Abstract
Die vorliegende Erfindung betrifft neue, spezifische und effektive Prodrug-Verbindungen der allgemeinen Formel A-B-C in der A jede Aminosäure, B die chemische Bindung zwischen A und C oder jede Aminosäure vorzugsweise ausgewählt aus Prolin, Hydroxyprolin, Thiazolidincarbonsäure, Dehydroprolin, Pipecolinsäure, Azetidincarbonsäure oder Aziridincarbonsäure, verbunden über Peptidbindungen mit A und C, sein kann, und C stabile Inhibitoren der Serinpeptidase Dipeptidyl Peptidase IV (DP IV), wie z. B. Aminoacyl Pyrrolidide, Aminoacyl Thiazolidide, N-Dipeptidyl, O-Acyl Hydroxylamine sein kann. DOLLAR A Solche Prodrug-Verbindungen dienen einem zell-, gewebs- bzw. organspezifischen Transport durch biologische Membranen, einer zeitgerechten und zielgerichteten in vivo Aktivierung von mittels der Prodrugform chemisch maskierten Inhibitoren der DP IV und können zur Behandlung von Erkrankungen eingesetzt werden.
Description
Die vorliegende Erfindung betrifft neue, spezifische und effektive Prodrug-Verbindungen der
Serinpeptidase Dipeptidyl Peptidase IV (DP IV). Solche Prodrug-Verbindungen dienen einem
zielgerichteten, zell-, gewebs- bzw. organpezifischen Transport durch biologische Membra
nen, einer zeitgerechten und zielgerichteten in vivo Aktivierung von mittels der Prodrugform
chemisch maskierten Inhibitoren der DP IV und können zur Behandlung von verschiedenen
Erkrankungen insbesondere von mit Diabetes mellitus im Zusammenhang stehenden Stoff
wechselerkranktingen eingesetzt werden.
Es ist bereits bekannt, daß Aminoacyl Thiazolidide, Aminoacyl Pyrrolidide, N-Dipeptidyl, O-
Acyl Hydroxylamine und andere Verbindungen als Inhibitoren des Serumenzyms Dipeptidyl
Peptidase IV (DP IV) und analoger Enzyme wirken [vgl. DEMUTH, H.-U., J. Enzyme Inhi
bition 3, 249 (1990); DEMUTH, H.-U., HEINS, J., in Dipeptidyl Peptidase IV (B. Fleischer,
Ed.) R.G. Landes, Biomedical Publishers, Georgetown, 1995, 1-37].
Es wurde gefunden, daß mittels stabiler Inhibitoren der Dipeptidyl Peptidase IV die T-Zell-
vermittelte Immunantwort z. B. bei Transplantationen beeinflußt werden [vergl. KOROM, S.,
DEMEESTER, I., STADLBAUER, T.H.W., CHANDRAKER, A., SCHAUB, M., SAYEGH,
M.H., BELYAEV, A., HAEMERS, A., SCHARPE, S., KUPIEC-WEGLINSM, J.W., Inhibi
tion of CD26/dipeptidyl peptidase IV activity in vivo prolongs cardiac allograft survival in rat

19, 15 (1997)] unterdrückt werden kann.
Es wurde gefunden, daß durch Verabreichung von Effektoren der Dipeptidyl Peptidase IV-
bzw. DP IV-analoger Enzymaktivität im Blut eines Säugers, durch deren damit verbundene,
temporäre Aktivitätsreduktion, in kausaler Folge die endogenen (oder zusätzlich exogen ver
abreichten) insulinotropen Peptide Gastric Inhibitory Polypeptide 1-42 (GIP1-42) und Gluca
gon-Like Peptide Amide-1 7-36 (GLP-17-36) (o.a. GLP-17-37 oder deren Analoga) durch DP IV-
und DP IV-ähnliche Enzyme vermindert abgebaut werden und damit die Konzentrations
abnahme dieser Peptidhormone bzw. ihrer Analoga verringert bzw. verzögert werden (vgl.
Die durch die Wirkung von DP IV-Effektoren erzielten, erhöhten Stabilität der (endogen vor
handenen oder exogen zugeführten) Incretine oder ihrer Analoga, die damit vermehrt für die
insulinotrope Stimulierung der Incretin-Rezeptoren der Langerhansschen Zellen im Pankreas
zur Verfügung stehen, verändert sich u. a. die Wirksamkeit von körpereigenem Insulin, was
eine Stimulierung des Kohlehydratstoffwechsels des behandelten Organismus nach sich zieht.
Als Resultat sinkt der Blutzuckerspiegel unter die für Hyperglykämie charakteristische Glu
kosekonzentration im Serum des behandelten Organismus. Dadurch können mittels
DP IV-Inhibitoren Stoffwechselanomalien wie Übergewicht, Glukosurie, Hyperlipidämie sowie
mögliche schwere metabolische Azidosen, Diabetes mellitus, die eine Folge längerer, erhöhter
Glukosekonzentrationen im Blut sind, verhindert bzw. gemildert werden [vgl.

Mit Hilfe von DP IV-Inhibitoren kann auch das Eindringen von HIV in CD 26 (DP IV) po
sitive Zellen experimentell verhindert werden [vergl. WAKSELMAN, M., NGUYEN, C.,
MAZALEYRAT, J.-P., CALLEBAUT, C., KRUST, B., HOVANESSIAN, A.G., Inhibition of

Weiterhin wurde gefunden, daß DP IV neuroaktive Peptide wie Neuropeptid Y und CLIP in
ihrer Aktivität modulieren kann [vergl. MENTLEIN, R., DAHMS, P., GRANDT, D., KRU-

Diese vielseitigen Wirkungen von DP IV-Inhibitoren implizieren, daß sich deren Wirkungen
bei Anwendungen in einem bestimmten pathophysiologischen Zustand eines Gewebes auf
andere, normale physiologische Zustände z. B. in anderen Organen auswirken können. Diese
Auswirkungen können positive als auch negative Folgen für den Zielorganismus haben.
Daher ist es von Interesse, eine hohe Bioverfügbarkeit von stabilen DP IV-Inhibitoren selek
tiv im gewünschten Zielgewebe oder Zielorgan zu erreichen. Wobei stabil erfindungsgemäß
eine Halbwertszeit der biologischen Wirkung der DP IV-Inhibitoren von mehr als 30 Minuten
bedeutet. Eine veränderte Pharmakokinetik und Bioverfügbarkeit von Enzyminhibitoren wird
üblicherweise durch die chemische Maskierung von Enzyminhibitoren durch sogenannte Pro
drugs erreicht [vergl. BUNDGAARD, H. Design of Prodrugs. Elsevier Science Publishers,
Amsterdam, 1985].
Mit Hilfe des Prodrug-Konzeptes lassen sich pharmakokinetische, pharmakodynamische, to
xikologische und Stabilitätseigenschaften von Enzyminhibitoren in fast jede gewünschte
Richtung, insbesondere aber auch Transporteigenschaften von Peptidwirkstoffen im gastro
intestinal Trakt beeinflussen [GANGWAR, S., PAULETTI, G.M., WANG, B., SIAHAAN,
T.J., STELLA, V.J., BORCHARDT, R.T., Prodrug strategies to enhance the intestinal ab
sorption of peptides. Drug Discoverv Today 2, 148 (1998)].
Die Freisetzung von biologisch aktiven Wirkstoffen (u. a. Pharmaka, Peptidhormonen oder
auch Enzyminhibitoren) erfolgt dabei durch chemische oder enzymatische Mechanismen.
Beispielsweise dienen Esterasen, Proteasen und Peptidasen (u. a. auch DP IV selbst) zur
Wirkstoff-Freisetzung aus Prodrugs oder zum Processing von aktiven Hormonen aus Propep
tidhormonen. [vergl. WO 97/45117, US 5433955, US 5614379, US 5624894,]. Dadurch kann
neben der gewünschten zell-, gewebs- oder organspezifischen Freisetzung des aktiven Wirk
stoffes gleichzeitig eine erwünschte Wirkungsverstärkung in vivo erreicht werden.
Bei Analyse der Dosis-Wirkungs-Beziehungen des DP IV-Inhibitors Isoleucyl Thiazolidid
als Modulator der Blutglukosekonzentration im Säugetierorganismus bei entweder enteraler,
oraler Applikation des Wirkstoffes oder parenteraler, intravenöser Applikation an der Wistar
ratte kann man eine Sättigung bezüglich der oral vermittelten Aufnahme des Wirkstoffs ge
messen an der Inhibierung des Serumenzyms beobachten, während im Serum die Aktivität des
Enzyms durch parenteral verabreichten Inhibitor vollständig gehemmt werden kann. Das ist in
Tabelle 1 beispielhaft belegt:
Dieses differente pharmakodynamische Verhalten des gleichen Wirkstoffs bei den verschie
denen Applikationsverfahren kann neben einer intestinalen Transportbarriere aber auch auf
die Hydrolyse der Verbindungen durch die in den Dünndarm-Mucosazellen, im Serum und in
Leberzellen vorhandenen Enzyme Prolin Aminopeptidase und Prolidase zurückgeführt wer
den. Um eine wünschenswerte erhöhte Bioverfügbarkeit nach oraler Applikation des Wirk
stoffes, eine zielorientierte sowie zeitabhängige Freisetzung und damit eine antizipierte Wir
kungsverbesserung potentieller DP IV-Inhibitoren zu erzielen, wurden exemplarisch Prodrug
verbindungen eines bekannten DP IV-Inhibitors (Isoleucyl Thiazolidid, auch Ile-Thia) darge
stellt und an die Auswahl der neuen Verbindungen folgende Prämissen geknüpft:
- 1. Die Prodrugverbindungen sollen Di- oder Tripeptidderivate sein, die durch die bekann ten intestinalen bzw. zellulären Peptidtransporter PepT1 bzw. PepT2 erkannt werden [DANIEL, H., Perspectives in Physiology, J. Physiol. 498, 561 (1997)].
- 2. Die Freisetzung des aktiven DP IV-Inhibitors soll vorzugsweise durch im Serum von Säugetieren vorhandene Aminopeptidasen erfolgen, um eine zielgerichtete Freisetzung des Inhibitors zu erreichen.
- 3. Die zeitliche Freisetzung des aktiven DP IV-Inhibitors soll durch die Auswahl der Komponenten A und B innerhalb der Prodrug-Verbindung A-B-C steuerbar sein.
- 4. Durch die Auswahl von u. a. der DP IV selbst als freisetzendem Enzym, soll aufgrund der bekannten Hydrolysedaten für niedermolekulare Dipeptidderivate, die unter 2. und
- 3. aufgeführten Eigenschaften in besonders einfacher Weise steuerbar sein. Beispiels weise liegen die Geschwindigkeitskonstanten der DP IV-katalysierten Freisetzung von Dipeptiden der Struktur A-B aus DP IV-Substraten zwischen ca. 1 sec-1 und 100 sec-1. Insbesondere die Freisetzung von H-Xaa-Pro-OH (H-Xaa steht für A, Pro-OH steht für B) aus H-Xaa-Pro-Yaa (Yaa steht für Amine wie Aminosäuren) ist bekanntermaßen ab hängig von der Natur des Aminosäurerestes Xaa. Dabei werden die Substrate mit hy drophoben und verzweigten Seitenketten in Xaa deutlich langsamer hydrolysiert. Für die DP IV-katalysierte Hydrolyse von Substraten des Typs H-Xaa-Pro-Yaa wurde folgende Reihenfolge für kcat in Abhängigkeit von Xaa für DP IV-Substrate des Typs Xaa-Pro- Yaa empirisch gefunden: Ile<Val<Phe<Pro<Ala<Gly. Dabei variieren die Geschwin digkeitskonstanten kcat zwischen etwa 1 sec-1 bis 100 sec-1 [vgl. DEMUTH, H.-U., J. En zyme Inhibition 3, 249 (1990); DEMUTH, H.-U., HEINS, J., in Dipeptidyl Peptidase IV (B. Fleischer, Ed.) R.G. Landes, Biomedical Publishers, Georgetown, 1995, 1-37]. Da mit soll für das Design der antizipierten Prodrugs ein Spielraum von um zwei Größen ordnungen differierender Freisetzungsgeschwindigkeiten zur Verfügung stehen.
- 5. Da prolinspezifische Aminopeptidasen wie z. B. Pyroglutamyl Aminopeptidase, Prolyl Aminopeptidase neben der prolinspezifischen Dipeptidyl Peptidase IV im Blut von Säu getieren vorkommen, und die durch intestinalen Transport aufgenommenen Prodrugver bindungen im Serum hydrolysieren können und damit erfindungsgemäß den durch die Prodrugverbindung chemisch maskierten DP IV-Inhibitor freisetzen können, sollten vorzugsweise Prodrugverbindungen zum Einsatz kommen, deren Struktur aus der Sub stratspezifität dieser Enzyme abgeleitet wurde.
Überraschenderweise wurde gefunden, daß erfindungsgemäße stabile Prodrugs der Dipeptidyl
Peptidase eine deutlich verstärkte Glukosetoleranz im Glukosetoleranztest bewirken als der
korrespondierende, unmaskierte, DP IV-Inhibitor.
Die Herstellung und die Verwendung von Prodrug-Verbindungen der allgemeinen Formel
A-B-C ist durch die folgenden Beispiele belegt (vergleiche Tabelle 2: Analysedaten von Pro
drugs der Dipeptidyl Peptidase IV):
6,5 mM Boc-Pro-Ile-OH (ein Equivalent = 1 eq.) werden mit N-Hydroxybenzotriazol (1 eq.)
und Thiazolidin (1 eq.) in 30 ml Dichlormethan (DCM) suspendiert. Bei -10°C wird unter
Rühren die equivalente Menge einer 1 M Dicyclohexylcarbodiimid-Lösung zugetropft. Man
läßt zunächst eine Stunde bei -10°C und über Nacht bei Raumtemperatur (RT) rühren. Zur
Aufarbeitung wird gründlich vom ausgefallenen Dicyclohexylharnstoff abfiltriert, DCM im
Vakuum abgezogen und der erhaltene Rückstand in Essigester aufgenommen. Die Essige
sterlösung wird mindestens dreimal mit gesättigter Bicarbonat-Lösung, einmal mit gesättigter
NaCl-Lösung, dreimal mit verdünnter KHSO4-Lösung und nochmals mit NaCl-Lösung gewa
schen. Die Essigesterphase wird über Na2SO4 getrocknet, einrotiert und das verbleibende
Rohprodukt mit Essigester/Pentan umkristallisiert. Boc-Pro-Ile-Thia kristallisiert nach 1-2
Tagen bei 4°C (Ausbeute 80%). Boc-Pro-Ile-Thia wird mit 1,1 N HCl/Eisessig-Lösung (3 ml
pro mmol Peptid) versetzt. Man läßt zwei Stunden bei RT rühren, versetzt mit absolutem
Ether und dampft überschüssige Abspaltungslösung am Rotationsverdampfer ab. Das Hydro
chlorid kristallisiert quantitativ unter absolutem Ether über Nacht bei 4°C. Man saugt die Kri
stalle schnell unter mehrfachem Waschen mit absolutem Ether ab und bewahrt das Produkt im
Exsikkator über KOH oder Phosphorpentoxid auf.
Boc-Gly-OH (1 eq.) wird in 20 ml ml Tetrahydrofuran (THF) gelöst, auf -10°C gekühlt und
unter Rühren nacheinander mit N-Methylmorpholin (1 eq.) und Chlorameisensäureisobutyl
ester (1 eq.) versetzt. Man läßt etwa 20 min aktivieren. Parallel dazu wird Pro-Ile-Thia × HCl (1
eq.) in 10 ml THF suspendiert auf -10°C temperiert und mit N-Methylmorpholin (1 eq.) zur
Neutralisation versetzt. Nach beendeter Aktivierungszeit werden beide Lösungen miteinander
vermischt, nach ein bis zwei Stunden auf Raumtemperatur erwärmt und über Nacht gerührt.
Danach wird das Reaktionsgemisch mit wenig Wasser versetzt und THF im Vakuum abde
stilliert. Der verbleibende Rückstand wird in Essigester aufgenommen und mindestens drei
mal mit gesättigter Natrium Bicarbonatlösung, einmal mit gesättigter NaCl-Lösung, dreimal
mit verdünnter KHSO4 und nochmals mit NaCl-Lösung gewaschen. Man trocknet die Essige
sterphase über Na2SO4, rotiert ein und kristallisiert das Product Boc-Gly-Pro-Ile-Thiazolidid
mit Essigester/Pentan um (Ausbeute 85%). Die Boc-Abspaltung wird analog der Synthese
von H-Pro-Ile-Thia/HCl durchgeführt (Ausbeute < 95%).
Die Affinität verschiedener Peptide, DP IV-Inhibitoren und Prodrugs von Inhibitoren der DP
IV zum Peptitidtransporter PepT1 wurde mittels Verdrängung des radioaktiv markierten Sub
strates D-Phe-Ala analysiert [AMASHEH, S., WENZEL, U., WEBER, W.M., CLAUSS, W.,
DANIEL, H., Electrophysiological analysis of the function of the mammalian renal peptide
transporter expressed in Xenopus laevis oocytes. J. Physiol. 504, 169-174 (1997)]. Dabei zeigt
sich, daß beispielsweise das Tripeptidderivat Ile-Pro-Ile-Thia, vergleichbar bzw. besser als
ausgewählte Aminosäure Derivate an das Transporterprotein PepT1 gebunden und im Ver
gleich zu ausgewählten Aminosäure- bzw. Peptidanaloga ähnlich oder besser transportiert
wird (Tabelle 3).
Zweck der Entwicklung von Prodrugs von DP IV-Inhibitoren ist eine retardierte Freisetzung
des aktiven DP IV-Inhibitors im Zielkompartiment, z. B. im Blutkreislauf. Die Freisetzung soll
durch die geeignete Auswahl von Bestandteilen A-B der allgemeinen Formel A-B-C chemisch
maskierter Inhibitoren der DP IV modulierbar sein. Beispielsweise bietet die Maskierung ei
nes DP IV-Inhibitors mittels einer Aminosäure oder eines Dipeptides die Möglichkeit, den DP
IV-Inhibitor zielgerichtet durch Enzyme wie Aminopeptidasen, sofern sie im Zielkomparti
ment vorhanden sind, freizusetzen. In Abb. 1 ist die erfindungsgemaße, zeitabhängig
differenziert verlaufende Inhibierung von humaner Blut-DP IV durch Freisetzung des Inhibi
tors Isoleucyl Thiazolidid exemplarisch dargestellt. Die Freisetzung des maskierten DP IV-
Inhibitors im Blut kann im Falle der ausgewählten Beispiele (Pro-Pro-Ile-Thia = PPIThia,
Gly-Pro-Ile-Thia = GPIThia) durch DP IV selbst bzw. durch Aminopeptidasen (pGlu-Ile-Thia
= pEIThia, Pro-Ile-Thia = PIThia) erfolgen (Abb. 1). Die Freisetzung des DP IV-
Inhibitors Isoleucyl Thiazolidid aus den Prodrugverbindungen erfolgt im Blut bei gleicher
eingesetzter Konzentration der Prodrugverbindung mit differenzierter Effizienz, wobei sich
für Pro-Pro-Ile-Thia und pGlu-Ile-Thia eine im Vergleich zu Pro-Ile-Thia und Gly-Pro-Ile-Thia
mehr retardierte Wirkstoff-Freisetzung zeigt.
Durch Überführung des aktiven Wirkstoffes Isoleucyl Thiazolidid in erfindungsgemäße Pro
drugs beobachtet man nach deren oraler Applikation bei der Wistar Ratte ein deutlich verbes
sertes Wirkprofil (Abb. 2). Die erwünschte Reduktion des Blutglukosespiegels durch
DP IV-Inhibitoren im untersuchten Zeitintervall wird durch Einsatz der Prodrugs erfindungs
gemäß gegenüber dem unmaskierten Wirkstoff Ile-Thia etwa um 30% verstärkt (Tabelle 4):
Claims (8)
1. Prodrug-Verbindungen der allgemeinen Formel A-B-C in der A jede Aminosäure, B die
chemischen Bindung zwischen A und C oder jede Aminosäure vorzugsweise ausgewählt
aus Prolin, Hydroxyprolin, Thiazolidincarbonsäure, Dehydroprolin, Pipecolinsäure, Azeti
dincarbonsäure oder Aziridincarbonsäure, verbunden über Peptidbindungen mit A und C,
sein kann, und C stabile Inhibitoren der Serinpeptidase Dipeptidyl Peptidase IV (DP IV)
wie z. B. Aminoacyl Pyrrolidide, Aminoacyl Thiazolidide, N-Dipeptidyl, O-Acyl Hy
droxylamine sein kann.
2. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
zum zielgerichteten zell-, gewebs- und organspezifischen Transport durch biologische
Membranen von mittels der Prodrugform chemisch maskierten Inhibitoren der DP IV.
3. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
zur zeitgerechten in vivo Aktivierung von mittels der Prodrugform chemisch maskierten
Inhibitoren der DP IV.
4. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
zur zielgerichteten, d. h. zell-, gewebs- und organspezifischen in vivo Aktivierung von
mittels der Prodrugform chemisch maskierten Inhibitoren der DP IV.
5. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
zur Behandlung von Erkrankungen von Säugern, die durch Modulation der DP IV-
Aktivität in verschiedenen Zellen, Geweben und Organen beeinflußt werden können.
6. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
zur Behandlung insbesondere von Stoffwechselerkrankungen des Menschen.
7. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 1
in pharmazeutischen Formulierungen.
8. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch 7
in pharmazeutischen Formulierungen insbesondere zur enteralen und parenteralen Ver
abreichung.
