Abstract

スライス厚みを制御することはいくつかの理由のために重要であり得る。

− サンプルの3D性質を考えると、複数の特徴は、隣接するスライスでさえ、横切って変化し得、そして自動化された特徴抽出、およびたわみ矯正は、より大きなサンプルでは困難である。
代表的な連続切片顕微鏡法では、スライスはヒトの注釈またはアルゴリズムを用いて整列され、そこでは、マウンティングの間のたわみが再構築の前に矯正される。このプロセスは、代表的には、隣接する切片を横切って存在する点である位置合わせマークの選択を含む。この位置合わせ点は、画像中の画素を整列にもたらすために両方の画像に適用される変換関数を計算するために用いられ得る。一連の画像を横切る位置合わせマークを選択することは困難であり得る。なぜなら、スタック中の画像の各々は次とは異なるからである。複数の特徴はまた、切断面に正確に垂直に交差することは希であり、位置合わせマークのために同じ特徴が選択される場合、これらの特徴は、切片を横切って空間的にドリフトし得、そしてそれ故、例えば、傾斜にある目的物は、垂直であると誤解釈され得る。

ワークフローのさらなる実施形態は、マウントするステップと染色するステップとの間のさらなる撮像ステップとともに、上記のワークフロー200に類似し得る。

Claims (14)
- サンプルを分析する方法であって:
該サンプルの複数の画像を生成する工程;
該サンプルを複数の切片にスライスする工程;
該複数の切片を染色する工程;
複数の染色された切片を撮像することにより、該サンプルの複数の二次画像を生成する工程;および
該サンプルの複数の二次画像を互いと同時位置合わせする工程、を含む方法。 - 前記サンプルが、組織サンプルを含む、請求項1に記載の方法。
- 前記サンプルの個々の一次画像が、該サンプルの個々の切片がスライスされる前に生成され、該サンプルの個々の切片が該個々の一次画像と関連する、請求項1に記載の方法。
- 前記複数の一次画像に基づき、前記サンプルの仮想モデルを生成する工程をさらに含む、請求項1に記載の方法。
- 前記サンプルの同時位置合わせされた複数の二次画像を、前記仮想モデルに同時位置合わせする工程をさらに含む、請求項4に記載の方法。
- 前記サンプルの仮想モデルおよび前記同時位置合わせされた複数の二次画像の同時位置合わせに応答して、診断を生成する工程をさらに含む、請求項5に記載の方法。
- 個々の一次画像を、対応する個々の二次画像と同時位置合わせする工程をさらに含む、請求項1に記載の方法。
- 前記個々の一次画像に基づき、対応する個々の二次画像を歪める工程をさらに包含する、請求項7に記載の方法。
- 前記サンプルが、ナイフエッジ走査顕微鏡(KESM)でスライスされる、請求項1に記載の方法。
- 前記サンプルを複数の切片にスライスする工程が、該サンプルのブロック面をスライスすることを含む、請求項1に記載の方法。
- 前記複数の切片を、複数のスライドにマウントする工程をさらに含む、請求項1に記載の方法。
- 前記複数の切片が、抗原またはタンパク質染色で染色される、請求項1に記載の方法。
- 伸張、剪断、並進、回転、スプライン、非線形補間、または光学矯正変換の1つ以上を用いて、前記二次画像を歪める工程をさらに含む、請求項1に記載の方法。
