Claims (15)
1. Automatizirani multimodularni instrument za obradu stanica, naznačen time, da obuhvaća:
kućište (100) koje je konfigurirano za smještaj svih ili nekih modula;
spremnik (626a) koji je konfiguriran za prihvat stanica;
jedan ili više spremnika (626b) koji su konfigurirani za prihvat nukleinskih kiselina;
transformacijski modul (110c) koji je konfiguriran za uvođenje nukleinskih kiselina u stanice;
modul za oporavak (11a, 110b) koji je konfiguriran tako da prihvaća stanice iz transformacijskog modula i da omogući oporavak stanicama nakon transformacije stanica u transformacijskom modulu;
modul za uređivanje uz pomoć nukleaze (922) koji je konfiguriran tako da omogućava uvedenim nukleinskim kiselinama da uređuju nukleinske kiseline u stanicama; i
procesor (126) koji je konfiguriran za upravljanje automatiziranim multimodularnim instrumentom za obradu stanica, na temelju korisničkog unosa i/ili odabira predprogramirane skriptne naredbe;
sustav robotske manipulacije (108) za pomicanje materijala prema ili između modula; i koji sadrži automatizirani sustav za manipulaciju s tekućinom (120) za pomicanje tekućina izravno iz jednog od modula za oporavak, transformacijskog modula ili modula za uređivanje uz pomoć nukleaze, prema drugom od modula za oporavak, transformacijskog modula ili modula za uređivanje uz pomoć nukleaze, bez intervencije korisnika.
2. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da nukleinske kiseline u jednom ili više spremnika sadrže okosnicu i kazetu za obradu, i automatizirani multimodularni instrument za obradu stanica nadalje obuhvaća modul za asembliranje nukleinske kiseline.
3. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da automatizirani sustav za manipulaciju s tekućinom sadrži usisni uređaj (518) ili pipetu.
4. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 2, naznačen time, da je modul za asembliranje nukleinske kiseline (400) konfiguriran da izvršava asembliranje izotermalne nukleinske kiseline.
5. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da su modul za uređivanje i modul za oporavak sastavljeni u jednostruki modul.
6. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da nadalje obuhvaća modul za rast (850) koji je konfiguriran za rast stanica, opcionalno gdje modul za rast mjeri optičku gustoću rastućih stanica, opcionalno gdje je modul za rast konfiguriran da neprekidno mjeri optičku gustoću rastućih stanica.
7. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 6, naznačen time, da je procesor konfiguriran za prilagodbu uvjeta rasta u modulu za rast tako da stanice dosegnu ciljanu optičku gustoću u vremenskom periodu koji je odredio korisnik.
8. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da su spremnik koji je konfiguriran za prihvat stanica i jedan ili više spremnika koji su konfigurirani za prihvat nukleinskih kiselina, sadržani unutar patrone za reagense (104), opcionalno gdje se neki ili svi reagensi koji su potrebni za obradu stanica, prihvaćaju putem patrone za reagense.
9. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 8, naznačen time, da se reagensi koji su sadržani unutar patrone za reagense, mogu locirati preko skriptne naredbe očitane putem procesora.

automatizirani sustav manipulacije s tekućinom pomiče tekućine izravno iz jednoga od modula za asembliranje nukleinskih kiselina, transformacijskog modula ili modula za uređivanje uz pomoć nukleaze, prema drugom od modula za asembliranje nukleinskih kiselina, transformacijskog modula ili modula za uređivanje uz pomoć nukleaze, bez intervencije korisnika.
14. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da nadalje obuhvaća najmanje jednu patronu za reagense koja sadrži reagense u svrhu izvođenja obrade stanica u automatiziranom multimodularnom instrumentu za obradu stanica, opcionalno gdje su spremnici za stanice i nukleinske kiseline položeni unutar patrone za reagense.
15. Automatizirani multimodularni instrument za obradu stanica prema patentnom zahtjevu 1, naznačen time, da nadalje obuhvaća:
modul za asembliranje nukleinske kiseline koji je konfiguriran tako da
a) asemblira okosnicu i kazetu za obradu, i b) desalinizira asemblirane nukleinske kiseline nakon asembliranja;
modul za rast koji je konfiguriran za rast stanica;
filtracijski modul koji je konfiguriran tako da koncentrira stanice i da učini stanice elektrokompetentnima; i
kombinaciju modula za oporavak i modula za uređivanje uz pomoć nukleaze, koja je konfigurirana tako da omogući oporavak stanicama nakon elektroporacije u transformacijskom modulu i da omogući nukleinskim kiselinama da obrade stanice;
i time, da:
transformacijski modul sadrži protočni elektroporator za uvođenje asembliranih nukleinskih kiselina u stanice; i
automatizirani sustav manipulacije s tekućinom pomiče tekućine izravno iz jednoga od modula za rast, filtracijskog modula, transformacijskog modula, ili kombiniranog modula za oporavak i uređivanje uz pomoć nukleaze, prema drugom od modula za rast, filtracijskog modula, transformacijskog modula, ili kombiniranog modula za oporavak i za uređivanje uz pomoć nukleaze, bez intervencije korisnika.
