Description

발명의 상세한 설명
I. Et 743 대사 연구
A. 대사 혼합물-ETM의 제조:
Et-743 (50 μM)을 NADPH 생성 시스템(0.4 mM NADP+, 25 mM 글루코스-6-포스페이트, 0.5 U/ml 글루코스-6-포스페이트 디하이드로겐아제 및 3.3 mM 염화마그네슘)을 함유하는 0.1 M Tris-HCl 버퍼(pH 7.4)에서 0.4 mg/ml의 인간 림프아구(lymphoblast)-발현 CYP3A4 이소폼(isoform: Gentest Corporation, Woburn, MA)과 함께 배양하였다. 37℃에서 4 시간후에, 반응을 빙냉 아세토니트릴로 중지시키고, 고체를 원심분리(12,000 g, 4 분)에 의해 제거하였다. 상청액을 HPLC에 의해 분석하였다.
B. ETM 305 및 ETM 775의 정제
2.6 mg의 ETM(상기 A.에서 생성된)을 소량의 CHCl3에 용해시키고, 실리카 겔 칼럼(실리카겔/CHCl3 슬러리로 충전된 8 x 100 mm 글래스 칼럼)에 로딩하였다. 우선, 칼럼을 CHCl3 에 이어서 CHCl3/MeOH 혼합물(98, 96, 94, 92 및 90%)에 의해 용출(elute)하였다. 총 10 개의 시험관을 수집하고(각각 3 mL), TLC의 기초상에 결합시켜 4개의 분획을 수득하였다(표 1). 덜 극성이고 비-세포독성인 분획(ETM-SiOH-1, 2 mg)은 1H NMR 스펙트럼(도 1)에서 보여지듯이 Et 743에 구조적으로 관련되지 않은 리피드로 구성되었다.
잔류 세포독성 분획을 HPLC(Phenomenex-Ultracarb ODS, 10 μm, 10 x 150 mm, 3:1 MeOH/H2O 0.02 M NaCl, 1 mL/분, Da 검출: 210, 220, 254 및 280 nm)에 의해 더욱 정제하였다. 최대 극성 분획(ETM-SiOH-4, 0.2 mg)은 0.1 mg의 ETM 775(도 2)를 산출하였다. ETM-SiOH-3은 0.3 mg의 ETM305 (도 3)를 산출하였고, ETM-SiOH-2는 미량 대사산물의 복합 혼합물 (도 4)로 구성되었다.

C. ETM 305의 구조
ETM 305(IC50 0.2 μm/mL 대 L1210 세포)는 HRFAB/MS(도 5)에 의해 306.0977에서 분자 이온을 보여주었다. 이 데이타는 분자식 C15H16NO6(△ 0.1 mmu)과 일치한다. ESI/MS 분석은 ETM 305의 분자량을 확인시켰고(도 6); m/z 306에서의 분자 이 온이 그 나트륨 부가생성물(m/z 328)과 함께 관찰되었다. ETM 305의 1H NMR 스펙트럼(도 7)은 구조성 부여(structural assignment)에 매우 중요하였다. δ2.04, 2.28 및 6.09에서의 공명(resonances)은 각각 Et 743에서 Me-6(δ2.03), -OCOCH3(δ2.29) 및 디옥시-메틸렌 프로톤(δ6.11 및 6.01)의 것들과 거의 일치하였다.
더욱이, -CH=CH-NHCHO 단위(δ7.09, d, 1H, J=15 Hz; δ8.04, s, 1H), 및 추가적 메틸기(δ2.52, s, 3H)에 해당하는 공명도 관찰되었다. 이 메틸기의 화학적 시프트(shift)는 아세토페논(δ2.55)상의 메틸기의 것과 매우 잘 매치한다. ETM 305의 1H NMR 스펙트럼이 -NH-CHO 결합주위의 회전 형성체(conformers)때문에 두 세트의 공명들(4:1 비율)로 구성된다는 것도 흥미있다. 1H NMR 데이타는 MS 데이타와 함께 ETM 305 가 도식1에 보여지듯이 비닐-포름아미드 단위 및 메틸케톤에 부착된 Et 743의 B-단위 방향족 고리 시스템을 가진다는 것을 제시하였다. m/z 306상에서의 FAB/MS/MS는 제안된 구조(도 8)를 뒷받침하였다.

D. ETM 775의 구조
ETM 775(IC50 0.2 ug/mL 대 L1210 세포)는 HRFAB/MS(도 11)에 의해 776.2489에서 분자 이온을 보여주었다. 이 데이타는 분자식 C39H42N3O12S(△ 0.0 mmu)와 일치하며, 이는 ETM 775가 Et 743의 산화 생성물임을 가리킨다. 양성 및 음성 모드 둘다의 ESI/MS 스펙트럼은 ETM 775의 분자량을 확인시켰다(도 12 및 13). ETM 775의 제한된 양때문에 구조성 부여는 주로 그 매스 스펙트럼 데이타의 해석에 의해 수행되었다. ETM 775(m/z 776)의 M + H상의 FABMS/MS는 m/z 276 및 260(276-산소)에서의 피크에 의해 보여지듯이 N-옥시드의 형성에서 N-2상에 위치한 여분(extra) 산소의 위치를 결정하는데 중요하였다. Et 743에서 관찰되지 않는 m/z 232에서의 분획 이온은 카르비놀 아민 산소가 아미드로 산화된다는 것을 제시하였다(도식 3). ETM 775에서 A- 및 C-단위의 구조는 m/z 204(A-단위), 및 m/z 224 및 250(C-단위)에서의 특징적 매스 스펙트럼 피크의 존재에 의해 보여지듯이 완전한 상태로 남아있다. 750 750 Mhz 1H NMR (도 16) 및 UV (도 17) 스펙트럼 둘다는 Et 743의 것들과 유사하였다.

II. Et 743 - Mayo 대사 연구
A. M1 대사산물 (ETM 305)
ETM 샘플을 C18 셉-팩 및 질소 스트림하에 농축된 용출제(3:1 MeOH/H2O)를 통해 여과하였다. HPLC(상기와 동일한 조건)에 의한 결과 잔류물의 정제는 ETM 305의 것과 동일한 보유 시간(retention time)을 갖는 화합물의 존재를 보여주었다(도 18). M1의 UV(도 19) 및 ESI/MS(도 20) 스펙트럼 둘다는 ETM 305의 것들과 일치하였다. 따라서, M1 대사산물은 ETM 305와 동일한 화학적 구조를 가진다는 결론에 이르렀다.
B. M2 대사산물 (ETM 204)
제공된 샘플을 C18 셉-팩 및 질소 스트림하에 농축된 용출제(3:1 MeOH/H2O)를 통해 여과시키고, 결과의 잔류물을 FAB/MS, ESI/MS 및 1H NMR에 의해 분석하였다.
C. ETM 204(M2)의 구조
ETM 204는 HRFAB/MS에 의해 204.1024에서 분자 이온을 보여주었다. 이 데이타는 분자식 C12H14NO2(△ 0.0 mmu)와 일치한다. ESI/MS 분석은 분자량을 204로 확인시켰다(도 21). 분자식은 Et 743에서 a-단위의 분자식과 매치되었다. 따라서, ETM 204의 화학 구조는 도식 3에서 보여지는 방향족 암모늄 염 유도체일 것으로 제안되었다. 이 단순한 화합물(다른 대사산물뿐만 아니라)은 쉽게 모니터되어 생체내(in vivo)에서 Et 743의 분해를 측정할 수 있다.

ETM 204의 1H NMR 스펙트럼(도 22)은 제안된 구조를 뒷받침하는 공명들을 나타내었다: 4개의 방향족 시그날(δ9.2, s; δ7.8, d, J=5 Hz, 및 δ6.8, s) 및 3개의 메틸 싱글릿(δ4.2, δ3.9 및 δ2.4). ETM 204의 ESI/MS/MS(도 23)는 N-메틸기(204-CH3)의 명백한 결실에 해당하는 189에서의 돌출된 피크 이온을 보여주었다.
ETM-305 및 ETM-775의 생물학적 연구
ETM-305 및 ETM-775의 화합물이 다음 종양 세포주에 대한 표준 프로토콜을 이용하여 분석하였다: P-388(쥐 백혈병); A-549(인간 폐 암종); HT-29(인간 결장 선암종); 및 MEL-28(인간 악성 흑색종). 예컨대, Bergeron et al. Biochem. Biophys. Res. Comm., 1984, 121 (3) 848-854 및 Schroeder et al., J. Med. Chem., 1981, 24 1078-1083을 참조한다. 이들 결과는 하기 표 2에 보여진다.

치료 방법
본 발명은 생활성 화합물을 포함하며, 따라서, 본 발명의 구체예는 그러한 화합물을 이용한 치료 방법에 관한 것이다. 상기와 같이, 본 발명의 화합물은 시험관내(in vitro)에서 종양 세포주에 대한 세포독성을 나타내었다. 이들 시험관내 활성은 생체내(in vivo) 유용성에도 똑 같이 확장될 수 있다고 예상된다.
이들 화합물은 실질적으로 순수 형태, 즉 그것의 물리적 및 생물학적 특성화를 허용하기에 충분한 순도로 단리되었다. 이들 화합물은 특이적 항종양 활성을 가진다고 밝혀졌으며, 그래서 그들은 포유동물, 특히 인간에서 의학적 약제로서 유용할 것이다. 따라서, 본 발명의 또 다른 태양은 상기 활성 화합물을 함유하는 약학적 조성물 및 그 약학적 조성물을 이용한 치료 방법에 관한 것이다.
상기와 같이, 본 발명의 활성 화합물은 항종양 활성을 나타낸다. 따라서, 본 발명은 또한 이들 화합물에 민감한 악성 종양을 앓고 있는 어떤 포유동물의 치료 방법을 제공하며, 이 방법은 병에 걸린 개체에 치료학적 유효량의 활성 화합물 또는 화합물의 혼합물, 또는 그 약학적 조성물을 투여하는 것을 포함한다. 본 발명은 또한 활성 성분으로서 본 발명의 하나 이상의 화합물을 함유하는 약학적 제제 및 그 제조방법에 관한 것이다.
약학적 조성물의 예는 적합한 조성물 또는 경구, 국소 또는 장외적 투여를 갖는 어떤 고체(정제, 알약, 캡슐, 과립 등) 또는 액체(용액, 에멀션의 현탁액)도 포함하며, 그들은 순수 화합물 또는 다른 약학적 활성 화합물의 어떤 담체와 조합하여 포함할 수 있다. 이들 조성물은 장외적 투여시 멸균될 필요가 있을 수 있다.
본 발며에서 "단위 투여량"이란 용어는 동물의 단위 투여에 적합한 물리적으로 구별되는 단위를 의미하며, 각 단위는 필요한 희석제, 즉 담체 또는 부형제와 연합되어 원하는 항종양 효과를 생성하도록 계산된 활성 물질의 예정된 양을 포함한다. 본 발명의 신규 단위 투여량에 대한 상세는 (a) 활성 물질의 독특한 특성과 성취될 특정 항종양 효과, 및 (b) 동물에서 항종양용으로 그러한 활성 물질을 화합분야에 있어서의 내재된 제한에 의해 결정되며 직접적으로 의존한다.
단위 투여 형태는 전형적으로 수성 조성물을 형성하기 위해 물, 염수 또는 포스페이트-버퍼된 염수와 같은 생리적으로 견딜 수 있는(즉, 허용가능한) 희석제 또는 부형제에서의 분산에 의해 냉동 또는 건조 활성 화합물(또는 그의 염)로부터 제조된다. 그러한 희석제는 당업계에 잘 알려져 있으며, 예컨대 Remington's Pharmaceutical Sciences, 16판, Mack Publishing Company, Easton, PA (1980) 페 이지 1465-1467에 논의되어 있다.
투여 형태는 또한 희석제의 일부로서 보조제(adjuvant)를 포함할 수 있다. 완전 Freund's 보조제(CFA), 불완전 Freund's 보조제(IFA) 및 명반(alum)과 같은 보조제는 당업계에 잘 알려진 물질들이고 여러 공급원으로부터 상업적으로 구입가능하다.
투여될 활성 화합물의 양은 특히, 치료될 동물종, 피험 동물의 크기, 종양의 크기(알려진 경우), 존재하는 종양의 유형(예컨대, 고체), 및 피험체가 활성 화합물을 이용하는 능력에 의존한다. 투여될 필요 활성 화합물의 정확한 양은 시술자의 판단에 의존하며, 특히 인간이 치료 동물인 경우 각 개체에 따라 특유적이다. 그러나, 투여량 범위는 치료적으로 유효한 혈액 농도에 의해 특성화될 수 있으며, 약 0.01 μM 내지 약 100 μM, 바람직하게는 약 0.1 μM 내지 10 μM의 농도 범위일 수 잇다.
초기 투여 및 부스터(booster) 주사에 대한 적합한 섭생법은 또한 가변적이나, 전형적으로는 초기 투여후 한 시간 이상 간격으로 후속 주사 또는 기타 투여에 의해 반복 투여된다. 이와 달리, 치료적으로 유효한 혈중 농도를 유지하기에 충분한 연속적인 정맥내 주사가 고려될 수 있다.
