Description
空タンパク質マイクロカプセルからなる診断剤の製造に
関する。
事実はいつのまにか知られるようになった。泡含有液は
この目的のため血液中に注入でき〔コラーゲン膜で泡を
安定化させたOphir et al(1980)“Ultrasonic Imagin
g",2,67−77、米国特許第4,446,442号(Schering)及び
欧州特許出願第131540号(Schering)参照〕、欧州特許
出願第224934号及び欧州特許出願第324938号明細書では
アルブミン溶液を音波処理することでつくられた泡の用
法について開示している。しかしながら、泡のサイズ分
布は明らかにコントロールできず、泡は左心室で出会う
圧力に付された場合に消失する(Shapiro et al(199
0)J.Am.Coll.Cardiology,16(7),1603−1607)。
を中に混入させた固体粒子について開示しており、ガス
は血流中でその粒子から放出される。
リラクチド及びポリグリコリドのような合成ポリマーの
界面重合により空気又はガス充填微小中空球を製造する
プロセスについて開示している。国際出願第91/12823号
明細書(Delta)ではアルブミンを用いた類似プロセス
について開示している。Wheatley et al(1990)Biomat
erials,11,713−717では30μm径以上の微小泡を形成す
るためにアルギネートのイオノトロピー性ゲル化につい
て開示している。国際出願第91/09629号明細書では超音
波造影剤として使用のリポソームについて開示してい
る。
いで形成されるマイクロカプセルを不溶化するプロセス
が、改善された製品をもたらすことをここに発見した。
Przyborowskiら(1982,Eur.J.Nucl.Med.7,71−72)は放
射性同位元素標識のためスプレードライすることによる
ヒト血清アルブミン(HSA)微小球の製造とその後で肺
のシンチグラフィー画像処理における用法について開示
していた。その微小球は中空でないと言われ、我々の反
復研究では固体微小球のみが得られる。粒子が中空でな
いならば、それらは心エコー検査に適さない。更に、そ
の微小球は我々が心エコー検査に適したマイクロカプセ
ルを製造する上で適さないことを発見した1ステッププ
ロセスで製造された;微小球から未変性アルブミンを除
去することは従来プロセスで必要であった(我々のプロ
セスでは不要である);更に篩分けステップが必要とな
るような広いサイズ範囲の微小球が明らかに得られた。
このため、Przyborowskiらのプロセスは超音波画像処理
で有用なマイクロカプセルの製造用に選択する上で明白
なプロセスでないばかりか、産生される粒子もその目的
にとり不適切である。我々はその従来のプロセスに著し
い改善を加えた。
ブミン粒子を得る方法の2つの初期開示に関する。Aldr
ich&Johnston(1974)Int.J.Appl.Rad.Isot.25,15−18
では3〜70μm径粒子を形成してから熱油中で変性させ
るためスピニングディスクの使用について開示した。そ
の油は除去され、粒子は放射性同位元素で標識される。
Raji et al(1978)Isotopenpraxia,14(2),57−61で
も同様のスプニングディスク技術を用いたが、但し粒子
を単に加熱することでアルブミンを変性させた。いずれ
のケースでも中空マイクロカプセルについて言及され
ず、製造された粒子は心エコー検査に適さなかった。
質の溶液又は分散液を噴霧する第一ステップからなるプ
ロセスを提供する。

よく又は第一ステップの中間生成物は集められて、第二
ステップで別に処理してもよい。これら2つの可能性は
1ステップ及び2ステッププロセスとして以下呼ばれ
る。

かつ無免疫原性であるように選択されるべきである。壁
形成物質はデンプン誘導体、tert−ブチルオキシカルボ
ニルメチルポリグルタメート(米国特許第4,888,398
号)のような合成ポリマー又はポリデキストロースのよ
うな多糖である。
生理学上適合しうるポリマーから選択できる。このよう
なポリマーとしては低水溶性の多糖、ポリアクチド及び
ポリグリコリドとそれらのコポリマー、ラクチドとεカ
プロラクトン、δ−バレロラクトンのようなラクトンと
のコポリマー、ポリペプチド並びにゼラチン、コラーゲ
ン、グロブリン及びアルブミンのようなタンパク質を挙
げることができる。他の適切なポリマーとしてはポリ
(オルト)エステル(例えば、米国特許第4,093,709
号、米国特許第4,131,648号、米国特許第4,138,344号、
米国特許第4,180,646号明細書参照);ポリ乳酸、ポリ
グリコール酸及びそれらのコポリマー、例えばデキソン
(DEXON)(J.Heller(1980)Biomaterials,1,51参
照);ポリ(DL−ラクチド−コ−δ−カプロラクト
ン)、ポリ(DL−ラクチド−コ−δ−バレロラクト
ン)、ポリ(DL−ラクチド−コ−γ−ブチロラクト
ン)、ポリアルキルシアノアクリレート;ポリアミド、
ポリヒドロキシブチレート;ポリジオキサノン;ポリ−
β−アミノケトン(Polymer,23(1982),1693);ポリ
ホスファジン(Science,193(1976),1214);ポリ酸無
水物がある。生物分解性ポリマーに関する言及はR.Lang
er et al(1983)Macromol.Chem.Phys.C23,61−125でみ
られる。ポリグルタミン酸及びポリアスパラギン酸のよ
うなポリアミノ酸とそれらの誘導体、即ち低級アルコー
ル類又はグリコール類との部分エステルも使用できる。
このようなポリマーの1つの有用な例はポリ(t−ブチ
ルグルタメート)である。メチオニン、ロイシン、バリ
ン、プロリン、グリシン、アラニン等のような他のアミ
ノ酸のコポリマーも可能である。最近、コントロールさ
れた生物分解性を有するポリグルタミン酸及びポリアス
パラギン酸のいくつかの新規誘導体が報告された(参考
のためここに組み込まれる国際出願第87/03891号;米国
特許第4,888,398号及び欧州特許出願第130,935号明細書
参照)。これらのポリマー(及び他のアミノ酸とのコポ
リマー)は下記タイプの式を有する: −(NH−CHA−CO)x(NH−CHX−CO)y 上記式中xはアミノ酸残基の側鎖を表す;Aは式−(CH
2)nCOOR1R2OCOR(II)の基であり、ここでR1及びR2は
H又は低級アルキル、Rはアルキル又はアリールであ

例えば欧州特許出願第322094号の場合及びモノマーアル
ブミンのポリマーを含む)を発現する技術は例えば欧州
特許第201239号及び欧州特許出願第286424号明細書で開
示されている。すべての参考文献は参考のためここに含
まれる。HAの“アナログ及び断片”には(i)本発明の
プロセスでマイクロカプセルを形成できる及び(ii)ア
ミノ酸配列の少くとも50%(好ましくは少くとも75%、
80%、90%又は95%)の連続領域がヒトアルブミンの少
くとも50%(好ましくは75%、80%、90%又は95%)の
連続領域で少くとも80%相同性(好ましくは少くとも90
%、95%又は99%相同性)であるすべてのポリペプチド
を含む。組換えDNA技術で産生されるHAが特に好まし
い。このため、HAは酵母又は他の微生物でHAコードヌク
レオチド配列を発現させて当業界で知られているように
生成物を精製することにより産生してもよい。このよう
な物質は血清由来物質に伴う脂肪酸を欠く。好ましく
は、HAは脂肪酸を実質上含有しない;即ちそれは血清由
来物質で1%以下の脂肪酸レベルを有する。脂肪酸はHA
で検出できないことは好ましい。
という用語はこれが我々が好んだものであることから用
いられているが、但し他の生物適合性壁形成物質も前記
のように使用できることが理解される。
ブミンである場合に、好ましくは0.1〜50%w/v、更に好
ましくは約5.0〜25.0%タンパク質である。約20%が最
適である。壁形成物質の混合物も用いてよく、そのケー
スにおいて最後の2文におけるパーセンテージは壁形成
物質の全含有率に関する。
リア液体以外の物質を含有してもよい。このため、水相
は糖及び安定剤としてのポリマー、例えばポリビニルア
ルコール(PVA)、ポリビニルピロリドン(PVP)、ポリ
エチレングリコール(PEG)、ゼラチン、ポリグルタミ
ン酸及びデンプン、デキストラン、寒天、キサンタンガ
ム等の多糖のような水溶性親水性化合物を1〜20重量%
含有してもよい。類似した水相も最終微小球生成物が使
用前に懸濁されるキャリア液体として使用できる。ほと
んどの生理学上許容される乳化剤、例えば卵レシチン又
は大豆レシチンあるいは飽和合成レシチン、例えばジミ
リストイルホスファチジルコリン、ジパルミトイルホス
ファチジルコリンもしくはジステアロイルホスファチジ
ルコリン又は不飽和合成レシチン、例えばジオレイルホ
スファチジルコリンもしくはジリノレイルホスファチジ
ルコリンのような合成レシチンを含めた乳化剤が使用で
きる(0.1〜5重量%)。乳化剤には、遊離脂肪酸、脂
肪酸とポリオキシアルキレン化合物、例えばポリオキシ
プロピレングリコール及びポリオキシエチレングリコー
ルとのエステルのような界面活性剤;脂肪アルコールと
ポリオキシアルキレングリコールとのエーテル;脂肪酸
とポリオキシアルキル化ソルビタンとのエステル;石
鹸;グリセロール−ポリアルキレンステアレート;グリ
セロース−ポリオキシアルキレンリシノレエート;ポリ
アルキレングリコールのホモ及びコポリマー;ポリエト
キシル化大豆油及びヒマシ油と水素添加誘導体;スクロ
ース又は他の炭水化物と脂肪酸、脂肪アルコールとのエ
ーテル及びエステル(これらは場合によりポリオキシア
ルキル化されている);飽和もしくは不飽和脂肪酸、グ
リセリド又は大豆油及びスクロースのモノ、ジ及びトリ
グリセリドも含む。

ルトン)ポリグリコリド又はポリラクチドを配合するこ
とで有利に改質する(柔軟性及び生物分解性を高める)
ことができる。中〜低Mwのポリエチレングリコール(例
えば、PEG2000)も有用な柔軟性添加剤である。

く使用されない;他のケースにおいては壁の約20重量%
に達する添加剤の量が可能である。
液又は分散液(好ましくは溶液)は1.00〜50.0μm径の
個別のマイクロカプセルを与えるいずれか適切な技術に
より噴霧及びスプレードライされる。これらの数値はマ
イクロカプセルの総数の少くとも90%に関し、径はコー
ルター・マスター・サイザーII(Coulter Master Sizer
II)で測定される。“マイクロカプセル”という用語
は空間を封じる中空粒子を意味し、その空間は何らかの
固体物質ではなくガス又は蒸気で充填される。
国で販売される菓子に似たニーコーム化粒子は形成され
ない。空間が全体的に包囲されることは不要であり(但
しこれが好ましい)、マイクロカプセルが正確に球状で
ある必要はないが、但しそれらは通常球状である。マイ
クロカプセルが球状でない場合、上記径は非球状マイク
ロカプセルと同一の質量を有して同一の中空空間容量を
包囲する対応した球状マイクロカプセルの径に関する。
加圧下で少くとも1つのオリフィスから調製液を押出す
か又はその中で遠心アトマイザーを用いることによりタ
ンパク質調製液のエアゾールを形成する。その室は乾燥
前に最大射出液滴が壁をたたかないほど十分に大である
べきである。室内におけるガス又は蒸気は心エコー検査
においてマイクロカプセルの投与に伴う量で血流中に投
与された場合にクリーン(即ち、好ましくは無菌及び無
発熱物質)及び無毒性である。タンパク質調製液からの
液体の蒸発速度は中空マイクロカプセルを形成する上で
十分高いが、但しマイクロカプセルを破裂させるほど高
くてはならない。蒸発速度はガス流速、タンパク質調製
液中におけるタンパク質の濃度、液体キャリアの性質、
溶液の供給速度と最も重要なことにはエアゾールで出会
うガスの温度を変えることによりコントロールしてもよ
い。水中15〜25%のアルブミン濃度の場合、少くとも約
100℃、好ましくは少くとも110℃の入口ガス温度であれ
ば中空性を確保する上で通常十分であり、その温度はカ
プセルが破裂しないのであれば250℃ほどの高温でもよ
い。約180〜240℃、好ましくは約210〜230℃、最も好ま
しくは約220℃が少くともアルブミンの場合で最適であ
る。その温度は本発明のプロセスの1ステップバージョ
ンにおいて壁形成物質の少くとも一部(通常外側)、多
くは壁形成物質の実質上全部を不溶化する上で十分であ
る。エアゾールで出会うガスの温度はエアゾールが放出
される速度及びタンパク質調製液の液体分にも依存する
ことから、出口温度は室内で適度の温度を確保するため
にモニターされる。40〜150℃の出口温度が適切である
とわかった。しかしながら、このファクターは別にし
て、流速のコントロールは他のパラメーターのコントロ
ールほど有用でないとわかった。
は典型的には96〜98%のモノマーHAを含み、超音波画像
処理で限られたインビボ寿命を有する。しかしながら、
それらは超音波画像処理で用いても又はそれらは2ステ
ッププロセスの第二ステップが実施される前に貯蔵及び
輸送してもよい。したがって、それらは本発明のもう1
つの面を形成する。
で製造された中間マイクロカプセルは固定されて、それ
らが長時間存続するように低水溶性とされるが、一方そ
れらが生物分解性でないほど不溶性及び不活性ではな
い。このステップでは更にそれらが投与、血管内剪断及
び心室内の苛酷さにもっとよく耐えうるようにマイクロ
カプセルを強化する。マイクロカプセルが破裂すると、
それらはエコー源性が少なくなる。Schneider et al(1
992)Invest.Radiol.27,134−139では、従来の音波処理
アルブミン微小泡がこの強度を有さず、左心室で典型的
な圧力に付された場合にそれらのエコー源性を急速に失
うことを示した。本プロセスの第二ステップではタンパ
ク質を架橋するため熱(例えば慣用的なオープン中にお
ける、例えばマイクロ波熱、輻射熱又は熱風)、電離線
(例えば、10.0〜100.0kGy線量のγ線による)又は例え
ばホルムアルデヒド、グルタルアルデヒド、エチレンオ
キシドもしくは他の物質を用いる化学的架橋を用いても
よく、第一ステップで形成された実質上乾燥した中間マ
イクロカプセルで又はマイクロカプセルが不溶性である
液体、例えば適切な溶媒中このようなマイクロカプセル
の懸濁液で実施される。本プロセスの1ステップバージ
ョンにおいて、グルタルアルデヒドのような架橋剤はス
プレードライ室内にスプレーしても又はスプレー手段の
すぐ上流でタンパク質調製液中に導入してもよい。一
方、室内の温度はマイクロカプセルを不溶化するほど十
分高くてもよい。
ところ所望であれば0.05〜50.0μmの径を有するマイク
ロカプセルからなるが、但し0.1〜20.0μm、特に1.0〜
8.0μmの範囲が本発明のプロセスで得られ、心エコー
検査にとり好ましい。我々は約0.5〜3.0μmの範囲が低
コントラスト画像の作製及びカラードップラー画像処理
用に特に適しており、一方約4.0〜6.0μmの範囲がシャ
ープな画像の作製にとり良いことを発見した。第一ステ
ップで作製されたサイズを調べる上で第二ステップがマ
イクロカプセルのサイズを変えるかもしれないという事
実を考慮する必要がある。

る;微小球の少くとも90%、好ましくは95%又は99%が
40〜500nm、好ましくは100〜500nmの壁厚を有する;微
小球の壁中におけるタンパク質の少くとも50%が架橋さ
れているタンパク質壁の中空微小球を提供する。好まし
くは、タンパク質の少くとも75%、90%、95%、98.0
%、98.5%又は99%は1%HC1溶液で2分間の抽出に抵
抗しうるほど十分に架橋されている。抽出されたタンパ
ク質はクマシーブルータンパク質アッセイ、ブラッドフ
ォード(Bradford)を用いて検出される。架橋度はタン
パク質の加熱、照射又は化学処理を変えることによりコ
ントロールされる。架橋プロセス中に、タンパク質モノ
マーは下記例8で示されたゲル透過HPLC又はゲル電気泳
動で検出されるように架橋されて、単純な溶解プロセス
で直ちに利用不能になる。継続処理は既に架橋された物
質を更に架橋させ、その結果それは前記HC1抽出で利用
不能になる。175℃で加熱中に本発明によるrHA微小球は
20分間の過程でHC1抽出性タンパク質の約99%を失う
が、一方150℃で20分間の加熱では約5%のHC1抽出性タ
ンパク質のみを除去し、30分間で47.5%を除去し、40分
間で83%、60分間で93%、80分間で97%及び100分間で9
7.8%のHC1抽出性タンパク質を除去する。したがって、
良好な架橋レベルを達成するため、微小球は175℃で少
くとも17〜20分間、150℃で少くとも80分間及び他の温
度でそれに対応してより長い又は短い時間にわたり加熱
される。
水に懸濁された場合に破裂、崩壊又は水侵入なしに2.66
×104Pa圧力の0.25秒間適用から残存しうる、主に径が
1.0〜10.0μmの中空微小球を提供する。ヒト左心室に
おける一過性最大圧力は約200mmHg(2.66×104Pa)であ
る。好ましくは50%、75%、90%又は100%が前記試験
で2.66×104Paの上記0.25秒間適用から残存し、即ちエ
コー源性のままである。インビボにおいて、同率が心臓
の両室に1回の通過中エコー源性のままであることが好
ましい。

る。添加剤としては、0.1〜25重量%のマンニトール、
ガラクトース、ラクトースもしくはスクロースのような
水溶性の生理学上許容される化合物又はデキストラン、
キサンタンガム、寒天、デンプン、PVP、ポリグルタミ
ン酸、ポリビニルアルコール(PVA)及びゼラチンのよ
うな親水性ポリマーから選択することができる。注入用
液体キャリア相中における微小球の有用な寿命、即ち有
用な超音波検査シグナルが観察される期間は必要性に応
じて数分間〜数ヵ月間続くようにコントロールできる;
これは壁の多孔度、溶解度又は架橋度のコントロールに
より実施できる。これらのパラメーターは壁形成物質及
び添加剤を適正に選択してスプレードライ室における蒸
発速度及び温度を調整することによりコントロールでき
る。
カプセルは0.5mmスクリーン装備フリッチュ(Fritsch)
遠心ピンミル又はグレン・クレストン(Glen Creston)
エアーインパクトジェットミルを用いて適切な不活性賦
形剤と共に粉砕することができる。適切な賦形剤がラク
トース、グリコール、マンニトール、ソルビトール、ガ
ラクトース、マルトース又は塩化ナトリウムのような静
脈内使用で不活性かつ適切な微粉砕粉末である。粉砕さ
れると、マイクロカプセル/賦形剤混合物は非機能性/
欠陥マイクロカプセルの除去を容易化するため水性媒体
に懸濁させることができる。水相中における再調製で
は、凝集を防止するため痕跡量の界面活性剤を含有させ
ることが望ましい。この目的に適したアニオン系、カチ
オン系及びノニオン系界面活性剤としてはポロオキサマ
ー、ソルビタンエステル、ポリソルベート及びレシチン
がある。
液体で満たしてもはやエコー源性でないようにする表面
欠陥を有した欠陥粒子を浮遊させても又は沈降させるた
め遠心してもよい。

aCl 0.01mMトリスのような静注用に適した緩衝液で再調
製される。その懸濁液は凍結乾燥とその後に例えばγ−
線照射、乾熱又はエチレンオキシドによる滅菌のために

〜5.0×109粒子/mlのマイクロカプセル濃度が適切であ
る。

スでは人体の心臓領域の視覚画像を生じる。典型的に
は、変換器は画像化される領域の皮膚上に直接おかれ
る。スキャナーは超音波変換器を含めた様々な電子部品
を収納している。変換器は心臓領域のセクター走査を実
施する超音波を生じる。超音波は心臓領域の様々な部分
により反射され、受信変換器により受信され、当業界で
公知のパルス−エコー法に従い処理される。処理後、シ
グナルは観察のため画像処理装置(当業界でも周知)に
送られる。
ナーが適所におかれた後、マイクロカプセル懸濁液は例
えば腕血管から注入される。造影剤はその血管から心臓
の右静脈側に流れ、主肺動脈を経て肺に、更に肺から毛

腎臓を含めた多くの臓器系の画像を作製するために使用
できるが、心臓の組織及び主要血管の視覚化のために多
く用いられる。

させるため(ストレプトキナーゼのような)血栓溶解剤
の使用;又は(4)最近の心臓発作による灌流欠陥又は
変化のような介在による心筋組織灌流の変化を示す上で
有用である。

大6バールの空気圧でエアタービンにより駆動される遠
心アトマイザー(タイプM−02/Bマイナー)からなる。
6バールで約33,000rpmのアトマイザーホイール速度に
達する。アトマイザーへの圧縮空気のオン−オフ制御は
計器盤に設置されたバルブにより行われる。アトマイザ
ーへの圧縮空気の最大消費は6バールの圧力で17Nm3/h
である。液体フィード及び粉末と接触するすべての部分
はステンレススチールAISI 316から構成されるが、但し
ポンプ供給チューブ及びアトマイザーホイールはステン
レススチールAISI 329から構成されて高遠心力に耐える
ように造られている。

板でカバーされている。乾燥室は操作中における検査用
に側灯及び観察窓を備えている。乾燥室の屋根は内面が
ステンレススチールAISI 316及び外面がステンレススチ

る空気の分配に用いる。ステンレススチールAISI 316か
ら構成されるエアダクトはステンレススチールAISI 316
から構成されて粉末及び空気を分離するように考えられ
たサイクロンに排気及び粉末を横輸送する。
ーンゴムのガスケットを有するバタフライ弁タイプの閉
鎖バルブはスプリング装置によりサイクロン下にしっか
りと設置された粉末回収ガラスジャー中へのサイクロン
下における粉末放出のために用いられる。

から構成されるファンは乾燥室及びサイクロンから空気
及び粉末を吸引する。

圧力で8〜15kg/h アトマイザー装置への壁形成調製液フィードの輸送用
に適した供給ポンプは蠕動ポンプである。そのポンプは
モーター(1×220V,50Hz,0.18kW)及び手動調節用の連
続可変ギアを備えている。シリコーンホースから構成さ
れる供給パイプは供給タンク(局部供給)から供給ポン
プを経て噴霧装置に続いている。
ディ及び絶対エアフィルターからなる絶対エアフィルタ
ーは導入乾燥空気を完全にクリーンにするその処理のた
めに用いられる。
Aの20%溶液を前記市販スプレードライユニットに設置
された二液ノズルアトマイザーのノズルにポンプ導入し
た。蠕動ポンプ速度は220℃の入口空気温度のとき出口
空気温度が95℃に維持されるように約10ml/minの速度で
維持した。

はインビトロエコー源性半減期を数秒間から30分間を超
えるまでに増加させたことであった。インキュベートの
温度及び時間を変えることにより、固定率を5〜100%
で変動させることができる。様々な温度の熱固定特性の
例は図2で示されている。
解凝集して水中に分散させた。方法1では最初に熱固定
された球を等重量の微粉砕ラクトース(平均径5μm)
とミックスさせた。次いで混合物を0.5mmスクリーン及
びは12歯ローターを装備したフリッチュ遠心ミルに通し
た。粉砕された球を集め、完全混合が生じたことを確実
にするため再度ミルに通した。次いで粉砕された粉末を
1mg/mlプルロニック(Pluronic)F68含有水に再懸濁し
た。典型的には10gのマイクロカプセル及びラクトース
を水100ml及びプルロニックF68に加えた。解凝集に関す
る方法2ではプルロニックF68 100mg含有水100mlに熱固
定マイクロカプセル5gを加える。マイクロカプセルはシ
ルバーソン(Silverson)ホモゲナイザー(2.54cmチュ
ーブ状ホモゲナイズプローブ及び高剪断スクリーンを装
備したモデルL4R)を用いて60秒間ホモゲナイズするこ
とにより分散させた。

程度はこの方法により5〜100%で変動させて、1〜2
分間から1時間を超えるまでインビトロエコー源性半減
期を伸ばすことができる。

て、マイクロカプセル形成及びポリマー物質の不溶化は
スプレードライプロセス中で同時に行われる。

口から揮発性溶媒を取り扱える特別に適合化されたスプ
レードライユニットに設置された二液ノズルアトマイザ
ーに直接供給した。スプレードライの条件は例1で示さ
れたとおりであった。マイクロカプセルを室温で乾燥イ
ンキュベートしてタンパク質内架橋を形成させ、しかる
後5%エタノールアミン含有エタノールに懸濁して残留
架橋剤を不活化させた。マイクロカプセルの十分な洗浄
を行い、最後にマイクロカプセルを真空乾燥させて残留
溶媒を除去した。

カプセル100mgに加え、30秒間音波処理した。この懸濁
液にH2O19mlを加えた。

ウム緩衝液(pH7.0)を用いて1ml/minの流速でTSKゲル
透過カラムでクロマトグラフィーに付すことにより実施
した。

ために用いた。
モノマー濃度を測定して非固定マイクロカプセルのモノ
マー濃度のパーセンテージとしてこの数値を表すことに
より計算した。結果は図2で示されている。
レードタイマイクロカプセルの加熱は検出しうるモノマ
ーの量を減少させる(図2参照)。検出しうるモノマー
rHAに関するこの減少は前記HPLC法で調べることができ
ない不溶性ポリマー中へのモノマーrHAの変性及び架橋
に起因する。

液について圧力増加は3回実施した。密閉容器における
圧力はRS把持マノメーターで評価した。

イクロカプセルの分類に関しても良い結果を出すことが
できる。

燥空気温度を350℃以内にする電気エアヒーターであ
る。乾燥空気温度はパーセンテージタイマースイッチを
用いて連続的に調節できる。最大粉末消費は7.5KWであ

Claims (12)
- 【請求項1】懸濁液を形成するために非経口注入媒体中
に懸濁でき、静脈内投与による超音波造影に適した中空
マイクロカプセルからなる粉末の形成方法であって、 (a)液体中で壁形成物質の溶液又は分散液を作り、 (b)上記液体が蒸発するように無菌および無毒性の加
熱ガス中に上記溶液又は分散液を噴霧して、上記壁形成
物質の中空マイクロカプセルを形成させ、そして (c)上記壁形成物質の水溶性を減少させるために上記
マイクロカプセルを加熱して、粉末として上記マイクロ
カプセルを得る(その粉末は20.0μm以下の径を有した
中空マイクロカプセルからなる) ことを含む方法。 - 【請求項2】粉末が0.05〜20.0μmの径を有した中空マ
イクロカプセルからなる、請求項1に記載の方法。 - 【請求項3】粉末が0.5〜20.0μmの径を有した中空マ
イクロカプセルからなる、請求項1に記載の方法。 - 【請求項4】粉末が滅菌条件下でステップ(a)〜
(c)を行うことにより滅菌される、請求項1に記載の
方法。 - 【請求項5】壁形成物質が水溶性である、請求項1〜4
のいずれか一項に記載の方法。 - 【請求項6】壁形成物質がタンパク質である、請求項5
に記載の方法。 - 【請求項7】タンパク質がコラーゲン、ゼラチン又はヒ
トアルブミンである、請求項6に記載の方法。 - 【請求項8】タンパク質が組換えDNA技術により製造さ
れた又はヒト血清から単離されたヒトアルブミンであ
る、請求項6に記載の方法。 - 【請求項9】タンパク質溶液又は分散液が10.0〜30.0%
