Description
Gegenstand der Erfindung sind N-Acyl-1-aminoalkan-l.l-diphosphonsäuren
der Formel:

wobei R, R', R" = H oder Alkyl mit C1-C3, R1 -R4 = H
oder Alkali bedeuten, mit der Einschränkung: wenn R und R" = CH3, dann ist R' nicht H und wenn R" = H,
dann ist R = H und R' = Alkyl mit Ci-C3.
Die neuen Verbindungen besitzen eine ausgezeichnete und insbesondere im Vergleich mit den bekannten an
der Aminogruppe nicht acylierten Phosphonsäuren eine wesentlich verbesserte steinverhütende und komplexbildende Wirkung. Darüber hinaus sind die erfindungsgemäßen
Verbindungen leicht in Wasser löslich, was für den Einsatz der Verbindungen sehr wesentlich ist und
woran bisher die Verwendung der entsprechenden nicht acylie/ten Phosphonsäuren in vielen Fällen gescheitert
ist.
Die erfindungsgemäßen Verbindungen lassen sich durch Acylierung der Alkalisalze der entsprechenden
l-Aminoalkan-U-diphosphonsäuren auf einfache Weise
herstellen.
Geeignete Acylierungsmittel sind Säureanhydride, Säurechlorids oder Ameisensäure.
Zur Herstellung der Verbindungen arbeitet man Vorteilhaff so, daß man das Phösphonsäuresälz und das
AcyiierungsmiUet miteinander vermischt und dann bei
gleichzeitigem Rühren unter Rückfluß erhitzt. Das Salz der Phosphonsäure kann in fester Form zugegeben
werden, man kann aber auch das Salz erst im Reaktionsgemisch durch Zugabe von Lauge herstellen
und dann gleich acylieren,
Der Verlauf der Acylierung kann anhand eines Dünnschichtchromatogrammes verfolgt werden, da sich
die Acylierungsprodukte von den Ausgangssubstanzen im RF-Wert stark unterscheiden.
Man erhält eine Reaktionslösung, aus der das Reaktionsprodukt durch Einengen im Vakuum zur
Trockene oder durch Ausfällen mit einem geeigneten Lösungsmittel wie Alkohole, z. B. N-Butar.ol, Propanol-(2);
Eisessig, Aceton u. a. in fester Form gewonnen wird.
Es ist aber auch möglich, die Reaktionslösung ohne Isolierung des Reaktionsproduktes direkt zu verwenden.
Man kann auch gegebenenfalls die Salze der Phosphonsäuren auf übliche Weise durch Neutralisation
oder durch Behandlung mit Austauschern in die freien Säuren überführen.
Die so erhaltenen N-Acyl-l-aminoalkan-l.l-diphosphonsäuren
bzw. deren Salze verhindern schon in unterstöchiometrischen Mengen (Impfmengen) die
Abscheidung steinbildender Ablagerungen in wäßrigen Systemen. Die neuen Phosphonsäuren besitzen darüber
hinaus ein gutes Komplexbindevermr^en gegenüber
zwei- und mehrwertigen Metallionen z. B. Calcium, Magnesium, Eisen, Chrom, Mangan und anderen und
zeigen auch eine korrosionsinhibierende Wirkung. Sie können in feste und flüssige Produkte eingearbeitet
werden, die in wäßrigen Medien einzusetzen sind. Auch sind die neuen Phosphonsäuren mit den üblichen
Waschrohstoffen verträglich und können Wasch- und Reinigungsmitteln zugesetzt werden. Besonders vorteilhaft
ist auch ihr Einsatz beispielsweise in automatisch arbeitenden Flaschenspülmaschinen oder bei der Tank-
und Container-Reinigung.
In der nachfolgenden Tabelle 1 wird die überraschend bessere Impfwirkung der erfindungsgemäßen N-AcylM*
äminoalkän-M-diphosphonsäüreri Im Vergleich zu den
nichtacylierten i-tAminoalkan-l,i-diphosphonsäuren im

Zur Bestimmung der Impfwirkung wurde in einem Substanz 1000-ml-Becb.erglas eine bestimmte Menge der zu
testenden Substanz in 11 Wasser von 18,9°dH gelöst
und 12 g Ätznatron zugegeben. Das Becherglas wurde
mit einem Uhrglas bedeckt und bei Zimmertemperatur stehengelassen. Es wurde dann geprüft, ob sich am
Glasstab bzw. an der Wandung des Becherglases Kristalle abgesetzt hatten.

Zur Bestimmung der Wirkung bei pH 7 wurden 100 ml Wasser bekannter Härte mit 2,0 mg Substanz
so (berechnet als freie Säure) versetzt, auf pH 7 eingestellt
und in einem Wärmeschrank 16 Stunden bei 8O0C gehalten. Dann wurde mit destilliertem Wasser auf
100 ml aufgefüllt, durch ein doppeltes Faltenfilter filtriert, im Filtrat die Resthärte bestimmt und gemäß
DIN 19 640 in m val Erdalkaliionen im Liter umgerechnet (1 m val Erdalkaliionen = 2,8° dH).
Die Komplexbindefähigkeit gegenüber Calciumionen wird in Tabelle 3 gezeigt.

Zur Bestimmung des Komplexbindevermögens gegenüber Caicium-ionen wurde i g der zu prüfenden
Substanz in 70 ml destilliertem Wasser gelöst und unter Rühren durch Zugabe von 1 η NaOH ein pH = 11,5
eingestellt Die klare Lösung wurde mit 10 ml 2°/oiger Sodalösung versetzt und dann tropfenweise eine
0,25molare Calciumacetatlösung zugegeben bis eine permanente Trübung erreicht wurde, d. h., die Zahlen
oder Buchstaben auf einer hinter dem Becherglas aufgestellten Karte nicht mehr gelesen werden konntea
19,1 g (0,1 Mol) Aminomethandiphosphonsäure werden
in 9 ml Wasser (0,5 Mol) und 12 g NaOH (0,3 Mol)
gelöst Danach gibt man 130,14 g (1 Mol) Propionsäureanhydrid dazu und kocht 3—4 Stunden unter ieichtem
Rückfluß. Man erhält die N-Propionylaminomethandiphosphonsäure
in Lösung. Der Verlauf der Acylierung kann anhand eines Dünnschichtchromatogrammes
verfolgt werden. Die N-Acylverbindung besitzt einen kleineren RF-Wert als die nichtacylierte Phosphonsäure.
Laufmittel: 350 ml Propanol-(2), 50 ml Wasser, 20 g Trichloressigsäure gelöst in 80 ml Wasser, 0,5 ml
konzentrierter Ammoniak.
Zur Isolierung der Verbindung wird die Lösung im Vakuum eingeengt Den teilweise kristallisierten dicken
Sirup rührt man in 60 ml n-Butanol an und saugt den reinweißen Niederschlag ab. Nach dem Trocknen erhält
man 28,4 g (90,8% der Theorie) N-Propionylaminomethandiphosphonsäure-tri-Natriumsalz.
Analyse:
Berechnet: N 4,47%, P 19,8%, Na 22,05;
gefunden: N 4,60%, P 19,5%, Na 21,00%.
gefunden: N 4,60%, P 19,5%, Na 21,00%.
20,5 g (0,1 Mol) l-Aminoäthen-l,1-diphosphonsäure
gibt man unter Rühren in eine kalte NaOH-Lösung (18 g Wasser und 4 g NaOH). Man erhält für 2-3 Sekunden
eine klare Lösung des Mono-Natrium-Salzes. Diese erstarrt dann sofort zu einer polymerisatähnlichen
Paste. Diese fast feste Masse trägt man dann in 169,2 g (1,3 Mol) Propionsäureanhydrid ein, kocht 3—4 Stunden
unter Rückfluß und filtriert dann ab. Das Filtrat enthält die N-Propionyl-l'aminoäthan-l.l-diphosphonsäure als
Mono-Natrium-Salz. Der Verlauf der Acylierung wird
anhand eines Dünnscliichtchromatogrammes verfolgt Die N-Acylverbindung besitzt einen kleineren RF-Wert
als die nichtacylierte Phosphonsäure,

Zur Kristallisation kühlt man dk > lösung auf 100C ab.
Dabei erhält man das kristalline Phobpt Dnsäuresaiz.
Analyse:
Berechnet: N 4,95%, P 21,9%, Na 8,1%;
gefunden: N 5,25%, P 23,0% Na 8,2%.

24,9 g (0,1 Mol) N-Methylaminomethan-diphosphonsäure-di-Natrium-Saiz
werden in 51 g (0,5 Mol) Essigsäureanhydrid 2 Stunden unter Rückfluß gekocht
Die klare Lösung des Reaktionsproduktes ist ca. 40%ig und kann direkt eingesetzt werden.
Zur Herstellung des festen Phosphonsäuresalzes engt man die Lösung im siedenden Wasserbad unter Vakuum
ein. Der Rückstand wird pulverisiert in 200 ml Äthylalkohol kurz aufgekocht, abgesaugt und bei 130° C
im Vakuum über Nacht getrocknet
Ausbeute: 27,6 g (95,0% der Theorie) N-Acetyl-N-
methylaminomethan-Diphosphonsäure-di-Natrium-SaIz.
Analyse:
Berechnet: N 4,8%, P 21,3%, Νε 15,8%:
gefunden: N 53b, P 21,8%, Na 15.2%.
gefunden: N 53b, P 21,8%, Na 15.2%.
21,9 g (0,1 Mol) 1-Aminopropan-l, 1-diphosphonsäure
gibt man zu einer kalten Lösung aus 81,7 g (0,8 Mol) Essigsäureanhydrid und 13 g verdünnter NaOH (4 g
NaOH in 9 g H2O), erwärmt unter Rühren und kocht
3-4 Stunden unter Rückfluß. Die entstandene Lösung wird im Vakuum eingeengt, der Rückstand pulverisiert
in Λ Ikohol aufgeschlämmt und getrocknet
Ausbeute: 243 g (88% der Theorie) N-Acetyl-1-aminopropan-1,1
-dipiiosphonsäure-mono-Natriurn-Salz.
Analyse:
Berechnet: N 4,95%, P 215%, Na 8.1%;
gefunden: N 5,1%, P 21,5%. Na 9,1%.
gefunden: N 5,1%, P 21,5%. Na 9,1%.
20,5 g (0,1 Mol) N-Nethylaminomethandiphosphonsäure
suspendiert man in einer verdünnten NaOH aus 9 ml Wasser und 4 g (0,1 Mol) NaOH und gießt dazu
46,03 g (1 Mol) Ameisensäure. Danach bringt man dieses Gemisch zum Sieden. Nach dem Erreichen der
Rückflüßtemperatur bildet sich eine klare Lösung. Die Reaktion ist nach 2,5 Stunden beendet Die Lösung wird
dann unter Rühren in 400 ml Aceton getropft
Nach dem Absaugen und Trocknen des reinweißen Niederschlags erhält man 23,9 g (93,7%) der N-Förmyl·
N-methylaminomethan-diphosphonsäure als Mono-Natrium-Salz.
Analyse:
Berechnet: N 5,5%, P 24,3%, Na 11,1%;
gefunden: N 5,9%, P 24,1%, Na 10,5%.

22,7 g (0,1 Mol) l-Aminoäthan-l.l-diphosphonsäUfemonö-Natfiumsalz
werden in 30,6 g (03 Mol) Essigsäureanhydrid
etwa 30 Minuten unter Rückfluß gekocht. Dabei entsteht eine klare Lösung. Nach Erkalten erhält
man einen dicken gelblichen Sirup. Den Sirup kann man
mit Wasser^verdünnen und die wäßrige Lösung direkt
einsetzen. Zur Kristallisation rührt man den Sirup in 40 ml Eisessig und saugt das entstehende Kristallisat ab.
Nach dem Trocknen beträgt die Ausbeute 20,3 g an
N-Acetyl- 1-aminoäthan-1,1 -diphosphonsäüre-rnono-Natriumsaiz
(75,4% der Theorie).
Analyse:
Berechnet: N 5,2%, P 23,1%, Na 8,55%;
gefunden: N 5,3%, P 24,0%, Na 93%.

Der Verlauf der Acylierung läßt sich dünnschichtehromatographisch
verfolgen. Die N-Acyiverbindung besitzt einen kleineren RF-Wert als die nichtäcylierte
Phosphonsäure.
19,1 g (0,1 Mol) Aminometharldiphosphonsüure werden
zunächst in 9 ml Wasser Und 4 g (0,1 Mol) NaOH
gelöst. Dann gibt man 46,03 g (1,0 Mol) Ameisensäure
idazu und kocht 1 -2 Stunden unter leichtem Rückfluß. Anschließend wird die klare Lösung Unter Rühren in
:etwä 500 ml Methanol getropft. Man erhält 21,3 g
N-Forrnylaminömethändiphosphönsäufe-fnörio-Nätriümsalz
(88,5% der Theorie). ·( .
Analyse:
Berechnet: N 5,8%, P 26,1%, Na 9,5%;
gefunden: N 6,1%, P 25,8%, Na 10%.

Claims (3)
- Patentansprüche:I. N-Acyl-l-aminoalkan-l.l-diphosphonsäuren der Formel:PO3R1R2R'R —C —N4
\CO-R"
PO3R5R4
wobei R, R', R"=H oder Alkyl mit Ci-C3, R1 — R4=H oder Alkali bedeuten, mit der Einschränkung: wenn R und R"=CH3, dann ist R' nicht H und wenn R"=H, dann ist R=H und R'=AIkyl mit C1-C3. - 2. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß man in an sich bekannter Weise die Alkalisalze der l-Aminoalkan-l.l-diphosphonsäurenacyliert.
- 3. Verwendung der Verbindungen nach Anspruch 1 als Komplexbildner für Metallionen oder zur Verhinderung der Steinabscheidung in wäßrigen Medien oder als Korrosionsinhibitoren.
