Description
Basisch substituierte Theophyllin-Abkömmlinge
Die Erfindung betrifft neue basisch substituierte Theophyllin-Derivate,
die wertvolle pharmakologische Eigenschaften besitzen.
Es ist bekannt, dass bestimmte, basisch substituierte Xanthin-
Abkömmlinge wertvolle pharmakologische Eigenschaften besitzen. So ist beispielsweise in dem DBP 1 237 578 das
1-[N-Methyl-N-(3·,4'-dimethoxybenzyl)-ß-aminoäthyl]-theobromin
beschrieben, das coronarerweiternde und blutdrucksenkende Wirkungen besitzt.
Es wurde nun gefunden, dass Verbindungen der allgemeinen

sie am Benzolkern Alkoxy-, Methylendioxy-, Benzyloxy-
oder Benzylcarboxy-Gruppen tragen, einer Ätherspaltung unterwirft.
Bei der Verfahrensweise a) arbeitet man zweckmässig in alkoholischer
Lösung mit Alkalialkoholaten als Kondensationsmittel oder in alkoholisch-wässriger Lösung mit Natronlauge,
wobei man die Reaktionspartner einige Zeit unter Rühren am

Rückfluss erhitzt. Ale besondere vorteilhaft hat es sich erwiesen,
eine Suspension von Theophyllin, trockenem Kaliumcarbonat
und der Verbindung II in einem inerten Lösungsmittel - vorzugsweise Aceton oder Butanon - bei Siedetemperatur
zu rühren.
Bei dem Verfahren b) hat es eich bewährt, «ils halogenwasserstoffbindendes
Mittel ein zweites Hol der Base III zuzufügen und die Mischung der beiden Reaktionspartner einige
Stunden im Ölbad auf etwa 1400C zu erhitzen. Fügt man darauf ein geeignetes Lösungsmittel (z.B. Toluol oder Xylol)
in der Wärme zu, so scheidet sich ein Mol Halogenwasserstoffsalz der Base III kristallin aus und kann abgetrennt
werden. Das Filtrat enthält dann das basische Verfahrensprodukt.
Es ist auch möglich, die Umsetzung in siedendem Xylol, oder bei reaktionsfähigem Halogen in Toluol
durchzuführen. Man kann in diesem Falle statt eines zusätzlichen
Mols der Base III auch trockenes Kaliumcarbonat oder tertiäre Basen wie Pyridin oder Triäthylamin als
säurebindendes Mittel einsetzen.
Die gleichen Arbeitsbedingungen gelten für das Verfahren c).
Die Einführung der Methylgruppe in die sekundären Basen der Formel V zur Überführung in die erfindungsgemässen

Verbindungen I kann gemäss Verfahrensweg d) durch Behandeln
mit Methylhalogeniden oder Dimethylsulfat erfolgen,
ferner durch Kochen mit Formalin bzw. Paraformaldehyd und Ameisensäure. Die Methylierung kann auch in der Weise vorgenommen
werden, dass man die mit Formaldehyd versetzte alkoholische Lösung der sekundären Base V in Gegenwart
von Palladium- oder Nickelkatalysator mit Wasserstoff schüttelt, oder mit aiaalgamiertem Aluminium oder Natriumboranat
oder einem anderen geeigneten Reduktionsmittel behandelt.
Um zu den erfindungsgemässen Produkten mit freien phenolischen
Hydroxylgruppen zu gelangen, kann man Verbindungen der Formel I, falls R^ und/oder R„ eine Alkoxygruppe,
eine Benzyloxy- oder Benzylcarboxy-Gruppe oder - abweichend von der Formel I - gemeinsam eine Methylendioxygruppe
bedeuten, mit wässrigen Halogenwasserstoffsäuren in der Wärme behandeln. Dabei kann man, um die Löslichkeit
zu verbessern, erforderlichenfalls ein geeignetes Lösungsmittel wie Eisessig oder Acetanhydrid zufügen.
Wählt man als Entmethylierungsmittel Salzsäure, so ist es in manchen Fällen notwendig, unter Druck zu erhitzen.
Handelt es sich bei den Ausgangsstoffen um. Benzyloxy-
oder Benzylcarboxy-Verbindungen, so können diese auch durch durch Hydrogenolyse mit Palladium-Katalysatoren
in die phenolischen Verfahrensprodukte übergeführt werden.

Eine andere >kjglichkeit der Verfahrensdurchführung besteht
in der Vertvendung gebundener Halogenwasserstoff sauren in Form von Salzen wie beispielsweise Pyridin-ilydrochlorid.
Dabei verfahrt man in der V/eise, dass man die Reaktionskouponenten,
{^ogebenonfallü unter Zusatz eines geeigneten
LöüunGsraittols wie iCiceßsig oder Essigsäureanhydrid bei
erhöhter Temperatur zuuaiunionaclunilzt.
Man kann die Ätherspaltung auch mit organischen Säurehalogeniden bei Abwesenheit oder in Gegenwart von Katalysatoren
durchführen, xrobei die Reaktionsfähigkeit in der
Reihenfolge Saurechlorid - Säurebroraid - Säurejodid zunimmt. Als Katalysatoren verwendet man Metall- bzw. Nichtmetallhalogenide.
Zinkclilorid, Zinn-(IV)-chlorid und Bortrifluorid
sind besondere gut geeignet. Eine weitere Möglichkeit besteht in der Verwendung elektrophiler Metallhalogenide
wie beispielsweise Aluminiumchlorid oder -bromid.

Wirkungsspektrum, das sich gegen endogene (anaphylakcische)
und exogene Histaminreaktionen richtet. Diese
Eigenschaften lassen sie als wertvolle Heilmittel bei der Behandlung einer ganzen Reihe von Krankheiten, z.B.
Asthma' bronchiale, Urticaria, Heufieber, Hauterscheinungen und Allergosen erscheinen.

Am isolierten Ileerschweinchenileum zeigen die neuen Verbindungen
gleichfalls eine hohe histaminolytisehe Wirksamkeit*
wie nach der Methode von Magnus [li. Pflügers Arch, ges. Physiol. 102, 123 (I904)] gezeigt werden kann.
In der Spalte (B) der Tabelle sind die Konzentrationen in jig/ml Badflüssigkeit aufgeführt, die eine vollständige
Hemmung des Ausgangsspasmüs bewirken.
Für die therapeutische Anwendung können die neuen Stoffe als Ampullen, Tabletten, Dragees und Inhalat eingesetzt
werden. Die Dosis beträgt etwa 0,1 - 1,0 mg/kg oral bzw. 0,01 - 0,1 mg/kg parenteral pro Tag.

Die für die Herstellung der neuen Verbindungen benötigten Ausgangsstoffe sind zum Teil noch nicht beschrieben. Sie
lassen sich wie folgt herstellen:
Die Ilalogenalkylamine der Formel II erhält man durch Kon
densation der entsprechenden kernsubstituierten ß-Phenyläthylamine
mit 3-CIilorpropanol. Die entstandenen sekundären
Aminoalkohole werden darauf in üblicher Weise, z. B. mit Hilfe von Formaldehyd und Ameisensäure! am Stickstoff
methyliert, und anschliessend wird die alkoholische Hydroxylgruppe
durch Behandeln mit Thionylchlorid gegen Chlor ausgetauscht. Verwendet man an Stelle der ß-Phcnyläthylamine
ihre N-Monomethyl-Derivate (Formel III)* so gelangt
man in einen Arbeitsgang zu den tertiären Aminoalko holen, und es entfallt die ilethylierung an Stickstoff.
Die Herstellung der 7-(3-Halogcnpropyl)-theophylline sowie
des 7-(3-Aminopropyl)-theophyllino sind bereits in
der Literatur beschrieben.
7-(3-Methylaminopropyl)-theophyllin als freie Base lässt
sich aus 7-(3-IIalogenpropyl)-theophyllin darstellen. Dieses
wird zunächst mit N-Methyl-benzylamin zu 7-[3-(H-IJethyl-N-benzyl)-aminopropyl]-theophyllin
kondensiert und darauf die Benzylgruppe mit Pd/H2 entfernt.

Die Verbindungen der Formel III sind auf verschiedenen Wegen zugänglich. So kann man kernsubstituierte Benzole häufig
durch Behandeln mit Formaldehyd und Salzsäure (Chlormethylierung) in die analogen Benzylchloride überführen.
Diese Benzylchloride können auch aus aromatischen Carbonsauren oder ihren Estern durch Reduktion mit Lithiumalanat
oder aus kernoubstituierten Aldehyden durch katalytische
Hydrierung oder Behandlung mit Reduktionsmitteln und anschliessender Chlorierung der Benzylalko-hate dargestellt
werden Aus den Benzylchloriden sind die entsprechenden Benzylcyanide leicht erhältlich z.B. durch Umsetzung mit
Alkalicyanide!* in Dimethylformamid oder Dimethylsulfoxid.
Aus ihnen gewinnt man dann in bekannter Weise durch katalytische
Hydrierung die ß-Phenyläthylamine. Von den auf
diese Welse erhaltenen primären ß-Phenyläthylaminen gelangt
man dann durch Behandeln ihrer Benzalverbindungen mit Dimethylsulfat oder Jodmethan oder auch durch Reduktion ihrer
N-Formylverbindungen mit Lithiumalanat zu den Aminen
der Formel III.
Die Verbindungen der Formel IV erhält man aus den oben beschriebenen
kernsubstituierten Benzylcyaniden. Diese werden
zu den Phenylessigsäuren verseift, die bei der Reduktion mit Lithiumalanat ß-Phenyläthanole liefern, die z.B.
mit Thionylchlorid in die gewünschten Verbindungen übergeführt werden.

Die Verbindungen der Formel V für den Verfahrensweg d)
sind auf verschiedene Weise erhältlich. Sie lassen sich beispielsweise aus kernsubstituierten ß-Phenyläthylaminen
durch Umsetzung mit 7-(3-Halogenpropyl)-theophyllin
herstellen.
Die Herstellungsverfahren werden durch nachstehende Ausführungsbeispiele
erläutert.
Die Temperaturen sind in °Celsius angegeben. Die Schmelzpunkte sind korrigierte Werte.

Beispiel 1
7-rN-Methyl-N-(ß-3,4-dichlorphenyläthyl)-3-aminopropyl1-theophyllin
Α) Herstellung des Ausgangsmaterials
I. N-Methyl-ß-3,4-dichlorphenyläthylamin
Eine Lösung von 41,2 g (0,217 Mol) ß-3,4-Dichlorphenyläthylamin
(F. Benington et al. J. Org. Chem. 2$ (i960), 2066) wird mit 11,8 g (1,2 χ 0,217 Mol) Ameisensäure
v.ersetzt und 4 Stunden am Wasserabscheider unter Rückfluß gekocht. Dabei werden 8,4 ml Wasser
entfernt. Man verdampft das Toluol, destilliert den Rückstand im Vakuum und erhält 41 ,0 g (90 $ der Theorie)
der N-Formyl-Verbindung vom Kp = I68 - 171°/o,O2
Torr. 41,0 g (0,188 Mol) des so erhaltenen Formamide werden in 200 ml Tetrahydrofuran gelöst und zu einer
im Dreihalskolben siedenden Suspension von 11 g (2 χ
0,75 χ 0,188 Mol) Lithiumalanat in 200 ml Tetrahydrofuran
unter Rühren getropft. Man hält weitere 4 Stunden am Sieden und zersetzt durch Eintropfen von Wasser
und Natronlauge. Man saugt vom Aluminiumhydroxid

ab, dampft das Filtrat ein, löst den Rückstand in 2 N-Salzsäure und schüttelt mit Toluol. Die wässrige
Lösung wird mit konzentrierter Natronlauge versetzt, die Base in Toluol aufgenommen, das Lösungsmittel
verdampft und der Rückstand im Vakuum destilliert. Bei 96 - 97°/0,3 Torr gehen 32,3 g (86 # der Theorie) N-Methyl-ß-S.U-dichlorphenyläthylamin vx ηβ 5
über. Das Hydrochlorid (aus Λlkohoi und Äther)
schmilzt bei 197 - 200°.
II. N-Methyl-N-(ß-3.4-dichlorphenyläthyl)-3-aminochlorpropan
133,5 g (2 χ 0,328 Mol) N-Methyl-ß-3,4-dichlorphenyläthylamin
werden mit 31 ^ (0,328 Mol) 3-Chlorpropanol
in Toluol 2k Stunden unter Rückfluß und Rühren gekocht* Nach Abtrennen dos ausgeschiednen N-Methyl-ß-3,4-dichlorphenyläthylamin-Hydrochlorids
wird das N-Methyl-N-(3-hydroxypropyl)-ß-3,4-dichlorphenyläthylamin,
das nach Abdampfen des Lösungsmittels als Öl zurückbleibt, in Chloroform aufgenommen, tropfenweise mit 43»0 g
(1,1 χ 0,328 Mol) Thionylchlorid versetzt und 2 Stunden unter Rückfluß gekocht. Man nimmt das Reaktionsprodukt nach dem Abdampfen des Chloroforms in heißem
Aceton auf. Beim Abkühlen fällt das Hydrochlorid des

Β) Herstellung des Endproduktes
158,6 g (0,5 Mol) N-Methyl-N-(ß-3,4-dichlorphenyläthyl)-3-aminochlorpropan-Hydrochlorid
werden in die freie Base übergeführt, diese in 100 ml Toluol gelöst und innerhalb
30 Minuten zu einer unter Rückfluß siedenden Mischung von
250 ml Isopropanol, 20 g (0t5 Mol) Natriumhydroxid, 14 ml
Wasser und 90,5 g (0,5 Mol) Theophyllin getropft. Man kocht weitere 3 Stunden, saugt vom ausgeschiedenen Natriumchlorid
ab und dampft das Filtrat ein. Der ölige Rückstand wird in Toluol gelöst. Chlorwasserstoffgas wird bis zur
stark sauren Reaktion eingeleitet. Es scheidet sich das 7-[N-Methyl-N-(ß-3,4-dichlorphenyläthyl)-3-aminopropyl]-theophyllin-Hydrochlorid
als weißes kristallines Pulver aus. Nach dem Umlösen aus Isopropanol erhält man 200 g
(83,4 # der Theorie) des Salzes vom Fp = 225 - 228°. Dieses
Salz enthält 1,5 Mol HCl pro Mol Base und seine wässrige
Lösung reagiert stark sauer. Vermeidet man bei der Herstellung des Salzes Überschuß an Chlorwasserstoff sondern

neutralisiert genau, so scheidet sich das Salz in der Zusammensetzung Base χ 1 HCl aus, das den gleichen
Schmelzpunkt besitzt und dessen wässrige Lösung neutral reagiert.
Die vorstehend beschriebene Kondensation von Theophyllin mit N-Methyl-N-ft-(3i4-diclilorphenyiathyl)-3-aminochlorpropan
lässt sich auch so durchführen, dass man als Lösungsmittel Aceton oder Butanon und als halogenwasserstoffbindendes
Mittel trockenes Kaliumcarbonat verwendet.
A) Herstellung des Ausgangsmaterials
I. N-Itethyl-ß-2,6-dichlorphenyläthylamin
Man orhält die Verbindung aus ft-Dichlorphenyläthylamin
(J. Augstein et al., J. Med. Chem. 10 (1967) 399) durch Reduktion ihrer N-Formylverbindung mit
Lithiumalanat gemäss Beispiel 1 A) X. Das Hydrochloric!
schmilzt bei 171 - 174°.
II. N-Methyl-N-(ß-2,6-dichlorphenyläthyl)-3-aminochlorpropan

Diese Verbindung wird aus N-l-iethyl-ß~2,6-dichlorpheiiyläthylarnin
analog Beispiel 1 A) III durch Kondensation mit Chlorpropanol und anschließende Behandlung
des Aminoalkohols mit Thionylchlorid erhalten. Das
Hydrochlorid schmilzt bei 168 - 170°.
B) Herstellung des Endproduktes
7-f N-Me thyl-N-(ß-2,6-dichlorphenyläthyl)-3-aminopropyl]-theophyllin
90,3 S (0|25 Mol) des N-Methyl-N-ß-2,6-dichlorphenyläthyl)■
^-aminochlorpropan-Hydrochlorids und 45,0 g (0,25 Mol) Theophyllin werden in gleicher Weise wie unter Beispiel
1 B) beschrieben kondensiert und aufgearbeitet. Die Base wird zur Reinigung in das Bioxalat übergeführt, das nach
dem Umlösen aus Isopropanol bei 192 - 195 schmilzt.
Ausbeute: 91,2 g (7I $ der Theorie).
Die aus dem Bioxalat in Freiheit gesetzte Base gibt in Toluollösung mit Chlorwasserstoffgas das Hydrochlorid,
das bei 257 - 260° schmilzt.

durch Kondensation mit 3-Chlorpropanol und anschließende Behandlung des
Aminoalkohols mit Thionylchlorid erhalten. Ihr Bioxalat
schmilzt bei 152 - 15^0.
B) Herstellung des Endproduktes

Priewe et al., Chem. Ber. 90 (1957) 191) und 142,7 g (2 χ
0,42 Mol) N-Methyl-ß-2-chlorphenyläthylamin (Η. Huisgen et
al., Chem. Ber. 92 (1959) 210) zusammengegeben und in Stickst

bis zum Erkalten. Man sougt von 100,2 g N-Methyl-ß-2-cblorphenyläthylamin-Hydrobromid ab, das sich als weißes Kristallpulver abgeschieden hat (Fp β 105 - 107 )» und wäscht das
Toluol-Filtrat mit Wasser· Der Toluol-Phaee wird darauf
durch Schütteln mit 500 ml 1 N-HCl die Base entzogen, die
wässrig-snlzsaure Lösung mit konzentrierter Natronlauge versetzt, die Base in Toluol aufgenommen und nach Trocknen über Kaliumcarbonat im Vakuum eingedampft. Man erhält als Rückstand 133 β (81 io der Theorie) 7-[N-Methyl-N-(ß-2-chlorphenyläthyl)-3-aminopropyl]-theophyllin, das beim Stehen kristallin erstarrt. Eine Probe der Substanz schmilzt nach dem Umlösen aus Xther bei 85 - 90°. Bei der Destillation im Kugelrohr geht sie bei 200 - 210° Luftbadtemperatur/o,001 Torr
bis zum Erkalten. Man sougt von 100,2 g N-Methyl-ß-2-cblorphenyläthylamin-Hydrobromid ab, das sich als weißes Kristallpulver abgeschieden hat (Fp β 105 - 107 )» und wäscht das

Bioxalat: Aus der Lösung der Substanz in Methanol/Äthanol
(1+2) scheidet sich nach Zugabe von Oxalsäure das Bioxalat aus,' das nach dem Umlösen aus Methanol bei 184,5 188°

(C. M. Suter et al., J. Am. Chem. Soc. 63, 609 (1941)). Das abgeschiedene N-Methyl-ß-4-fluorphenyläthylamin-Hydrobromid
wird abgetrennt und die erhaltene Base durch Vakuumdestillation gereinigt. Bei 226°/ü,03
Torr gehen 280,5 g (75 # der Theorie) 7-[N-Methyl-N-(ß-U-fluor-phenyläthyl)-3-aminopropyl]-theophyllin

Löst- man die erhaltene Verbindung in Methanol und leitet
Chlorwasserstoff im Überschuß ein, eo scheidet sich ein
lockeres, weißes Salz aus, das pro Mol Base 2 Mol HCl enthält und bei raschem Erhitzen bei 242 - 244 schmilzt. Seine
wässrige Lösung reagiert stark sauer (5 $» pH 0,85). Neutralisiert
man dagegen die Methanollösung der Base mit genau 1 Äquivalent Chlorwasserstoff, so scheidet sich das Monohydrochlorid
aus, das nach der UmIösung aus Methanol bei
237 - 239 schmilzt (rasches Erhitzen) und dessen wässrige
Lösung neutral reagiert (5 #» pH 6,θ)..
Das Bioxalat wird aus Äthanol erhalten und umgelöst. Fp = 176 - 177,5°.

(K. Kraft, Ber. ök, 150 (1950) und 146 g (1,1 χ 2,02
Mol) 90 %ige Ameisensäure Werden in 1 1 Toluol gelöst
und durch Kochen unter Rückfluß das Reaktionswasser entfernt.
Der Abdampfrücketand der Toluollösung liefert nach
Destillation 532 β (96 % der Theorie) N-Formyl-ß-3-fluor-4-methoxy-phenyläthylamin
vom Kp = 1o2°/o,03 Torr und n-1,5288.
Davon werden 265,0 g (1,3^3 Mol) in 1 1 Tetrahydrofuran
gelöst. Die Lösung wird innerhalb 1 Stunde unter einer Stickstoffatmosphäre und unter Rühren in eine Mischung
von 69 g (1,8 χ 0,75 x 1.343 Mol) Lithiumalanat in 3 1
Tetrahydrofuran getropft, die unter Rückfluß siedet. Man hält 6 Stunden am Sieden und zersetzt die Reaktionslösung
anschließend durch Eintropfen von Wasser und Natronlauge. Das Aluminiumhydroxyd wird abgesaugt, das Filtrat eingedampft,
der Rückstand in Salzsäure aufgenommen und durch Schütteln mit Toluol von Verunreinigungen befreit. Auf
Zugabe von ko %iger Natronlauge scheidet sich die Base
wieder aus, die in Benzol aufgenommen und nach Abdampfen
dea Lösungsmittels destilliert wird. Bei 139 - Ι4θ°/ΐ6
Torr gehen 206 g (84 % der Theorie) N-Methyl-ß-3-fluor-

Α) Herstellung des Ausgangsraaterials
I. fl-3-Tri fluorine thyl-phenyläthylamin
233 e (I,26 Mol)^-Trifluormethyl-benzylcyanid (B. E.
Rosenkranz et al., J. Cham. Eng. Data 8, 237 (1963))
werden in 1,2 1 Methanol, das etwa 80 g Ammoniak enthält,
gelöst und mit Raney-Kobalt bei 100°/100 at
mit Vaseerstoff hydriert. Die vom Katalysator abgetrennte Lösung wird eingedampft, der Rückstand in
Salzsäure aufgenommen und durch Schütteln mit Toluol von Verunreinigungen befreit. Man versetzt die Lösung
mit Natronlauge, nimmt die entstandene Base in Benzol auf, dampft das Lösungemittel ab und destilliert
den Rückstand. Die Verbindung ist ein wasserhelles 01 vom Kp = 88°/14 Torr und n^5 1,4609·
Ausbeute« 195 g (82 $. der Theorie).
II. N-Methyl-ß-3-trifluomtethyl-phenyläthylamin
g (1,ü8 Mol) ß-3-Trifluormethyl-phenyläthylamin

172,8 g (0,8 Mol) der vorstehenden N-Formylverbindung
werden in 1 1 Tetrahydrofuran gelöst und gemäß Beispiel
1 A) I mit 245,5 g (2 χ 0,75 x 0,8 Mol) Lithiuraalanat
in 2 1 Tetrahydrofuran reduziert. Nach dem Aufarbeiten
erhält man 131,5 g (81 $ der Theorie) N-Methyl-ß-3-trifluormethyl-phenyläthylamin
vom Kp = 96 - 100 / 14 Torr und η 1,4598. Das Hydrochlorid, aus Äthanol/
Äther ./'inlten und aus Äthylacetat umkristallisiert,
schmilzt bei 143 - 144°.
B) Herstellung des Endproduktes
Die Umsetzung von 80,1 g (O,27 Mol) 7-(3-Brompropyl)-theophyllin
mit 1θ8,3 g (2 χ 0,27 Mol) des vorstehend
erhaltenen N-Methyl«ß-3-trifluorraethyl-phenylä<t;hylamin

sende Reduktion des Amide mit Lithiumalanat. Man erhält
62 $ der Theorie (bezogen auf primäre Base) der N-Monome

In einem Dreihalskolbeii werden 4?.,2 £ (O,14 Mol) 7-(3-Bromprojjyl)-theophyllin
in 200 ml Toluol gelöst. Es werden 28,7 g (0,i4 Mol) N-Methyl-ß-2,6-dichlorphenylathylamin
(aus dem nach Ä) erhaltenen Hydroehlorid regeneriert), und 14,2 g (O,14 Mol) Triäthylamin zugefügt.
Anschließend wird 16 Stunden unter Rückfluß gekocht. Man saugt vom ausgeschiedenen Triäthylamin-Hydrobrc* id ab,
dampft das Filtrat im Vakuum ein, löst den Rückstand in 100 ml warmem Isopropanol und versetzt mit einer Lösung
von 12,8 g Oxalsäure in Isopropanol· Beim Erkalten scheidet
sich das Bioxalat des gewünschten Produktes aus. Nach Umlösen aus Isopropanol erhält man 28,4 β (46 $ der Theori·)
7-[N-Methyl-N-(fi-2,6-dlchlorph«nyläthyl)-3-aainopropyl]-theophyllin-Bioxalat
vom Fp « 192 - 195 . Anstelle von Triäthylamin kann auch N-Kethyl-ß-2,6-dichlorphenyläthylamin
als säurebindend®s Mittel verwendet werden, von
dem dann die doppelte stöchiomatrischs Menge einzusetzen

Λ) Herstellung des Ausgangsiuaterlals
I. fl-3t5-Dichlorphenyläthylamin
46 g (0,3 Mol) 3,5-Dichiorbenzylcyanid (M. B. Pybue
et al., Ann. Appl. Biol. k7, 593 (1959)) werden in
250 ml Methanol gelöst, das etwa 20 g Ammoniak enthält und bei 5O°/7O at H mit Raney-Kobalt hydriert.
Man trennt vom Katalysator ab, dampft das Filtrat ein, nimmt den Rückstand in Salzsäure auf und entfernt
nichtbasische Verunreinigungen durch Schütteln mit Toluol. Die Base wird durch Zugabe von Natronlauge
wieder frei gesetzt, in Xther aufgenommen, mit Natriumhydroxid
getrocknet und durch Einleiten von Chlorwasserstoffgas das Hydrochlorid abgeschieden.
Nach Auskochen mit Aceton schmilzt das Salz bei 233 - 2360. Die Ausbeute beträgt 39 g (89 £ der
Theorie).
II. N-Methyl-ß-S.S-dichlorphenyläthylamin
Aus dem vorstehend erhaltenen Hydrochlorid wird die Base regeneriert und gemäß Beispiel 1 A) I in die
Formyl-Verbindung übergeführt. Durch Reduktion die-

B) Herstellung des Endproduktes
32,4 g (0,11 Mol) 7r(3-Brompropyl)-theophyllin und 22 g
(0,11 Mol) des vorstehend erhaltenen N-Methyl-ß-315-dichlorphenyläthylainir.s
werden analog Beispiel 9 mit äquivalenten Mengen Triäthylamiii kondensiert. Man neutralisiert
das Toluol-Filtrat vom Triäthylamin-Hydrobromid-Niederschlag
mit Chlorwasserstoff gas, wobei sich das 7-* [N-Methyl-N-(ß-3,5-dichlorphenyläthyl)-3-aminopropyl]-theophyllin-Hydrochlorid
als weißes Kristallpulver ausscheidet. Mach UmIöβen aus Methanol erhält man 28,8 g

ml Tetrahydrofuran gelöst und innerhalb 30 Hinuten
zu einer unter Rückfluß siedenden Mischung von J8 g
(1,6 χ 0,75 x 0,81 Mol) Lithiumalanat und 2 1 Tetrahydrofuran
getropft. Man kocht 30 Minuten nach und zersetzt durch aufeinanderfolgendes Eintropfen von
Wasser und Natronlauge* Das ausgeschiedene Aluminiumhydroxid vird abgesaugt und das Filtrat eingedampft.

Als Rückstand werden I32 g (95 5b der Theorie) 2-Chlor-3-inethoxy-benzylalkohol
als hellgelbes dickes öl erhalten.
II. 2-Chlor-3Tmethoxybenzylchlorid
Die vorstehend erhaltenen 132 g (0,76 Mol) 2-Chlor-3-tnethoxy-benzylalkohol
werden in 300 ml Toluol gelöst und unter Rühren und Eiskühlung innerhalb 1 Stunde
tropfenweise mit 100 g (1,1 χ 0,76 Mol) Thionylchlorid versetzt. Anschließend kocht man die Lösung 1
Stunde unter Rückfluß, dampft im Vakuum ein und reinigt den Rückstand durch fraktionierte Destillation.
Man fängt die zwischen 110 und 12O°/1 Torr übergehen·
.de Fraktion vom xÜ^ 1,5610 auf und erhält so 7k g
(51 % der Theorie) 2-Chlor-3-ιnethoxy-benzylchlorid.
III. 2-Chlor-3-iaethoxy-benzylcyanid
**9 β (0»26 Mol) des vorstehend erhaltenen 2-Chlor-3-methoxy-benzylchlorids
werden unter Rühren innerhalb 30 Minuten bei 30 in eine Suspension von 14 g (1,1 χ

0,262 Mol) Natriumcyanid in 66 ml Dirnethylsulioxid getropft. Anschließend wird 2 Stunden nachgerührt. Daβ
Keaktionsgeraisch wird in 250 iul Wasser gegossen und
mit Äther extrahiert. Die vereinigten Ätherlösungen hinterlassen nach Trocknen und Abdampfen das rohe Nitril, das durch Vakuumdestillation gereinigt wird. Man
erhält 36 g (77 ic der Theorie) 2-Chlor-3-methoxy-benzylcyanid vom Kp = 1 ho /θ, 1 Torr.
Durch katalytische Hydrierung von 3° β (°»2 Mol) des
vorstehend erhaltenen 2-Chlor-3-«eethoxy-benzylcyanids
gemäß Beispiel 8 A) I erhält man 36,5 g (°ö % der Theorie ) ß-2-Chlor-3-methoxy-phenyläthylamin-Hydrochlorid
vom Fp = 124 - 126°.
36,5 g (0,164 Mol) des vorstehend erhaltenen Hydro·
Chlorids werden in die Base verwandelt. Diese wird

analog Beispiel 1 A) I mit Ameisensäure in das Formamid verwandelt, welches anschliessend durch Behandeln
mit Lithiumalanat in N-Methyl-ß^-chlor-S-methoxy-phenyläthylamin
übergeführt wird. Man erhält 24» 5 e (74 % der Theorie) vom Kp =· 90 - 95°/0,3 Torr
und n^ 1,5394· Das Hydrochlorid schmilzt bei 192 195°.
B) Herstellung des Endproduktes
33,2 g (0,12 Mol) 7-(3-Brompropyl)-theophyllin und 22,0 g (0,12 Mol) N-Methyl-ß-2-chlor-3-methoxy-phenyläthylamin
werden gemäss Beispiel 9 mit Hilfe von Triethylamin kondensiert und aufgearbeitet. Die Toluollösung
der Base scheidet nach Neutralisation mit Chlorwasserstoffgas das Hydrochlorid ab, das nach Umlösen
aus Äthanol bei 245 - 248° schmilzt.
Ausbeute: 32,2 g (64 % der Theorie) 7-[ü-Methyl-N-(ß-2-chlor-3-methoxy-phenyläthyl)-3-aminopropyl]-theophyllin-Hydrochlorid.

Beispiel 17
7-rN-Methyl-N-(fl-3.4-dichlorphenyläthyl)-3-aminopropyl1-theophyllin
A) Herstellung der Ausgangsstoffe
I· 7-Γ3-(N-Hethyl-N-benzyl)-aminopropyl!-theophyllin
301,2 g (1 Mol) 7-(3-Brompropyl)-theophyllin und 242,3 g
(2x 1 Mol) N-Methyl-benzylamin werden in Stickstoffatmosphäre
2 Stunden unter Rühren auf 145 erhitzt. Nach Absinken der Temperatur auf etwa 110° lässt man 2 1 Toluol
zufliessen und erhitzt die Mischung 30 Minuten unter Rückfluss. Es scheidet sich ein fein kristalliner
Niederschlag von 202 g N-Methyl-benzylamin-Hydrobromid
(Fp = l60°) ab, der abgetrennt wird. Die Toluollösung
wird mit Wasser gewaschen und ihr dann durch Schütteln mit 1050 ml 1 N-Salzsäure die Base entzogen. Die wässrig
salzsaure Phase scheidet auf Zugabe 40#iger Natronlauge
die Base wieder ab. Sie wird in Toluol aufgenommen und nach Abdampfen des Löeungsmxttels im Vakuum
destilliert. Bei 2 2O°/°»O3 Torr gehen 311 g (91 % der
Theorie) 7-[3-(N-Methyl-N-benzyl)-aminopropyl]-theo-
2 K phyllin als dickes, hellgelbes Öl vom n^ 1,5748 Über.
Die Base scheidet sich aus Äthanol kristallin aus und

schmilzt bei 86 - 88°. Das Hydrochlorid, aus Äthanol erhalten, schmilzt nach dem Umlösen aus Äthanol bei
214,5 - 216°.
II. 7-(3-Methylaminopropyl)-theophyllin
10 g Palladium-Kohle-Katalysator (10 %) werden in
100 ml Äthanol vorhydriert. Man fügt darauf die Lösung von 204,6 g (0,6 Mol) 7-[3-(N-Methyl-N-benzyl)-aminopropyl]-theophyllin
in 400 ml Äthanol hinzu und schüttelt bei 40° mit Wasserstoff. Die berechnete Wasserstoff
menge wird innerhalb 6 Stunden aufgenommen. Nach Abtrennen des Kontaktes und Abdampfen des Äthanols
verbleibt ein Öl, das man im Vakuum destilliert. Bei 192 - 194°/°>°3 Torr gehen 137 g (91 % der Theo-

Zu einer Lösung von 424,0 g (1,1 χ 3,24 Mol) Thionylchlorid in 300 ml Diäthyläther tropft man unter Rühren
innerhalb 2 Stunden die Mischung Von 618,5 g (3,24 Mol) ß-3,4-Dichlorphenyläthanol (R. Fuchs, J.
Ata. Chem. Soc. 78, 5612 (1956)), 256,2 g (3,24 Mol)

trockenem Pyridin und 300 ml Diäthyläther. Die Mischung erwärmt sich dabei zum Siedenr, wobei gleichzeitig Xther
abdestilliert ι Man erwärmt schliesslich im Wasserbad bis etwa 500 ml Destillat Übergegangen sind, wobei die
Innentemperatur auf etwa 85° ansteigt. Nach Zugabe von 42 g Thionylchlorid rührt man bei dieser Temperatur
noch 2 Stunden nach. Das erkaltete Reaktionsgut wird in 1 1 Äther gelöst, in 1 1 Eiswasser gegossen und die
wässrige Schicht nach Abtrennen der Ätherlösung noch zweimal ausgeäthert. Die vereinigten Ätherauszüge werden mit Wasser und Natriumhydrogencarbonat-Lösung gewaschen, mit Kalziumchlorid getrocknet und eingedampft.
Der Rückstand geht bei 142 - 1450/11 Torr als farbloses Öl vom n^ 1,5631 Über. Die Ausbeute beträgt 602 g
(89 % der Theorie).
I5818 g (2 χ 0,316 Mol) des nach II erhaltenen 7-(3-Methylaminopropyl)-theophyllin und 66,1 g (0,316 Mol)
ß-3,4-Dichlorphenyläthylchlorid (nach III) werden zusammengegeben und in einer Stickstoffatmosphäre unter Rühren 3 Stunden auf 145° erhitzt. Man lässt auf 110° abkühlen, fügt 1 1 Toluol hinzu und kocht 30 Minuten unter Rückfluss. Das ausgeschiedene 7-(3-Methylaminopro-

pyl)-theophyllin-Hydrochlorid (90 g) wird abgesaugt. Es
schmilzt bei 263 - 265°. Das Toluol-Filtrat wird zunächst mit Wasser gewaschen und ihm dann durch Ausrühren
mit 2,7 1 heisser N/l0-Salζsäure das Endprodukt entzogen.
Man versetzt die wässrig-salζsaure Lösung mit
40$iger Natronlauge, nimmt die abgeschiedene Base in Toluol
auf und trocknet Über Kaliumcarbonat. Nach Abdampfen
des Lösungsmittels verbleiben 103 g 7-[N-Methyl-N-(ß-3,4-dichlorphenyläthyl)-3-aminopropyl]-theophyllin.
Eine Probe geht bei der Destillation im Kugelrohr bei 210 - 220°/0,001 Torr Luftbadtemperatur ale blassgelbes
sehr zähes Öl über. n£5 1,5840.
Hydrochlorid: Löst man 92 g der Base in einer warmen Mischung
von 150 ml Äthanol und 300 ml Äthylacetat und leitet Chlorwasserstoffgas bis zur schwach sauren Reaktion
ein, so scheiden sich 95 S des Salzes aus. Man löst aus
Methanol um und erhält 91 g Hydrochlorid vom Fp =» 226 228
, das sich als chromatographisch rein erweist.

werden gemäss Arbeitsvorschrift von Beispiel 17 A) III durch Behandeln mit Thionylchlorid und Pyridin
in Diäthyläther-Lösung chloriert. Nach dem Aufarbeiten erhält man 770,6 g (94 % der Theorie) ß-3-Chlor-4-methoxy-phenyläthylchlorid vom Kp «■ 102°/0,005 Torr
und n*5 1,5555.
132,2 g (2 χ 0,27 Mol) 7-(3-Methylaminopropyl)-theophyllin und 55,6 g (0,27 Mol) ß-3-Chlor-4-methoxy-phenyläthylchlorid werden gemäßs Beispiel 17 B) kondensiert
und aufgearbeitet. Man erhält 111 g 7-[N-Methyl-N-(ß-3-chlor-4-methoxyphenyläthyl)-3-aminopropyl]-theophyllin.
Eine Probe der Substanz geht bei 225 - 23O°/O,OO1 Torr

A) Herstellung des Vorproduktes
7~ F N-(fi-3»4-dichlorphenyläthyl)-3-aminopropyl1-theophyllin
In einem Dreihalskolben werden 38,0 g (0,2 Mol) ß-3,4-Diclilorphenyläthylamin
(F. Benningfcon et al., J. Org. Chera. 25, 2066 (I960)) und 20,2 g (0>2 Mol) Triäthylamin mit
100 ml Toluol zusammen gegeben. Unter Rühren lässt man
nun bei Zimmertemperatur innerhalb 90 Minuten die Suspension aus 6O,2 g (0,2 Mol) 7-(3-Brompropyl)-theophyllin
und 300 ml Toluol zufliessen und kocht anschliessend 16 Stunden unter Rückfluss. Man saugt von dem abgeschiedenen
Triäthylamin-Hydrobromid (gegebenenfalls vom ß-3,4-Dichlorphenyläthylamin-HydrobPomid)
ab, wäscht das Filtrat mit Wasser und dampft im Vakra» ei»<>

wird in 100 ml Isopropanol gelöst. Nach Zugabe von 15 ml 48#iger wässriger Bromwasserstoffsäure scheiden sich Kristalle aus, die isoliert und aus Methanol umgelöst werden.
Man erhält 57,5 g (59 % der Theorie) 7-[N-(ß-3,4-Dichlorphenyläthyl)-3-aminopropyl]-theophyllin-Hydrobromid vom
Fp - 243 - 245°.
40,4 g (0,1 Mol) 7-[N-(ß-3,4-Dichlorphenyläthyl)-3-aminopropylj-theophyllin (aus dem vorstehend erhaltenen Hydrobromid freigemacht) werden in 370 ml Äthanol gelöst und
mit 7,4 g (1,6 χ 0,1 Mol) Ameisensäure versetzt. Man fügt
darauf 11,1 g (1,3 χ 0,1 Mol) 35$ige wässrige FormalinLösung hinzu und kocht die Mischung 2 Stunden unter Rückfluss. 0er Abdampfrückstand der Reaktionslösung wird in
verdünnter Salzsäure aufgenommen und mit Toluol geschüttelt. Aus der wässrig-salζsauren Lösung scheidet man mit
Natronlauge die Base ab, die in Toluol aufgenommen und getrocknet wird. Beim Einleiten von Chlorwasserstoffgas
in die Toluol15sung scheidet sich das 7-[N-Methyl-N-(ß-3,4-dichlorphenyläthyl)-3-aminopropyl]-theophyllin-Hydrochlorid aus. Es wird aus Isopropanol umkristallisiert.
Man erhält 39,3 g (82 % der Theorie) des Salzes vom Fp
= 225 - 228°.

Zum gleichen Ergebnis gelangt man, wenn man die alkoholische
Lösung des 7-[N-(ß~3,4-Dichlorphenyläthyl)~3-aminopropyl]-theophyllin
mit der berechneten Menge Formalin-Lösung versetzt und mit Natriumboranat 2 Stunden
auf dem Wasserbad erhitzt,. oder mit Palladium-Kohle-Katalysator
bei 40° mit Wasserstoff schüttelt*
7- [N-Methyl-N- (ß- S-chlor^-hydroxy-phenyläthyl )-3-aminqpropyl!-theophyllin
56,1 g (0,13 Mol) 7-[N-Methyl-N-(ß-3-chlor-4-methoxy-phenyläthyl)-3-aminopropyl]-theophyllin
(Herstellung siehe Beispiele 14 und 18) werden mit 120 ml 48^iger wässriger Bromwasserstoffsäure
(D = 1,5) entsprechend 96,4 g HBr (2,18 χ 2 χ 0,13 Mol) 2 Stunden unter Rückfluss gekocht. Man fügt weitere
60 ml 48<ige Bromwasserstoffsäure (4,1 χ 0,13 Mol) hinzu
und kocht noch 1 Stunde. Das Reaktionsgemisch wird in 1 Eiswasser gegossen und mit 4Obiger Natronlauge genau neutralisiert.
Dio Phenolbase scheidet sich dabei als Öl ab, das beim Stehen kristallisiert. Man löst die Substanz in 2 Ν-3ε1ζ-säure,
worauf sich ihr Hydrochlorid in Form weisser Kristalle ausscheidet. Nach dem Umlösen aus Aceton erhält man 25,2 g

Analog Beispiel 20 werden 10,7 g (0,03 Mol) 7-[N-Methyl-N-(ß-3-methoxy-4-chlor-phenyläthyl)-3-aminopropyl]-theophyllin
(Herstellung siehe Beispiel 15) mit 34,4 ml 48#iger wässriger Bromwasserstoffsäure der Ätherspaltung unterworfen.
Die nach der Aufarbeitung erhaltene rohe Phenolbase wird in Aceton aufgenommen und mit der Lösung von 3,5 g Maleinsäure
in Aceton versetzt. Die nach kurzer Zeit abgeschiedenen Kristalle werden aus Aceton umgelöst. Man erhält 43 g (32 %
der Theorie) 7-[N-Methyl-N-(ß-3-hydroxy-4-chlor-phenyläthyl)-3-aminopropyl]-theophyllin-Maleinat
vom Fp «= 190 193°.

das nach dem Umlösen aus Aceton bei 195 - 196° schmilzt.
Beispiel 23
7-[N-Methyl-N-(ß-3j 4-dihydroxy-phenyläthyl)-3-aminopropyl1-theophyllin
Im Dreihalskolben versetzt man 226 g (0,5 Mol) 7-(N-Methyl-N-homoveratryl-3-aminopropyl)-theophyllin-Hydrochlorid
(hergestellt nach DBP 1 237 578) mit 672 ml 48$iger wässriger
Bromwasserstoff säure (D = 1,5) = 485 g HBs* (4 x 3 x 0,5
Mol) und bringt die Mischung in Stickstoffatmosphäre zum
Sieden. Bereits bei 80° beginnt eine lebhaft© Broisssaethan-Entwicklung,
die nach Sstündigem Kochen unter Rückfluss
zum Stillstand kommt. Danach sind 96 % der berechneten Brommethan-Menge
abgespalten. Die klare Lösung erstarrt nach dem Erkalten zu einem dicken Kristallbrei der abgesaugt
und mit Isopropanol gewaschen wird. Man erhält 248 g kristallwasserhaltiges

Hydrochlorid: 100 c des vorstehenden Hydrobromids werden in
2,5 1 heissem Wasser gelöst. Nach dem Erkalten fügt man bei
Zimmertemperatur unter Stickstoff 500 ml 2 N-Ammoniak hinzu,
worauf sich eine schwach rosa gefärbte Substanz ölig abscheidet. Man dekantiert die wässrige Phase ab, wäscht
die Substanz mit Wasser und löst sie in 2 1 warmem Äthanol. In die klar filtrierte Lösung leitet man Chlorwasserstoffgas
im Überschuss ein, worauf sich 78 g 7-[N-Methyl-N-(ß-3,4-dihydroxy-phenyläthyl)
3-aminopropyl]-theophyllin-Dihydrochlorid als weisses Kristallpulver vom Fp = 208 - 213°
abscheiden. Die Ausbeute der Reaktion beträgt 84 % der Theorie. Versetzt man die 5/^ige wässrige Lösung des Dihydrochloride
(pH 1,0) mit 1 Äquivalent Natronlauge, so erhält man die wässrige Lösung des Monohydrochloride (pH 6,7). Das Monohydrochlorid
gibt in Wasser oder Methanol mit Eisen (III)-chlorid eine tief dunkelgrüne Färbung, die auf Zugabe von
Natronlauge in violett umschlägt.

die erkaltete farblose Lösung mit dem gleichen Volumen Diäthyläther, worauf sich das 7-[N-Methyl-N-(ß-3-fluor-4-hydroxy-phenyläthyl)-3-aminopropyl]-theophyllin-Hydrobromid
kristallin abscheidet. Nach Umlösen aus Isopropanol erhält man 59»4 g (92 % der Theorie) des Salzes als weisses Kristallpulver
vom Fp = 72,5 - 75°· Bei weiterem Erhitzen erstarrt die Schmelze bei etwa 145° und schmilzt dann bei
225 - 227° erneut.
53»6 g (0,11 Mol) des vorstehenden Hydrobromids werden in
1,3 1 warmem Wasser gelört und in Stickstoffatmosphäre
durch Eintropfen von 114 ml 1 N-Natronlauge auf den pH-Wert
7>0 eingestellt. Es scheiden sich 43,5 g Base als weisses Kristallpulver vom Fp = 130 - 132,5° ab, der sich
nach Umlösen aus Isopropanol nicht ändert. Die Lösung der Base in Methanol gibt mit Eisen(lll)-chlorid eine gelbrote Färbung.
Löst man die trockene Base in Methanol und leitet Chlorwasserstoff
