Abstract
Description
本発明は、病的細胞と免疫細胞の両方に共通して見られる抗原マーカー(例えば、CD38)を標的とするキメラ抗原受容体を備える、免疫療法のための遺伝子操作された、好ましくは、非アロ反応性の免疫細胞を開発する方法に関する。
エクスビボで生成された自己の抗原特異的免疫細胞の移入を含む養子免疫治療は、ウイルス感染および癌を処置するための有望な戦略である。養子免疫治療のために使用されるT細胞は、例えば、抗原特異的T細胞の増大または遺伝子改変を通したT細胞の向け直しのいずれかによって生成され得る(Park,Rosenberg et al. 2011)。

本明細書において特に定義されない限り、使用される技術用語および科学用語は、全て、遺伝子治療、生化学、遺伝学、および分子生物学の領域の当業者によって一般的に理解されるのと同一の意味を有する。
(a)免疫細胞において、抗原マーカーの発現もしくは提示に関与する遺伝子を遺伝子不活化するかまたは変異させる工程であって、前記抗原マーカーが前記T細胞の表面と病的細胞の表面の両方に存在することが知られている、工程、;
(b)前記免疫細胞の中で、前記病的細胞の表面に存在する前記抗原マーカーに対して向けられたキメラ抗原受容体をコードするトランスジーンを発現させる工程
を含む。
Cas9はCsn1(COG3513)とも呼ばれ、crRNAの生合成および侵入DNAの破壊の両方に関与する大きなタンパク質である。Cas9は、S.サーモフィラス(S.thermophiles)、リステリア・イノクア(Listeria innocua)(Gasiunas, Barrangou et al. 2012;Jinek, Chylinski et al. 2012)、およびS.ピヨゲネス(S. Pyogenes)(Deltcheva, Chylinski et al. 2011)などの様々な細菌種において述べられている。この大きなCas9タンパク質(>1200アミノ酸)は、タンパク質の中央にある2つの推定ヌクレアーゼドメイン、すなわち、HNH(McrA様)ヌクレアーゼドメインと、分割されたRuvC様ヌクレアーゼドメイン(RNアーゼHフォールド(RNase H fold))(Makarova, Grishin et al. 2006)を含有する。
RuvCドメインおよびHNHドメインの以前の特徴付けに促されて、本発明者らはスプリットCas9タンパク質を作り出すようにCas9タンパク質を操作した。驚いたことに、本発明者らは、これらの2つのスプリットCas9が一緒になって、または別々に、核酸標的を処理できることを示した。この観察から、スプリットCas9タンパク質を用いて新たなCas9系を開発することが可能になる。それぞれのスプリットCas9ドメインは別々に調製および使用することができる。従って、このスプリット系は、さらに短い、および/または不活性のタンパク質の送達を可能にする、RNAガイドエンドヌクレアーゼをT細胞に向けかつ送達するための、いくつかの利点を示し、かつ望ましい時間でT細胞においてゲノム操作を誘導するのに特に適しており、従って、組み込まれたCas9ヌクレアーゼの潜在的な毒性を制限する。
レアカットエンドヌクレアーゼは、メガヌクレアーゼの名前でも知られるホーミングエンドヌクレアーゼでもよい。このようなホーミングエンドヌクレアーゼは当業者に周知である(Stoddard 2005)。ホーミングエンドヌクレアーゼは高度に特異的であり、長さが12〜45塩基対(bp)、通常、長さが14〜40bpのDNA標的部位を認識する。本発明によるホーミングエンドヌクレアーゼは、例えば、LAGLIDADGエンドヌクレアーゼ、HNHエンドヌクレアーゼ、またはGIY-YIGエンドヌクレアーゼに対応してもよい。本発明による好ましいホーミングエンドヌクレアーゼはI-CreI変種でもよい。「変種」エンドヌクレアーゼ、すなわち、自然界で天然には存在せず、遺伝子工学またはランダム変異誘発によって得られるエンドヌクレアーゼは、野生型エンドヌクレアーゼが認識するDNA配列とは異なるDNA配列に結合することができる(国際出願WO2006/097854を参照されたい)。
当初、インビトロでDNAを切断するために開発された「ジンクフィンガーヌクレアーゼ」(ZFN)は、IIS型制限酵素であるFokIの切断ドメインと、3つ以上のC2H2ジンクフィンガーモチーフを含有するDNA認識ドメインとの融合である。2つのZFNそれぞれが正確な方向および間隔でDNAの特定の位置でヘテロ二量体化すると、DNAの二本鎖切断(DSB)が起こる。このようなキメラエンドヌクレアーゼの使用は、Urnov et al.(Genome editing with engineered zinc finger nucleases(2010) Nature reviews Genetics 11:636-646)により概説されるように当技術分野において広範囲にわたて報告されている。
「TALE-ヌクレアーゼ」または「MBBBD-ヌクレアーゼ」とは、典型的には転写アクチベーター様エフェクタータンパク質(TALE)に由来するDNA結合ドメインまたはモジュラー塩基対塩基結合ドメイン(MBBBD)と、エンドヌクレアーゼ活性を有する触媒ドメインとの融合に起因する操作されたタンパク質を指す。このような触媒ドメインは、通常、例えばI-TevI、ColE7、NucA、およびFok-Iなどの酵素に由来する。TALE-ヌクレアーゼは、選択された触媒ドメインに応じて単量体または二量体の形で形成することができる(WO2012138927)。このような操作されたTALE-ヌクレアーゼは、商品名TALEN(商標)(Cellectis, 8 rue de la Croix Jarry, 75013 Paris, France)で市販されている。
本発明者らは、エンドヌクレアーゼを発現するポリヌクレオチド、この可能性のあるコエフェクター(例えば、Cas9またはアルゴノートヌクレアーゼに結合するガイドRNAまたはガイドDNA)、ならびにキメラ抗原受容体を細胞内に、または前記細胞の細胞下区画に送達するために当技術分野において公知の、あらゆる手段を考えた。これらの手段には、非限定的な例として、ウイルス形質導入、エレクトロポレーションが含まれ、リポソーム送達手段、ポリマー担体、化学的担体、リポプレックス、ポリプレックス、デンドリマー、ナノ粒子、エマルジョン、天然のエンドサイトーシスまたはファゴサイトース(phagocytose)経路も含まれる。

本発明によれば、キメラ抗原受容体をT細胞に入れて発現させるには、レトロウイルスベクター、より好ましくはレンチウイルスベクターの使用が特に適している。ウイルス形質導入のための方法は当技術分野において周知である(Walther et al.(2000) Viral Vectors for Gene Transfer. Drugs. 60(2):249-271)。組込みウイルスベクターを用いると、T細胞ゲノムにポリヌクレオチドを安定して組み込み、長期間にわたってキメラ抗原受容体を発現させることが可能になる。
本発明の方法は、遺伝子療法の一環として、例えば、血液循環中で、特異的レアカットエンドヌクレアーゼを発現する遺伝子配列と、CARを発現する他の遺伝子配列を含む、T細胞を標的とするウイルスベクターを用いることによってインビボで行うことができるが、本発明の方法は、さらに一般的には、患者またはドナーから入手可能な培養T細胞に対してエクスビボで実施されることが意図される。エクスビボで操作された、操作されたT細胞は、自家処置の一環として、これが得られた患者に再移植されてもよく、同種異系処置の一環として用いられてもよい。この後者の場合では、細胞が確実に正しく生着するように非アロ反応性になるように細胞をさらに操作することが好ましい。従って、本発明の方法は、ドナーからT細胞を獲得し、例えば、WO2013/176915に記載のように、MHC認識に関与する、および/または免疫抑制薬物の標的であるT細胞遺伝子を不活化する、さらなる工程を含んでもよい。
癌療法および同種異系T細胞の選択的生着を改善するために、操作されたT細胞に薬物耐性を付与して、操作されたT細胞を化学療法または免疫抑制剤の毒性副作用から保護することができる。実際に、本発明者らは、ほとんどの患者が、T細胞免疫療法を受ける前に治療の標準として化学療法および免疫枯渇剤(immune depleting agent)による処置を受けたことを観察した。本発明者らは、処置前に細胞をエクスビボで増殖させるために培養培地中に化学療法薬物を添加すること、または化学療法もしくは免疫抑制処置を受けている患者においてインビボで操作されたT細胞の選択的増殖物を得ることのいずれかによって、これらの処理を利用して操作されたT細胞の選択を助けることができることも発見した。
(a)T細胞を準備する工程、
(b)少なくとも1種類の薬物を選択する行程、
(c)薬物耐性を前記T細胞に付与するようにT細胞を改変する工程、
(d)前記操作されたT細胞を前記薬物の存在下で増殖させる工程
を含む、免疫療法のために同種異系かつ薬物耐性のT細胞を操作する方法に関し、任意で、前述の工程は、以前に述べたように、本方法の工程と組み合わせてもよい。
免疫抑制剤は、いくつかの作用機序の1つによって免疫機能を抑制する薬剤である。言い換えると、免疫抑制剤は、免疫応答の程度および/または貪食を減らす能力によって示される化合物が果たす役割である。非限定的な例として、免疫抑制剤は、カルシニューリン阻害剤、ラパマイシン標的、インターロイキン-2α鎖遮断薬、イノシン一リン酸デヒドロゲナーゼ阻害剤、ジヒドロ葉酸還元酵素阻害剤、コルチコステロイド、または免疫抑制性代謝拮抗物質でもよい。古典的な細胞傷害性免疫抑制剤はDNA合成を阻害することによって作用する。その他の免疫抑制剤はT細胞を活性化することによって作用してもよく、ヘルパー細胞の活性化を阻害することによって作用してもよい。本発明による方法を用いると、T細胞内にある免疫抑制剤の標的を不活性化することによって、免疫療法のためにT細胞に免疫抑制耐性を付与することが可能になる。非限定的な例として、免疫抑制剤の標的は、免疫抑制剤の受容体、例えば、CD52、グルココルチコイド受容体(GR)、FKBPファミリー遺伝子メンバー、およびシクロフィリンファミリー遺伝子メンバーでもよい。
本発明によれば、T細胞は、炎症性Tリンパ球、細胞傷害性Tリンパ球、制御性Tリンパ球、またはヘルパーTリンパ球からなる群より選択することができる。別の態様において、前記細胞はCD4+Tリンパ球およびCD8+Tリンパ球からなる群に由来してもよい。前記細胞は血液から抽出されてもよく、幹細胞に由来してもよい。幹細胞は成人幹細胞、胚性幹細胞、さらに具体的には、非ヒト幹細胞、臍帯血幹細胞、前駆細胞、骨髄幹細胞、人工多能性幹細胞、全能性幹細胞、または造血幹細胞でもよい。代表的なヒト細胞はCD34+細胞である。本発明の細胞の増殖および遺伝子組換えの前に、様々な非限定的な方法によって対象から細胞供給源を得ることができる。T細胞は、末梢血単核球、骨髄、リンパ節組織、臍帯血、胸腺組織、感染部位に由来する組織、腹水、胸水、脾臓組織、および腫瘍を含む多数の非限定的な供給源から得ることができる。本発明のある特定の態様では、入手可能であり、かつ当業者に公知の任意の数のT細胞株を使用することができる。別の態様において、前記細胞は健常ドナーから得られてもよく、癌と診断された患者または感染症と診断された患者から得られてもよい。別の態様において、前記細胞は、異なる表現型特徴を示す細胞の混合集団の一部である。本発明の範囲には、以前に述べられた方法による形質転換T細胞から得られた細胞株も包含される。

本発明に従ってT細胞に導入されるキメラ抗原受容体は、単鎖CARまたは多鎖CARなどの異なる設計を採用することができる。これらの異なる設計を用いると、標的となった病的細胞に対する特異性および結合効率を改善するための様々な戦略が可能になる。これらの戦略の一部を本願の図に示した。単鎖CARは、当技術分野における最も古典的なバージョンである。特異性および強度の点でT細胞の活性の調整が可能になるので、多鎖CAR構造が出願人によって開発された。多サブユニットは、さらなる共刺激ドメインを保護することできるか、またはこのようなドメインを遠ざけることができ、同様に、他のタイプの受容体も保護することできるか、または遠ざけることができるのに対して、古典的な単鎖構造は、時として、多重特異性相互作用に対して感度が高すぎる、および許容性が低すぎるとみなされることがある。
養子免疫療法は、エクスビボで作製された抗原特異的自己T細胞の導入を伴い、ウイルス感染症および癌を処置するのに有望な戦略である。養子免疫療法に用いられるT細胞は抗原特異的T細胞の増殖によって作製されてもよく、遺伝子工学によるT細胞の再誘導によって作製されてもよい(Park, Rosenberg et al. 2011)。ウイルス抗原特異的T細胞の導入は、移植に関連するウイルス感染症および稀なウイルス関連悪性疾患の処置に用いられる十分に確立した手順である。同様に、腫瘍特異的T細胞の単離および導入は黒色腫の処置においてうまくいくことが示されている。
T細胞に導入される先行技術のキメラ抗原受容体は、シグナル伝達ドメインの連続付加を必要とする単鎖ポリペプチドから形成された。しかしながら、シグナル伝達ドメインがその自然の膜近傍位置から動くと、その機能が妨げられる可能性がある。この欠点を克服するために、出願人は、最近、全ての関係するシグナル伝達ドメインの正常な膜近傍位置を可能にするために、FcεRIに由来する多鎖CARを設計した。この新たな構造では、FcεRIα鎖の高親和性IgE結合ドメインは、T細胞特異性を細胞標的に再誘導するためにscFvなどの細胞外リガンド結合ドメインと交換され、共刺激シグナルを正常な膜近傍位置に配置するためにFcεRIβ鎖のN末端テールおよび/またはC末端テールが用いられる。

本発明による方法は、一般的に、T細胞を活性化および/または増殖させる、さらなる工程を含む。これは、例えば、米国特許第6,352,694号;同第6,534,055号;同第6,905,680号;同第6,692,964号;同第5,858,358号;同第6,887,466号;同第6,905,681号;同第7,144,575号;同第7,067,318号;同第7,172,869号;同第7,232,566号;同第7,175,843号;同第5,883,223号;同第6,905,874号;同第6,797,514号;同第6,867,041号;および米国特許出願公開第20060121005号に記載の方法を用いて、T細胞の遺伝子組換えの前または後に行われてもよい。これらの方法によれば、本発明のT細胞は、CD3 TCR複合体関連シグナルを刺激する薬剤およびT細胞表面にある共刺激分子を刺激するリガンドが取り付けられた表面と接触させることによって増殖させることができる。
上記の様々な方法によって得られたT細胞は、中でも癌、感染症、または免疫疾患を、必要とする患者において処置するための医薬として使用することが意図される。
本発明者らは、細胞下局在を含むBioGPS(Human Primary Tumors(U95))uniprotデータからもダウンロードすることができる、1パネルの正常組織(Human U133A/GNF1H Gene Atlas)癌マイクロアレイデータからBioGPSマイクロアレイデータを使用した。
この研究のために、RNA-seqデータは利用できず、従って、本発明者らは、11の研究所が参加したMILE(Microarray Innovations in Leukemia)コンソーシアムによる大規模研究から得られたマイクロアレイデータを使用した(http://www.ngrl.org.uk/wessex/downloads/tm08/TM08-S4-1_KenMills.pdf-Haferlach et al. 2010、http://www.ncbi.nlm.nih.gov/pubmed/20406941)。この生データには、ALL(急性リンパ芽球性白血病)、AML(急性骨髄性白血病)、CLL(慢性リンパ芽球性白血病)およびCML(慢性骨髄性白血病)およびMDS(骨髄異形成症候群)の結果が含まれる。本発明者らは、細胞下局在については、いつものようにuniprotデータも使用した。
本発明をさらに深く理解するために、白血病CD38陽性細胞に対して向けられたT細胞を生成するために従う工程の一例を以下に示した。
1.患者1人1人または血液バンクからの細胞培養物または血液試料からT細胞を準備し、抗CD3/C28アクチベータービーズ(Dynabeads(登録商標))を用いて前記T細胞を活性化する。このビーズは、T細胞の活性化および増殖に必要な一次シグナルおよび共刺激シグナルを両方とも供給する。
2.前記細胞に、抗CD38抗体の可変鎖をコードする配列と融合した、CD3ζ活性化ドメイン、4-1BB共刺激ドメイン、膜貫通ドメイン、およびCD28に由来するヒンジの融合からなるキメラ抗原受容体をコードするトランスジーンを含むレトロウイルスベクターを形質導入する。形質転換されたT細胞の安全性を改善するために、リツキシマブに対して感受性のある自殺遺伝子が、WO2013/153391に記載のように、レンチウイルスベクターに、T2A分割配列で分けられて、さらに導入されてもよい。
3.(任意で)非アロ反応性および/または耐性のT細胞を操作する:
(a)WO2013/176915に示したプロトコールを従うことによって、前記細胞のTCRαを不活化して、TCRを前記細胞の表面から無くし、同種異系のTCRにより宿主組織が異物と認識されるのを阻止し、従って、GvHDを回避することができる。
(b)移植されたT細胞に影響を及ぼすことなく移植片拒絶反応を阻止するために、前記細胞が免疫抑制処置または化学療法処置に対して耐性になるように、免疫抑制剤または化学療法薬物の標的をコードする1種類の遺伝子を不活化することもできる。本実施例では、免疫抑制剤の標的はCD52であり、免疫抑制剤は、WO2013/176915に記載のようなヒト化モノクローナル抗CD52抗体(例えば、アレムツズマブ)である。
4.特異的TAL-エンドヌクレアーゼ(TALEN(商標)-Cellectis, 8 rue de la Croix Jarry, France)をコードするmRNAを用いてT細胞をエレクトロポレーション処理することによって、遺伝子不活化が行われる。不活化されたT細胞を、磁気ビーズを用いて分別する。例えば、標的にされた遺伝子(例えば、CD38、CD70、およびCD70)を依然として発現するT細胞は固体表面に固定されることで除去することができ、不活化された細胞は、カラムを通過するストレスに曝露されない。この穏やかな方法によって、適切に操作されたT細胞の濃度が上昇する。
5.CD3複合体刺激によって、操作されたT細胞を患者に投与する前にインビトロで増殖させるか、または患者に投与した後にインビボで増殖させる。投与工程前に、患者は、ヒト化モノクローナル抗CD52抗体であるCAMPATH1-Hなどの免疫抑制治療剤に供することができる。
6.任意で、操作された細胞を標的抗原に接近させるために、患者に投与する前に、前記細胞を二重特異性抗体にエクスビボで曝露するか、または患者に投与した後に、前記細胞を二重特異性抗体にインビボで曝露する。
特に、キメラ抗原受容体(CAR CD38)が提供されたときに、操作されたT細胞の抗腫瘍活性を示す能力がゲノム操作によって影響を受けなかったことを検証するために、操作されたT細胞を、表面にCD38を発現するダウジ細胞と4時間インキュベートした。Tリンパ球による細胞傷害性顆粒放出(脱顆粒と呼ぶ)のマーカーであるCD107aの細胞表面アップレギュレーションをフローサイトメトリー分析によって測定した(Betts, Brenchley et al. 2003)。
CD38標的-サイクリックADPリボース加水分解酵素の提示
CD38は、大多数の患者において高レベルのCD38を発現する多発性骨髄腫(MM)細胞を含む、多くの免疫細胞の表面に見出される糖タンパク質である。多くの研究によって、抗CD38 mAbを用いて、CDCおよびADCCによって患者由来CD38+MM細胞およびCD38+MM細胞株が効率的に死滅することが示されているので(Ellis, J. H. K. et al, Journal of Immunology, 1995, 155 (2), 925-937)、CD38はMMの検証された標的である。ダラツムマブは、多発性骨髄腫および他の血液腫瘍の死滅を誘導する治療用ヒトCD38モノクローナル抗体である(De Weers, M. et al, J Immunol 2011 186:1840-1848)。いくつかの研究において、CD38は活性化T細胞によって高発現されることも示されている(Sandoval-Montes CJ et a:, 2005, Leukoc Biol. 77(4):513-21)。
CD3/CD28ビーズおよびIL-2を用いた刺激後のT細胞によるCD38発現を、17日間にわたって3〜4日毎にFACSによって分析した。活性化後、6日目と17日目との間に、90%を超えるT細胞が発現することが観察された(図10B)。
CD38遺伝子内にある、13pbスペーサーで分けられた2つの17pb長配列を標的とするヘテロ二量体TALE-ヌクレアーゼを設計および作製した。それぞれの半標的は、以下の表15および図10Aに列挙した半TALE-ヌクレアーゼの反復によって認識される。

CAR抗CD38の構造および組成物
表17には、scFv抗CD38のVH鎖およびVL鎖を示した。SEQ ID NO:10〜11はヒト化抗CD38抗体ダラツムマブ(Genmab)に対応し、SEQ ID NO:12〜13は、米国特許第8,263,746B号に記載のようなMOR202(またはMOR03087)に対応する。

活性化後、4日目に、CD38 TALENをトランスフェクトする。3日後に、CD38欠損細胞を負の選択によって分別し、3日後に、抗CD38 CAR mRNAを用いてトランスフェクトする。次いで、CAR mRNAを一過的にトランスフェクトしたら、作製されたCAR分子を、発現および標的細胞株発現CD38に対する脱顆粒活性についてスクリーニングする。活性試験にには、異なる発現レベルのCD38を発現する標的細胞株(図11B):
-抗CD38マイクロビーズを用いた磁気分離によって入手したU266 CD38+およびU266 CD38-
-中間レベルのCD38を発現する多発性骨髄腫細胞株であるL363
-高レベルのCD38を発現するバーキットリンパ腫由来細胞株であるダウジ
-慢性骨髄性白血病に由来する細胞株CD38陰性細胞株であるK562
を使用する。
CS1標的の提示
多発性骨髄腫(MM)は、骨髄内でのプラズマ細胞(PC)の異常なクローン増殖を特徴とするB細胞悪性疾患であり、2012年に米国ではMMにより推定21,700件の新規症例と、10,710件の死亡が特定されている(Siegel R, et al. Cancer J Clin 2012 62:10-29)。2013年には、米国において22,350人の個体がMMと新たに診断され、10,710人がMMで死亡したと見積もられており、これは全血液悪性腫瘍による死亡の20%を占める。全生存期間を改善したプロテアソーム阻害剤および免疫調節薬物の使用にもかかわらず(Palumbo A, et al. Leukemia 2009 23:449-456)、MMは、新規の治療アプローチが緊急に必要な不治の悪性疾患のままである(Podar K, et al. Leukemia 2009 23:10-24)。
抗CD38抗原標的単鎖CARについて実施例1で説明したように、ヒンジ、膜貫通ドメイン、同時活性化ドメインおよび伝達ドメイン(例えば、図11Aに図示し、配列を表18に示した)の点で同じ成分を用いて同じ構造V1、V2、およびV3を設計する。
CS1は、多発性骨髄腫患者に由来する形質細胞様細胞において高レベルで発現しており、そのため、CARを開発するための興味深い標的である。T細胞、特に、CD8サブセットは低レベルのCS1を発現する。T細胞、特に、CD8サブセットが抗CS1 CARを発現したときに死滅する可能性があるので、この低レベルのCS1はT細胞CAR開発に不利な点である。
CAR CS1+発現
図16の結果から、モックトランスフェクトした細胞の形質導入効率は、CS1遺伝子を標的とするTALEnでトランスフェクトした細胞より高いことが分かる。これは、おそらく、形質導入されていないCS1発現T細胞の特異的細胞死滅によるものであるが、この集団は、TALENによる遺伝子破壊の結果として細胞がCS1を発現しなくなったときに影響を受けない。
CAR形質導入の8日後に、これらの細胞の細胞傷害活性を、等量のT細胞をCS1発現細胞株(L363細胞)またはCS1発現を欠く負の対照細胞株(MOLM13)と共培養することによって評価した。細胞共培養を開始して4時間後に、標的細胞株の生存率をフローサイトメトリーによって測定した。図15Aに示した結果から、CAR+T細胞と共培養したときにCS1(+)細胞の細胞生存率が低下したのに対して、CS1(-)細胞生存率に及ぼす影響は観察されなかったことが分かる。特異的細胞溶解をフローサイトメトリーデータを用いて計算した。T細胞を、CS1遺伝子を標的とするTALEnでトランスフェクトした後に、CARを形質導入したときに、特異的細胞溶解は2倍高くなった(図15B)。細胞をモックトランスフェクトしたときには、共培養中に存在するCAR+T細胞の量は多いので、この影響は、さらに大きい可能性があると考えるべきである(図16からのフローサイトメトリーデータを参照されたい)。実験結果は以下の通りである。
- モック/NTD試料の場合、BFP+細胞の%は0.1%であり、CD8+細胞の量は53.9%である。
- TALEn/NTD試料の場合、BFP+細胞の%は0.2%であり、CD8+細胞の量は49.5%である。
- モック/L90-2試料の場合、BFP+細胞の%は94%であり、CD8+細胞の量は1.8%である。
- TALEn/L90-2試料の場合、BFP+細胞の%は61%であり、CD8+細胞の量は8.3%である。
TALEnでトランスフェクトしたT細胞ではCS1遺伝子が破壊されたことを確認するために、形質導入後、11日目に、様々な試料を、CD3/CD28ビーズを用いて再活性化した。再活性化の72時間後に、細胞を抗CD8抗体および抗CS1抗体で染色し、発現をフローサイトメトリーによって分析した。
- モック/NTD試料の場合、BFP+細胞の%は0.01%であり、CS1は細胞の65.2%で発現しており、CD8+細胞の量は80.7%である。
- TALEn/NTD試料の場合、BFP+細胞の%は0.2%であり、CS1は細胞の9.7%で発現しており、CD8+細胞の量は78.8%である。
- モック/L90-2試料の場合、BFP+細胞の%は94%であり、CS1は細胞の37.5%で発現しており、CD8+細胞の量は16%である。
- TALEn/L90-2試料の場合、BFP強度は61%であり、CS1発現は8.5%であり、CD8発現は68.5%である。
CD70標的の提示
CD70は、CD27に結合するサイトカインであり、TNFファミリーの一部である(Goodwin.G. et al, 1993, Cell 73:447-456)。このタンパク質はT細胞適応応答において役割を果たし、共刺激されたT細胞の増殖を誘導し、細胞溶解性T細胞の生成を強化する。そのアクセッション番号はP32970(Uniprot)である。Schurch, C. et al.(J. Clin. Invest., 2012;doi:10.1172/JCI45977)などの、いくつかの研究から、CD27-CD70相互作用のブロッキングは慢性骨髄性白血病(CML)の処置を助ける可能性があることが示唆されている。
CD70遺伝子をKOするために、実施例1および実施例2と同じ戦略を行う。CD70遺伝子内にある、15pbスペーサーで分けられた2つの49pb長配列を標的とするヘテロ二量体TALE-ヌクレアーゼ、および23pbスペーサーで分けられた57pb長配列を標的とする1種類のTALE-ヌクレアーゼを設計および作製した。それぞれの半標的は、以下の表22に列挙した半TALE-ヌクレアーゼの反復によって認識される。
CAR抗CD70を発現させるために実施例1および実施例2と同じ戦略を行う。
Claims (43)
- (c)免疫細胞において、抗原マーカーの発現または提示に関与する遺伝子を遺伝子不活化するかまたは変異させる工程であって、該抗原マーカーが該免疫細胞の表面と病的細胞の表面の両方に存在する、工程、
(d)該免疫細胞において、該病的細胞の表面に存在する該抗原マーカーに対して向けられたキメラ抗原受容体をコードするトランスジーンを発現させる工程
を含む、病的細胞に対する免疫療法のための免疫細胞、好ましくはT細胞を調製する方法。 - 抗原マーカーが、表4に列挙した抗原マーカーより選択される、請求項1記載の方法。
- 抗原マーカーがCD38またはその免疫反応性断片である、請求項1記載の方法。
- 免疫細胞を活性化させかつ増殖させるさらなる工程を含む、請求項1〜3のいずれか一項記載の方法。
- 細胞表面にマーカー抗原を提示する細胞を排除することによって結果として得られた免疫細胞を精製するさらなる工程を含む、請求項1〜4のいずれか一項記載の方法。
