Description
熱帯果物バイオマスであるココナッツ殻、パーム殻及びOPEFBを、平均面積0.5〜5cm2または平均長さ0.1〜5cmに粉砕した後、40〜80℃で12〜24時間乾燥した。
0.2〜5.0%硫酸水溶液で酸処理する代わりに、0.2〜5.0%の塩酸水溶液を利用して酸処理したことを除いては、実施例1と同様に行った。得られたキシロースとアラビノースの回収率及び純度を表3に示した。
0.2〜5.0%硫酸水溶液で酸処理する代わりに、0.2〜5.0%のシュウ酸水溶液を利用して酸処理したことを除いては、実施例1と同様に行った。得られたキシロースとアラビノースの回収率及び純度を表3に示した。
0.2〜5.0%硫酸水溶液で酸処理する代わりに、0.2〜5.0%の苛性ソーダ水溶液を利用したアルカリ処理後、塩酸でpH5.0〜7.0に中和させることを除いては、実施例1と同様に行った。得られたキシロースとアラビノースの回収率及び純度を表3に示した。
実施例1〜3及び比較例1により得られた単糖類とキシロース及びアラビノースを含む、イオン精製された糖液の糖組成を調べた。その結果を表4に示した。
実施例1〜3及び比較例1のキシロース及びアラビノースの製造過程で発生した副産物である加水分解残余物を収去し、40〜100℃で24〜48時間乾燥した後、10kg重量部を基準とし、500〜700℃で72〜168時間炭化させた後、成分含量を調べた。対照群として、酸加水分解を施さなかったココナッツ殻とパーム殻を、上記と同様な方法により製造した後、成分含量を比較し、その結果を表5に示した。表5から分かるように、対照群に比べ、炭化収率及び固定炭素が増加して、灰分量が減少したことが分かる。
実施例5のキシロース及びアラビノースの製造過程で発生した副産物である加水分解残余物の炭化後、800〜1,000℃の温度範囲で炭素を酸化反応させることにより、炭化物の表面浸食及び炭化物の微孔構造を生成させた。その後、ガス(水蒸気、二酸化炭素、空気などの酸化性ガス)活性化を通じて製造された活性炭を対照群として使用して、酸加水分解を施さなかったココナッツ殻とパーム殻を、上記と同様な方法を使用して活性炭を製造した後、成分含量を比較し、その結果を表6に示した。表6から分かるように、熱帯果物バイオマスであるココナッツ殻、パーム殻の加水分解残余物を使用した場合、対照群に比べ、ガス吸着力に優れていることが分かる。
加水分解糖化液には、微生物生育を抑制するフルフラール(furfural)、5−ヒドロキシメチル(5-hydroxymethly;HMF)、アセテート(acetate)、ヒドロキシベンズアルデヒド(hydroxybenzaldehyde;HBA)、バニリン(vanillin)などが存在し、今までのキシロースの転換収率は、58%に過ぎなかった。したがって、微生物の生長とキシリトールの収率の改善のために、本発明では、キシロース含量が20%である加水分解糖化液が炭素源として構成された固体培地で、20回の継代を通じて、キシリトール発酵微生物を、菌を阻害する成分に適応させた。
実施例7の培地と同様な培地を使用して、生物培養器で培養をした。その後、キシロースが枯渇される直前に、反応の完了された培養液を、生物反応器に取り付けられた減圧式微細ろ過管に移した。使用した菌体と培養濾液を分離した後、分離された菌体を濃縮した。新しい培地2Lを注入した生物反応器に、前記濃縮された菌体を搬送し、再培養した。培養条件は、前記実施例8と同様にした。
Claims (6)
- 熱帯果物バイオマスを酸加水分解する段階と、
前記酸加水分解によりキシロース及びアラビノースを含む加水分解糖化液を得る段階と、
前記加水分解糖化液を、沈殿法と電気透析工法とイオン交換法を順に利用して分離精製する段階と、
を含むことを特徴とする、キシロースとアラビノースとを含む加水分解糖化液の製造方法。 - 前記熱帯果物バイオマスが、ココナッツ殻(Coconut shell)またはパーム殻(Palm shell)またはパームオイル副産物(Oil Palm Empty Fruit Bunch;OPEFB)であることを特徴とする、請求項1に記載の加水分解糖化液の製造方法。
- 前記酸加水分解を、酸濃度0.2〜5%、温度100〜200℃、及び30分〜10時間の範囲で行うことを特徴とする、請求項1に記載の加水分解糖化液の製造方法。
- 前記電気透析工法が、イオン交換膜、電極板、流量調節ポンプ及び整流器を含んで構成される電気透析装置を用いることを特徴とする、請求項1に記載の加水分解糖化液の製造方法。
- 前記イオン交換膜が、陽イオン交換膜と陰イオン交換膜から構成されることを特徴とする、請求項4に記載の加水分解糖化液の製造方法。
- 請求項1〜5のいずれか1つの製造方法を含み、当該方法により製造された加水分解糖化液を醗酵する段階を含むことを特徴とする、キシリトールの製造方法。
