Abstract
Se proporcionan composiciones y métodos para editar nucleótidos y/o alterar sitios diana en el genoma de una célula. Los métodos y composiciones emplean una construcción de ADN recombinante que comprenden un promotor de Pol-II unido de forma funcional a un polinucleótido que codifica un ARN guía simple, donde el ARN guía puede formar un complejo de ARN guía/endonucleasa Cas, donde el complejo puede unirse a y escindir una secuencia de sitio diana en el genoma de una célula, tal como una célula eucariota.
