Die Erfindung bezieht sich auf neue substituierte Phenylessigsäuren und betrifft insbesondere gewisse 2-Amino-3-(und
5-)benzoylphenylessigsäuren, Zusammensetzungen davon und ihre Herstellung,

Die Erfindung betrifft insbesondere 2-aminophenylessigsäuren
der allgemeinen Formel:

in der R Wasserstoff oder einen niederen Alkylrest und
R Wasserstoff, Halogen oder einen niederen Alkyl-, Nitro- oder Trifluormethylrest bedeuten.
Die Verbindungen der allgemeinen Formel I sind wirksam hinsichtlich der Verhinderung des entzündlichen Ansprechens
auf Reizung der Pleura (des Ansprechens durch Entzündung auf Brustfellreizungen). Die Aktivität- wurde unter
Verwendung einer Modifikation des Evans-Blue-Carrageenan-Pleural-Effusionstests
von Sancilio, L.F., J. Pharmacol. Exp.
Ther. 168_# 199-2o4 (1969) gezeigt.
Es ist demgemäß Aufgabe der Erfindung neue und brauchbare Verbindungen und Zusammensetzungen davon sowie Verfah-

ren zu ihrer Herstellung zu schaffen, um wirksam Entzündungen bei Brustfeilreizungen verhindern zu können.
Bei der Definition der Symbole in den Formeln und wo sie sonst auftreten, haben die Ausdrücke die folgende Bedeutung:
Der Ausdruck "niederes Alkyl" wie er hier benutzt wird,
schließt gerad- und verzweigtkettige Reste mit bis zu 6 Kohlenstoffatomen, vorzugsweise nicht mehr als A Kohlenstoff- ·
atomen, ein und wird beispielsweise durch solche Gruppen wie Methyl, Xthyl, Propyl, Isopropyl, Butyl, sec. Butyl, tert.
Butyl, Amyl, Isoamyl, Hexyl od.dgl. dargestellt.
Wenn "Halogen" genannt wird, wird vorzugsweise^Jedoch
nicht notwendigerweise,ein Halogen mit einem Atomgewicht nicht größer als 80 angewendet.
Die Herstellung der neuen 2-Amlno-3-benzoyl- und 2-Amino-5-benzoylpheny!essigsauren
(I) der Erfindung kann durch Hydrolyse

von 5-Benzoyl-und 7-Benzoyl-indolin-2—onen (II) In wäßriger
basischer Lösung mit nachfolgender Neutralisation der basischen Reaktionsmischung ausgeführt werden. Die Reaktionsfolge
kann wie folgt veranschaulicht werden:

wobei R und R die oben angegebene Bedeutung haben.
Die Hydrolyse des Indolin-2-on (II) wird in einer verdünnten
wäßrigen Base, z,Be Jn-Natriumhydroxydlösung während
einer Zeitdauer von etwa ο,5 bis 1 Stunde ausgeführt. Die
Hydrolyse kann in einer inerten Atmosphäre unter Verwendung von Stickstoff durchgeführt werden. Die Hydrolysenmischung
kann filtriert werden, um basenunlösliche Materialien zu entfernen, und der pH-Wert der basischen Lösung wird auf 6 bis
durch Zusatz einer-schwachen organischen Säure,wie Essigsäure

oder einer verdünnten Mineralsäure, wie Salzsäure, eingestellt.
Wenn die Verbindung der allgemeinen Formel I eine 2-Amino-5-benzoylessigsäure ist, kann das Produkt leicht
aus einem geeigneten Lösungsmittel umkristallisiert werden. Wenn die Verbindung der allgemeinen Formel I eine 2-Amiho-3-benzoylessigsäure
1st, kann die Umkristalllsation zu einer teilweisen Cyclisierung des 7-Benzoylindolin-2-ons führen.
Daher wird bei der Herstellung einer 2-Amino-3-benzoylessigsäure (I) das Produkt gev/3hnlich nicht umkristallisiert
und es wird durch sorgfältige Ansäuerung der filtrierten basischen Hydrolysenmischung isoliert.
Die Ausgangsmaterialien für das Verfahren gemäß der Erfindung sind die in geeigneter V/eise substituierten
5-benzoylindolin-2- one und die 7-benzoylindolin-2-one (II).
Die entsprechenden Ausgangsmaterialien können durch die folgende Reaktionsfolge hergestellt werden.

Die Indolin-2-one (III) sind entweder im Handel erhältlich
oder sie können nach bekannten Verfahren hergestellt werden, wie sie 2.B4 von Wenkert u.a. in J» Am. Chem.

Die l-Aminoindolin-2-on-Ausgangsmaterialien (IV)
können durch geeignete bekannte Verfahren hergestellt werden, wie sie z.B. durch Baumgarten u.a. in J. Am. Chem.
Soc. 8£, 3977-3982 (i960) beschrieben sind. Das 1-Aminoindolin-2-on
(IV) wird mit einem Phenylaceton zu einem l-( oC-methylphenäthylidenimino)indolin-2-on (V) umgesetzt,
das in äthanolischer Salzsäure au Äthyl-aC-.(2-methyl-3-phenylindol-7-yl)acetat
(VI) cyclisiert wird. Der Ester wird mit Ozon in Essigsäurelösung behandelt, um ein Äthyl-2-acetamido-3-benEoyIphenylaeetat
(VII) zu ergeben, das in verdünnter Mineralsäure zu einem 7-Benzoylindolin-2-on (II) cyclisiert
wird*
Alternativ kann Äthyl-oC-(2-methyl-3-phenylindol-7-yl)-acetat
(VI) in wäßriger basischer Lösung zu einer oC-(2-Methyl-3-phenylindol-7-yl)essigsäure
(VIII) hydrolisiert werden, die mit Ozon in Essigsäurelösung behandelt wird, um eine 2-Acetamido-3~benzoylphenyles.sigsäure (IX) zu ergeben«
Die Säuren werden in verdünnter Säure zu den 7-Benzoylindolin-2-onen
(II) cyclisiert .

Die Herstellungsweisen und Beispiele, die nachstehend
gegeben sind, dienen der näheren Erläuterung der Erfindung.
5-Benzoylindolin-2-on
Ein gerührter Schlamm von 66 g (o,5 Mol) Aluminiumchlorid
und 42,5 g (o,3 Mol) Benzoylehlorid wurde auf 15o°C erhitzt und dann wurden 133 g (o,l Mol) Indolin-2-on langsam '
in einem solchen Verhältnis zugesetzt, daß die Temperatur der gerührten Reaktionsmischung auf 18o bis 185 C gehalten
wurde. Nach der Zugabe wurde die Reaktionsmischung 5 min bei 1850C gerührt, abgekühlt und in Eiswasser gegossen. Das
sich abscheidende 5-Benzoylindolin-2-on wurde gesammelt und aus Methanol umkristallisiert; es schmolz bei 2o4 bis 2o5°C.
Die Ausbeute betrug 17,5 g (73$).

5-Benzoyl-3-methylindolin-2-on
Wenn bei Anwendung des Verfahrens gemäß der Herstellungsweise
1 eine äquimolare Menge an 3-Methylindolin-2-on anstelle von Indolin-2-on zur Anwendung gelangte, wurde 5-Benzoyl-3-methylindolin-2-on
erhalten.

Herstellungsweise 3
Methylphenäthylidenimino)indolin-2-on
Eine Mischung von Io g (o,o7 Mol) l-Aminoindolin-2-on
und 9,o5 g (o,o7 Mol) Phenylaceton in 65 ml absolutem Äthanol
wurde erhitzt, bis das gesamte l-Aminoindolin-2-on aufgelöst war. Die Lösung wurde mit o,5 ml Essigsäure behan- ·
delt und auf einem Dampfbad weitere 15 min erhitzt. Bach dem Abkühlen wurde das Produkt abfiltriert und das Piltrat
wurde mit Wasser behandelt. Das sich aus dem Piltrat abscheidende weitere Produkt wurde mit dem Ausgangsmaterial
vereinigt und aus absolutem Alkohol umkristallisiert. Die Ausbeute betrug 16 g (9o$); F Io2 bis Io4°0.

Herstellungsweise 4
Äthy1-& -(2-methyl-5-phenylind ol-7-yl)acetat
Eine Lösung von äthanolischem Chlorwasserstoff wurde dadurch hergestellt, daß man trockenen Chlorwasserstoff in
loo ml absolutem Äthanol in Blasen durchführte, bis die Lösung zu kochen begann. Bei diesem Punkt wurden Io g (o,o4
Mol) l-(OC-Methylphenäthylidenimino)indolin-2-on zugesetzt
und die Mischung wurde 3o min gerührt. Weiterer Chlorwasserstoff wurde in Blasen durch die heiße Mischung geführt, bis

die Dünnschichtchromatographie anzeigte, daß kein Ausgangsmaterial zurückgeblieben war. Die Reaktionsmischung wurde
abkühlen gelassen und der sich aus der abgekühlten Reaktionsmischung abscheidende Peststoff abfiltriert. Der Peststoff
wurde als l-Aminoindolin-2-on identifiziert. Das FiI-trat
wurde eingeengt und das kernmagnetische ReBonanzspektrum
zeigte, das das rückständige braune Öl eine Mischung aus Phenylaceton und Produkt ist. Die Mischung wurde bei 165 bia
1750C (o,5 mm) abdestilliort. Das ölige Destillat verfestigte
sich beim Abkühlen. Der Feststoff wurde aus Ligroin zu Äthyl-cx'-(2-methyl-3-phenylindol-7-yl)acetat umkriatallisiert,
das bei Io8 bis Io 90C schmolz und ein Gewicht von 2,2 g (21$)
hatte.

Herstellungsweise 4 oC-(2-Methyl-3-phenylindol-7-yl)essigsäure
Einer Lösung von 8 g Kaliumhydroxyd in loo ml Wasser wurden 6 g (o,o2 Mol) Äthyl-oC-(2-methyl-3-phenylindol-7-yl)acetat
zugegeben. Die Mischung wurde 2 Stunden unter Rückfluß
erhitzt. Die abgekühlte Reaktionsmischung wurde filtriert und das Piltrat mit einer gleichen Menge Wasser verdünnt.
Die Aneäuerung der basischen Lösung mit 3n-Saizsäure ergab <X-(2-Methyl-3-phenylindol-7-yl)essigsäure, die aus

Benzol umkristallisiert wurde. Die Ausbeute betrug 3,7 g
(67$)S F 1650C (Zersetzung).

Herstellungsweise 6

Eine Lösung, von 5. g (o,ol7 Mol) Äthyl-oc-(2-methy1-3-phenylindOl-7-yl)
acetate und 75 ml Essigsäure wurde 25 isin
mit Ozon behandelt. UaG-Ii der Qzonbehandlung wurde die Essigßäurelö.aung
mit Wasser Yerdünnt und mit Äther extrahiert. Die Ätäierextrakte wurden mit. Wasser und 5$igem Natriumcarbonat
geviasehen., getrocknet (Magnesiumsulfat) und eingeengt,
Die Umkristallisation aus Isopropanol ergab 2,6 g (47^) des
Produkts, das bei 133 bis 1340G schmolz.

Herstellungsweise 7
2-Acetamido-3-benzoylphenylessigsäure
Eine Lösung von 2g oc-(2~Methyl-3-phenylindol-7-yl)-essigsäure
in 60 ml Essigsäure wurde 15 min mit Ozon behan-

delt. Die Reaktionsmischung wurde mit Io ml Y/asser behandelt
und über Nacht verdampfen gelassen. Der Rückstand (1,7 g) wurde aus Isopropanol umkristalliciert. Die Ausbeute
betrug 1,6 g (71$£); P 188 bis 19o°C.

Heratellungsweise 8 7-Benzoylindolin-2-on
Methode A
Eine Mischung von 2,5 g (o,oo77 Mol) Äthyl-2-acetamido-3-benzoylphenylacetat
in 5o ml 3n-Salzsäure wurde 1 Stunde unter Rückfluß erhitzt. Die Reaktionsmischung wurde filtriert
und das Filtrat in eine Mischung aus Eis und Wasser gegossen. Der Niederschlag wurde gesammelt und aus Aceton umkristalliüiert.
Die Ausbeute betrug 1 g (55$); P 1540C.

Methode B
Eine Lösung von 1,3 g (o,oo44 Mol) 2-Acetamido-3-benzoylphenylessigsäure
in 15 ml 3n-Salzsäure und 15 ml

Essigsäure wurde 3 Stunden unter Rückfluß erhitzt. Die abgekühlte Lösung wurde in Eiswasser gegossen und das sich
abscheidende 7-Benzoylindolin-2-on wurde gesammelt und getrocknet.
Herstellungsweise 9 Andere 5-Benzoylindolin-2-one;
5-(p-Chlorbenzoyl)-indolin-2-on, 5-(o-Fluor"benzoyl)-indolin-2-on,
5-(p-Methoxybenzoyl)indolin-2-on und 5-(m~ Irifluormethylbenzoyl)indolin-2~on werden entsprechend
der Herstellungsweise 1 aus Indolin-2-on und dem entsprechenden
substituierten Benzoylchlorid hergestellt.
Herstellungsweise Io Andere l-(qC -Methylphenäthylidenimino)indolin-2-one:
1_(c^-Methyl-p-chlorphenäthylidenimino)indolin-2-on,
l~(<iC -Methyl-o-f luorphe'näthylidenimino) indolin-2-on und

Andere 2-Acetamido-3-benzoylphenylessif;säuren:
2~Acetamido-3-(p-chlorbenzoyl.) phenylessigsäure,
2-Acetamido-3-( o-f luorb.enzoyl)phenylessigsäure und 2-Acetamido-?- (m-trifluormethylbenzoy1)phenylessigsäure werden
entsprechend der Herstellungsweise 6 durch Ozonbehandlung·
der entsprechenden °C-(2-MetIiyl-3-phenylindol-7-yl) -essigsauren
hergestellt.
Herstellungsweise 15 Andere 7-Benzolindolin-2-one:
7-(p-Chlorbenzoyl)-indolin-2-on} 7-(o-Fluorbenzoyl)~
indQlin-2-on und 7-(m-!Brifluormethylbenzoyl)indolin-2-on
werden entsprechend der Herstellungsweise 7 durch Cyclisierung des entsprechenden Äthyl-2-acetamido-3~benzoylphenylacetats
oder der 2-Acetamido-3-benzoy!phenylessigsäure
hergestellt.

Beispiel 1
2~Amino-5-benzoy!phenylessigsäure
Eine Mischung von l,o g (o,oo4 Mol) 5-Benzoylindolin-2-on
und 3o ml 3n-Natriumhydroxyd wurde o,5 Stunde unter Rückfluß erhitzt. Die Reaktionsmischung wurde abgekühlt,
mit 3n-Chlorwasserstoff auf einen pH-Wert von 6 angesäuert,
und die schwach saure Lösung wurde mit Chloroform extrahiert. Die Chloroformextrakte wurden getrocknet (Magnesiumsulfat)
und zu einem Peststoff eingeengt. Die Umkristallisation
aus Äthanol und Wasser ergab o,9 g (84»5$)
eines reinen Produkts, das bei 143 bis 1450C schmolz.

für C15H13NO3: ber.: C 7o,57; H 5,13; N 5,49
gef.: C 7o,8o; H 5,18} N 5,55

Wenn bei Anwendung des Verfahrens gemäß Beispiel 1
eine äquimolare Menge an 5-Benzoyl-3-methylindolin-2-on an-

stelle von 5-BenzQyl-indolin-2-on zur Anwendung gelangte,
wurde 2~Amino-5-benzoyl-o<: -methy!phenylessigsäure erhalten,,
Beispiel 3
2-Amino-3-benzoylphenylessijs;säure
2-Amino-3-benzoylphenylessijs;säure
Eine Mischung von l,o g (o,oo4 Mol) 7-Benzoylindolin-2-on
wurde zu 3o ml 3n.-N"atriumhydroxyd zugegeben und die basische
Lösung 45 Minuten unter Stickstoff am Rückflußkühler erhitzt. Die Mischung wurde "filtriert und das Filtrat mit
Bisessig neutralisiert. Der Niederschlag wurde abfiltriert,
mit Wasser gewaschen und getrocknet. Das Material hatte einen Schmelzpunkt von 1220C (Zez'setzung). Die Ausbeute betrug
o,8 g (72?$).

In der gleichen Weise wie in Beispiel 1 beschrieben,
wurden

2-Amino-5-(p-raG'thoxybenzoyl)phenylessigsäure und

5-(p-I.Iethoxybenzoyl) indolin-2-on und 5-(m-Trifluorraethylbenzoyl)indolin-2-on
hergestellt.
Beispiel 5
In der gleichen Weise wie in Beispiel 2 beschrieben,
wurden
2-Amino-3-(p-ehlorbenzoyl)phenylessigsäure
2-Amino-3-(o-fluorbenzoyl)phenylessigsäure und
2-Amino-3-(m-trifluormethylbenzoyl)phenylessigsäure
7-(p-Chlorbenzoyl)indolin-2-on
7-(o-Fluorbenzoyl)indolin-2-on und
7-(m-Trifluormethylbenzoyl)indolin-2-on
hergestellt. .
Die vorliegende Erfindung betrifft auch neue, die erfindungsgemäßen
Verbindungen als aktive Bestandteile enthaltende Zubereitungen.. Bei der Herstellung der erfindungsgemäßen
neuen Zubereitungen wird der aktive Bestandteil in einen geeigneten Träger, beispielsweise in einen pharmazeutischen
Träger, eingearbeitet. Beispiele für geeignete phar-

mazeutische Träger, die zur Formulierung der erfindungsgemäßen
Zubereitungen verwendet werden können, sind Stärke, Gelatine, Glucose, Magnesiumcarbonat, Lactose, Malz od.dgl.
Flüssige Zubereitungen fallen ebenfalls in den Rahmen der vorliegenden Erfindung, und Beispiele für geeignete flüssige
pharmazeutis.ehe Träger sind Äthylalkohol,Propylenglykol,
Glycerin, Glucosesirup od.dgl.
Die fo3.genden Beispiele sollen erfindungsgemäß hergestellte
Zubereitungen erläutern.
1. Kapseln
Es wurden Kapseln mit 5o mg und loo mg aktivem Bestandteil
hergestellt.

Weitere Kapselformulierungen enthalten vorzugsweise einen höheren Anteil an Wirkstoff und sind nachstehend

In jedem Pail wird der ausgewählte aktive Bestandteil
mit Lactose, Stärke und Magnesiumstearat gleichmäßig gemischt,
und die Mischung wurde eingekapselt.
2. Tabletten
Eine typische Formulierung für eine. Tablette, die pro Tablette 5o,o mg aktiven Bestandteil enthält, ist folgende:
Die Formulierung kann durch Einstellung des Gewichtes an Dicalciumphosphat für andere Stärken von aktivem Bestandteil
verwendet werden.

Der aktive Bestandteil, Lactose, die Milostärke und die Maisstärke werden gründlich miteinander gemischt. Die-

se Mischung wird unter Verwendung von Wasser als Granulierungsmedium
granuliert. Die feuchten Körnchen werden durch ein Sieb mit einer lichten Masehenweite von 2,36 mm
(8 mesh) passiert und über Nacht bei 6o bis 710C (I4o bis
16o°F) getrocknet. Die getrockneten Körnchen werden durch ·
ein Sieb mit einer lichten Maschenweite von 1,65 mm (Io meeh)
geführt und mit der richtigen Menge an Calciumstearat gemischt, und diese Mischung wird dann in einer geeigneten
Tablettenpresse in Tabletten überführte
sprechens auf Reizung der Pleura können als einzigen
Wirkstoff 2-Amino~3-(oder 5-)benzoy!phenylessigsäure der
angegebenen allgemeinen Pormel I enthalten.

Claims (5)
- PatentansprücheW) 2-Amino-3-(und 5-)benzoylphenylessigsäuren der allgemeinen FormelCHRCOOH NHoin der R Wasserstoff oder einen niederen Alkylrest und R Wasserstoff, Halogen oder einen niederen Alkyl-, Nitro- oder Trifluormethylrest bedeuten.
- 2. 2-Amino-3-"benzoylphenylessigsäure.
- 3. 2~Amino-5-benzoylphenylessig8äure.
- 4- Verfahren zur Herstellung von 2-Amino-3-(und 5-)-benzoylphenylessigsäuren gemäß Anspruch 1, dadurch gekennzeichnet, daß man 5-Benzoyl- oder 7-Benzoylindolin-2-one in wäßriger Lösung hydrolysiert und die basische Reaktionsmischung danach neutralisiert.
- 5. Mittel zur Verhinderung des entzündlichen Ansprechens auf Reizung der Pleura, dadurch gekennzeichnet, daß es als Wirkstoff eine 2-Amino-3-(oder 5-)benzoylphenylessigsäure der allgemeinen Formel309848/1200CHRCOOH (I)enthält, in aer R Wasserstoff oder einen niederen Alkylrest und R Wasserstoff, Halogen oder einen niederen Alkyl-, Nitro- oder Trifluormethylrest bedeuten.309848/1200
