本出願は、2018年2月15日にされた米国仮出願第62/631,390号の優先権を主張し、その教示は、あらゆる目的のために、その全体が参照により本明細書に組み込まれる。
この技術は、Bioorganic & Medicinal Chemistry Lettersに掲載され、2018年2月24日にオンラインで公開された学術論文「A chemically stable fluorescent marker of the ureter(化学的に安定な尿管の蛍光マーカー)」に記載されており、その全体が参照により本明細書に組み込まれる。
本発明は、ヘプタメチン系シアニン化合物及びその胆道系及び腎臓系の蛍光マーカーとしての使用に関する。
分子医学の目覚しい進歩にもかかわらず、外科的介入は、通常、記憶や視覚・触覚による手がかりのみで行われる。重要な構造を認識し、かつ、正確に切開し、また修復することは、外科的プロセスの中核である。意図しない損傷は、短期及び長期の合併症、入院の長期化並びに高額の医療費をもたらす。イメージングによる知見の追加が、さまざまな医療機器で検討されている。蛍光ガイド手術は、比較的単純な光学的情報のみによるリアルタイムのイメージを提供する。この方法は、さまざまな疾患において臨床で使用する方向に進んでいる。しかしながら、臨床の大半は、50年以上前にFDAが承認した化合物であるインドシアニングリーンを使用している。蛍光ガイド手術の幅広い適応を可能にするために、現場での特定の課題に対処する新しい色素が必要である。

飽和炭素鎖を有する炭化水素基。鎖は、分枝、非分枝又は環状(シクロアルキル)であってもよい。特に明記しない限り、アルキルという用語は、置換及び非置換アルキルを包含する。

1以上の置換基が結合した、芳香族化合物若しくは脂肪族化合物などの基本的な化合物又はそのラジカルで、通常、基本的な化合物の水素原子が置換基に置き換わる。たとえば、限定されるものではないが、置換された芳香族化合物は、芳香環に結合した脂肪族基を有していてもよく、トルエンなどである。また、たとえば、限定されるものではないが、長鎖の炭化水素は、ヒドロキシル基を結合していてもよい。

標的化剤を含む立体配座が制限されたシアニン フルオロフォアが特異的に結合することが予定された分子。標的の例には、試料組織中のタンパク質や核酸配列が含まれる。標的領域は、標的分子が存在しているか、存在している可能性がある領域である。

本発明のある態様によれば、本明細書に開示される事項は、式I又は式IIで表されるヘプタメチン系シアニン化合物の製造方法である。
ある態様によれば、本発明のヘプタメチン系シアニンは、生きている細胞の可視化及び追跡に有用である可能性がある。さらに、探査及び診断での使用は、本発明の範囲内である。

ある態様によれば、本発明は、キットについてさらに説明する。キットの態様は、少なくとも1つの式I又は式IIで表されるヘプタメチン系シアニン化合物を含む。ある態様では、キットには、少なくとも1つの化合物を溶解又は懸濁できる溶液を含んでいてもよい。キットは、使い捨てのバイアル又は注射器などの1以上の容器を含んでいてもよい。キットは、化合物を使用するための説明書をさらに含んでいてもよい。
特に明記しない限り、一般的に、反応は、オーブンで乾燥したガラス器具を使用し、窒素又はアルゴン雰囲気下で無水溶媒(活性アルミナカラムを通過)を使用して行った。全ての市販の試薬は、そのまま使用した。N-[(3-(アニリノメチレン)-2-クロロ-1-シクロヘキセン-1-イル)メチレン]アニリン・塩酸塩はSigma-Aldrich(St. Louis, MO)から購入した。IR-800CWは、Li-Cor Biosciences(Lincoln, NE)から購入した。
マグネチックスターラー回転子が入ったマイクロ波反応用容器にインドレニン1(3.0g、10.8mmol;Park et al., Bioconjugate Chem. 2012, 23:350)、MeCN(12mL)及びヨウ化物1(3.0g、10.8mmol;Lawal et al., Supramol. Chem. 2009, 21:55)を加えた。容器をアルゴン中で封止し、淡褐色のスラリーを砂浴中120℃で22時間加熱したところ、反応物は濃い赤/ピンク色に変化した。反応物を冷却し、溶媒をロータリーエバポレーターで留去した。水(10mL)を赤色の粗生成物に加え、逆相クロマトグラフィー(C18 Aq、0→30%MeCN/水)で精製した。生成物含有画分を合わせ、溶媒をロータリーエバポレーターで留去し、化合物3(2.1g、収率45%)を赤いゴム状固体として得た。
第一に、BL-760中間体を製造した。マグネチックスターラー回転子入りマイクロ波反応用チューブを用い、市販のIR Dye 783(120mg、0.16mmol)を乾燥DMF(2mL)に溶解し、密封した。アルゴンを2分間流し、続いて2-(メチルアミノ)エタノール(65μL、0.8mmol)を加えた。溶液を砂浴中で80℃に加熱したところ、緑から青色に変化し、LC-MSは所望の生成物の形成を示した。反応混合物を冷却し、Et2O中に沈殿させ、4500rpmで3分間遠心分離した。ペレットに水(2mL)及びNaHCO3飽和水溶液(2mL)を加え、残留物を逆相クロマトグラフィー(C18、0→40%MeCN/水)で精製した。生成物含有画分を合わせ、凍結乾燥し、BL-760中間体(99mg、収率79%)を青色の固体として得た。

フラッシュカラムクロマトグラフィーは、CombiFlash(登録商標)Rf200iシステム(Teledyne Isco, Inc.)の逆相(100Å、粒子径20〜40μ、RediSep(登録商標)RfGold(登録商標)逆相C18又はC18Aqカラム)で行った。高分解能LC/MSは、陰イオンモードのIon MAX APIエレクトロスプレーイオンソースを備えたThermo-Fisher LTQ-Orbitrap-XLハイブリッド質量分析システムで行った。分析LC/MSは、Phenomenex, Inc.から入手したKinetex 2.6μm C18 100Å(2.1×50mm)カラムを用い、Shimadzu LCMS-2020シングル四重極システムで行った。0→90%MeCN/0.1%ギ酸水溶液のグラジエントを用い、流速0.2mL/minで4.5分かけて分析を行った。1H NMR及び13C NMRスペクトルは、ブルカー分光計(400若しくは500MHz又は100若しくは125MHz)で測定し、重水素化溶媒のシグナルと比較して報告する。1H NMRスペクトルのデータは、化学シフト(δppm)、多重度、結合定数(Hz)及び積分値で報告する。13C NMRスペクトルのデータは、化学シフトにより報告する。IRスペクトルは、Jasco FT/IR-4100分光計で測定し、吸収周波数(cm−1)により報告する。量子収率の吸収曲線は、Shimadzu UV-2550分光光度計でUVProbe 2.32ソフトウェアを用いて測定した。蛍光は、励起及び発光スリット幅5nm、インテグレーション比(integration rate) 0.1s並びに有効な発光補正を備えたPTI QuantaMaster定常状態分光蛍光光度計で、FelixGX 4.2.2ソフトウェアを用いて記録した。データ分析及びカーブフィッティングには、Microsoft(登録商標)Excel 2011及びGraphPad Prism 7を使用した。光の強度は、S120VC標準Siフォトダイオードパワーセンサー(200〜1100nm、50nW〜50mW)を備えたThorlabs PM200光パワー及びエネルギーメーターで測定した。フローサイトメトリーはCCR Flow Cytometry Core(NCI-Frederic
k)で行い、顕微鏡検査はOptical Microscopy and Analysis Laboratory(NCI-Frederick)で行った。略号については、JOC Standard Abbreviations and Acronyms(http://pubs.acs.org/userimages/ContentEditor/1218717864819/joceah_abbreviations.pdf)を参照。
式I及び式IIで表される本発明のヘプタメチン系シアニンの実施の態様は、対象の腎臓系又は胆道系の少なくとも一部のin vivoでの可視化であってもよい。ヘプタメチン系シアニンを含む適切な医薬組成物は対象に投与することができる。投与後、対象の腎臓系及び/又は胆道系の少なくとも一部に有効な量の近赤外線を照射することができ、画像が得られ、これにより、腎臓系及び/又は胆道系の器官及び/又は構造が可視化される。照射は外部から、又は内部で行うことができる。外部からの照射では、光を照射する範囲は、マイクロレンズ、フレネルレンズ又はディフューザーの利用などの適切な光照射装置で制御することができる。内部からの照射では、内視鏡又は光ファイバーカテーテルを使用してもよい。腎臓系の可視化では、ヘプタメチン系シアニンは式I又は式IAで表される構造であってもよい。胆道系の可視化では、ヘプタメチン系シアニンは式II又は式IIAで表される構造であってもよい。
Children’s National Hospitalの動物の管理及び使用のための委員会がプロトコルを承認した(IACUC # 30597)。全ての手順はChildren’s National HospitalのResearch Animal Facilityで行われた。この実験では、Charles River Laboratories(Wilmington, Massachusetts, USA)の、体重250〜300gの雌のSprague-Dawleyラットを使用した。鎮静と拘束のために、4%イソフルランを3分間吸入した。麻酔は、キシラジン2mg/kg及びケタミン75mg/kgを筋肉内注射して維持した。無菌状態を確保した後、開腹した。蛍光イメージングでは、24Gカテーテルを尾静脈に留置し、90μg/kgの注射を行い、その後、直ちにイメージングした。
開腹手術では、正中開腹術を行った。蛍光イメージングでは、24Gカテーテルを尾静脈に留置し、90μg/kgを注入し、その後、直ちにイメージングした。画像化機器を配置し、最大で2時間のビデオを記録した。全ての撮影は33ミリ秒の露光時間で行った。ImageJソフトウェアを用い、関心領域の蛍光シグナルとバックグラウンドシグナルの比を定量化した。コントラスト−バックグラウンド比(CBR)を計算し、時間軸に対してプロットした。結果を、平均+/−標準誤差(SEM)で表した。
この実施例では腹腔鏡手術におけるUL-766の有用性を評価した。さらに、この実施例では、激しい炎症の下で尿管の可視化を支援するUL-766の能力を評価した。臨床で利用可能な腹腔鏡蛍光システムを用いた、腹部に炎症を有する豚における尿管の可視化を支援する色素の能力。
