Bei dem in der Patentschrift 105050 beschriebenen Verfahren zur Darstellung von
Paraxanthin (1, 7-DimethyIxanthin) geht man aus von einem 1,7- Dimethyl - 3 - chlormethyl-'
8-chlorxanthin von der Formel

welches dadurch entsteht, daß man (8)-Chlorcaffein bei ungefähr 1 500 der chlorierenden Einwirkung
eines Gemisches von Phosphorpentachlorid und Phosphoroxychlorid unterwirft.
Es wurde nun gefunden, daß man die Chlorierung des Chlorcaffeins in andere Wege
leiten kann, wenn man eine Lösung von Chlor in einem geeigneten Medium, wie Nitrobenzol,
Phosphoroxychlorid usw., bei einer Temperatur von 500 oder darüber auf das Chlorcaffein einwirken
läßt. In diesem Falle wird nicht ein Wasserstoffatom der am Stickstoff (3) sitzenden
Methylgruppe durch Chlor ersetzt, sondern vielmehr ein Wasserstoffatom der (7) Methylgruppe,
so daß man ein (71, 8) - Dichlorcaffein von der Formel

erhält.
In dem 71, 8-Dichlorcaffein ist, ähnlich wie
bei dem in der Patentschrift 105050 beschriebenen Isomeren, die Chlormethylengruppe leicht
abspaltbar. Erwärmt man den Körper mit Wasser, verdünnten Säuren oder Alkalien, so
wird er unter Bildung von Formaldehyd und Salzsäure hydrolysiert und es entsteht ein

Das so erhaltene Chlortheophyllin läßt sich durch Reduktion leicht in das therapeutisch
wertvolle Theophyllin überführen.
I. a) Darstellung von 71, 8-Dichlorcaffein.
23 g trockenes Chlorcaffein . werden mit 100 ecm Phosphoroxychlorid, das 9 g Chlorgas
gelöst enthält, im geschlossenen Gefäß 9 Stunden auf ioo° erhitzt. Man läßt darauf
erkalten und destilliert das Phosphoroxychlorid im Vakuum ab, wobei das Dichlorcaffein in
fast reinem Zustande zurückbleibt.
Aus heißem Methylalkohol kristallisiert es in langen, zu Warzen vereinigten Nadeln, die bei
1450 schmelzen.
b) Darstellung von 8-Chlortheophyllin.

ίο Teilen Wasser längere Zeit gekocht; es tritt
starker Formaldehydgeruch auf und allmählich ' erfolgt vollständige Lösung. Aus der Lösung
; wird durch Konzentration das entstandene 8-Chlortheophyllin gewonnen, welches die von
i'o E. Fischer und L. Ach (Ber. d. d. ehem.
Ges. 28, S. 3139) angegebenen Eigenschaften zeigt. Es wird auf bekannte Weise zu Theophyllin
reduziert.
Natürlich braucht man das Dichlorcaffein nicht erst zu isolieren, sondern kann in einer
Operation vom Chlorcaffein zum Chlortheophyllin gelangen, wie folgendes Beispiel
zeigt:
II. Man suspendiert 45 g Chlorcaffein in 400 ecm Nitrobenzol und leitet in das auf 500
erwärmte Gemisch unter beständigem Rühren
einen Überschuß von Chlor. Der Zusatz von 0,5 g Jod zur Flüssigkeit wirkt vorteilhaft.
Nach kurzer Zeit erfolgt Lösung; es wird dann der größte Teil des Nitrobenzols im Vakuum
abdestilliert, der Rest mit Wasserdampf abgetrieben und das ausgeschiedene Produkt noch
so lange mit Wasserdampf behandelt, bis bloß noch, schwacher Formaldehydgeruch wahrnehmbar
ist. Dabei geht Chlortheophyllin neben wenig Chlorcaffein in Lösung, während die
Hauptmenge des unveränderten Chlorcaffeins ungelöst bleibt. Man filtriert und dampft das
Filtrat zur Trockne ein. Dem Rückstand wird das Chlortheophyllin mit wäßrigem Ammoniak
entzogen und aus der ammoniakalischen Lösung durch Säure gefällt.
Claims (1)
- Patent-Anspruch :Verfahren zur Darstellung von Chlortheophyllin, darin bestehend, daß man das durch Einwirkung von Chlor auf 8-Chlorcaffein bei Gegenwart eines geeigneten Lösungsmittels erhältliche 71J 8-Dichlorcaffein hydrolysiert.
