本発明の一般的手法は、癌と反応でき破壊できる免疫リンパ球の移入を使用する、癌を有する患者のための治療の開発である。単離された腫瘍浸潤リンパ球は培養液中で多くの数まで成長し、患者内へ注入される。本発明において、少なくとも1種のサイトカインをコードするアデノウイルスベクターがリンパ球の効果を増加させるために利用される。養子細胞治療組成物およびアデノウイルスベクターの個別投与に先立って、しばしば、骨髄破壊的(myeloablating)または非骨髄破壊的(non-myeloablating)プレコンディショニング化学療法および/または放射線療法が行われる。養子細胞療法処置は患者における癌を低減または排除することを意図する(図21)。
ウイルスベクター
本発明に使用される腫瘍退縮性アデノウイルスベクターはヒトまたは動物を治療するのに適した任意のアデノウイルスベクターであってもよい。本発明の一態様において、アデノウイルスベクターはヒトウイルスのベクターであり、Ad5、Ad3およびAd5/3ベクターからなる群から選択されていてもよい。別の態様において、ベクターはAd5またはAd5/3ベクターである。
腫瘍特異的腫瘍退縮性アデノウイルスを生成するための1つの手法は、E1の定常領域2(CR2)に影響を与える24塩基対欠失(D24)を操作することである。野生型において、アデノウイルスCR2は、合成(S)期、すなわちDNA合成または複製期を誘導するための、細胞性Rb腫瘍抑制因子/細胞周期調節タンパク質を結合するのに関与している。pRbとE1Aとの間の相互作用はE1Aタンパク質保存領域の8アミノ酸121〜127を必要とし、このE1Aタンパク質保存領域は本発明において欠失している。本発明のベクターは、Heise C.ら(2000、Nature Med 6、1134−1139)に従ってベクターのアミノ酸122〜129に対応するヌクレオチドの欠失を含む。D24を有するウイルスは、G1−Sチェックポイントを克服することにおける低い能力を有し、この相互作用が必要でない細胞、たとえばRb−p16経路において欠損のある腫瘍細胞においてのみ効果的に複製することが知られており、これは全てではないが大部分のヒト腫瘍を含む。(図17、33および34を参照のこと)
たとえば腫瘍特異的プロモーターによってE1A内因性ウイルスプロモーターを置きかえることも可能である。本発明の特定の態様において、hTERTプロモーターがE1A内因性ウイルスプロモーターの代わりに利用される。
これらの欠失の両方は本発明者らの発明に関して驚くべき利点を提供する。本発明者らは養子移入T細胞に対する腫瘍エピトープの提示のためのHLA/MHCの発現を回復しようと試みているので、gp19kタンパク質の発現は非生産的であり、実際にHLA/MHCの上方制御はgp19kの欠失を必要とする。6.7kに関して、本発明者らの発明の態様はウイルス由来のTNFアルファの産生であり、その抗腫瘍活性の1つは、(形質導入と非形質導入バイスタンダー細胞の両方で)直接的な抗腫瘍アポトーシス促進効果があるので、6.7kの存在は非生産的である。
本発明の別の特定の態様において、腫瘍退縮性アデノウイルスベクターはアデノウイルス血清型3(Ad3)核酸骨格に基づき、以下:E3領域における欠失、およびE3の欠失領域の代わりに導入遺伝子(たとえばCD40L)を発現するための腫瘍特異的プロモーター(たとえばCMVまたはE2F)を含む。本発明の一態様において、アデノウイルスベクターはヒトアデノウイルスに基づく。(図37および38を参照のこと、ウイルスベクターAd3−hTERT−E3del−CMV−CD40LおよびAd3−hTERT−E3del−E2F−CD40Lの対応するヌクレオチド配列は配列番号30および31に示される)
アデノウイルス3における早期領域(E3)タンパク質の正確な機能は知られていない。一般的にアデノウイルスにおいて、それらは、欠失した場合、複製を損なうようには見えず、それらはアデノウイルスに対する抗ウイルス宿主反応に影響を与えるように見える(Woldら、1999)。ヒトアデノウイルスゲノムのE3はヒトに見出される6種(A〜F)のアデノウイルスの間で最高レベルの遺伝的多様性を含有する。遺伝的内容のこの多様性は、遺伝子の種特異的アレイがコードされる、高度に保存されたE3−gp19KとE3−RIDαオープンリーディングフレーム(ORF)との間に主に位置する(Burgert and Blusch、2000)。
癌
本発明の組換えベクターは腫瘍細胞内で複製可能である。本発明の一態様において、ベクターは細胞内で複製可能であり、その細胞はRb経路、具体的にはRb−p16経路を欠損している。これらの欠損細胞には動物およびヒトにおける全ての腫瘍細胞が含まれる。ここに使用される場合、「Rb経路の欠損」は、経路のいずれかの遺伝子またはタンパク質における変異および/または後成的変化を指す。これらの欠損に起因して腫瘍細胞はE2Fを過剰発現するので、通常、有効な複製に必要とされるE1A CR2によるRbの結合は不要である。さらに選択性はE2Fプロモーターによって媒介され、そのE2Fプロモーターは、Rb/p16経路欠損細胞において見られるように、遊離E2Fの存在下でのみ活性化する。遊離E2Fの非存在下ではE1Aの転写は生じず、ウイルスは複製しない。E2Fプロモーターの包含は正常組織内でE1Aの発現を阻止するのに重要であり、その正常組織は、E3プロモーターから導入遺伝子発現を可能にすることにより、直接と間接の両方で毒性を引き起こし得る。
医薬組成物
本発明の医薬組成物は本発明の少なくとも1種類のウイルスベクターを含む。さらに組成物は、少なくとも2種、3種または4種の異なるベクターを含んでいてもよい。ベクターに加えて、医薬組成物はまた、他の治療的に有効な薬剤、任意の他の薬剤、たとえば薬学的に許容される担体、緩衝剤、賦形剤、アジュバント、添加剤、防腐剤、充填剤、安定剤および/もしくは増粘剤、ならびに/または対応する製品に通常見出される任意の構成成分を含んでいてもよい。組成物を製剤化するための好適な成分および適切な製造方法の選択は当業者の一般的知識に属する。
投与
本発明のベクターまたは医薬組成物は、植物、動物およびヒトからなる群から選択される任意の真核生物対象に投与されていてもよい。本発明の特定の態様において、対象はヒトまたは動物である。動物は、ペット、家畜および生産動物からなる群から選択されていてもよい。
材料および方法
B16−OVA動物モデル:オボアルブミンを発現するB16細胞(B16−OVA)を、RPMI、10%FBS、5mg/mlのG418、20mMのL−グルタミン、1×Pen/Strep溶液(GIBCO)中に維持した。4〜7週齢のC57BL/6免疫応答性メスマウスに、50ulのRPMI、0%FBS中の2.5×105B16−OVA細胞を、一匹のマウスにつき1つの腫瘍で右脇腹に皮下移植した。腫瘍生着のおおよそ10日後(腫瘍が注射可能、約3mmの最小直径になったとき)、マウスを群に分け、いくつかの実験において、50ulのPBSまたは50ulのPBS中の腫瘍退縮性アデノウイルスの1×109ウイルス粒子(VP)のいずれかの腫瘍内注射により6連続日で処置した。他の実験において、0、2および4日目に3回注射を与えた。マウス細胞はヒトアデノウイルスに対して非許容性であるので、ウイルス複製により誘導される炎症を模倣するために複数の腫瘍内ウイルス注射を使用した(Blairら、1989)。
本発明を支持する実験
実験1(腫瘍内アデノウイルス注射によって誘導されるサイトカインおよびケモカイン):
アデノウイルス注射がサイトカインおよびケモカイン発現を生じ得るかどうかを調べるために、本発明者らは、皮下B16−OVA腫瘍を保有するマウスに、0、1、2、3、4および5日目に、PBSまたは5/3キメラ腫瘍退縮性アデノウイルスのいずれかを腫瘍内に注射した。1つの治療群につき3匹のマウスから腫瘍を抽出し、0日目(ウイルス注射の前=ベースライン対照)にサイトカイン分析のために処理し、6、10、14および18日目の時点ごとに3匹の他のマウス由来の腫瘍を処理した。
実験2(養子T細胞療法のアデノウイルスにより媒介される増強):
養子T細胞療法のアデノウイルス処置の影響を調べるために、マウスB16−OVA黒色腫を、5/3キメラ腫瘍退縮性アデノウイルス単独、または腫瘍特異的OT−I細胞の養子移入との組合せで処置し、腫瘍内PBS注射を受けているマウスと比較した。図2にまとめた3回の独立した実験の結果は、一方で、ウイルス注射それ自体で(ヒトアデノウイルスはマウス細胞において生産的に複製しないことに留意されたい)、小さい腫瘍の成長制御を生じ、処置後14日まで続き、その後減少したことを表す(図2A)。他方で、処置したマウスに5×105または2×106OT−I細胞を養子移入した場合、PBSとAd群との間に統計的に有意な相違が2つの別の実験において観察された(それぞれ図2Bおよび2C)。
実験3(in vivoでの免疫細胞集団の質および量のアデノウイルスにより媒介される変化):
実験2の養子移入細胞の輸送および増殖を調べるために、OT−I細胞を、5μMのカルボキシフルオレセインスクシンイミジルエステル(CFSE)を用いてex vivoで染色した。この蛍光細胞染色色素を全ての細胞分裂に伴って希釈し、したがって本発明者らは、各細胞分裂(最大7回の分裂、ここでM0−7と標識した)における蛍光信号強度の約1/2のわずかな減少を分析することにより、フローサイトメトリーによってリンパ球増殖を追跡できる。移入の1日後、結果は、腫瘍内で移入OT−I細胞(CD8+ CFSE+二重陽性集団)のウイルスにより誘導される蓄積を示し、血液中のこれらの細胞の低減が同時に起こった(図3A)。その後、また、全CD8+ T細胞数が、PBS注射した腫瘍と比べてウイルス処置した腫瘍において多いように見え、14日目に全CD8+ T細胞数はウイルス処置したマウスのリンパ器官において増加した。

新規ウイルス構築物
以下の新規ウイルス構築物を本発明者らの提案した技術の例として提示する:C5aおよびTNF−α発現腫瘍退縮性ウイルス
本発明者らは、6.7K/gp19遺伝子領域の代わりに導入遺伝子として相補体構成成分C5aまたはヒトTNF−αの活性部分を有する新規腫瘍退縮性Ad5/3アデノウイルスを生成した(図17)。
実験5(C5aコードアデノウイルスベクターからの導入遺伝子発現):
概念の実証として、腫瘍退縮性アデノウイルスが、養子細胞療法を増強させるために提案されている選択されたサイトカインを発現できるかどうかを確認するために、本発明者らは、C5aをコードするアデノウイルスの10VP/細胞にて培地中でヒトA549細胞を感染させ(図24)、感染後の異なる時点にて細胞培養上清中でC5aレベルをELISAによって評価した。実際に結果は、所有しているアデノウイルス構築物が選択されたサイトカインを保有しているという仮定を実証し、支持している。
実験6(新規アデノウイルスベクターによる単球移動に対する効果):
本発明者らは、in vitro走化性アッセイを使用して単球を動員するC5aの能力を試験した:A549細胞を、C5aを発現するアデノウイルスもしくは非武装対照ウイルス(10VP/細胞−同様のウイルス間の感染単位)のいずれかで感染させた、またはPBSで処置し、感染の48時間後に培地を採取し、製造業者の指示書(Millipore QCM、cat.no.ECM512)に従ってトランスウェル走化性アッセイにおいてヒト単球細胞株THP1を動員するために使用した。結果は、非感染細胞または非武装対照ウイルスに感染した細胞由来の培地による以上に、C5a発現ウイルスに感染した細胞由来の上清によって、有意に多い単球の誘引を表す(図25)。
実験7(C5a武装アデノウイルスの抗腫瘍効果):
非細胞溶解性腫瘍感染の状況においてC5aの抗腫瘍有効性を評価するために、本発明者らは、PBS、C5a発現または非武装対照ウイルスのいずれかを用いて、0、2および4日目にC57BL/6マウスにおいて樹立されたB16−OVA腫瘍を処置した。結果はC5a発現ウイルスによる強力な抗腫瘍効果を表す(図26)。
実験8(C5aウイルスによる増加した抗腫瘍T細胞増殖):
C5a発現ウイルスの抗腫瘍効果の観察された増加(図24)がT細胞に関連しているかどうかを評価するために、免疫優勢オボアルブミンペプチドSIINFEKL(ProImmune、USA)が負荷されたMHC Iを認識するTCRに特異的なAPCがコンジュゲートした五量体で染色することによって検出された、オボアルブミン特異的CD8+ T細胞についてフローサイトメトリーによって腫瘍を分析した。実際に、C5aウイルス群における腫瘍は、対照ウイルスまたはPBSを注射した腫瘍より有意に多い割合の腫瘍特異的CD8 T細胞を含有した(図27)。
実験9(TNF−αコードアデノウイルスからの導入遺伝子発現):
C5aウイルス(図24および実験5)と同様に、本発明者らは、最適な導入遺伝子の分泌を媒介する、hTNF−αを発現する腫瘍退縮性アデノウイルスの能力を試験した。結果により、発現が確認される(図18)。
実験10(発現された導入遺伝子の生物学的効果は腫瘍退縮性アデノウイルスにおいて保持される):
アデノウイルス発現導入遺伝子がその生物学的効果を維持するかどうかを評価するために、対照非武装ウイルスまたはTNF−アルファ発現ウイルス(様々なVP/細胞、72時間 p.i.)に感染させたA549細胞由来のウイルスを含まない(100kD濾過した)上清を、TNF−アルファに対して感受性があるWEHI−13VAR(ATCC CRL−2148)細胞上に適用し、これらの細胞を上清への曝露の72時間後に生存について評価した。(たとえばEspevik Tら J Immunol Methods.1986;95(1):99−105が方法を記載している。)結果は、腫瘍退縮性アデノウイルスから発現したTNF−アルファが効力のある生物学的効果を保持することを表す(図19)。
実験11(腫瘍退縮性サイトカイン発現ウイルスはin vitroで細胞殺傷能力を保持する):
TNF−アルファは抗ウイルス効果を有する可能性があるので、TNF−アルファを発現するアデノウイルスの腫瘍退縮効果が、癌細胞に感染し、殺傷する、その能力を保持することを確認することは重要であった。培養物中のいくつかの癌細胞株をTNF−アルファ発現または対照ウイルスに感染させ、製造業者の指示書(Promega、USA)に従って、CelltiterGlo AQ MTSアッセイによって生存について評価した。結果は、ウイルスがin vitroで腫瘍退縮性であることを示す(図19〜20)。
実験12(放射線療法とTNFアルファを発現する腫瘍退縮性ウイルスとの間の相乗効果):
本発明者らは、付随する焦点外部ビーム放射線(XRT)を用いて、または用いずに皮下A549異種移植片を有するヌードマウスを腫瘍内においてウイルス処置した(図21)。RDは複製欠損ウイルスを示し、非武装ウイルスはTNFアルファを有さない腫瘍退縮性ウイルスである。
実験13(免疫応答性宿主内のTNF−アルファ発現アデノウイルスの増加した抗腫瘍効果):
マウス細胞内で複製しない腫瘍退縮性アデノウイルスが依然として、免疫応答性マウスにおいてin vivoで抗腫瘍効果を引き起こすことができるかどうかを試験するために、樹立したB16.OVA腫瘍を有するマウスに、図26と同様にTNF−アルファ発現もしくは非武装対照ウイルスまたはPBSを腫瘍内に注射した。結果は、対照と比較してTNF−アルファ発現ウイルスにより強力な全体の腫瘍制御を示し(図22)、ヒトTNFアルファがマウスにおいて部分的に活性であることを示唆し、強力な抗腫瘍効果を達成する武装ウイルスの考えを支持する。
実験14(TNFα−ウイルスによる増加した抗腫瘍T細胞増殖):
実験11と同様に、本発明者らは、TNF−アルファ発現ウイルスの観察された抗腫瘍効果が、腫瘍特異的細胞溶解性T細胞反応の誘導に関連するかどうかを試験することを望んだ。本発明者らは腫瘍を抽出し、それらを実験11のようにフローサイトメトリー分析のために処理した。結果(図23)により実際に、同様にTNF−アルファ発現が腫瘍部位における腫瘍特異的T細胞の増殖を促進することが確認され、提案された技術を支持する強力な根拠が示される。

クローニング手順:
1.対応する発現カセットを含有するAd3 3’末端プラスミドの構築、このプラスミドはAd3ゲノムの3’ITRを含有し、Ad3ゲノムの29,892〜30,947のE3領域を発現カセットと置きかえた。(注記:本発明者らは、3’末端に近接するAd3ゲノムにおけるEcoRI制限部位を利用する)
2.Ad3 5’末端プラスミドの構築、このプラスミドは5’ITRおよびhTERT−E1を含有する。(注記:本発明者らは、Ad3ゲノムにおける特有の制限部位NotIおよび5’末端に近接するNheI制限部位を利用する)

1.PCRにより、pKBS2−hTERT(Ad3−hTERT−E1Aペーパー由来のプラスミド)由来のAd3ゲノムの5’末端を増幅する、フォワードプライマー5’gtcagtttaaacttaggccggccctatctatataatataccttatagatggaatgg3’(配列番号28)、リバースプライマー5’CTTCATCAGCAGCTAGCAGCATAGAATCAG3’(配列番号29)。pGem−T→pGemT−Ad3−5’末端−hTERT内にクローニングした。
1.プラスミドpWEA−Ad3−hTERT−E2F−は3’末端ゲノムを除去するためにEcoRIを用いて消化し、pWEA−Ad3−3’末端−CMV−、pWEA−Ad3−3’末端−CMV−CD40LおよびpWEA−Ad3−3’末端−E2F−CD40L由来の発現カセットを含有する対応する断片とライゲーションする。
Ad3ウイルスの機能性をin vitroで試験し、その結果を図41に示す。全ての新規ウイルスは機能的であり、腫瘍細胞株に感染できる。
AD3ベクターのin vivoでの結果
全ての動物実験は、ヘルシンキ大学および南フィンランド州政府の動物実験委員会により承認された。マウスをそれらの健康状態について頻繁にモニターし、疼痛または苦痛の兆候に気付くとすぐに安楽死させた。メスのfox chase重症複合免疫不全マウス(Charles River)を使用した。
MTS細胞増殖アッセイ(図42〜44)
1日目に、105細胞/ウェル(A549、PC3−MM2またはSKOV3−luc)を、5%のFBSを含有した100μlの成長培地(GM)入りの96ウェルプレート内に播種した。2日目に、5%のFBSを含有するGMを用いて単層を1回洗浄した。次いで細胞を、100、10、1、0.1および0ウイルス粒子/細胞の用量にて異なるウイルスに感染させた。その後、細胞を振盪機械上で1時間インキュベートし、次いでGMで洗浄した。新たに5%GMを加えた後、細胞をインキュベータに残し、GMを4日ごとに置きかえた。試験したウイルスの1つの細胞変性効果が最高濃度で100%に到達した後、mts試薬(Promega)を加えることによって試験を終了した。インキュベーションの2時間後、吸光度を490nmフィルターにて測定した。次いでバックグラウンドを減算し、結果を分析した。
免疫応答性マウスにおいてマウスCD40Lをコードする腫瘍退縮性アデノウイルスの治療域
免疫応答性動物において、ウイルスゲノムは、静脈注射後、腫瘍内に存在する(図46)。アルビノC57マウスにマウスB16−ova細胞を皮下接種し、マウスCD40Lをコードする5つの異なるウイルス用量のAd5に基づいたウイルスを用いて静脈内で処置した(実験4および15を参照のこと)。1群当たり3匹の動物の腫瘍を採取し、−80℃にて保存した。全DNAを抽出し、ウイルスDNA負荷を定量的PCRを用いて調べた。ウイルスE4コピー数を、マウスB−アクチンプライマーを用いてゲノムDNAに正規化した。図46において、各記号は1つの腫瘍を表し、水平線は群の中央値を示す。偽:n=5;用量5:n=4;用量4:n=4;用量3:n=4;用量2:n=6;用量1:n=2;用量2 i.t.:n=4。用量5:1×1011VP/マウス;用量4:3×1010VP/マウス;用量3:1×1010VP/マウス;用量2:1×109VP/マウス;用量1:1×108VP/マウス;陽性対照(用量2腫瘍内。)
用量5に関して、マウスの67%は肝臓毒性の兆候を有した。用量4は、静脈送達後、肝臓毒性の兆候なしに良好な腫瘍形質導入を達成できた。

[24] E3領域における欠失およびE3の欠失領域の代わりに導入遺伝子を発現するための腫瘍特異的プロモーターを含む、血清型3(Ad3)腫瘍退縮性アデノウイルスベクター。
[25] 前記サイトカインが[13]に記載のサイトカイン群から選択される、[24]に記載の腫瘍退縮性アデノウイルスベクター。
[26] 前記腫瘍退縮性アデノウイルスベクターが2種以上のサイトカインをコードする、[23]〜[25]の何れか1項に記載の腫瘍退縮性アデノウイルスベクター。
[27] 前記腫瘍退縮性アデノウイルスベクターが、[15]に記載の第1のサイトカイン群から選択される任意の2種以上のサイトカインをコードする、または前記腫瘍退縮性アデノウイルスベクターが、IL−2と[15]に記載の第2のサイトカイン群から選択される1種のサイトカインまたは複数種のサイトカインとをコードする、[26]に記載の腫瘍退縮性アデノウイルスベクター。

[32] 前記方法または使用が、同時もしくは連続する放射線療法、モノクローナル抗体、化学療法または他の抗癌薬もしくは介入の対象への適用をさらに含む、[1]〜[17]、[20]または[30]〜[31]の何れか1項に記載の方法または使用。
Claims (32)
- 対象における癌を治療する方法であって、養子細胞治療組成物および少なくとも1種のサイトカインをコードする腫瘍退縮性アデノウイルスベクターの対象への個別投与を含む、方法。
- 癌の治療に使用するための別個の養子細胞治療組成物と併用される少なくとも1種のサイトカインをコードする、腫瘍退縮性アデノウイルスベクター。
- 前記養子細胞治療組成物が、腫瘍浸潤リンパ球(TIL)、T細胞受容体改変リンパ球およびキメラ抗原受容体改変リンパ球からなる群から選択される細胞型を含む、請求項1〜2の何れか1項に記載の方法または使用。
- 前記養子細胞治療組成物が、T細胞、CD8+細胞、CD4+細胞、NK細胞、デルタ−ガンマT細胞、調節性T細胞および末梢血単核細胞からなる群から選択される細胞型を含む、請求項1〜3の何れか1項に記載の方法または使用。
- 前記養子細胞治療組成物がT細胞を含む、請求項1〜4の何れか1項に記載の方法または使用。

- 前記対象がヒトまたは動物である、請求項1〜6の何れか1項に記載の方法または使用。
- 養子細胞治療組成物および少なくとも1種のサイトカインをコードする腫瘍退縮性ウイルスベクターの対象への投与を、同時にまたは任意の順序で連続して行う、請求項1〜7の何れか1項に記載の方法または使用。
- 養子細胞治療組成物および少なくとも1種のサイトカインをコードする腫瘍退縮性ウイルスベクターの連続投与の間隔が、1分から4週間の期間である、ならびに/または養子細胞治療組成物および腫瘍退縮性ウイルスベクターを複数回投与する、請求項8に記載の方法または使用。
- 前記養子細胞治療組成物および/または少なくとも1種のサイトカインをコードする腫瘍退縮性ウイルスベクターの投与を、腫瘍内、動脈内、静脈内、胸膜内、膀胱内、腔内もしくは腹膜注射、または経口投与により行う、請求項1〜9の何れか1項に記載の方法または使用。
- 前記アデノウイルスベクターが、Ad5、Ad3またはAd5/3から選択される、請求項1〜10の何れか1項に記載の方法または使用。
- 前記アデノウイルスベクターが、Ad5核酸骨格およびAd3繊維ノブまたはAd5/3キメラ繊維ノブを含むAd5/3である、請求項11に記載の方法または使用。

- 前記養子細胞治療組成物および腫瘍退縮性アデノウイルスベクターを、同時にまたは任意の順序で連続して対象に投与する、請求項1〜17の何れか1項に記載の方法または使用。
- 対象における養子細胞療法またはT細胞療法の効果を増加させる際に使用するための腫瘍退縮性アデノウイルスベクター。
- 腫瘍退縮性アデノウイルスベクターを、それを必要とする対象に投与することによって、対象における養子細胞療法またはT細胞療法の効果を増加させる方法。
- 養子細胞治療組成物および少なくとも1種のサイトカインをコードする腫瘍退縮性アデノウイルスベクターを含む医薬キットであって、前記養子細胞治療組成物が第1の製剤に製剤化され、少なくとも1種のサイトカインをコードする前記腫瘍退縮性アデノウイルスベクターが第2の製剤に製剤化される、医薬キット。
- 前記第1および第2の製剤が同時にまたは任意の順序で連続して対象に投与されるためのものである、請求項21に記載の医薬キット。
- 1)5/3キメラ繊維ノブを含むアデノウイルス血清型5(Ad5)核酸骨格、

- 前記腫瘍退縮性アデノウイルスベクターが、請求項15に記載の第1のサイトカイン群から選択される任意の2種以上のサイトカインをコードする、または前記腫瘍退縮性アデノウイルスベクターが、IL−2と請求項15に記載の第2のサイトカイン群から選択される1種のサイトカインまたは複数種のサイトカインとをコードする、請求項26に記載の腫瘍退縮性アデノウイルスベクター。
