葉酸NIRコンジュゲートの合成および特徴付けは、実質的に実施例1に記載したとおりに行った。
KB細胞(ヒト鼻咽頭細胞株)をAmerican type culture collection(Rockville、MD)から入手し、少なくとも6継代の間5%二酸化炭素:95%空気−加湿雰囲気中37℃で、10%加熱不活性ウシ胎児血清(Atlanta Biological、GA)および1%ペニシリンストレプトマイシン(Gibco、NY)を含有する葉酸塩不含1640RPMI培地を用いて単層として増殖させ、その後、それらをアッセイに使用した。
OTL−0039、OTL−0038、または葉酸に対して、研究から導かれた解離定数(KD)を計算すると、それぞれ81.8nM、10.4nM、および7.4nMであった。OTL−0039、OTL−0038、および葉酸に対して計算した相対結合親和性はそれぞれ0.09、0.71、および1であった。すべての3種の試験品は、[3H]−葉酸と定量的に匹敵した。

細胞培養および動物の準備

7週齢の雌nu/nuマウスに、肩上にKB細胞(葉酸塩不含RPMI1640培地中1.0×106個/マウス)を皮下接種した。キャリパを用いて2日毎に垂直方向で腫瘍の増殖を測定し(体重は、同じスケジュールでモニターした)、腫瘍の容量を0.5×L×W2(ミリメートルで、L=最長軸であり、W=Lに垂直な軸である)として計算した。腫瘍が容積で400〜500mm3に達すると直ちに、動物(5匹のマウス/群)に、リン酸緩衝生理食塩水(100μL)中10nmolのOTL−0038またはOTL−0039を静脈内注射した。2.5時間後、動物をCO2窒息により安楽死させた。次いで、Living Image 4.0ソフトウェア付きのCaliper IVIS Lumina II Imaging Station(Perkin Elmer Inc、MA)を用いて、全身画像化(無傷腫瘍)実験を行った。画像化のための設定:ランプレベル:中;励起:745nm;発光:ICG(インドシアニングリーン);epi照射;ビニング:4(M)、FOV=12.5;f−stop=2;捕捉時間=1秒。
全身画像化に続いて、動物を切開し、前の通りのIVIS撮像装置を用いて蛍光活性について、選択した組織(心臓、肺、肝臓、脾臓、腎臓、胃、小腸、大腸、筋肉、皮膚、腫瘍)を分析した。画像化のための設定:ランプレベル:中;励起:745nm;発光:ICG;epi照射;ビニング:4(M)、FOV=12.5;f−stop=2;捕捉時間=1秒。
全身画像化
図14に見られるように、OTL−0038は、葉酸受容体陽性腫瘍において主に蓄積したが、他の組織において実質的な蛍光活性はなかった。
組織生体分布の分析は、各マウスを安楽死させ、それらの臓器を取り出して、IVIS撮像装置を用いてそれらを画像化することによって、全身画像化を施した同じ動物に対して行った。図15に見られるように,最大の蛍光強度が、FR陽性腫瘍で認められ、腎臓を除く他の組織には蓄積されていなかった。OTL−0038の腎臓取り込みは、腎臓の近位尿細管の頂端膜が高いレベルの葉酸受容体を発現することが公知であったので、予期された。さらに、プローブが、それらの低い分子量および半減期(大部分の葉酸コンジュゲートは<30分の半減期を有する)のために、腎臓を通って排泄されることが可能である。
OTL−0038は、葉酸受容体陽腫瘍異種移植および腎臓に主に蓄積した。他のすべての正常な組織は、最小レベルまたは取り込みなしを示し、結果として優れた腫瘍と正常な組織との蛍光比をもたらした。

KB細胞(ヒト鼻咽頭細胞株)をAmerican type culture collection(Rockville、MD)から入手し、少なくとも6継代の間5%二酸化炭素:95%空気−加湿雰囲気中37℃で、10%加熱不活性ウシ胎児血清(Atlanta Biological、GA)および1%ペニシリンストレプトマイシン(Gibco、NY)を含有する葉酸塩不含1640RPMI培地を用いて単層として増殖させ、その後、それらをアッセイに使用した。
7週齢の雌nu/nuマウスに、肩上にKB細胞(葉酸塩不含RPMI1640培地中1.0×106個/マウス)を皮下接種した。キャリパを用いて2日毎に垂直方向で腫瘍の増殖を測定し(体重は、同じスケジュールでモニターした)、腫瘍の容積を0.5×L×W2(ミリメートルで、L=最長軸であり、W=Lに垂直な軸である)として計算した。腫瘍が容積で400〜500mm3に達すると直ちに、動物(2匹のマウス/群)に、リン酸緩衝生理食塩水(100μL)中10nmolの試験品(OTL−0038、葉酸−LS288、葉酸−IR800、葉酸−ZW800)を静脈内注射した。2.5時間後、動物をCO2窒息により安楽死させた。次いで、Living Image 4.0ソフトウェア付きのCaliper IVIS Lumina II Imaging Station(Perkin Elmer Inc、MA)を用いて、全身画像化(無傷腫瘍)実験を行った。画像化のための設定:ランプレベル:中;励起:745nm;発光:ICG(インドシアニングリーン);epi照射;ビニング:4(M)、FOV=12.5;f−stop=2;捕捉時間=1秒。
全身画像化に続いて、動物を切開し、前の通りのIVIS撮像装置を用いて蛍光活性について、選択した組織(心臓、肺、肝臓、脾臓、腎臓、胃、小腸、大腸、筋肉、皮膚、腫瘍)を分析した。画像化のための設定:ランプレベル:中;励起:745nm;発光:ICG;epi照射;ビニング:4(M)、FOV=12.5;f−stop=2;捕捉時間=1秒。
全身画像化
図14に見られるように、OTL−0038(L−異性体)、葉酸−LS288、葉酸−IR800、葉酸−ZW800は、葉酸受容体陽性腫瘍において主に蓄積したが、他の組織において実質的な蛍光活性はなかった。さらに、直接の比較は、OTL−0038注射マウスの腫瘍蛍光強度が、他の葉酸コンジュゲーテッド近IR色素で処置したマウスよりも明るい(蛍光強度がより高い)ことを実証した(図15)。
組織生体分布の分析は、各マウスを安楽死させ、それらの臓器を取り出して、IVIS撮像装置を用いてそれらを画像化することによって、全身画像化を施した同じ動物に対して行った。図16に見られるように、最大の蛍光強度は、FR−陽性腫瘍および腎臓において認められた。腎臓取り込みは、腎臓の近位尿細管の頂端膜が高いレベルの葉酸受容体を発現することが公知であったので、予期された。さらに、プローブが、それらの低い分子量および半減期(大部分の葉酸コンジュゲートは<30分の半減期を有する)のために、腎臓を通って排泄されることが可能である。
腫瘍蓄積蛍光強度において、OTL−0038は、葉酸−LS288、葉酸−IR800、および葉酸−ZW800と比べて有利な態様を有する。OTL−0038は、LS288、IR800、およびZW800などの他の市販の近IR色素よりも明るくなり得る。

表1および図14に示されているように、0.3nmol用量を除くすべての用量が、葉酸受容体陽性腫瘍においてより高い腫瘍の取り込みを有した。他方では、より高い腎臓取り込みも用量範囲0.3〜10nmolに対して認められ、より低い腎臓取り込み(腫瘍取り込みと比べて)が用量範囲30〜90nmolに対して認められた。さらに、より高い非特異的取り込みが60および90nmol用量において認められた。
0.3nmol用量において認められた、葉酸受容体陽性腫瘍におけるより低い取り込み(弱い蛍光強度)は、腫瘍細胞上の葉酸受容体の不完全な飽和に起因し得る。他方では、腎臓において認められたより高い蛍光強度は、腎臓を通ったプローブの排除に起因し得る。さらに、腎臓の近位尿細管の頂端膜は、高いレベルの葉酸受容体を発現することが公知である。1.0〜30.0nmolの間の用量範囲は、腫瘍取り込みおよび腫瘍と正常な組織との優れた比(信号対バックグラウンド比)を示していた。より高い腎臓取り込みは、腎臓を通ったプローブの排除(30nmol用量を除く)および腎臓上での葉酸受容体の発現に起因し得る。用量レベル60.0nmolおよびそれ超は、より高い非特異的取り込みを示している。しかし、それらは高い腫瘍取り込みおよびより少ない腎臓取り込み(30nmol用量を含む)を依然として有する。より少ない腎臓取り込みは、肝臓および腸を通ったプローブの代替の排除に起因し得る。したがって、OTL−0038は、このより高い濃度で凝集を形成し得る。1.0nmolが、腫瘍の画像化に対して非侵襲性態様を維持しながら(図22)、腫瘍とバックグラウンドとの良好な比を得るために投与される最も低い用量であり、30nmolが、腫瘍とバックグラウンドとの最も良い比を得るために投与される最大の用量であると結論づけることができる。
細胞培養物およびマウスの準備

全身画像化
図14に見られるように、OTL−0038は、葉酸受容体陰性腫瘍において蓄積せず、腎臓を除く他の組織において実質的な蛍光活性はなかった。
侵襲性腫瘍および腎臓取り込み
OTL−0038は、葉酸受容体に対して極めて特異的である。
無胸腺雌ヌード(nu/nu)マウス(6週齢、18〜20g)をHarlan Laboratories(Indianapolis、IN)から購入し、正常な食餌(Teklad、WI)で維持した。無菌の覆い隠したラックの障壁中5匹/ケージで動物を飼った。オートクレーブした水道水および餌を必要に応じて与えた。研究期間中標準的な12時間明暗サイクルで無菌環境において動物を飼った。耳パンチによりマウスを個別に識別した。動物手順のすべては、Purdue Animal care and Use Committeeにより承認された。動物のケアおよび研究は、動物の人道的処置についての国および国際ガイドラインに従って行った。
OTL−0038またはOTL−0039の注入直後、動物の皮膚は緑色になった。しかし、この緑色は24時間以内に消えた。試験品の投与後動物は活動的であり、研究を通して正常に挙動した。図14に見られるように、研究を通して体重は安定したままであった。IHCデータに従い(図15)、いかなる組織にも病変は識別されなかった。
1μmol(1000×臨床的用量)のOTL−0038またはOTL−0039は、動物に対して毒性を持たず、これは、OTL−0038(1nmol)およびOTL−0039(1nmol)が、病院でヒトに対して毒性を持たないことを示唆している。

c)前記赤外光による励起の際に蛍光を発する領域の外科的切除を行うステップ

- 対象における疾患を診断する方法であって、
a)診断を必要としている対象に、ある量の請求項1に記載の化合物を、前記化合物の標的細胞型の少なくとも1個の細胞への結合を可能にする時間および条件下で投与するステップ、
b)生物試料中に存在する前記請求項1に記載の化合物からの信号を測定するステップ、
c)b)で測定された前記信号を少なくとも1つの対照データセットと比較するステップであって、前記少なくとも1つの対照データセットは、前記標的細胞型を含まない生物試料と接触した前記請求項1に記載の化合物からの信号を含むステップ、および
d)ステップc)における前記比較が前記疾患の存在を示す、疾患の診断を提供するステップ
を含む方法。 - 請求項1に記載の化合物を含むキット。
