Die Zubereitungen der Erfindung, welche zur örtlichen
Anwendung dargeboten werden, schliessen Salben, Cremes und Lotionen ein, welche Verbindungen der obigen all«
gemeinen Formel oder ihrer Derivate enthalten. Gebräuchliche Salben und Cremes köiinen der Eigenschaft
nach Wasser-mischbare oder Y/asser~nicht-mischbare sein
und schliessen Emulsionen ein, welche aus emulgierenden «■/achsen und Ölen und ebenso jenen, welche aus Wassermischbaren Polyäthylenglykolen hergestellt werden» Die
Lotionen nach der Erfindung können eine Lösung der Wirkstoffe der obigen allgemeinen Formel in einem ge«

vorzugsweise ein niederer aliphatischer Alkohol ist,
welcher einen kleinen Anteil Wasser enthalten kann«,

Die Wirkstoffe der vorliegenden Erfindung können auch
in neue Zubereitungen mit anderen therapeutisch allgemein
bekannten wirksamen Verbindungen einverleibt weraen,
Der üntersuchungstext, welcher zur Feststellung der Antientzündungseigenschaften
benutzt wurde, war der von Adams und Cobb in Nature, 181, 773, 1958 beschriebene.
Analgetiöche und antipyretische Eigenschaften der Verbindungen
wurden auch festgestellt, wie ihre Toxizität' an verschiedenen Tierarten, nämlich Mäusen, Hatten,
Meerschweinchen, Katzen und Hundene Wie zu erwarten war,
wechselten, die relative Wirksamkeit in weitem Bereich und zur Bestätigung der pharmakologisehen Wirksamkeit
in einem klinischen Versuch, wurde eine Verbindung ausgewählt, welche eine gute allseitige Wirksamkeit mit
niederer 'foxizitat hatte,, nämlich 4-Isobuty!phenylessigsäure.
Die akute LDcq dieser Verbindung bei Mäusen
war 1300 mg/kg oral und 600 mg/kg i.p.; bei Hatten war die orale Menge grosser als 1200 mg/kg. Keine toxische
Wirkungen und keine pathologischen Veränderungen wurden festgestellt, wenn bei Hatten täglich 200 mg/kg 4-Isobutylphenylessigsäure
8 Wochen lang gefüttert wurden* In gleicher Weise wurden keine toxischen Wirkungen bei
Hunden festgestellt, wenn 50 mg/kg täglich 6 Wochen lang gefüttert wurden, BAD

Der klinische Anfangsversuch wurde durchgeführt in. einer kontrollierten Form unter Verwendung der "doppelt
Blind-Techniky wobei weder der Patient noch der medizinische
Beobachter die Droge kennen, die während der Zeitdauer der Beobachtung gegeben wird«. Zwölf Patienten
mit akuter rheumatischer Arthritis, einschliesslich
multipler G-elenkbeschwerden und mit systematischer fiebriger Reaktion, wurden für eine Zeitdauer von verschiedenen

4-Isobutylphenyle essigsäure (75 g), reiner Alkohol
(500 ml) und konzentrierte Schwefelsäure (15 ml) wurden
4 Stunden lang unter Rückfluss gehalten» Der überschüssige Alkohol wurde im. Vakuum, destilliert, der
Hackstand mit Wasser verdünnt und der Ester in jither
abgetrennt, mit Natriumkarbonatlösung gewaschen, dann
mit Wasser gewaschen, bevor er getrocknet und destilliert wurde; sein Siedepunkt war 10Ö bis 11O°C/O,6 mm»
(Gefunden: 76,7$ C, 9,2% H; die Brut ιοί ormel 0-14H20O2
erfordert 76,4% C, 9,1% H.)
Auf die gleiche Weise wurden die folgenden Verbindungen hergestellt:
Äthyl-4-cyklohexylphenylacetat; Siedepunkt 140 G/1 mm.
(Gefunden: 78,5$ C, 9,2$ H; die Bruttoformel C16H22O2
erfordert 78,0$ C, 8,9% H.)

Bin inniges Gemisch, wurde aus gleichen Teilen 4-Tsobuty!phenylessigsäure
und einem Ta/blettengrundstaffy
welcher aus Stärke mit der Zugabe -von Vj0 Magnesiumstearat
als Gleitmittel bestand, hergestellte Das.Gemisch wurde in tabletten gepresst, welche 2 1/2 grains
4-Isobutylphenylessigsäure enthielten»
Ahnliche Tabletten wurden auch, hergestellt unter Verwendung
anderer Verbindungen der vorliegenden Erfindung als Wirkstoff, wie 4-Isobutylphenylxjropionsäure oder
4-Cyklohexylphenylessigsäure.
Ein inniges Gemisch wurde aus 5 Teilen 4-Isobuty!phenylessigsäure
und 3 Teile eines Tablettengrundstoffes hergestellt, wobei der Tablettengrundstoff Stärke mit
der Zugabe von Vh Magnesiumstearat als Gleitmittel
umfassteβ Das Gemisch wurde in Tabletten gepresst, welche
5 grains 4-Isobutylphenylessigsäure enthielten,
Ähnliche Tabletten wurden auch hergestellt unter Ver«
wendung anderer Verbindungen der Erfindung als Wirkstoff, wie 4-Isobutylpheny!propionsäure oder 4-Cyklohexylphenylessigsäure.
