細胞株での実験に加えて、個々のCARユニットの機能を試験するために、患者の試料についての研究も実施した。侵攻性の急性骨髄性白血病(AML)であるAML-9は、CD33CD123 cCARの有効性を試験するために用いられた。AML-9サンプル中の複数の細胞型を含む患者細胞集団の不均一性のために、白血病芽球は、2つの陽性マーカーであったCD34およびCD33でゲートされた。白血病細胞のこのCD33 + CD34 +集団の枯渇は、GFPコントロールと比較してCAR T細胞では48%であることが観察された(図6)。
ナチュラルキラー(NK)細胞はCD56+ CD3-であり、CD8+ T細胞のような感染細胞および腫瘍細胞を効率的に殺すことができる。CD8+ T細胞とは異なり、NK細胞は、細胞を殺すために活性化を必要とせずに腫瘍に対して細胞傷害性を発揮する。NK細胞は、サイトカインストームによる潜在的に致命的な合併症を回避できるので、より安全なエフェクター細胞である。しかしながら、CD33またはCD123または両方のCAR NK細胞を用いて、白血病を死滅させることに関しては完全に未開発である。
NK-92細胞に、CD33CD123 CARレンチウイルス上清を、2度にわたる連続したオーバーナイトでの形質導入方で形質導入を行い、方法としてはレトロネクチンおよびウイルスをコーティングしたプレートをそれぞれの形質導入時に交換した。形質導入した細胞を3または4日間増殖させ、次いでCARの発現をフローサイトメトリーによって分析した。細胞を採取し、ヤギ抗マウスF(Ab ')2と約1:250の割合で約30分間インキュベートした。細胞を洗浄し、懸濁させ、ストレプトアビジン-PEで約30分間染色した。細胞を洗浄し、2%ホルマリン中に懸濁させ、フローサイトメトリーにより分析した。次いで、CD33CD123cCARを発現するNK-92細胞を上記のように標識し、FACSAria上で選別し、F(Ab ')2でラベルされた細胞の上部0.2%を収集し、培養した。選別して培養した細胞のその後の標識では、抗マウスF(Ab ')2についてNK-92細胞は約89%の陽性を示した(図8)。
最初に、我々は、共培養においてHL-60癌細胞株を死滅させる能力を測定することにより、CD33CD123 cCAR NK-92細胞の機能を試験した。実質的にすべてのHL-60細胞はCD33を高発現するが、この細胞株でのCD123発現は10%(弱)未満である。したがって、CD33CD123cCARの殺傷能力は、cCARがCD33を適切に標的化している能力 に依存する可能性が高い。
3つのcCARは、上記のCD33CD123 cCARと同様の方法を用いて作製された(図13)。
CD38、CS1、CD138、B細胞成熟抗原(BCMA)およびCD38を含む標的抗原に対するcCARsの前臨床研究が展開されている。CD19 CARはまた、第I相臨床試験において多発性骨髄腫の治療に対していくつかの有効性を示した。しかし、表面抗原発現の不均一性が悪性形質細胞で一般的に生じることから(Ruiz-Arguelles and San Miguel 1994)、単一の標的がこの疾患を治療するのに十分であるとは考えにくい。BCMA CS1 cCAR、BCMA CD19 cCAR、BCMA CD38 cCARおよびBCMA CD138 cCARが作製され、実験デザインは上記のCD33CD123 cCARのものと同様であった。
BCMA CS1 cCAR(BC1cCAR)の作製および特性評価
BC1cCARのモジュールデザインは、自己切断P2Aペプチドを挟んで、抗CD269(BCMA、B細胞成熟抗原)一本鎖可変断片(scFv)領域と抗CD319(CS1)scFvと融合した 、CD8由来のヒンジ(H)および膜貫通(TM)領域、そしてタンデム4-1BB共活性化ドメイン、ならびにCD3ζシグナル伝達ドメインに連結した形を含むように設計される(図14A)。強力な脾臓フォーカス形成ウイルスプロモーター(SFFV)およびCD8リーダー配列を、BC1cCARのCAR分子のT細胞表面上での効率的な発現のために使用した。2つのユニットCARは、同じ共刺激ドメインである、4-1BBを使用する。トランスフェクトされたBC1cCAR HEK293T細胞を、compound構造の確認のために、ウエスタンブロット分析を行った。抗CD3モノクローナル抗体を用いたイムノブロットにおいて、compound CAR CD3融合タンパク質での予測される大きさのバンドを示した(図14E)。重要なことに、P2Aペプチドの高切断作用の成功により同様の強度の2つの別個のバンドのシグナルが観察された。期待されたように、GFPコントロールベクターについてはCD3の発現は見られなかった。
臍帯血(UCB)バフィーコートから単離したT細胞を、2日間の活性化後にBC1cCARレンチウイルスを用い形質導入を行った。2つのCARsユニットは、同じ共刺激ドメイン4-1BBを使用した。BC1cCARの形質導入効率は、フローサイトメトリーによって 約15%であると決定された(図14B)。BC1cCAR T細胞を、骨髄腫マーカーであるBCMAおよびCS1らが陰性であるCML(慢性骨髄性白血病)細胞株で最初に試験した。予想通り、野生型K562に対して、コントロールT細胞またはBC1cCAR T細胞のいずれからも溶解させなかった(図14C)。BCMA-K562(Kochenderfer、NIH)は、BCMAを発現するcDNAで形質導入されたK562細胞で、細胞集団の80%以上でBCMAを発現する。BC1cCAR T細胞をこの細胞株と2:1および5:1のE:T比で共培養し、コントロールと比較して30%を超える溶解を示した(図14C)。これらの結果は、CS1CAR T細胞のように、その他のCARに対する抗原を形質導入した細胞株で行われた他の1共培養実験の結果と適合する。
BC1cCAR T-cells による細胞障害活性を評価するため、MM1S(BMCA + CS1 +)、RPMI-8226(BCMA + CS1-)およびU266(BCMA + CS1dim)らの骨髄腫細胞株と共培養アッセイを行った。標的細胞を溶解するBC1cCAR T細胞の能力は、フローサイトメトリー解析によって定量化し、標的細胞をCytotracker色素(CMTMR)で染色した。24時間の共培養において、BC1cCARはMM1S細胞の実質的に完全な溶解を示し、2:1のE:T比で標的細胞の90%以が減少し、および5:1のE:Tでの95%以上の減少を示した(図15)。RPMI-8226細胞において、BC1cCARはE:T比2:1で70%以上のBCMA +標的細胞を溶解し、5:1のE:Tで75%以上を溶解した(図16)。U266標的細胞との24時間の共培養において、BC1cCARはBCMA+U266細胞の80%を2:1のE:T比で溶解し、飽和状態に達した(図17)。
MM10-G患者サンプルのフローサイトメトリー分析で、明瞭かつ一貫したBCMA +およびCS1 +集団サブセットを明らかにする(図18)。MM7-Gサンプルは完全なBCMA + CS1 +表現型を示し、一方、MM11-Gは骨髄吸引物であるという性質に起因すると思われるノイズの多いBCMAdim CS1dimの表現型を示す。24時間後、BC1cCAR T細胞は、E:T比5:1で75%以上の溶解を示し、10:1で85%以上に増加し(図19)、MM7-Gの一次患者試料の強いアブレーションを示す。MM11-G(図20)に対して、BC1cCAR T細胞はE:T = 10:1でBCMA + CS1 +集団の45%以上を溶解することができた。
C1cCAR T細胞のインビボでの抗腫瘍活性を評価するために、我々は、ルシフェラーゼ発現MM.1S細胞(多発性骨髄腫細胞株)を致死量以下で照射したNSGマウスに静脈注射し、測定可能な白血病形成を誘発することができる、異種マウスモデルを開発した。腫瘍細胞の注射3日後に、マウスに、8×106個のBC1cCAR T細胞またはベクターコントロールT細胞を単回用量で静脈注射した。3日目、6日目、および8日目に、マウスにRediJect D-Luciferin(Perkin Elmer)を皮下注射し、腫瘍負荷をIVISイメージングにより測定した(図21)。BC1cCAR T細胞またはベクターコントロールT細胞で治療した後の腫瘍細胞のパーセンテージを比較するため、測定した平均光強度をそれぞれのマウスで比較した。Unpaired T検定(P = 0.0001)では、2つのグループ間で8日目までに、BC1cCAR T細胞の注射グールプでは、コントロールグループと比較し、光強度がより少なく、したがって腫瘍負荷が少ない非常に有意な差を示した(p <0.0001)。1日目およびその後1日おきに、腫瘍サイズ面積を測定し、2つのグループ間の平均腫瘍サイズを比較した(図21)。要約すると、これらのインビボデータは、CD269-CS1-BBCAR T細胞が、ベクターコントロールT細胞と比較した場合、MM.1Sを注射したNSGマウスにおける腫瘍負荷を有意に低下させることを示す。
目的の遺伝子を標的とするために、3対のsgRNAをCHOPCHOPで設計する。次いで、遺伝子特異的sgRNAを、ヒトCas9およびE2A自己切断リンカーと連結したピューロマイシン耐性遺伝子を発現するレンチウイルスベクター(Lenti U6-sgRNA-SFFV-Cas9-puro-wpre) にクローニングする。U6-sgRNAカセットはCas9エレメントの前にある。sgRNAおよびCas9puroの発現は、それぞれU6プロモーターおよびSFFVプロモーターによって誘導される(図23)。

レンチウイルスに保有された遺伝子特異的sgRNAを用いてNK-92細胞を形質導入した。NK-92細胞におけるCD45発現の消失は、フローサイトメトリー解析によって判定した。NK-92細胞のCD45陰性集団を選別し、増殖させた(図25)。選別後に増殖させたCD45陰性NK-92細胞を用いてCD45CAR NK細胞を作製した。得られたCD45CAR NK細胞を用いて、CD45 +細胞を殺傷させる能力においてテストした。
我々は、CD45のCRISPR / Casヌクレアーゼによる不活性化後、NK45i-92細胞の細胞増殖が野生型NK-92細胞の増殖と同様であることを実証した(図26)。CD45の不活性化は、NK-92の細胞増殖に全く有意に影響しなかった。さらに、NK45i-92細胞の溶解能は、細胞を白血病細胞であるCCRFと共培養した場合、野生型NK-92の溶解能と一致していることを示した(図27)。
我々は次に、NK45i-92細胞中のCD45CARが白血病細胞中のCD45抗原に応答することを調べる。我々はCD45CARを作製した。CD45CARは、CD3ζシグナル伝達ドメインが連結された抗CD45一本鎖可変断片(scFv)領域、CD8由来ヒンジ(H)および膜貫通(TM)領域およびタンデムCD28および4-1BB共活性化ドメインからなる29A)。強力な脾臓フォーカス形成ウイルスプロモーター(SFFV)およびCD8リーダー配列を使用した。CD45CARタンパク質は、CD45CARレンチウイルスプラスミドでトランスフェクトされたHEK293-FT細胞を用いたウエスタンブロットで適切なベクターコントロールと比較によって明らかにされた。さらに、抗CD3ゼータモノクローナル抗体を用いたイムノブロットにより、ベクターコントロールはバンドが見られなかったが、CD45CARタンパク質の予測されるサイズのバンドが目的のサンプルからみられた(図29B)。
蛍光活性化細胞選別(FACS)によるCD45NK45i-92細胞が多く含まれる領域を回収後、ソーティング後にけるCD45CAR NK-92導入効率は87%であるとフローサイトメトリー(図30)によって測定された。CD45NK45i -92細胞のFACS収集後、CD45CAR発現レベルは、少なくとも10継代にわたって一貫して安定したままであった。
CD45CAR NK45i -92抗白血病活性を評価するために、T-ALL細胞株である、CCRF-CEMおよびJurkat、ならびにNK細胞株およびNK-92細胞を用いた共培養アッセイを実施した、何故ならばそれら細胞はCD45を発現しているからである(図31、32、33)。本発明者らは、我々はCD45CAR NK45i -92細胞が一貫して白血病細胞の強力な溶解を示したことを実証した。CD45CAR NK45i -92細胞は、標的細胞に対する低有効性(E:T比5:1)で6時間インキュベートした後、60%を超えるCCRF-CEM細胞を効果的に溶解した(図31)。6時間の共培養後、CD45CAR NK45i -92細胞はまた、E:T、2:1または5:1の比で約60%のJurkat細胞を殺傷することができた(図32)。6時間の共培養の後、CD45CAR NK45i-92細胞は、E:T比2:1で20%CD45陽性NK-92細胞を効率的に溶解し、E:T = 5:1で60%に近い溶解度を示した(図33A~33C)。
最近の研究により、T細胞の持続性がCAR T細胞の治療効果と良好に相関することが実証されている。最近の試験は、強力かつ持続性の高腫瘍活性を持つように作製された高品質の少数のCAR T細胞を注入することが、量を注入することよりもむしろ 抗腫瘍活性に寄与する上でより重要であることを実証している。
CD4IL15RA-CARが作製され、それはIL15RAに連結した第3世代のCD4CARを含む(図34)。CAR(第3世代)、sushi / IL-15の組み合わせを発現ベクター上に組み立て、それらの発現をSFFVプロモーターにより誘導する(図34)。sushi/ IL-15を含むCARは、P2A切断配列と連結している。sushi/ IL-15部分は、sushiドメインにIL-2シグナルペプチドが融合され、26アミノ酸のポリ - プロリンリンカーを介してIL-15に連結されたものからなる(図34)。
NK-92細胞にCD4IL15RA-CARレンチウイルス上清で形質導入を行った。5日間のインキュベーション後、細胞を採取し、ヤギ抗マウスF(Ab ')2と1:250の割合で30分間インキュベートした。細胞を洗浄し、懸濁させ、ストレプトアビジン-PEで30分間染色した。細胞を洗浄し、2%ホルマリン中に懸濁させ、フローサイトメトリーにより分析した結果、形質導入細胞のほぼ70%でCD4IL15RA-CARを発現したことがわかった (図36の丸で囲んだ領域)。CD4IL15RA-CARのさらなる実験的試験には、インビトロおよびインビボでの白血病/リンパ腫殺傷アッセイ、ならびにCD4CARで形質導入された細胞とでの標的殺傷能力および細胞増殖速度の比較検討が含まれる。本発明者はまた、CD19IL15RA-CARを作製するために上記に述べられた、同じ方法を用いた。
ヒト臍帯バフィーコート細胞にCD4IL15RA-CARレンチウイルス上清を形質導入した。5日間のインキュベーション後、細胞を採取し、ヤギ抗マウスF(Ab ')2と1:250の比率で30分間インキュベートした。細胞を洗浄し、懸濁させ、ストレプトアビジン-PE抗体で30分間染色した。細胞を洗浄し、2%ホルマリン中に懸濁させ、フローサイトメトリーで分析し、その結果、形質導入細胞の63%がCD4IL15RA-CARを発現する ことがわかった(図37の丸で囲んだ領域 )。CD4IL15RA-CARのさらなる実験的試験には、インビトロおよびインビボでの白血病/リンパ腫殺傷アッセイ、ならびにCD4CARで形質導入された細胞とでの標的殺傷能力および細胞増殖速度の比較検討が含まれる。
Karpas 299細胞株は、患者由来の未分化大型T細胞リンパ腫である。CD4を発現するMOLT4細胞株は、19歳の患者の末梢血から樹立された急性リンパ芽球性白血病(T-ALL)である。4時間の共培養実験の間に、エフェクター:標的の5:1比率において、CD4IL15RA CAR NK細胞は、CD4CAR NK細胞の殺傷能力(82%、図38)よりもさらに高い割合で、Karpas 299細胞への著しい殺傷能力(95%)を示した。同様に、MOLT4細胞と1:1で共培養した場合、CD4IL15RA CAR NK細胞は、一晩アッセイ(図39)においてCD4CAR NK細胞よりも高い速度(84%〜65%)で標的細胞を溶解した。これらの結果は、CD4IL15 CAR NK細胞が、CD4CAR NK細胞同様に腫瘍細胞をアブレートすることができることを示す。
CD4CARおよびCD4IL15RACAR T細胞のインビボでの抗腫瘍活性を評価し、CD4IL15RA CAR T細胞がCD4CAR T細胞に対して、より持続性が増加する可能性を検討するため、本発明者らは、致死以下に照射したNSGマウスに 100%CD4である急性骨髄性白血病細胞株(M5)であるルシフェラーゼ発現MOLM13細胞を静脈注射することで測定可能な腫瘍形成を可能とする異種マウスモデルを開発した。腫瘍細胞注射の3日後、6匹のマウスに、8×10 6個のCD4CAR、CD4IL15RACAR T細胞またはベクターコントロールT細胞を1コースで静脈注射した。3、6、9および11日目に、マウスにRediJect D-Luciferin(Perkin Elmer)を皮下注射し、腫瘍量を IVISイメージングにて測定した(図40)。6日目において、CD4CAR T細胞での治療マウスは、コントロールと比較して腫瘍量が52%低かったが、一方、CD4IL15RA CAR T細胞での治療マウスは腫瘍負荷量が74%低かった(図41)。11日目に、ほとんどすべての腫瘍細胞がこれらの群の両方において溶解された。Unpaired T test解析では、コントロールと比較して、CD4CARおよびCD4IL15RACAR T細胞での治療グループは、9日目までで光強度が弱く、より少ない腫瘍量であり、非常に有意な統計差(P = 0.0045)を示した。
HEK293FT細胞に、SFFV、EF1またはCAGプロモーターの下でGFPを発現するレンチウイルスプラスミドでトランスフェクトした。トランスフェクションの約60時間後、上清をそれぞれから回収した。相対的ウイルス力価は、最初に、3つのプロモーターのそれぞれからの上清をHEK293細胞に導入することによって決定した。HEK-293FT細胞から得られた、それぞれ3つのGFPウイルス上清を用いてHEK-293細胞に形質導入した。ポリブレンを4μL/ mLまで添加した。16時間後に培地を交換し、ウイルス上清またはポリブレンを含まない培地と交換した。形質導入の3日後、細胞を回収、洗浄し、2%ホルマリン中に懸濁し、GFP発現(FITC)の状態をフローサイトメトリーにより分析した。GFP発現は各試料で見られたが、SFFVプロモーターを用いて作製したウイルスで形質導入した細胞で最も高かった。
本発明者らの各上清中のウイルスベクター粒子の機能的力価を決定するために、HEK293細胞に、12ウェル組織培養処理プレートの1ウェルあたり30μL(低)、125μL(中)または500μL(高)のいずれかの量で、EF1-GFPまたはSFFV-GFPウイルス上清のいずれかで形質導入した。翌朝、培地を10%FBSを含む DMEMに変更した。
初代T細胞におけるプロモーター形質導入効率および表面発現の持続性を決定するために、活性化された臍帯血バフィーコートT細胞を、レトロネクチン(Clontech)でプレコートした12ウェルの組織培養プレート中で50μLのSFFV-GFPまたは1mLのEF1-GFP ウイルス上清のいずれかを用い形質導入した。2度にわたるオーバーナイトでの形質導入の後、細胞を300 IU/mLのIL-2(Peprotech)を含むT細胞培養培地で培養し、1.0〜4.0×106/mLの細胞密度を維持した。細胞を洗浄し、ホルマリン中に懸濁させ、形質導入後7、14、21および28日におけるGFP +細胞の割合をFITCチャンネルを用いてフローサイトメトリー分析を行うことで決定した。GFP +細胞のパーセンテージは、SFFV-GFPで形質導入されたT細胞の方がEF1-GFP形質導入されたT細胞(図44A)と比較して一貫して高かった。さらなる比較により、より多い量(1mL)のEF1-GFP上清で形質導入されたT細胞が、より少ない量(50μLまたは20倍少ない)のSFFV-GFP上清で形質導入された細胞よりも、7日目と28日目の間で、GFP+の細胞の割合が60%超から40%未満まで減少した(図44B)。このことは、SFFVプロモーターを用いた形質導入が、形質導入された細胞の形質発現のより良い持続性を維持することを示唆している。
BCMAまたはTACIまたはBAFF-R NK細胞またはT細胞の少なくとも1つによるCARによる細胞毒性能力を評価するために、BCMAまたはTACIまたはBAFF-Rの少なくとも1つを発現する細胞株または初代ヒト細胞を用いて共培養アッセイを行う。前述のCAR NK細胞またはT細胞が標的細胞を溶解する能力をフローサイトメトリー分析によって定量し、標的細胞はCytotracker色素(CMTMR)で染色した。溶解は、24時間の長い培養時間で観察される。
CAR中のキメラ抗原受容体は、BCMAまたはTACIまたはBAFF-Rのリガンドである。

(i.) 第1の抗原認識ドメイン、第1のシグナルペプチド、第1のヒンジ領域、第1の膜貫通ドメイン、第1の共刺激ドメイン、および第1のシグナル伝達ドメインをからなる第1のキメラ抗原レセプターポリペプチド、そして

- 請求項1〜15および29〜34のいずれかにおいて記載の操作細胞を、それを必要とする患者に投与する、第1の抗原認識ドメインの標的がCD33を含み、第2の抗原認識ドメインの標的はCD123を含む、慢性骨髄性白血病を治療する方法。
- 請求項1〜15において記載の操作細胞を、それを必要とする患者に投与する、第1の抗原認識ドメインの標的がCD19を含み、第2の抗原認識ドメインの標的はCD123を含む、B細胞急性リンパ芽球性白血病(B-ALL)を治療する方法。
- 請求項1〜6において記載の操作細胞を、それを必要とする患者に投与する、第1の抗原認識ドメインの標的がCD38、CS1、BCMA そしてCD38のグループから選ばれ、第2の抗原認識ドメインの標的はCD38、CS1、BCMA そしてCD38のグループから選ばれることを含む、多発性骨髄腫を治療する方法。
- 請求項1〜15のいずれかにおいての記載の方法にて、それを必要とする患者に、第1の抗原認識ドメインの標的がBCMA、TAC1、CS1、およびBAFF-Rの中から選ばれ、第2の抗原認識ドメインの標的は、BCMA、TAC1、CS1、およびBAFF-Rからなるグループから選択される、細胞増殖性疾患を治療する方法。
- 細胞増殖性疾患が、リンパ腫、白血病および形質細胞新生物からなるグループから選択される、請求項41に記載の細胞増殖性疾患の治療方法。
- 形質細胞新生物が、形質細胞白血病、多発性骨髄腫、形質細胞腫、アミロイドーシス、waldestromのマクログロブリン腫、重鎖疾患、孤立性骨芽細胞腫、不確定性のモノクローナル性ガンマパシー(MGUS) 、および多発性骨髄腫のくすぶりを含む中から選ばれ、請求項42に記載の細胞増殖性疾患を処置する方法。

- 細胞増殖性疾患を治療する方法であって、それを必要とする患者に、インターロイキン6受容体、NY-ESO-1、アルファフェトプロテイン(AFP)、グリピカン-3(GPC3)、BCMA、BAFF-R、TACI、LeY、CD5、CD13、CD14、CD15、CD45、CD19、CD20、CD22、CD33 、CD41、CD61、CD64、CD68、CD117、CD123、CD138、CD267、CD269、CD38、Flt3受容体、およびCAR増強剤を含む、これらグループから選択される標的に対して選択的な抗原認識ドメインを有するキメラ抗原レセプターポリペプチドを含む操作された細胞を投与することを含む方法。
- 前述のCAR増強剤が、免疫チェックポイント経路を標的とする薬剤、コロニー刺激因子-1受容体(CSF1R)、PD-1、PD-L1、IL-2、IL-12、IL-15、CSF1R、CTAL-4、TIM-3、およびTGFRベータからなるグループより選択される、請求項45に記載の細胞増殖性疾患の治療方法。
- 請求項13〜23のいずれかにおいて記載の操作されたポリペプチドを含む操作された細胞を、それを必要とする患者に投与することを含む、細胞増殖性疾患を治療する方法。
- 操作されたキメラ抗原レセプターポリペプチドであって、シグナルペプチド、CD45抗原認識ドメイン、ヒンジ領域、膜貫通ドメイン、少なくとも1つの共刺激ドメイン、およびシグナル伝達ドメインを含むポリペプチド。

- ポリヌクレオチドがベクター中に存在する、請求項55に記載の操作されたポリヌクレオチド。
- 請求項55〜56のいずれかにおいて記載のポリヌクレオチドを含む操作された細胞。
- 請求項48〜54のいずれかにおいて記載のポリペプチドを含む操作された細胞。
- 細胞がT細胞またはナチュラルキラー細胞である、請求項57~58のいずれかにおいて記載の操作された細胞。
- T細胞がCD4 T細胞またはCD8 T細胞である、請求項59に記載の操作された細胞。
- ナチュラルキラー細胞がNKT細胞またはNK-92細胞である、請求項59に記載の操作された細胞。
- 標的細胞の数を減少させる方法であって、以下の方法に含まれる、
i. 請求項1〜14、29〜34、および57〜61のいずれかにおいて記載の、有効量の操作細胞と前述での標的細胞を接触させること、そして
ii. 場合により、前述の細胞の数の減少をアッセイすること、標的細胞が、インターロイキン6受容体、NY-ESO-1、アルファフェトプロテイン(AFP)、グリピカン-3(GPC3)、BCMA、BAFF-R、TACI、LeY、CD5、CD13、CD14、CD15、CD19、CD20、CD22、CD33、CD41、CD45、CD61、CD64、CD68、CD117、CD123、CD138、CD267、CD269、CD38、Flt3レセプター、およびCS1からなるグループから選択される少なくとも1つの細胞表面抗原を含む、操作された細胞。 - 第1の抗原認識ドメインの標的がBCMAを含み、第2の抗原認識ドメインはCS1を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD19を含み、第2の抗原認識ドメインの標的がBCMAを含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD19を含み、第2の抗原認識ドメインの標的がCD22を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD19を含み、第2の抗原認識ドメインの標的がCD20を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD19を含み、第2の抗原認識ドメインの標的がCD123を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD33を含み、第2の抗原認識ドメインの標的がCD123を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD269を含み、第2の抗原認識ドメインの標的がCS1を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の抗原認識ドメインの標的がCD4を含み、エンハンサーはIL15RAを含む、請求項29〜34のいずれかにおいて記載の操作された細胞。
- TAC1抗原認識ドメインがAPRILリガンドまたはBAFFリガンドまたはその一部を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- BCMA抗原認識ドメインがAPRILリガンドまたはBAFFリガンドまたはその一部を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- BAFF-R抗原認識ドメインがBAFFリガンドまたはその一部を含む、請求項1〜6のいずれかにおいて記載の操作された細胞。
- 第1の共刺激ドメインおよび第2の共刺激ドメインが同じであることを含む、請求項1〜6及び11のいずれかにおいて記載の操作された細胞。
- 第1の共刺激ドメインおよび第2の共刺激ドメインが4-1BB共刺激ドメイン含む、請求項1〜6及び11のいずれかにおいて記載の操作された細胞。
