Claims (19)
1. Ein Material, das in einem Bindungstest zur Bestimmung des Vorliegens oder der Menge eines Analyten in einer
Testprobe nützlich ist, welches Material eine poröse Matrix aus Fasern und eine Mehrzahl von im wesentlichen kugelförmigen,
festen Teilchen mit einem mittleren Durchmesser von 0,1 bis 10 Mikron enthält, wobei die Teilchen in der
genannten Matrix auf den Fasern zurückgehalten werden lind immobilisiert sind.
2. Das Material von Anspruch 1, wobei die genannten Teilchen
auf ihren Oberflächen eine Substanz aufweisen, die mit dem Analyten in der Probe reagieren kann.
3. Das Material von Anspruch 1, wobei der mittlere Durch-
messer der Teilchen kleiner ist als die mittlere Porengröße der·
Matrix.
4. Eine FestphaseOf-Testeinrichtung, die in einem Bindungstest zur Bestimmung des Vorliegens oder der Menge eines Analyten
in einer flüssigen Probe nützlich ist, wobei die Einrichtung eine im wesentlichen ebene Schicht eines Materials mit
einer porösen Fasermatrix und einer Mehrzahl von im wesentlichen kugelförmigen, festen Teilchen mit einem mittleren
Durchmesser von 0,1 bis 10 Mikron enthält, wobei die Teilchen innerhalb der genannten Matrix auf den Fasern zurückgehalten
werden und immobilisiert sind, und wobei die im wesentlichen ebene Schicht eine erste, Proben-kontaktierende Oberfläche
und eine zweite, der ersten Oberfläche gegenüberliegende Oberfläche aufweist und in der Einrichtung derart angeordnet
ist, daß bei Benützung der Einrichtung bei der Ausführung des Tests wenigstens ein Teil der mit der ersten Oberfläche
in Berührung stehenden Probe durch die im wesentlichen ebene Schicht zu der zwei ten überfläche gelangt.

5. Die Testeinrichtung von Anspruch 4, wobei die Einrichtung zusätzlich Filtermittel enthält, die derart in bezug auf
die erste Oberfläche der im wesentlichen ebenen Schicht angeordnet sind, daß die Probenflüssigkeit durch die genannten
Filtermittel hindurchgeht, bevor sie mit der ersten Oberfläche in Berührung kommt.
6. Die Testeinrichtung von Anspruch 4, wobei die Einrichtung zusätzlich absorbierende Mittel enthält, die derart in
bezug auf die untere Oberfläche der im wesentlichen ebenen Schicht angeordnet sind, daß wenigstens ein Teil der durch
die im wesentlichen ebene Schicht hindurchgehenden Flüssigkeit durch die absorbierenden Mittel absorbiert wird.
7. Die Testeinrichtung von Anspruch 6, wobei die Einrichtung zusätzlich eine Sperrschicht enthält, die zwischen der
im wesentlichen ebenen Schicht und den absorbierenden Mitteln angeordnet ist, wobei die Sperrschicht derart
ausgebildet ist, daß die von den absorbierenden Mitteln zurückgehaltene Flüssigkeit an der Rückkehr zu der im
wesentlichen ebenen Schicht behindert wird.
8. Die Testeinrichtung von Anspruch 7, wobei die Sperrschicht
aus einein gewebten Material aus Polyethylen zusammengesetzt ist.
9. Ein Bindungstest zur Bestimmung des Vorliegens oder der Menge eines Analyten in einer Testprobe, umfassend
die Stufen:
a) des Inberührungbringens einer porösen Fasermatrix und einer Mehrzahl von im wesentlichen kugelförmigen,
festen Teilchen mit einem mittleren Durchmesser von 0,1 bis IO Mikron, wobei die Teilchen innerhalb der genannten
&Sgr;-Iatrix auf den Fasern zurückgehalten werden und immobilisiert
sind,mit der Probe, wobei der Analyt in der Probe an die Teilchen unter Bildung eines Analytkomplexes auf
den Teilchen gebunden wird;

b) des Inberührungbringens des den Komplex auf den Teilchen enthaltenden Materials mit einer zweiten Substanz,
die mit dem Analyten reagieren kann, und welche mit einer Substanz markiert ist, die eine feststellbare Antwort
in Gegenwart des Analyten und einer Indikatorsubstanz hervorbringen kann, wobei die markierte Substanz an den
Komplex auf den Teilchen gebunden wird;
c) des Entfernens von ungebundener, markierter Substanz aus dem Material;
d) des Inberührungbringens des Materials mit der Indikatorsubstanz; und
e) des Feststeilens der hervorgebrachten Antwort
als eine Funktion des Vorliegens oder der Menge des Analyten
in der Probe.

10. Der Test von Anspruch 9, wobei die feststellbare Antwort ein visuell feststellbares "+"-Zeichen für den Fall ist,
daß feststellbare Mengen von Analyt in der Probe vorliegen, und ein visuell feststellbares "-^"-Zeichen ist, falls
feststellbare Mengen von Analyt in der Probe nicht vorliegen.
11. Der Test von Anspruch 7, wobei der Analyt beta-hCG,
Strep-A-Antigen oder HBsAG ist.
12. Der Test von Anspruch 9, wobei die Teilchen auf ihrer
Oberfläche eine Substanz aufweisen, die mit dem Analyten in der Probe reagieren kann.
13. Ein Verfahren zur Erzeugung eines Materials, das zur Bestimmung des Vorliegens oder der Menge eines Analyten
in einer Flüssigkeit nützlich ist, umfassend die Stufen: a) des Bebrütens einer Mehrzahl von im wesentlichen
kugelförmigen, festen Teilchen mit einem mittleren Durchmesser
von etwa 0,1 bis etwa 5 Mikron mit einer Probe der Flüssigkeit, wobei die Teilchen auf ihren Oberflächen
eine Bindüngssubstanz immobilisiert aufweisen, welche mit dem Analyten reagieren kann, wodurch der Analyt an
die Substanz unter Bildung eines Analyt/BindungssubStanz-Komplexes
auf den Teilchen gebunden wird; und

b) des Inberuhrungbringens einer porösen Fasermatrix mit den bebrüteten Teilchen, wobei wenigstens ein Teil
der Teilchen innerhalb der Matrix auf wenigstens einem Teil der Fasern zurückgehalten und immobilisiert wird.

14. Eine Festphasen-Testeinrichtung zur Verwendung in
einem Bindungstest zur Bestimmung des Vorliegens oder der Menge eines Analyten in einer flüssigen Probe, umfassend
eine Reaktionsoberfläche mit einem negativen Kontrollbereich,
einem positiven Kontrollbereich und einem Analytbindungsbereich in derartiger Anordnung, daß der positive Kontrollbereich
mit dem Analytbindungsbereich unter Bildung eines ersten Begriffsymbols mit einer ersten Bedeutung für das
Vorliegen eines positiven Testergebnisses bei der Verwendung
der Einrichtung,und unter Bildung eines zweiten Begriffsymbols, das von dem ersten Begriffsymbol verschieden
ist, bei Vorliegen eines negativen Testergebnisses bei der Verwendung der Einrichtung in Wechselwirkung tritt.
15. Die Festphasen-Testeinrichtung von Anspruch 14, wobei der positive Kontrollbereich auf der Reaktionsoberfläche
in der Form eines rechteckigen Balkens gebildet wird.
16. Die Festphasen-Testeinrichtung von Anspruch 15, wobei der Analytbindungsbereich auf der Reaktionsoberfläche
in der Form von zwei rechteckigen Balken an entgegengesetzten Seiten des und senkrecht orientiert zu dem positiven
Kontrollbereich gebildet wird, wobei der Analytbindungsbereich zusammengenommen mit dem positiven Kontrollbereich
die Form eines "+"-Zeichens bildet, und wo der positive Kontrollbereich allein unter Bildung eines "-"-Symbols
bei Vorliegen eines negativen Testergebnisses wirksam wird.
17. Die Festphasen-Testeinrichtung von Anspruch 14, wobei der positive Kontrollbereich und der Analytbindungsbereich
in Form von Punkten gebildet werden.
18. Die Festphasen-Testeinrichtung von Anspruch 14, wobei der negative Kontrollbereich den positiven Kontrollbereich
und den Analytbindungsbereich umgibt.
19. Die Festphasen-Testeinrichtung von Anspruch 14, mit einer Flüssigkeitskammer zur Aufnahme einer flüssigen
Probe und von bei der Ausführung des Tests verwendeten Reagenzien, wobei die Flüssigkeitskammer durch wenigstens
eine Seitenwand und eine Basiswand begrenzt wird, wobei die Basiswand die Reaktionsoberfläche derart umfaßt, daß
sie mit einem einzigen Volumen der Flüssigkeitsprobe und der Reagenzien, die beim Test verwendet werden, gleichzeitig
in Berührung kommt.
