Description
KAISERLICHES
PATENTAMT.
Vorliegende Erfindung betrifft die Darstellung einer Methylencitronensäure, die vermöge ihrer
Eigenschaften therapeutische Verwendung finden soll.
Tollens und Henneberg (vergl. Liebig's Annalen 292, 31 ff.) haben Methylenderivate von
mehrwerthigen Säuren der Zuckergruppe erhalten, in denen das Methylen an die Stelle
von Hydroxylwasserstoffen getreten ist. Nur bei der Milchsäure tritt, analog der vorliegenden
Methylencitronensäure, bei der Einwirkung von Formaldehyd auch die Carboxylgruppe in
die Reaction ein, doch ist das Reactionsproduct ein Ester (Methylenlactat), wahrend bei der
Einwirkung von Formaldehyd auf Citronensäure eine zweibasische Estersäure resultirt.
Das Methylenlactat erhält man auch nur in sehr schlechter Ausbeute und mufs es erst
durch fractionirte Destillation im Vacuum isoliren; bei der Einwirkung von Formaldehyd
auf Citronensäure geht der Procefs sofort quantitativ vor sich, und man hat nur nöthig,
das Reactionsproduct in Wasser zu lösen, um sofort bei der !Crystallisation ein ganz reines
Product zu erhalten. Ferner ist das Methylenlactat aufserordentlich leicht zersetzlich und
spaltet Formaldehyd so rapid ab, dafs es infolge der dadurch bedingten Reizwirkung therapeutisch
unverwendbar ist. Thatsächlich ist es auch wegen seiner Toxicität therapeutisch nicht
verwendet worden. Methylencitronensäure dagegen ist völlig üngiftig.
Zur Darstellung der Methylencitronensäure verfährt man beispielsweise wie folgt:
21 kg Citronensäure werden mit-4 kg Paraformaldehyd
auf 140 bis i6o° so lange erhitzt, bis die Anfangs entstehende klare Lösung sich
zu trüben beginnt und Krystalle ausscheidet. Man läfst das Reactionsproduct dann allmählich
bis auf ca. ioo° abkühlen, löst es in 25 bis 30 kg Wasser und filtrirt. Die in der Kälte
abgeschiedenen Krystalle werden abgesaugt, gewaschen und getrocknet.
Man kann, wenn auch weniger vortheilhaft, so verfahren, dafs man auf Citronensäure in
Gegenwart eines Condensationsmittels, wie z. B. Salzsäure oder Schwefelsäure, Formaldehydlösung
einwirken läfst.
Die Methylencitronensäure hat einen Schmelzpunkt von 208 °. Sie ist ziemlich schwer in
kaltem, leicht in heifsem Wasser löslich. Gegen Säuren ist die wässerige Lösung ziemlich beständig,
empfindlicher ist die Verbindung gegen kohlensaure und kaustische Alkalien.
Es kommt ihr folgende Constitution zu:

Die Bindung erfolgt wahrscheinlich in dieser Weise, da nur eine Hydroxylgruppe in der
Säure vorhanden ist und an demselben Kohlenstoffatom eine Carboxylgruppe sitzt. Benachbarte
Carboxylgruppen sind nicht vorhanden; sie sind vielmehr durch C H^- Gruppen getrennt,
und dafs diese nicht reagiren, wurde durch Versuche mit anderen zweibasischen
organischen Säuren, ζ. BJ Malonsäure und Bernsteinsäure, erwiesen.
Die Analyse lieferte folgende Zahlen: 0,1998 g
der Substanz ergaben 0,0729 g Wasser und 0,3029 g Kohlensäure.
Berechnet für C7 Hs O7: 41,18 pCt. C,
3,92 pCt. H.
Gefunden: 41,34 pCt. C,
4,05 pCt. H.
Aus der Methylencitronensäure lassen sich therapeutisch werthvolle Salze darstellen, wie
z. B. das Silbersalz, Quecksilbersalz, Magnesiumsalz, Hexamethylentetraminsalz. Das letztere ist
leicht löslich in Wasser im Gegensatz zur Methylencitronensäure.

- Patent-Anspruch:Verfahren zur Darstellung von Methylencitronensäure, darin bestehend, dafs man gewöhnlichen oder polymeren Formaldehyd auf Citronensäure mit oder ohne Condensationsmittel einwirken läfst.
