る。

(12,000g、4分)で固体を除去した。上清をHPLCで分析した。

mのガラスカラム)中に配置した。最初に、カラムをCHCl3で溶出し、次い
でCHCl3/MeOH混合物(98,96,94,92及び90%)で溶出し

B/MSによって306.0977で分子イオンを示した(図5)。このデータ
は、分子式C15H16NO6と一致する(Δ0.1mmu)。ESI/MS分析に
より、ETM305の分子量を確認した(図6);m/z306での分子イオン
を、ナトリウムアダクト(m/z328)と共に観察した。ETM305の1H
NMRスペクトル(図7)は、構造分析について非常に重要であった。δ2.0
4、2.28及び6.09での共鳴は、Et743中のMe−6(δ2.03)

このメチル基の化学シフトは、アセトフェノン3(δ2.55)のメチル基のも
のと非常によくマッチした。ETM305の1H NMRスペクトルが、−NH−
CHO結合の周囲の回転子のために、二つのセットの共鳴(4:1比)より成る
ことに気付くと興味深い。MSデータと共に1H NMRデータは、ETM305
がスキーム1に示されたビニルホルムアミドユニット及びメチルケトンに付着し
たEy743のBユニット芳香環系を有することを示唆した。m/z306に対
するFAB/MS/MSは、提案された構造を支持した(図8)。
B/MSによって776.2489で分子イオンを示した。このデータは、分子
式C39H42N3O12Sと一致する(Δ0.0mmu)、それはETM775がE
t743の酸化生産物であることを示した。ポジティブ及びネガティブモードの
ESI/MSスペクトルの両者は、ETM775の分子量を確認した(図12及
び13)。制限された量のETM775のため、構造解析を主にマススペクトル
データの解釈により実施した。ETM775(m/z776)のM+Hに対する
FABMS/MSは、m/z276及び260(276−酸素)でのピークによ
って明らかにされるようにN−オキシドの形態でN−2で外部酸素が配置されて
いることを示すのに必須であった。Et743では観察されないm/z232の
断片イオンは、カルビノールアミン酸素が、アミドに酸化されていることを示唆
した(スキーム3)。ETM775中のA及びCユニットの構造は、m/z20
4(Aユニット)及びm/z224及び250(Cユニット)での特徴的なマス
スペクトルピークの存在によって明らかにされるように、完全なままであった1
。750 750Mhz 1H NMR(図16)及びUV(図17)の両者のスペ
クトルは、Et743のものと類似した1。

溶液、エマルションの懸濁液)が含まれ、それらは精製化合物若しくは薬理学的
