通常ヒト組織を通るキャピラリーベッドの特徴を再現するシステムが構築された。このシステムは、キャピラリーベッドの長さスケールで三次元(3D)の細胞モノカルチャーおよびヘテロタイプ細胞同時培養の灌流を容易にする。このシステムはまた、組織が形成された後、腫瘍細胞のような第2の細胞タイプの付加を可能にする。主要な特徴は、このシステムが、薬剤開発で一般に用いられるタイプのロボット工学に匹敵する高スループットアッセイに受け入れられる「マルチウェルプレート」フォーマット内で利用され得ることである。このシステムは、毒物学および代謝のためのアッセイを実施するための手段、そして肝炎、照射関連病理学、および癌を含む、肝疾患のようなヒト疾患のためのモデルとして提供される。癌適用は、原発性肝臓癌およびその他の癌からの転移、ならびに薬物代謝を抗腫瘍活性とリンクするためを含む。このシステムは、癌、ウイルス感染、および慢性肝臓線維症のような複雑かつ慢性の疾患のために特に有用である。このシステムはまた、疾患および先天的遺伝子欠陥を処置するための遺伝子治療アプローチを試験する手段として用いられ得る。
I.システム
システム、および製造の方法およびその使用が、組織または器官構造の微小マトリックスを通る細胞培養培地を循環するための高度に平行な手段を利用するように改変された灌流微小マトリックスアプローチを基に開発され、この技法を極めて高スループットアッセイのために適切にする。このシステムは、薬物毒性を試験すること、癌転移のモデル、幹細胞培養、およびその他のヒト疾患モデルにおける適用を有する。このシステムは、その基礎的構成要素として、細胞または組織培養のための灌流バイオリアクターおよびリザーバーペアの密なアレイ、および共通制御チャネルを経由して平行に作動されるバルブおよびポンプを有し、これら灌流バイオリアクターおよびリザーバーペアのアレイを通じて培地を再循環する。このアレイの各バイオリアクターは、バイオリアクターウェルおよびそれ自身のリザーバーウェルを含む。これらバイオリアクターウェルおよびリザーバーウェルは、細胞培養培地の再循環を可能にする流体チャネルによって連結される。各灌流バイオリアクター/リザーバーペアは、アレイ中の灌流バイオリアクター/リザーバーペアから流体的に隔離されている。アレイ中のすべてのバイオリアクターのバルブおよびポンプは、共通の水力または空気力制御チャネルを経由して平行に作動される。
センサーは、pH、酸素レベル、グルコースのような特定の代謝産物、ウイルスタンパク質のような指標分子の存在もしくは不在、または組織に対する影響の任意のその他の指標もしくはバイオリアクター内の組織に曝される物質における変化を検出するために用いられる。
II.適用
上記技術は、大スケール組込みを受ける。これは、大量の平行アッセイを実施することを可能にする。例えば、異なる細胞または細胞混合物が、各バイオリアクターウェル中に接種され得る。あるいは、異なる細胞培養培地が、すべてのバイオリアクター/リザーバーペアを通って循環され得るか、または各バイオリアクターウェル中の細胞/組織が異なる薬剤に曝され得る。初期の細胞タイプが添加され得(例えば、ヒト肝臓細胞単離物)、組織に安定化され、そして次に第2の細胞タイプ(例えば、癌細胞)が添加されて応答を調べる。
Claims (29)
- 細胞または組織培養のための灌流バイオリアクターおよびリザーバーペアの密なアレイ、および
共通制御チャネルを経由して平行に作動され、そして該バイオリアクターおよびリザーバーペアのアレイを通じて培地を再循環するバルブおよびポンプ、を備える、装置。 - 各バイオリアクターが、三次元の細胞/組織支持構造を備えるウェルを備える、請求項1に記載の装置。
- 前記細胞の足場またはキャリヤが、多孔性材料から作製される、請求項1に記載の装置。
- 前記細胞の足場が、微多孔性フィルターまたはメンブレンによって支持される中実フィルムまたはシート中の微小チャネルのアレイによって形成される、請求項2に記載の装置。
- 前記足場が、前記バイオリアクターウェルから射出され得る、請求項2に記載の装置。
- 前記アレイの各バイオリアクターが、バイオリアクターウェルおよびそれ自身のリザーバーウェルを備える、請求項1に記載の装置。
- 前記バイオリアクターウェルおよびリザーバーウェルが、細胞培養培地の再循環を可能にする流体チャネルによって連結される、請求項6に記載の装置。
- 前記アレイ中のすべてのバイオリアクター/リザーバーペアが、共通の取り外し可能なリッドによって覆われる、請求項1に記載の装置。
- 細胞/組織接種、薬剤添加、またはサンプル収集の手動またはロボット工学ピペット操作のためのデバイスを備える、請求項1に記載の装置。
- 各バイオリアクター/リザーバーペアが、前記アレイ中のすべての他のバイオリアクター/リザーバーペアから流体的に隔離されている、請求項1に記載の装置。
- 前記アレイ中のすべてのバイオリアクターのバルブおよびポンプが、共通の水力または空気力制御チャネルを経由して平行に作動される、請求項1に記載の装置。
- 前記バイオリアクター/リザーバーペアが、流体多岐管に製作または微細製作される、請求項1に記載の装置。
- 前記制御チャネルが、制御多岐管に製作または微細製作される、請求項1に記載の装置。
- ダイヤフラムバルブが、流体多岐管と制御多岐管との間にモノリシックエラストマーメンブレンを挟持することによって生成される、請求項1に記載の装置。
- 前記制御チャネルと流体チャネルとの間のメンブレンが、前記制御チャネルを通じて付与される水力または空気力作用によって偏向され得る、請求項14に記載の装置。
- 複数のバイオリアクター中の細胞培養の培地が、連続して連結されたバルブの逐次的作動によってポンプ輸送される、請求項1に記載の装置。
- 第1のセットの細胞をさらに備える、請求項1に記載の装置。

- 前記導入される第2のセットの細胞が幹細胞または前駆体細胞であって、そして該幹細胞または前駆体細胞が成熟組織によって分化するように誘導される、請求項20に記載の装置。
