部位の光強度データと前記第二の発光部位の光強度データとの各々に於いて、パルス状信号の存在する領域を検出し、前記検出されたパルス状信号の存在する領域に於ける光強度の時間変化に対する釣鐘状関数のフィッテングによって、前記検出されたパルス状信号の存在する領域に於ける光強度の時間変化が前記光検出領域内を相対的に移動する単一の発光粒子からの光に於いて想定されるプロファイルを有すると判定されたとき、その領域の光強度の時間変化を単一の発光粒子の光を表す信号として個別に検出する過程と、
前記第一の発光部位の光強度データと前記第二の発光部位の光強度データに於いて同時に発生した信号の各々に於いて、前記第一の発光部位の光の強度と前記第二の発光部位の光の強度との比率により単一の発光粒子を同定する過程と
を含むことを特徴とする方法。 - 請求項1の方法であって、前記第一の発光部位が前記試料溶液中の少なくとも一部の発光粒子が共通に有する共通発光部位であり、前記第二の発光部位が前記発光粒子の各々を特徴づける固有発光部位であることを特徴とする方法。
- 請求項1の方法であって、前記第一の発光部位の光の強度と前記第二の発光部位の光の強度との比率により前記単一の発光粒子の種類が識別されることを特徴とする方法。
- 請求項1の方法であって、前記第一の発光部位の光の強度と前記第二の発光部位の光の強度との比率により前記単一の発光粒子の大きさが決定されることを特徴とする方法。
- 請求項1の方法であって、前記第二の発光部位の種類が前記発光粒子の種類によって異なることを特徴とする方法。
- 請求項1の方法であって、前記第二の発光部位の数が前記発光粒子の種類によって異なることを特徴とする方法。
- 請求項6の方法であって、前記発光粒子の大きさが大きいほど単一の前記発光粒子の有する前記第二の発光部位の数が多くなることを特徴とする方法。
- 請求項1の方法であって、更に、前記同定された単一の発光粒子の数を計数する過程を含むことを特徴とする方法。
- 請求項1乃至8のいずれかの方法であって、前記光検出領域の位置が前記試料溶液中の発光粒子の拡散移動速度よりも速い速度にて移動されることを特徴とする方法。
- 請求項8の方法であって、更に、前記同定された単一の発光粒子の数に基づいて、該発光粒子の数密度又は濃度を決定する過程を含むことを特徴とする方法。
