Herstellung
Die Erfandung betrifft neue physiologisch aktive Verbindungen und
ein Fermentationsverfahren zu ihrer Herstellung.
Gegenstand der Erfindung ist eine neue Klasse von physiologisch aktiven Verbindungen der Formel
(D
in der R ein Wasserstoffatom, eine Hydroxygruppe oder eine 2-Methylbutyryloxygruppe
(eine -OCOCH(CH5)CH2CH5-GrUpPe) bedeutet.
Gegenstand der Erfindung ist außerdem ein Verfahren zur Herstellung
der Verbindungen der Formel (I) durch Züchtung eines Mikroorganismus der Art Penicillium.

Die erfindungsgemäßen neuen Substanzen werden auch als "ML-236"
bezeichnet.
Es wurde gefunden, daß die erfindungsgemäßen neuen Verbindungen der Bezeichnung ML-236 ausgezeichnete physiologische Aktivität
haben, die sie für therapeutische Zwecke geeignet machen. So zeigen sie insbesondere Aktivität zum Inhibieren der Biosynthese
von Cholesterin, Antiatheroskleroseaktivltät und Wirksamkeit gegen.Eyperlipämie.
Als eine Ursache von Stoffwechselerkrankungen, wie Atherpsklerose,
Hyperlipämie und ähnlichen Erkrankungen wird heute die Cholesterinablagerung im lebenden Körper angesehen, speziell in der
Intima von Arterien.
Unter diesen Voraussetzungen wurden systematische Untersuchungen
zur Auffindung von Substanzen gemacht, die aus Kulturbrühen von Mikroorganismen erhältlich sind und die inhibierende Aktivität
auf die Biosynthese von Cholesterin haben, im Hinblick darauf, daß die Inhibierung der Biosynthese von Cholesterin wirksam zur
Verhinderung und Behandlung dieser Krankheiten sein kann. Dabei wurde erfindungsgemäß festgestellt, daß die vorstehend beschriebenen,
als Ml-236 bezeichneten Substanzen aus der Kulturbrühe eines Mikroorganismus des Genus Penicillium isoliert werden
können und daß diese Substanzen ML-236 starke Wirkungen zur Verminderung des Cholesterin- und Lipoid- (Fett)- Spiegels im
Blut und in der Leber haben.
Der Erfindung liegt daher die Hauptaufgabe zugrunde, eine neue Gruppe von Substanzen der Bezeichnung ML-236 der vorstehenden
Pormel (I) zur Verfügung· zu stellen, die hochwirksam als bypocholesterinämische
Mittel und hypolipämische Mittel zur Behandlung von'Ätheroskleröse, Kyperlipämie und ähnlichen Krankheiten
sind.
Es ist ferner Aufgabe der Erfindung, eine Verfahren zur ferm.entatjven

zur Verfügung zu stellen.

.Andere Gegenstände und Vorteile der Erfindung sind aus der nachstehenden
Besenreibung ersichtlich.
Erfindungsgemäß werden einerseits die mit ML-236 benannten Substanzen
der Formel (I) geschaffen. Es wurde nun aufgeklärt, daß
die Substanzen ML-236 der Formel (I) drei verschiedene Einzelsubstanzen darstellen, die als »ML-236 A", "ML-236 B" und
"ML-236 C" bezeichnet werden und die Jeweilige Struktur jeder de: Verbindungen wurde mit Hilfe der IiMR-, Massen-, IR- und UV-Spektren und durch Röntgenbeugung aufgeklärt.

Wenn die erfindungsgemäßen, mit MI-236 bezeichneten Substanzen Patienten verabreicht werden, die an Krankheiten, wie Atherosklerose,
Hyperlipämie und ähnlichen Erkrankungen leiden, so können
sie oral oder parenteral in Form von Kapseln, Tabletten, injizierbaren Zubereitungen und dergleichen verabreicht werden und
es ist gewöhnlich wünschenswert, die Substanz auf oralem Weg zu verabreichen. Die Dosis kann in Abhängigkeit von dem Alter, der
Schwere des Palis, dem Körpergewicht und anderen Eigenschaften des Patienten variiert werden; gewöhnlich liegt jedoch die Tagesdosis
für einen Erwachsenen im Bereich von etwa 200 mg bis 2000 mg und wird in 3 oder 4 Teildosen verabreicht. Erforderlichenfalls
ist es jedoch auch günstig, höhere Dosen einzusetzen.
Wie bereits ausführlich erläutert wurde, haben die erfindungegemäßen
Substanzen der Bezeichnung ML-236 etarke Wirksamkeit zum Inhibieren der Cholesterinbiosynthese in vitro und in vivo.
(I) Die Aktivität zur Inhibierung der Cholesterinbiosynthese der Substanzen MI-236 in vitro kann nach folgender Methode geprüft
werden. Auf Scheiben von Rattenleber läßt man radioaktive Essigsäure bei 37° C 60 Minuten einwirken. Das dabei durch Biosynthese
gebildete radioaktive Cholesterin wird verseift und

als Niederschlag mit Digitonin abgetrennt. Dann wird seine Radioaktivität
gemessen, um die Menge des gebildeten Cholesterins festzustellen. Gesondert davon wird die gleiche Verfahrensweise
.wiederholt, mit der Abänderung, daß zu Beginn der Reaktion HL-236
zugesetzt wird, wobei wieder die Menge des durch Biosynthese gebildeten Cholesterins bestimmt wird. Auf diese Weise kann die
Wirkung von ML-236 quantitativ bestimmt werden (Bricker et al., The Journal of Biological Chemistry, 247, 4914, 1972).
Durch die vorstehend beschriebene Prüfmethode wurde gezeigt, daß
die Substanzen ML-236 A, ML-236 B und ML-236 C in Konzentrationen von 0,04/ag/ml, 0,01 ^g/ml bzw. 0,08/ug/ml eine etwa
50 $ige Inhibierung der Biosynthese von Cholesterin bewirken.
Außerdem wurde die akute Toxizität jeder Substanz ML-236 bei der Maus durch intraperitoneale Verabreichung zu 400 mg/kg oder mehr
bestimmt, was deutlich zeigt, daß die Substanz ML-236 eine niedere Toxizität hat.
(II) Die Wirksamkeit der erfindungsgemäßen Verbindungen ML-236 wurde durch die nachstehend beschriebene Prüfung ihrer Wirkung
zur Erniedrigung des Fettspiegels (Lipoidspiegel) in Blut und Leber von Tieren unter Anwendung verschiedener Testmethoden bestätigt.
Nachstehend werden repräsentative Testmethoden erläutert.
(l) Zum Test wurden zwei Gruppen von Ratten herangezogen, die
jeweils aus 5 Tieren mit einem durchschnittlichen Körpergewicht von etwa 200 g bestanden. Den Ratten einer der beiden Gruppen
wurden jeweils durch intravenöse Injektion 200 mg/kg Triton . WR 1339 (Produkt der Ruger Chemical Co,, Ltd., USA; mit Aktivität
zur Erhöhung des Cholesterinspiegels im Blut) gegeben und gleichzeitig wurden 20 mg/kg ML-236 B intraperitoneal verabreicht.
24 Stunden nach der Verabreichung wurden die Ratten durch Verbluten getötet, das Blut und die Leber wurden entnommen und der
Cholesterin- und Neutralfettspiegel wurde nach einer üblichen Methode bestimmt. Dabei wurde klar festgestellt, daß der
Cholesterinspiegel im Blut um 14,2 # und der Cholesterinspiegel in der Leber um 10,1 % vermindert waren, im Vergleich mit den

Tieren der anderen Gruppe, die ?;u.- durch intravenöse Injektion .
von Triton WR 1339 behandelt worden waren.
(2) Es wurden 4 Gruppen aus jeweils 5 Tieren verwendet. 2 Gruppen erhielten oral eine Dosis von 5 mg/kg ML-236 A als Suspension
in Gummi arabicum. 3 Stunden und 8 Stunden nach der Verabreichung wurden die Tiere durch Verbluten getötet und das Blut wurde gewonnen.
Im Blutserum wurden der Cholesterin- und Neutralfettspiegel mit Hilfe einer üblichen Methode bestimmt. Es wurde gezeigt,
daß nach 3 Stunden der Cholesterinspiegel um 13,5 $ und der
Neutralfettspiegel um 49,5 i° vermindert war und daß auch nach
18 Stunden der Cholesterinspiegel um 13,5 % vermindert war, während
nach 18 Stunden eine Verminderung des Neutralfettspiegels nicht beobachtet wux*de. Diese Ergebnisse wurden im Vergleich mit
den anderen beiden Gruppen erhalten, denen allein Gummi arabicum verabreicht worden war.
Zu dem nachstehenden Versuch wurden 2 Gruppen von jeweils 5 Ratten
verwendet. Einer dieser beiden Gruppen wurden oral 80 mg/kg pulverförmiges ML-236 B in Form einer physiologischen Kochsalzlösung
verabreicht. 3 Stunden nach der Verabreichung wurden in gleicher V/eise wie vorher der Cholesterin- und Neutralfettspiegel
im Blutserum bestimmt. Dabei wurde klar ersichtlich, daß, im Vergleich mit der anderen Gruppe, die eine von Ml-236 B freie
physiologische Kochsalzlösung erhalten hatte, der Cholesterin- und Neutralfettspiegel um 20,0 $ bzw. 44,2 $ vermindert war.
Aus den vorstehend erläuterten Ergebnissen ist ersichtlich, daß die erfindungsgemäßen Substanzen ML-236 hervorragende Wirkung .
zum Inhibieren der Biosynthese von Cholesterin im lebenden Körper haben und daß sie daher als Arzneimittel gegen Krankheiten, wie
beispielsweise Atherosklerose, Hyperlipämie und dergleichen geeignet sind.
Die Erfindung betrifft außerdem ein Verfahren zur Herstellung der physiologisch aktiven Substanzen ML-236 A, ML-236 B und KL-236 C,
das dadurch gekennzeichnet ist, daß ein ML-236-bildender Mikro-

Organismus des Genus Penicillium gezüchtet wird und diese
Substanzen danach aus der KuItür "brühe gewonnen werden.
Die für die Zwecke der Erfindung geeigneten Mikroorganismen sind Ml~236-bildende Stämme des Genus Penicillium. Ein Stamm, der sich
als besonders wirksam zur Bildung der erfindungsgemäßen Verbindungen erwiesen hat, ist beispielsweise Penicillium citrinum
SANK 18767, der unter der Hinterlegungsnuamer 2609 bei dem Technical Research Institute of Microbial Industry, Agency of
Industrial Science & Technology, Ministry of International Trade and Industry, Japan hinterlegt ist und unter der Nummer NRRl-8082
bei dem Northern Regional Research laboratory, Northern Central Region, Agricultural Research Service, United States
Department of Agriculture, in Peoria, Illinois, USA hinterlegt ist.
Die Erfindung wird zwar nachstehend hauptsächlich unter Bezugnahme
auf den Stamm SANK 18767 erläutert; es ist jedoch auf diesem Fachgebiet gut bekannt, daß verschiedene Eigenschaften aller
Mikroorganismen des Genus Penicillium rieht festgelegt sind, sondern daß Mikroorganismen der Art Penicillium durch natürliche
und künstliche Methoden leicht variiert verden können. Für die Zwecke der Erfindung können daher alle Stämme des Genus Penicillium
eingesetzt werden, die zur Bildung von ML-236 befähigt sind, einschließlich Varianten und Mutanien.
Der vorstehend genannte Stamm SANK 18767 ist als solcher bekannt und seine morphologischen Eigenschaften sind in der Veröffentlichung
von K.B. Raper und C. Thorn;. A Manual of the PenicillAa, the Williams and Wilkins Company, 1949 beschrieben.
Zur Durchführung des erfindungsgemäßen Werfahrens kann die Züchtung in zufriedenstellender Weise unter aeroben Bedingungen in
gleicher Weise durchgeführt werden, wie sie auf dem Fachbebiet zur Züchtung eines Stammes der Art PenioJLllium allgemein bekannt
ist. So können beispielsweise die Ml-23i-bildenden Mikroorganismen
in einem Kulturmedium gezüchtet werien, welches beispielswei-

und die so erhaltene Kultur kann in ein Kulturmedium eingeimpft
werden und dort gezüchtet werden, Greniä.t einer ancerer Methode
)cann der in einem Kulturmedium gezüchtete Mikroorganismus in einem
weiteren frischen Kulturmedium unter Bildung von KZ-2;3c gezüchtet
werden.
Als Bestandteile des Mediums können beliebige der gur bekannten
Nährstoffe für Penicillium eingesetzt werden. So eigren sich beispielsweise
als assimilierbare Kohlenstoff quellen Grlueoise, Glycerin,
Maltose, Dextrin, Stärke, lactose, Saccharose, Melassen, Sojabohnenöl, Baumwollsamenöl und dergleichen, wobei vorzugsweise
Glucose und Maltose verwendet werden können« Als assimilierbare
Stickstof !'quelle können Sojabohnenmehl, Erdnussmebi, SauiEWOllsamenmehl,
Fischmehl, Maisquellflüssigkeit, Pepton. Reiskleie,
Fleischextrakt, Hefe, Hefeextrakt, Natriumnitrat, Ammoniuiurjitrat,
Ammoniumsulfat und dergleichen eingesetzt werden. Außerdem können
anorganische Salze, wie Natriumchlorid, Phosphate, Ca.lciUD~
carbonat und dergleichen dem Kulturmedium zugefügt werden= Erforderlichenfalls
kann auch eine geringe Menge eines Metallsalzes zugesetzt werden. Darüber hinaus kann eine geringe Menge eines
Schwermetalls zugesetzt werden, falls erforderlich.
Zum Züchten des ML-236-bildenden Mikroorganismus unter aeroben Bedingungen
können vorteilhaft übliche aerobe Züchtungsmethoden angewendet werden, beispielsweise Kultur in fester Phase, Kultur
unter Belüftung und Rührung, Schüttelkultur und dergleichen.
Bei der Durchführung der Kultur unter Belüftung und Bewegung*kann
in geeigneter Weise eine Antischaummittel, beispielsweise Siliconöl, pflanzliche Öle, oberflächenaktive Mittel und dergleichen eingesetzt
werden.
Der pH-Wert des Medium kann gewöhnlich innerhalb eines Bereiches von 3 bis 9 liegen und befindet sich vorzugsweise innerhalb des
neutralen Bereiches oder nahe am neutralen Bereich. Die Züchtungstemperatur kann gewöhnlich bei 20 bis 30° C liegen, wobei speziell

eine Temperatur von etwa 28° C bevorzugt wird.
Die Züchtung kann fortgesetzt werden, bis ML-236 in den Kulturmedium im wesentlichen angereichert ist, was gewöhnlich 20 Stunden
bis 240 Stunden, vorzugsweise 48 Stunden bis 168 Stunden beträgt.
Nach der Züchtung kann jede der Substanzen ML-236 Kit
Hilfe einer geeigneten Kombination verschiedener Methoder; f in
Abhängigkeit von den erfindungsgemäß festgestellten Eigenschaften
dieser Substanzen aus der Kulturbrühe isoliert und gewonne-n
werden, wie in den nachstehenden Beispielen erläutert wird. So können beispielsweise die Extraktion mit einem organischer: lösungsmittel,
wie Äther, Äthylacetat oder Chloroform, das /.uf?.ösen
in einem stärker polaren Lösungsmittel, wie Acetom oder Alkoholen, das Entfernen von Verunreinigungen mit einen; veneer
polaren Lösungsmittel, z. B. Petroläther oder Hexan, Adsorptionschromatographie
mit Aktivkohle oder Silicagel, GeIfiltratior:
durch eine Säule aus Sephadex (Pharmacia Co. , Ltd. , USA) us\". angewendet werden. Durch Anwendung einer geeigneten Kombination
dieser Maßnahmen kann aus der Kulturbrühe die kristalline oder Ölige Form der erfindungsgemäßen Substanzen isoliert werden.
Die Erfindung wird durch die nachstehenden Beispiele weiter verdeutlicht,
ohne daß sie auf diese beschränkt sein soll. So kann das erfindungsgemäße Verfahren zahlreichen Abänderungen und Modifikationen
unterworfen werden, die leicht ersichtlich sind, insbesondere im Hinblick auf die Gewinnung von ML-236 A, ML-236 B
und ML-236 C aus der Kulturbrühe, wobei in jedem Fall die vorstehend aufgeführten, Eigenschaften dieser Substanzen ausgenützt
werden können. v
In einen Fermenter mit einem Volumen von 600 1 wurden 300 1 eines
Kulturmediums gegeben, das 2 % Glucose, 0,1 # Pepton (Kyo~
kuto der Kyokuto Seiyaku K.K. Japan) und 3 # Malzextrakt enthielt
und in dieses Medium wurde Penicillium citrinum SANK 18767 eingeimpft. Die Züchtung wurde bei einer Temperatur von

28° C, einer Belüftung von 3CO 1/;·.λώ unter'Hühr-n r. ϊ *· l'^O Upra
während 91 Stunden durchgeführt.
Die Kulturbrühe wurde durch eine Filterpresse filtriert, t wobei
280 1 des Filtrats erhalten wurden. Das so erhaltene Filtrat wurde mit 6 η Chlorwasserstoffsäure auf einen pK-Wert von 4,0
eingestellt und zweimal mit 250 1 Äthylacetat extrahiert.
Der Extrakt wurde unter vermindertem Druck zur Trockene eingedampft,
wobei 150 g des getrockneten Produkts erhalten wurden. 120 g des Produkts wurden an einer mit IO4O g Silicagel gefüllten
Säule adsorbiert (Wakogel C-200 der Wako Junyaku K.K. Japan),
die dann in der angegebenen Reihenfolge mit 5 1 Benzol, 7,5 1 Benzol-Äthylacetat (95:5) und dann mit 28 1 Benzol-Äthylacetat
(80:20) entwickelt wurde. Aktive Eluate befinden sich in 2 Fraktionen,
nämlich der ersten mit C bezeichneten eluierten Fraktion (die ML-236 C enthält) und der zweiten mit B bezeichneten eluierten
Fraktion (enthaltend ML-236 B). Dann wurde die Säule mit 2 1 Aceton entwickelt, wobei die dritte, mit A bezeichnete Fraktion
(enthaltend ML-236 A) eluiert wurde. Die zuerst genannten beiden Fraktionen wurden zur Trockene eingedampft, wobei aus der
Fraktion C 2,42 g und aus der Fraktion B 2,61 g der getrockneten Produkte erhalten wurden.
Die Fraktion B (2,61 g) wurde in 100 ml Benzol gelöst, die erhaltene
Lösung wurde mit 100 ml Wasser gewaschen und das Waschwasser v/urde verworfen. Die Benzolschicht wurde über Natriumsulfat
getrocknet und konzentriert und der Rückstand wurde über Nacht stehengelassen, wonach die so abgeschiedene kristalline Substanz
gewonnen wurde. Die Substanz wurde dann einmal aus Benzol uüd. anschließend einmal aus Äthanol umkristallisiert, wobei 232 mg
ML-236 B in reiner Form als weiße Kristalle erhalten wurden. Durch Dünnschichtchromatographie wurde bestätigt, daß es sich um eine
reine Substanz handelte. Sie zeigte auf die Biosynthese von Cholesterin in einer Konzentration von 0,01 yug/ml eine inhibierende
Aktivität von etwa 50 f>.

In einen Fermenter mit einem Fassungsvermögen von 6 t wurden .5000 1 des gleichen Kulturmediums wie in Beispiel 1 gegeben und
dann wurde Penicillium citrinum SANK 18767 eingeimpft. Die Züchtung
wurde in gleicher Weise wie in Beispiel 1 während 90 Stunden durchgeführt. Die Kulturbrühe wurde durch eine Filterpresse filtriert,
wobei 2590 1 Filtrat erhalten wurden. Das Filtrat wurde unter vermindertem Druck auf 450 1 eingedampft und das Konzentrat
wurde mit 6 η Chlorwasserstoffsäure auf einen pH-Wert von 4,0 eingestellt
und dann mit 450 1 Ä'thylacetat extrahiert. 390 1 des Extrakts wurden zur Trockene eingedampft, wobei 346 g einer öligen
Substanz erhalten wurden. Die Substanz wurde an einer mit 5,18 kg Silicagel (Wakogel C-200) gefüllten Säule adsorbiert, die dann
in der angegebenen Reihenfolge mit 98 1 n-Hexan, 60 1 n-Hexan-Aceton
(95 : 5) und schließlich mit 150 1 n-Hexan-Aceton (85 : 15)
entwickelt wurde. Die erhaltene aktive Fraktion (42 1, enthaltend ML-236 B und andere aktive Substanzen) wurde zur Trockene eingedampft,
wobei 40 g des trockenen Produkts erhalten wurden. Dieses Produkt wurde in einer geeigneten Menge Benzol gelöst und unlösliche
Bestandteile wurden abfiltriert. Das Filtrat wurde über
Nacht stehengelassen, wonach ML-236 B in Form von Kristallen abgeschieden war, während andere aktive Substanzen in der Lösung"
verblieben waren. Die Kristalle wurden durch Filtration gewonnen und dann einmal aus Benzol und schließlich einmal aus Äthanol
umkristallisiert, wobei 12,8 g der reinen Verbindung ML-236 B erhalten wurden.
In einen Fermenter mit einem Fassungsvermögen von 6 t wurden 3000 1 eines Kulturmediums gegeben, das 2 $ Glucose, 0,1 $ Pepton
(Kyokuto) und 3 % Malzextrakt enthielt, und in dieses Medium wurde
Penicillium citrinum SANK 18767 eingeimpft. Die Züchtung wurde bei einer Temperatur von 28° C, unter einer Belüftung von 3000 1/
min. und unter Rühren mit 145 Upm während 96 Stunden durchgeführt.

Die Kulturbrühe wurde durch eine Filterpresse filtriert, wobei 2900 1 Filtrat erhalten wurden. Das so erhaltene Filtrat wurde
unter vermindertem Druck auf 450 1 konzentriert und das Konzentrat
wurde dann mit 6 η Chlorwasserstoffsäure auf einen pH-Wert von 4,0 eingestellt, wonach es mit 500 1 Äthylacetat extrahiert
wurde. 500 1 des Extrakts wurden zur Trockene eingedampft, wobei 327 g einer öligen Substanz verblieben. Die Substanz wurde an
einer 5,5 kg Silicagel (Wakogel C-200) enthaltenden Säule adsorbiert, die dann nacheinander mit 10 1 n-Hexan, 60 1 n-Hexan-Aceton
(95 J 5) und dann mit 150 1 n-Hexan-Aceton (85 : 15) entwickelt
wurde. Es wurden zwei aktive Fraktionen erhalten, nämlich eine erste eluierte Fraktion, die ML-236 C enthielt und eine
zweite eluierte Fraktion, die MI-236 B enthielt.
Danach wurde die Säule mit 20 1 Aceton entwickelt, wobei die dritte aktive Fraktion, die ML-236 A enthielt, eluiert wurde.
Die erhaltenen Fraktionen wurden gesondert zur Trockene eingedampft
.
Das trockene Produkt (194 g) aus der ML-236 Α-Fraktion wurde in
2 1 Äthylacetat gelöst und die erhaltene lösung wurde dreimal mit 500 ml-Anteilen einer gesättigten wässrigen Natriumcarbonatlösung
extrahiert. Die Äthylacetatschicht wurde abgetrennt, weiter mit einer gesättigten wässrigen Natriumchloridlösung gewaschen,
über wasserfreiem Natriumsulfat getrocknet und dann zur Trockene konzentriert, wobei 70 g des getrockneten Produkts erhalten
wurden. Das Produkt wurde an einer 150 g Silicagel enthaltenden Säule (Wakogel C-200) adsorbiert, die danach mit 1 1 Benzolvund
3 1 Benzol-Äthylacetat (8 : 2) gewaschen wurde und mit 5 1 Benzol-Methanol
(95 : 5) eluiert wurde. Die Hauptfraktionen wurden zur Trockene eingedampft, wobei 9 g ML-236 A in Form einer öligen
Substanz erhalten wurden.
Das getrocknete Produkt ( 38 g ) aus der ML-236 B-Fraktion wurde in 500 ml Benzol gegeben und unlösliche Bestandteile wurden durch
Filtration entfernt. Das Filtrat wurde konzentriert und das Konzentrat wurde über Nacht stehengelassen, wonach ML-236 3 sich

in Form von Kristallen abgeschieden hatte. Die so abgeschiedenen Kristalle wurden durch Filtration gewonnen und einmal aus Benzol
und danach einmal aus Äthanol umkristallisiert, wobei 10,5 g der Substanz ML-236 B in reiner Form erhalten wurden.
Schließlich wurde das getrocknete Produkt (3,2 g) der ML-236 C-Fraktion
in einer kleinen Menge Dichlormethan gelöst und die erhaltene Lösung wurde an einer 50 g Silicagel (Wakogel C-200) enthaltenden
Säule adsorbiert, die dann nacheinander mit 500 ml Dichlormethan und dann mit Dichlormethan-A'thylacetat (95 : 5) entwickelt
wurde. ML-236 C enthaltende Fraktionen wurden gewonnen
und zur Trockene eingedampft, wobei 2,1g ML-236 C als ölige
Substanz erhalten wurden.

Gegenstand der Erfindung ist außerdem ein Arzneimittel, welches
mindestens eine der erfindungsgemäßen Verbindungen ML-236 A,
ML-236 B oder ML-236 C als Wirkstoff enthält und außerdem übliche pharmazeutisch geeignete Träger oder Exzipienten enthält.

Claims (6)
1. Verbindungen der Formel

in der R ein Wasserstoffatom, eine Hydrox; gruppe oder eine
2-Methylbutyryloxygruppe bedeutet.
2. Verfahren zur Herstellung der Verbindungen ML-236 A, ML-236 B
und MI-236 C der Formeln

dadurch gekennzeichnet , daß man einen Ml-236-bildenden Mikroorganismus
des Genus Penicillium in einem Kulturmedium unter aeroben Bedingungen züchtet und die Substanzen MI-236 in bekannter Weise
aus der Kulturbrühe gewinnt.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet,
daß man als Mikroorganismus Penicillium citrinum SANK 18767 verwendet.
4. Verfahren nach Anspruch 2 oder 3, dadurch gekennzeichnet , daß man die Züchtung bei einer Temperatur im
Bereich von 20 bis 30° C durchführt.
5. Verfahren nach einem der Ansprüche 2 bis 4, dadurch gekennzeichnet , daß man den pH-Wert des Kulturmediums
innerhalb eines Bereiches von 3 bis 9 hält.
6. Verfahren nach einem der Ansprüche 2 bis 5, dadurch gekennzeichnet , daß man die Züchtung während einer
Dauer von 20 bis 240 Stunden durchführt.
