Claims (16)
1. Polypeptid til anvendelse i en fremgangsmåde til behandling, hvilken fremgangsmåde omfatter etablering af kontakt mellem effektorceller, der udtrykker FcyRIIb, og polypeptidet, hvor polypeptidet binder med større affinitet til FcyRIIb-receptoren end et udgangspolypeptid og øger FcyRIIb- receptorens hæmmende aktivitet, og hvor polypeptidet omfatter en Fc-variant af udgangs-Fc-polypeptidet, og hvor Fc-varianten omfatter en aminosyresubstitution S267E ved sammenligning med udgangs-Fc-polypeptidet, hvor nummereringen er i henhold til EU-indekset.
2. Polypeptid til anvendelse i en fremgangsmåde til behandling, hvilken fremgangsmåde omfatter etablering af kontakt mellem effektorceller, der udtrykker FcyRIIb, og polypeptidet, hvor polypeptidet binder med større affinitet til FcyRIIb-receptoren end et udgangspolypeptid og øger FcyRIIb-receptorens hæmmende aktivitet, og hvor polypeptidet omfatter en Fc-variant af udgangs-Fc-polypeptidet, og hvor Fc-varianten omfatter en aminosyresubstitution S267D ved sammenligning med udgangs-Fc-polypeptidet, hvor nummereringen er i henhold til EU-indekset.
3. Polypeptid til anvendelse ifølge krav 1 eller 2, hvor Fc-varianten har en affinitet for FcyRIIb-receptoren, der er mere end 5 gange større end den for udgangs-Fc-polypeptidet.
4. Polypeptid til anvendelse ifølge krav 1 eller 2, hvor Fc-varianten har en affinitet for FcyRIIb-receptoren, der er mellem 5 gange og 300 gange større end den for udgangs-Fc-polypeptidet.
5. Polypeptid til anvendelse ifølge krav 1 eller 2, hvor polypeptidet er et antistof.
6. Polypeptid til anvendelse ifølge krav 5, hvor antistoffet er udvalgt fra gruppen bestående af et humant antistof, et humaniseret antistof og et kimært antistof.
7. Polypeptid til anvendelse ifølge krav 6, hvor antistoffet er et humaniseret antistof.
8. Polypeptid til anvendelse ifølge krav 6, hvor antistoffet er et kimært antistof.
9. Polypeptid til anvendelse ifølge et hvilket som helst af kravene 5 til 8, hvor antistoffet endvidere omfatter en forarbejdet glycoform.
10. Polypeptid til anvendelse ifølge et hvilket som helst af kravene 1 til 9, hvor forøgelsen af FcyRIIb-receptorens hæmmende aktivitet anvendes til at behandle en autoimmun sygdom.
11. Polypeptid til anvendelse ifølge et hvilket som helst af kravene 1 til 9, hvor forøgelsen af FcyRIIb-receptorens hæmmende aktivitet anvendes til at behandle en inflammatorisk sygdom.
12. Fremgangsmåde til forøgelse af bindingen af et Fc-polypeptid til FcyRIIb-receptoren, hvilken fremgangsmåde omfatter substitution i et udgangs-Fc-polypeptid af aminosyren Ser ved position 267 med aminosyren Glu for at tilvejebringe en Fc-variant af udgangs-Fc-polypeptidet, hvor nummereringen er i henhold til EU-indekset.
13. Fremgangsmåde til forøgelse afbindingen af et Fc-polypeptid til FcyRIIb-receptoren, hvilken fremgangsmåde omfatter substitution i et udgangs-Fc-polypeptid af aminosyren Ser ved position 267 med aminosyren Asp for at tilvejebringe en Fc-variant af udgangs-Fc-polypeptidet, hvor nummereringen er i henhold til EU-indekset.
14. Fremgangsmåde ifølge krav 12 eller 13, hvor polypeptidet er et antistof.
15. Fremgangsmåde ifølge krav 14, hvor antistoffet er udvalgt fra gruppen bestående af et humant antistof, et humaniseret antistof og et kimært antistof, og/eller hvor antistoffet endvidere omfatter en forarbejdet glycoform.
16. Fremgangsmåde ifølge et hvilket som helst af kravene 12 til 15, hvor aminosyresubstitutionen sker ved generering af en nukleinsyre, der koder for varianten.
